벤젠 노출 근로자의 생물학적 모니터링 지표로서의 요중 S-Phenylmercapturic Acid에 관한 연구 (Urinary S-Phenylmercapturic Acid as a Biomarker for Biological Monitoring in Workers Exposed to Benzene)

  • This study was to evaluate the associations between urinary S-Phenyl-mercapturic acid(S-PMA) as a new indicator of biological monitoring for low level of exposure to benzene and independent variables such as the air concentration of benzene in the breathing zone of workers, the years of work, and smoking. In this study the subjects were the total of 145 drawn from 53 workers who were occupationally exposed to benzene and 92 workers who were not. The results were as follows: 1. In the workplace geometric mean concentration of benzene in the breathing zone of workers was 0.31 ppm(0.02 - 3.26 ppm) for the spraying workers and 0.25 ppm(0.02 - 3.95 ppm) for the printing workers. 2. The geometric mean of uninary S-PMA for non exposed group was $8.9{\mu}g/g$ creatinine($0.6-72.3{\mu}g/g$ creatinine), 80.3% (74 workers) of the total non-exposed workers indicated less than $20{\mu}g/g$ creatinine of uninary S-PMA. The difference of uninary S-PMA by sex, age, smoking was not significant. 3. The geometric mean of urinary S-PMA for workers who were exposed to benzene was $37.2{\mu}g/g$ creatinine, and was four times higher than that of workers who were not exposed. And 79.3% (42 workers) of the total exposed workers indicated more than $20{\mu}g/g$ creatinine of urinary S-PMA. 4. Regarding the level of benzene in the air, urinary S-PMA was the highest level of $147.9{\mu}g/g$ creatinine in the workers who were exposed to air concentration of 0.5 ppm of benzene and was higher as the level in the air was increased. 5. The correlation coefficient between log urinary S-PMA and log benzene concentration in the breathing zone was 0.80, and the following linear equation was found between urinary log S-PMA and log benzene concentration in the breathing zone : log S-PMA(${\mu}g/g$ creatinine) = 0.564 log benzene in air(ppm) + 0.192 (n=53, r=0.80, p=0.000) In conclusion, the concentration of S-PMA in urine proved to be good parameter for biological monitoring benzene exposure at the workplace even at low level of benzene in air.

  • To investigate the exposure effect of polynuclear aromatic hydrocarbons (PAHs), we measured airborne total PAHs as an external dose, urinary 1-hydroxypyrene (1-OHP) as an internal dose of PAHs exposure, and analyzed the relationship between urinary 1-OHP concentration and PAHs exposure. The study population contained 44 workers in steel-pipe coating and paint manufacture industries. The airborne PAHs was obtained during survey day, and urine were sampled at the end of shift. Personal information on age, body weight, height, eniployment duration, smoking habit, and alcohol consumption was obtained by a structured questionnaire. Airborne PAHs were analyzed by the gas chromatograph with mass selective detector. Urinary 1-OHP levels were analyzed by the high performance liquid chromatograph with ultraviolet wavelength detector. For statistical estimation, t-test, ${\chi}^2$-test, analysis of variance, correlation analysis, arid regression analysis were executed by SPSS/PC (Windows version 10). The mean of environmental total PAHs was $87.8{\pm}7.81{\mu}g/m^3$. The mean concentration ($526.5{\pm}2.85{\mu}g/m^3$) of workers in steel-pipe coating industries using coal tar enamel was the higher than that ($17.5{\pm}3.36{\mu}g/m^3$) of workers in paint manufacture industries using coal tar paint. The mean of urinary 1-OHP concentration ($51.63{\pm}3.144{\mu}\;mol/mol$ creatinine) of workers in steel-pipe coating industries was the higher than that ($2.33{\pm}4.709{\mu}\;mol/mol$ creatinine) of workers in paint manufacture industries. The mean of urinary 1-OHP concentration of smokers was the higher than that of non-smokers. There was significant correlation between the urinary concentration of 1-OHP and the environmental concentration of PAHs (r=O.S48, p<0.001), pyrene(r=0.859, p<0.001), and urinary cotinine (r=0.324, p<0.05). The regression equation between the urinary concentration of 1-OHP in ${\mu}g/g$ creatinine($C_{1-OHP}$) and airborne concentration of PAHs (or pyrene) in ${\mu}g/m^3$ ($C_{PAHs}$ or Cpyrene) is: Log ($C_{1-OHP}$)=-0.650+0.889×Log($C_{PAHs}$), where $R^2=0.694$ and n=38 for p<0.001.Log ($C_{1-OHP}$)=1.087+0.707${\times}$Log(Cpyrene), where $R^2=0.713$ and n=38 for p<0.001. From the results of stepwise multiple regression analysis about 1-OHP, significant independents were total PAHs and urinary cotinine (adjusted $R^2=0.743$, p<0.001). In this study, there were significant correlation between the urinary concentration of 1-OHP and the airborne concentration of PAHs. The urinary 1-OHP was effective index as a biomarker of airborne PAHs in workplace. But it was influenced by non-occupational PAHs source, smoking.

