Antioxidative activities of Paulownia coreana extractives (오동나무 추출물의 항산화 효능)

  • Approximately each 5 kg dried Paulownia coreana leaves, seeds, outer barks, inner barks and wood were ground, extracted with acetone-$H_2O$ (7:3. v/v). concentrated under reduced pressure and successively fractionated using n-hexane. $CH_2Cl_2$, EtOAc and $H_2O$ on a separation funnel. Each fraction was then freeze dried and weighed. The extraction yield of each sample was investigated and the yield of inner barks was the highest, while the wood was the lowest. Antioxidative activities of crude extracts, precipitates and partitioned fractions were evaluated by DPPH radical scavenging method. The results indicated that all EtOAc soluble fractions, including some of $H_2O$ soluble fractions, showed significantly high antioxidative potential compared with ${\alpha}$-tocopherol and BHT used as controls.

Antioxidant Activity of Isolated Compounds from the Heartwoods of Rhus verniciflua (옻나무(Rhus verniciflua) 목질부에서 분리한 화합물의 항산화활성)

  • The heartwoods of Rhus verniciflua was extracted with $H_2O$ and the concentrated extract was partitioned with $CHCl_3$, EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc and n-BuOH fractions, four compounds were isolated through the repeated silica gel, ODS and Sephadex LH-20 column chromatographies. They were determined as sinapyl aldehyde (1), 2,4-dihydroxybenzaldehyde (2), 2,4-dihydroxybenzoic acid (3) and 2,4-dihydroxyphenylglyoxylic acid (4) on the basis of spectroscopic data, respectively. Among the isolated compounds, sinapyl aldehyde $(34.7{\pm}0.6%)$ and 2,4-dihydroxyphenylglyoxylic acid $(43.8{\pm}0.9%)$ showed the strong antioxidative activity than artificial antioxidant BHT $(14.4{\pm}0.3%)$ in DPPH radical scavenging activity.

Isolation and Identification of Sterol Compounds from the Red Kohlrabi (Brassica oleracea var. gongylodes) Sprouts (적콜라비 (Brassica oleracea var. gongylodes) 새싹으로부터 sterol 화합물의 분리 및 동정)

  • The sprouts of Brassica oleracea var. gongylodes were extracted with 100% MeOH and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc fraction, three sterols were isolated through the repeated silica gel and ODS column chromatographies. On the basis of physico-chemical and spectroscopic data including NMR, MS, and IR, the chemical structures of the sterols were determined as ${\beta}$-sitosterol (1), brassicasterol (2), and 7-ketobrassicasterol (3). Compound 1 is usually observed in plant. Compounds 2 is observed in Brassica sp., and compounds 3 have very rarely occurred in natural source including plant.

In Vitro Radical Scavenging Effect and Neuroprotective Activity from Oxidative Stress of Petasites japonicus (머위 분획물의 In Vitro 라디칼 소거능 및 신경세포의 산화적 스트레스 보호 효과)

  • This study was focused on the evaluation of radical scavenging effect and the protective activity against oxidative stress of the extract and fractions from Petasites japonicus. P. japonicus was extracted with methanol and then fractionated into 4 fractions [n-butanol, ethyl acetate (EtOAc), methylene chloride, and n-hexane]. The extract and fractions showed strong 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Among all the fractions, particularly, the EtOAc fraction showed the strongest effect with the $IC_{50}$ value of $0.02{\mu}g/ml$. In addition, the fractions also showed strong hydroxyl radical scavenging activity and nitric oxide scavenging activity as well. Furthermore, cell viability generated by the P. japonicus extract and 4 fractions were examined under C6 glial cellular model. The C6 glial cells showed high generation of reactive oxygen species (ROS) and decrease in cell viability by the treatment generator of hydrogen peroxide. However, the production of ROS formation was decreased by the treatment of the fractions of P. japonicus and also founded that the EtOAc fraction led to significant increase in the cell viability at concentration $100{\mu}g/ml$. Results from this work indicated that P. japonicus showed protective effects against oxidative stress and its EtOAc fraction may be served as a useful natural antioxidant.

  • The flower of Campsis granudiflora K. Schum. was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$. From the EtOAc fraction, four compounds were isolated through the repeated silica gel and ODS column chromatographies. From the result of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as linolenic acid methyl ester, linoleic acid methyl ester, ${\beta}-sitosterol$ and daucosterol. Daucosterol inhibited FPTase activity with $IC_{50}$ values of $14{\pm}0.04\;{\mu}M$.

  • The flower of Erigeron annuus L. was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc fraction, three compounds were isolated through the repeated silica gel, ODS column and high performance liquid chromatographies. From the result of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as ${\alpha}-amyrenone,\;{\alpha}-amyrin\;and\;{\beta}-amyrin$. These three compounds were isolated for the first time from the flower of Erigeron annuus L.

Anti-allergic and Anti-inflammatory Effect of Leonurus sibiricus Seed Ethyl Acetate Fractions (충울자 EtOAc 분획의 항알러지 및 항염증 효과)

  • In this study, we investigate anti-allergic and anti-inflammatory effects of Leonurus sibiricus seed (LSS) extract in basophilic leukemia RBL-2H3 cells. To identify anti-allergic actions of LSS, the degranulation was evaluated in IgE and DNP-BSA stimulated RBL-2H3 cells. At the concentration of $100{\mu}g/ml$ of methanol (MeOH) extract and Methylene chloride (MC) and Ethyl acetate (EtOAc) fractions, the degranulation was significantly inhibited 16.7%, 16.7% and 27.9% respectively. And then, to assess anti-inflammatory effects of LSS, IL-4 and IL-13 mRNA level were detected in PMA/ionomycin (PI)-induced RBL-2H3 cells and cell proliferation and IL-4 mRNA level in isolated splenocytes from Balb/c mice. LSS MeOH extract and MC and EtOAc fractions significantly decreased the level of IL-4 and IL-13 mRNA in PI-induced RBL-2H3 cells and showed inhibitory effects on cell proliferation and expression of IL-4 mRNA level in mouse splenocytes. Taken together, these results suggest that LSS has potential anti-allergic and anti-inflammatory effects and EtOAc fraction is the most effective in regulating immune responses.

