종모돈의 정액성상, 동결-융해 후 정자의 생존성, 혈청 중 FSH, LH, Estradiol-17$\beta$ 및 Testosterone 농도에 미치는 품종과 계절의 영향 (Effects of Semen Characteristics, Frozen-Thawed Sperm Viability and Serum FSH, LH, Estradiol-17$\beta$ and Testosterone Concentrations between Breeds and among Seasons in Boars)

  • 본 연구는 종모돈의 정액성상 동결-융해 후 정자의 생존성 그리고 혈청 중 FSH, LH, estradiol-17β 및 testosterone 농도에 미치는 품종과 계절의 영향을 조사하여 우수한 종모돈의 선발을 위한 기초자료를 얻고자 실시하였다. 요크셔종이 듀록종보다 봄, 여름, 가을, 겨울에서 정액량이 많았으며, 정액농도에서는 차이가 없었다. 계절별 정액량은 듀록 및 요크셔종에서 봄철이 여름, 가을 및 겨울철에 비하여 많았고, 정자농도는 차이가 없었다. 듀록종과 요크셔종에서 각각 봄철에 생산한 정자가 여름, 가을 및 겨울철에 생산한 정자보다 동결-융해 후 정자운동성 및 정상첨체 비율이 높았다. 한편 듀록종과 요크셔종에서 동결-융해 후 정자운동성은 모든 계절에서 요크셔종이 높게 나타났으나, 정상첨체에서는 차이가 없었다. 혈청 중 FSH의 농도를 비교한 결과 요크셔종이 듀록종보다 모든 계절에서 낮은 농도를 나타내었다. 그러나 두 품종 모두에서 각각 계절 간에 차이가 없었다. 혈청 중 LH와 estradiol-17β의 농도를 비교한 결과 요크셔종과 듀록종 간에 차이가 없었다. 또한 두 품종 모두에서 계절 간에 차이가 없었다. 종모돈의 품종별, 계절별 혈청 중 testosterone의 농도를 비교한 결과 요크셔종이 듀록종보다 모든 계절에서 높게 나타났다. 또한 두품종 모두에서 각각 봄철이 여름, 가을 및 겨울철에 비하여 혈청 중 testosterone의 농도가 높은 것으로 나타났다. 이상의 결과를 종합하여 보면, FSH의 농도가 낮을수록 정액생산량이 높은 것으로 나타났으며, 혈청 중 testosterone의 농도가 높을수록 동결-융해 정자의 운동성 및 정상첨체의 비율이 높은 것으로 나타났다.

팔진탕(八珍湯)이 자궁내막증(子宮內膜症)을 유발한 흰쥐에 미치는 영향 (Effects of Paljin-tang on Surgically Induced Endometriosis in Rats)

  • Purpose : This study examined effects of Paljin-tang on endometriosis. Methods : Sugically induced endometriosis in rats were given an oral dose of Paljin-tang for 40 days. The size of the ectopic uterine implants at the serosal wall and the concentration of progesterone, estradiol, tumor necrosis factor(TNF)-${\alpha}$ and interleukin(IL)-2, IL-4, IL-6, and IL-10 in the blood were examined and compared with the control group. Results : The size of the ectopic uterine implants in the experimental group was much smaller than of that in the control group. The concentration of estradiol was significantly lower in the experimental group than in the control group. The TNF-${\alpha}$, IL-4, IL-6 levels was significantly lower in the experimental group than the control group and IL-10 level was higher in the experimental group than the control group. The progesterone, IL-2 levels were similar in the experimental and control groups. Conclusion : These results indicate that Paljin-tang reduces the size of ectopic uterine implants at the serosal wall and inhibits the growth of ectopic uterine implants. This suggests that Paljin-tang is an effective treatment for endometriosis.

Effects of PGF2 α and GnRH during Different Ovarian Status at Onset of Puberty in Murrah Buffalo Heifers (Bubalus bubalis)

  • The objective of the investigation was to study the effect of intramuscular $PGF_2\;{\alpha}$ and GnRH on estrus behavior and ovarian response in Murrah buffalo heifers. Twelve Murrah buffalo heifers at 32 months of age that had not exhibited behavioral estrus symptom were included in the experiment. Out of 12,4 heifers were in follicular phase (plasma estradiol $57.05{\pm}12.52pg/ml$), another 4 heifers were in luteal phase (Plasma progesterone $2.24{\pm}0.25ng/ml$) while the ovaries of remaining four heifers were inactive (estradiol $23.70{\pm}1.66pg/ml$and progesterone $0.32{\pm}0.06ng/ml$). $PGF_2\;{\alpha}$ (25 mg, Lutalyse, im) and GnRH (200 ug, Fertagyl, iv) was administered to each heifer at interval of 10 days. The plasma progesterone concentration decreased within 48 hrs after $PGF_2\;{\alpha}$ injection and followed thereafter with follicular growth, estrus and ovulation. GnRH administration induced follicular growth, elevation of plasma estradiol concentration with subsequent exhibition of behavioral estrus in 2 out of 4 heifers having inactive ovary. The observation reveals that Murrah buffalo heifers at 32 months of age have developed receptors for $PGF_2\;{\alpha}$ and GnRH on ovarian and pituitary tissue respectively and response the single injection of $PGF_2\;{\alpha}$ and GnRH similar to the mature cycling animals.