Assessment of Environmental Pollution in Korean Stream Sediments by Chemical Analyses and Insect Immune Biomarkers

  • A comprehensive quality survey for PCDDs/PCDFs and coplanar PCBs as well as heavy metals (Cu, Zn, Cd and Pb) in sediments has been investigated in August 2006, Korea. Monitoring was undertaken at five streams representing different surrounding environments throughout Juwang and Gapyeong streams (reference sites), Jungrang stream (dense population site), Ansan stream (mixed small population and industrial site), and Siheung stream (heavy industrial site). The levels of heavy metal in samples were found to be significantly higher in sediment from Siheung stream compared to those of other stream sites. The heavy metal concentrations (dry weight basis) in sediment from Siheung stream were as follows; Cd (3.7 ${\mu}g$/g), Pb (1,295 ${\mu}g$/g), Cu (713.4 ${\mu}g$/g) and Zn (358.1 ${\mu}g$/g). Among 12 coplanar PCBs and 17 PCDDs/PCDFs selected as target compounds in this study, PCB (IUPAC no. 118) and OCDD were the most abundant congeners found in all sediment samples, followed by 1,2,3,4,6,7,8-HpCDD, OCDF and 1,2,3,4,6,7,8HpCDF as well as PCB (IUPAC no. 105). These results were shown to be in the same trend as the sediment samples of other countries. The levels of PCDDs/PCDFs/coplanar PCBs in sediment samples were expressed as concentrations and WHO- TEQ values. The PCDDs/PCDFs/coplanar PCBs concentrations and their WHO-TEQ values in sediment from Siheung stream were remarkably high. The levels detected were 788.16 pg/g and 36.080 pg WHO-TEQ/g dry weight for PCDDs/ PCDFs and 314 pg/g and 0.4189 pg WHO-TEQ/g dry weight for coplanar PCBs, respectively, beyond the safety level of sediment value 20 pg WHO-TEQ/g. Sediment samples of the five streams were also monitored by sensitive biomarkers using insect immune responses: hemocyte-spreading behavior and immune-associated enzyme activities of phospholipase A$_2$ (PLA$_2$) and phenoloxidase. Organic extracts of Siheung and Jungrang sediments significantly interfered with the hemocytespreading behavior, whereas those of Ansan, Gapyeong, and Juwang did not. These organic extracts did not inhibit the PLA$_2$ and phenoloxidase activities. However, phenoloxidase was highly susceptible to exposure to aqueous extracts in all site sediments. In comparison, PLA$_2$ activities of the hemocytes were significantly inhibited only by aqueous extracts of Siheung, Jungrang, and Gapyeong sediments, but not by those of Ansan and Juwang. Despite some disparity between bioand chemical monitoring results, the biomarkers can be recommended as a device warning the contamination of biohazard environmental chemicals because of a fast and inexpensive detection method.