Pancreatic Lipase Inhibitors Isolated from the Leaves of Cultivated Mountain Ginseng (Panax ginseng) (산양삼 잎으로부터 Pancreatic lipase 저해 활성물질의 분리)

  • Activity-guided fractionation of an ethyl acetate (EtOAc)-soluble portion of an ethanolic extract from the leaves of cultivated mountain ginseng, using pancreatic lipase inhibition assay, led to the isolation and identification of three flavonoids of a previously described structure, kaempferol-3-O-sophoroside (I), kaempferol-3-O-${\beta}$-Dglucopyranoside (astragalin, II) and kaempferol (III). All compounds (I.III) showed pancreatic lipase inhibitory activities, with $IC_{50}$ values ranging from $20.3{\pm}2.2$ to $9.1{\pm}1.5$ ${\mu}M$, kaempferol (III) showed the most potent inhibitory activity with an $IC_{50}$ of $9.1{\pm}1.5$ ${\mu}M$. The level of activity may depend on the number of C-3 glucosyl group(s) linked to the kaempferol backbone, and the isolated compounds may have promise as pancreatic lipase inhibitors.

Anti-inflammatory Effect of Flavonoids Kaempferol and Biochanin A-enriched Extract of Barnyard Millet (Echinochloa crus-galli var. frumentacea) Grains in LPS-stimulated RAW264.7 Cells (마우스 대식 세포주 RAW264.7에 있어서 LPS처리에 의해 유도되는 염증반응에 대한 식용피(Echinochloa crus-galli var. frumentacea)의 저해효과)

  • In order to compare the anti-inflammatory effects of five selected cereal grains-proso millet, hwanggeumchal sorghum, foxtail millet, barnyard millet, and adlay-the inhibitory activities of 80% ethanol (EtOH) extracts obtained from the individual grains on lipopolysaccharide (LPS)-induced nitric oxide (NO) generation were investigated in RAW264.7 cells. The EtOH extract of barnyard millet (Echinochloa crus-galli var. frumentacea) grains exhibited more potent anti-inflammatory activity than that of the other grains. When the EtOH extract of barnyard millet grains was sequentially fractionated with n-hexane, methylene chloride (MC), ethyl acetate (EtOAc), and n-butanol, the majority of the anti-inflammatory activity was detected in the MC fraction, followed by the EtOAc fraction. Pretreatment with the MC fraction caused downregulation of the expression levels of iNOS- and COX-2-specific transcripts and proteins, as well as proinflammatory cytokine gene transcripts (IL-$1{\beta}$, IL-6, and TNF-${\alpha}$) in LPS-stimulated RAW264.7 cells. Additionally, the MC fraction could suppress not only the LPS-induced nuclear translocation of cytosolic NF-kB, but also the LPS-induced activation of MAPKs, such as ERK, JNK, and p38MAPK. Further analysis of the MC fraction by HPLC identified kaempferol, biochanin A, and formononetin as the major phenolic components. Both kaempferol and biochanin A, but not formononetin, could exert anti-inflammatory effect at the same concentrations as those of the MC fraction. Consequently, these results indicate that kaempferol and biochanin A are among the most effective anti-inflammatory phenolic components in barnyard millet grains. This finding suggests that barnyard millet grains and the MC extract enriched in kaempferol and biochanin A could be beneficial functional food sources that have an anti-inflammatory effect.

Photoprotective Effect and Antioxidative Activity from Different Organs of Morus Bombycis Koidzumi (부위별 산뽕나무의 광보호효과 및 항산화 활성)

  • This study was investigated antioxidatve activity for the purpose of developing antioxidant from Morus bombycis Koidzumi. Antioxidant activities of four different organs of Morus bombycis Koidzumi such as fruit, leaf, stem, and root were examined by radical scavenging effect with 1,1-diphenyl-2-picrylhydrazyl (DPPH). 80% methanol extract from the stem showed strongly antioxidative activity and 80% Ethanol extracts from the root, stem, and fruit had high antioxidative activity among 24 samples tested. The 80% ethanol extract has strong absorbency at UVA region (350 nm). The ethyl acetate (EtOAc) fraction exhibited antioxidative activity with $IC_{50}$ of $15.0\;{\mu}g/ml$ similar to those of synthetic antioxidant, BHT The EtOAc fraction has a good absorbency property as synthetic filter. In the absorbance of various extracts, the 80% ethanol and ethyl acetate extracts from the root of Morus bombycis Koidzumi showed higher absorbancy at 285 nm. The ethyl acetate fraction from the root of Morus bombycis Koidzumi contained total phenolic compounds of 654.8 mg/100 g. These results indicate that phenolic compounds are the major was biological components in the root of morus bombycis Koidzumi extracts. Considering these biological activities, the extracts of Morus bombycis Koidzumi showed a possibility to be used as a new material for natural anti-oxidants and substitutes for synthetic UV sunscreen agents.