산삼약침이 자궁내막증 유발 흰쥐에 미치는 영향 (Effects of Cultivated Wild Ginseng Pharmacopuncture on Surgically Induced Endometriosis in Rats)

  • Purpose : This study was carried out to identified the effects of Cultivated Wild Ginseng Pharmacopuncture(CWG Pharmacopuncture) to the rats with surgically induced endometriosis. Methods : Endometrial tissue was implanted in the serosal wall of the small intestine in rats. The rats were divided randomly into an experimental and control group. The experimental group was treated with CWG Pharmacopuncture intravenous injection to the vein of tail and the control group was given same dose of normal saline for 4 weeks. And the size of the ectopic uterine tissue was estimated, and the serum progesterone, estradiol and cytokine($TNF-{\alpha}$, IL-2, IL-4, IL-6, IL-10) concentrations were analyzed. Results : 1. The size of the ectopic uterine implants in the experimental group was much smaller than that in the control group. 2. In analysis of serum hormone, the concentration of progesterone showed higher value than that of control group and estradiol showed lower value. 3. In analysis of cytokine, $TNF-{\alpha}$, IL-2 and IL-6 concentrations showed lower value than that of control group. 4. In analysis of cytokine, IL-4 concentration showed no significant difference and IL-10 concentrations showed higher value than that of control group. Conclusion : These results indicated that CWG Pharmacopuncture is useful treatment for endometriosis.

  • the aim of this study was to investigate whether osteoporosis can be advanced after cerebral infarction Of not and role of female sex hormone. all animals were classified 4 groups; group of ovariectomy (OVXgroup) group of cerebral infarction( INF group), group of combination ovariectomy and cerebral infarction(OVX + INF group), naturally intact group for control data (NOR group ). cerebral infarction was made by Chen,s method with some modification and ovariectomy was performed by Wayforth,s method. experimental data was collected at 15 days, 1 month, 3 months and 6months after starting observation. serum 17 ${{\beta}-estradiol\;(E_2)}$ was determined by radioimmunoassay, for comparision of osteoporosis formation, bone density, serum osteocalcin, Serum total calcium and phosphorus, Serum AST Concentration, Serum ALT Concentration, Creatinine Concentration were also calculated. we have found that cerebral ischemia decreases not only the serum concentration of 17 ${{\beta}-estradiol\;(E_2)}$ and inhibits but also the physiologically compensatory function of the ovariectomized rats and that the decreased estrogen concentration followed by cerebral infarction have not produced osteoporosis, regretfully.

첫발정과 종부시 체중, 연령, 체형, 등지방 두께 및 스테로이드 호르몬 농도에 미치는 종빈돈 품종의 영향 (Effects of breeds on body weight, age, body conformation, backfat thickness and steroid hormone concentrations at first estrus and mating of gilts)

  • This study was carried out to get a fundamental information for improvement of reproductive performance in gilt. We investigated the effects of breeds on body weight, age, body length, wither's height, girth and backfat thickness, and the serum concentrations of estradiol-17${\beta}$, cortisol and progesterone at first estrus and mating of gilts. A total of 47 gilts of Duroc, Landrace and Yorkshire, produced at Livestock Experiment Station, Chungnam livestock sanitation research institute from 2000 through 2002, were used for this experiment. Body weight, age and girth of Duroc at frist estrus and mating were higher than those of Landrace and Yorkshire. There were no differences on body length among the three breeds at frist and mating. Wither's height of Duroc and Yorkshire at first estrus and mating was higher than that of Landrace. Backfat thickness of Yorkshire was thinnest among the three breeds at first estrus, but there were no differences on backfat thickness among the three breeds at first mating. Serum estradiol-17${\beta}$ concentration of Landrace(45.0 pg/ml) at first estrus was higher than that of Yorkshire(27.4 pg/ml) and Duroc(21.8 pg/ml), but there were no differences on estradiol-17${\beta}$ concentration (from 18.5 to 31.9 pg/ml) among the three breeds at first mating. Serum cortisol concentration of Duroc at first esturs and mating was higher than that of Landrace and Yorkshire. There were no differences on serum progesterone concentration among the three breeds at first estrus and mating of gilt.