  • $estradiol-17\beta$ ${E_2}$에 노출된 미성숙 조피볼락의 혈장 내 vitellogenin (VTG), alkaline-labile protein phosphorus (ALPP), calcium (Ca), glutamate pyruvate transaminase (GPT) 및 hepatosomatic index (HSI)의 일시적인 변화가 조사되었다. 70% 에탄올에 녹여진 ${E_2}$ (5 mg/kg B.W.)를 조피볼락 치어의 복강에 주사한 후 0, 1, 3, 6, 9, 12 및 15일에 혈장이 수집되었다. 전기 영동상에서 주사 후, 3일째 약 170 kDa의 위치에 염색성을 나타내는 VTG 밴드가 검출되었다. 밴드의 염색성은 6일째 더욱 강하게 나타났지만, 시간의 경과와 더불어 흐려져 15일째에는 노출 전 처럼 VTG 밴드가 관찰되지 않았다. 혈장 내 ALPP와 Ca는 1일째부터 급격히 증가했으며 VTG 변동과 유사하게 6일째 가장 높은 농도를 나타낸 후, 시간의 경과와 더불어 점점 감소해 15일째는 노출 전과 유사한 값을 나타내었다. GPT는 투여 1일부터 급격히 증가해 3일에 최고치를 나타낸 이후 시간의 경과와 더불어 감소하는 경향을 나타냈으며 HSI의 경우도 1일째부터 증가해 3일째에 가장 높은 수치를 나타낸 후, 점점 감소해 ${E_2}$ 투여전과 유사한 수치를 나타내었다. 이들 결과로 E2에 노출 된 이후 조피볼락의 혈장 내 ALPP, Ca, GPT 및 HSI는 혈장 VTG와 유사한 증․감을 나타내는 것을 알 수 있었다. 따라서, ALPP, Ca, GPT 및 HSI는 VTG와 더불어 연안 생태계 내에서 외인성 ${E_2}$ 노출에 대한 생물학적 지표로서 사용 가능할 것으로 판단되어진다.

  • 무척추동물에서 척추동물에 이르기까지 대부분의 난생동물들의 난황 단백질의 전구체를 vitellogenin(VTG)이라 한다. 난생 척추동물에서 VTG는 간에서 합성되어 혈액을 통해 난세포로 전이된다. 암컷 어류는 정상적인 생식주기에서 난황단백전구체 형성이 시작되면 혈중 농도는 급격히 증가하게 된다. 수컷도 VTG의 유전자를 가지고 있기 때문에 낮은 수준의 내인성 에스트로겐으로 아주 적은 양의 단백질이 있을 수 있다. 그렇지만 수컷에 외인성에스트로겐 유사물질에 노출되면 그 양은 증가하게 된다. 따라서 어류에서 VTG는 내분비계 장애물질 효과를 조사하는 유용한 생물학적 추적자로 사용할 수 있다. 이 연구에서는 에스트로겐 유사물질에 오염된 지역에 서식하는 망둑어류의 혈중 VTG의 수준을 정량할 수 있는 방법을 제공하고자 한다. 이러한 목적을 위해 꾹저구(Chaenogobius annularis)의 VTG를 분리하고, 이에 대한 단클론 항체와 다클론 항체를 제작하여 혈중 VTG 수준을 정량할 수 있는 sandwich 경합 ELISA system을 만들었다. 난황 단백질에 대한 단클론 항체와 다클론 항체를 사용한 ELISA system에 대한 타당성 검토를 하였다. 순차적으로 희석한 암 컷의 혈청에 대한 흡광도 곡선은 VTG 표준농도의 곡선과 평행하였으나 수컷의 혈청에 대한 흡광도 곡선은 평행하지 않았다. 이 VTG에 대한 sandwich ELISA system으로 꾹저구(C. annularis)의 혈중 VTG의 수준을 정량뿐만 아니라 문절망둑(Acanthogobius flaviman)과 풀망둑(A. hasta)의 혈중 VTG 수준을 정량할 수 있다.

Radiofrequency에 노출된 Chlamydomonas sp.의 mycosporine-like amino acids 생합성 유전자 발현 (Expression of Mycosporine-like Amino Acids Biosynthetic Genes in the Chlamydomonas sp. Exposed to Radiofrequency)

  • Mycosporine-like 아미노산(MAAs)은 UV 흡수물질이며, 다양한 해양생물들은 MAAs의 합성과 축적을 통하여 환경 자외선의 직 간접적인 영향을 감소시키는 기능을 진화시켜 왔다. 이 연구에서 우리는 radiofrequency(RF) 발생장치를 제작하였고, 이를 미세조류 배양에 적용하였다. $0.35{\pm}0.05$ mHz의 RF를 Chlamydomonas sp. 배양기에 공급하였고, 정해진 시간(0, 0.5, 1 및 2 시간)에 시료를 채취하였다. MAAs 생합성 관련 유전자인 dehydroquinate synthase homolog (DHQS-like)와 nonribosomal peptide synthetase homolog (NRPS-like) 유전자를 Chlamydomonas sp.로부터 클로닝하였고, RF 노출에 대한 유전자 발현을 qRT-PCR을 이용하여 분석하였다. 연구결과 RF에 노출된 Chlamydomonas sp.의 DHQS-like와 NRPS-like 유전자의 발현은 노출 1시간에 각각 1.46 배 및 1.19 배 증가하였다. 이러한 결과는 DHQS-like와 NRPS-like 유전자가 Chlaydomonas sp.의 MAAs 생합성을 진단할 수 있는 좋은 바이오마커 후보가 될 수 있음을 의미한다.

국내 생강 에센셜오일이 만성 스트레스로 교감신경이 항진된 동물 모델에 미치는 영향 (The Effects of Korea Ginger Essential oil on Chronic Mild Stress-induced Rats Model of Sympathetic Hyperactivity)

  • 스트레스는 인체의 다양한 기관과 정신적 상태에 영향을 미치는 교감신경 항진을 야기시킨다. 본 연구의 목적은 만성 스트레스로 교감신경을 항진시킨 동물 모델에서 국내산 생강 에센셜오일이 스트레스 호르몬 및 뇌 조직 반응에 미치는 효과를 평가하였다. 평가 방법은 세포독성 평가 및 성분 분석을 수행하였으며, 혈청 바이오 마커와 뇌 조직의 병리학적 분석이 기초한 효과를 관찰하였다. 동물 실험에서 국산 생강 에센셜오일의 처리는 만성 스트레스로 교감신경을 항진시킨 동물 모델 제작 후 2주간 100 nl/㎖로 처리하였다. 그 결과, 국산 생강 에센셜오일은 100 nl/㎖ 농도 이하에서 독성이 없었으며, 6-진저롤 함량이 345 ppm으로 확인되었다. 국산 생강 에센셜오일의 처리는 대조군과 비교하여 혈청에서 부신피질호르몬, 코르티코스테론, 멜라토닌과 같은 스트레스 호르몬의 농도를 크게 줄였으며, 복측피개부(VTA) 및 흑색질 치밀부(SNpc) 부분에서 TH-면역 반응이 때때로 중단되는 것을 효과적으로 보존하였다. 이와 같은 결과는 국산 생강 에센셜오일이 교감신경 항진을 개선했음을 나타내고 있다. 따라서 국산 생강은 교감신경 항진에 대한 아로마오일의 새로운 원료로 활용될 수 있다.

A Study on 10 Metabolites Separated from DNA Adduce of Blood Lymphocytes in Rats Exposed Orally with 3,3-dichlorobenzidine(DCB) by GC/MS-SIM