  • A competitive type immunoassay method for 17$\beta$-estradiol(E2) based on the idiotypic anti-idiotypic antibody and time-resolved fluorescence is described. The anti-idiotypic antibody(Ab2) produced to E2 binding site of the primary idiotype antibody (Ab1) was labelled with europium and was allowed to compete with E2 standards or serum sample for the binding sites of Ab1 which was bound to 2nd antibody captured ontothe surface of microtitre plates. Fluorescence measured by time-resolved fluorometer was inversely proportional to the concentration of E2 over the range 5~500pg/well. The sensitivity of the assay was 5pg per well which was compatible with that ofradioimmunoassay using the same Ab1 and 3H-E2 as a tracer. One great advantage of this method described here was to enable antibodies to be labelled instead of haptens, and thus makes it easier to develop sensitive and robust immunoassay systems specially for haptens.

17$\beta$-estradiol 처리에 따른 해산 어류의 vitellogenin 생성 (Vitellogenin Production of Marine Fishes Exposed to E2)

  • A common used endpoint in bioassays testing the estrogenicity of chemicals including endocrine disruptors is the induction of egg yolk Precursor vitellogenin in male fish. Two marine fishes (Sebastes schlegeli and Paralichthys olivaceus) were exposed to the 17$\beta$-estradiol(E2) to determine the vitellogenin production. Vitellogenin was measured in fish blood using SDS-PAGE and Densimetry. Results showed that exposure to E2 caused vitellogenin in male fish. Especially, vitellogenin levels in young fish were about 4 times higher than in adult fish, which means young fish are more sensitive to E2 exposure. And plasma vitellogenin in fish increased related to E2 concentration and exposure duration.

High level of estradiol in a Holstein-Friesian cow with sacroiliac luxation after calving

  • This is the report of sacroiliac luxation in a Holstein-Friesian cow, describing the progression of the sacroiliac luxation from the onset of its development with changes in estradiol (E2) and serum chemistry. The high estrogen concentration was assumed to be a predisposing factor of the disease that relaxed the pelvic muscles and a secondary slippery finally caused the sacroiliac luxation. The E2 was present at levels four times higher in the cow with sacroiliac luxation (167 pg/mL) than in normal cows. Above normal levels of creatinine kinase, aspartate aminotransferase, and lactate dehydrogenase were also observed in cow with sacroiliac luxation.

황분(黃芬)의 경구와 직장투여가 실험적으로 유발된 자궁내막증에 미치는 영향 (Effects of Scutellaria baicalensis on Surgically Induced Endometriosis in Rats after Oral or Rectal Administration)

  • Purpose: Scutellaria baicalensis is a herb which has effect of eliminating heat, drying dampness, purging away fire, detoxicating. Clinically, has been used to remove fever and control of inflammation. We have studied in series to investigate the effective traditional oriental medicines of treatment for endometriosis. In this study, the effects of Scutellaria baicalensis administrated orally or rectally on surgically induced endometriosis in rat were investigated. Methods: The endometrial tissues of rats were autografted to their small intestine. Rats with surgically induced endometriosis were administered with Scutellaria baicalensis extracts for 40 days orally or rectally. We observed the tissue size of ectopic uterine implants at the mesenteric with the naked eye and examined the concentration of estradiol, progesterone, $TNF-{\alpha}$ and IL-2, IL-4, IL-6 and IL-10 in serum. Also, we inquired which was more effective between oral administration groups and rectal administration groups. Results: The size of ectopic uterine implants of orally or rectally treated groups was much smaller than those of control groups. The concentrations of estradiol, progesterone, $TNF-{\alpha}$, IL-2 and IL-6 were significantly decreased in oral administration experimental group compared with the control group. The concentrations of progesteron, $TNF-{\alpha}$, IL-2 and IL-6 were significantly decreased in rectal administration experimental group compared with the control group. The efficiency of oral administration group was better than that of rectal administration group at the serum concentration of estradiol and IL-4. Conclusion: Our data suggest that Scutellaria baicalensis inhibit the inflammation induced from ectopic uterine implants at the mesenteric wall and inhibit the growth of ectopic uterine implants. Judging from the above results, it can be suggested that Scutellaria. baicalensis should be a useful agent for inhibiting the inflammation induced from ectopic uterine implants and the proliferation of endometriosis. Maybe, oral administration of Scutellaria baicalensis is more effective than rectal administration.