  • 3.3'-Dichlorobenzidine(DCB) has be shown carcinogenic in several animals, and the development of non-invasive biomonitoring method in workers exposed with it is a very important subject. DNA adduct is a good biomarker for biomonitoring about carcinogens exposure, and lymphocytes is a good non-invasive samples. So we studied to analyze metabolites in blood lymphocytes of female Sprague-Dawley rats exposed orally with DCB(20, 30, and 40 mg/kg wt.) for 3 weeks. For analysis of them, we isolated DNA adducts from blood lymphocytes by using the enzymes method in /sup 32/P-postlabeling, and measured them by using gas chromatography/mass spectrometry-selected ion monitoring(GC/MS-SIM). 4-aminobiphenyl and phenanthrene-d/sub 10/ were added as internal standard for blank sample. Standard metabolites of DCB were synthesized with using pyridine and acetic acid which were promoter and controller in acetylation of DCB. And they were used for calibration curve. Our results showed two kinds of metabolites in DNA adducts of blood lymphocytes. They were N-acetyl 3,3'-dichlorobenzidine(acDCB) and N,N'-diacetyl 3,3'-dichiorobenzidine(di-acDCB ). They were combined with DNA at the same time as an acetyl of it was removed. So we measured DCB and acDCB for two kinds of metabolites in DNA adducts of blood lymphocytes. Our results showed the levels of DCB were 1.46∼2.26 times more than that of acDCB. And also the levels of metabolites in 20, 30 and 40 mg/kg wt. were gradually increased with going days from 1st to 3rd week. They are 1.66, 1.38 and 0.90 times in total metabolites, 1.76, 1.49 and 1.02 times in DCB, and 1.51, 1.22 and 1.28 times in acDCB. In conclusion, the results of this study showed DCB exposed to rats formed DNA adduct in blood lymphocytes after acetylated to N-acetyl 3.3'-dichloro benzidine(acDCB) and N,N'-diacetyl 3,3'-dichlorobenzidine(di-acDCB), and they could be analyzed by us ing gas chromatography/mass spectrometry-selected ion monitoring(GC/MS-SIM).

Mn-SOD와 GST 유전자 다형성에 따른 유아의 산화손상지표의 분포 (Oxidative damage biomarker levels according to Mn-SOD and GST gene polymorphisms in preschool children)

  • 우리나라 일부 건강한 유아를 대상으로 Mn-SOD Val16Ala, GSTP1 Ile105Val, GSTT1 present/null, GSTM1 present/null 유전자 다형성 분포를 살펴본 결과, Mn-SOD Val/Val형, GSTP1 Ile/Ile형, GSTT1 null 형, GSTM1 null 형이 주된 (major) 유전자형인 것으로 나타났다. 이 중 Mn-SOD Val/Val형은 Val/Ala 또는 Ala/Ala형에 비해 소변 8-OHdG 수준이 유의적이지는 않으나 높은 경향을 나타내었고, GSTP1 Ile/Ile형은 Ile/Val 또는 Val/Val형에 비해 소변 8-OHdG 수준이 유의적으로 낮았다. 간접흡연에의 노출 여부와 간접흡연-유전자 다형성의 상호 작용이 산화손상지표에는 유의적인 영향을 미치지 않는 것으로 나타났다. 이상의 결과로 볼 때 건강한 유아에서 GSTP1 Val allele 보유한 경우 산화적 손상에 대해 취약할 수 있음을 제시하며, 추후 대규모 연구를 통한 검증 및 이들 유전자형을 보유한 대상자를 위한 효과적인 영양 중재방안에 대한 고려가 필요할 것으로 사료된다.

Molecular Mechanism of Tetrabromobisphenol A (TBBPA)-induced Target Organ Toxicity in Sprague-Dawley Male Rats

  • Brominated flame retardants (BFRs) are present in many consumer products ranging from fabrics to plastics and electronics. Wide use of flame retardants can pose an environmental hazard, which makes it important to determine the mechanism of their toxicity. In the present study, dose-dependent toxicity of tetrabromobisphenol A (TBBPA), a flame retardant, was examined in male prepubertal rats (postnatal day 18) treated orally with TBBPA at 0, 125, 250 or 500 mg/kg for 30 days. There were no differences in body weight gain between the control and TBBPA-treated groups. However, absolute and relative liver weights were significantly increased in high dose of TBBPA-treated groups. TBBPA treatment led to significant induction of CYP2B1 and constitutive androstane receptor (CAR) expression in the liver. In addition, serum thyroxin (T4) concentration was significantly reduced in the TBBPA treated group. These results indicate that repeated exposure to TBBPA induces drug-metabolising enzymes in rats through the CAR signaling pathway. In particular, TBBPA efficiently produced reactive oxygen species (ROS) through CYP2B1 induction in rats. We measured 8-hydroxy-2'-deoxyguanosine (8-OHdG), a biomarker of DNA oxidative damage, in the kidney, liver and testes of rats following TBBPA treatment. As expected, TBBPA strongly induced the production of 8-OHdG in the testis and kidney. These observations suggest that TBBPA-induced target organ toxicity may be due to ROS produced by metabolism of TBBPA in Sprague-Dawley rats.
