Inflammation Relief Effect through ECS Control of Galgeunhwanggeumhwangryeon-tang Extract in Atopic Dermatitis (아토피피부염에서 갈근황금황련탕 추출물의 ECS 조절을 통한 염증 완화 효과)

  • Objective The purpose of this study was to confirm the effects of Galgeunhwanggeumhwangryeon-tang in reducing inflammation through the endocannabinoid system (ECS) control in atopic dermatitis. Methods 8-week-old Balb/C mice were divided into 4 groups: contorl group (Ctrl), lipid barrier elimination group (ADE), palmitoylethanolamide treated group after lipid barrier elimination (PEA), and Galgeunhwanggeumhwangryeon-tang applied group after lipid barrier elimination (GGRT). After inducing atopic dermatitis, cannabinoid receptor (CB) 1, CB2, CD68, phosphorylated inhibitor kappa B (p-IκB), inducible nitric oxide synthase (iNOS), substance P and serotonin were observed to confirm the regulation of the ECS, macrophage activity and mast cell activity. Results CB1 and CB2 showed higher positive reactions in the GGRT than in the LBE and PEA. CD68, p-IκB and iNOS showed higher positive reaction in the LBE, PEA and GGRT than in the Ctrl, but the increase in the positive reaction was lower in the GGRT compared to the LBE and PEA. Substance P and serotonin showed higher positive reaction in the LBE, PEA and GGRT than in the Ctrl, but the increase in the positive reaction was lower in the GGRT compared to the LBE and PEA. Conclusions The effects of Galgeunhwanggeumhwangryeon-tang were confirmed though the regulation of the ECS, macrophage activity and mast cell activity.

Th2 Differentiation Control through Formation of Skin Fat Barrier of Sabaek-san extract (사백산 추출물의 지방장벽 생성을 통한 Th2 분화 조절)

  • Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Sabaek-san extract may produce to improves skin lipid barrier damage. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DXT, n=10), and Sabaek-san extract treatment group after lipid barrier elimination (SBT, n=10). Sabaek-san extract was administered for 3 d after removal of the skin fat barrier in SBT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin, transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7), and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in SBT group than in LBE and DXT group. Also, pH and TEWL were significantly decreased with SBT group. However, filaggrin was significantly increased in SBT group compared to LBE, DXT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in SBT compared to the LBE and DXT. Conclusions As a result of this study, SBT administration decreased pH, TEWL, and increased filaggrin, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, SBT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.

Hydrolytic Reactivity and Holographic Quantitative Structure-Activity Relationship Analyses on the Melanogenesis Inhibitory Activities of Alkyl-3,4-dihydroxybenzoate and N-Alkyl-3,4-dihydroxybenzamide Derivatives (Alkyl-3,4-dihydroxybenzoate와 N-Alkyl-3,4-dihydroxybenzamide 유도체들의 가수분해 반응성과 멜라닌 생성 저해활성에 관한 홀로그램(H) QSAR 분석)

  • Holographic quantitative structure activity relationships (HQSAR) between the melanogenesis inhibitory activities of alkyl-3,4-dihydroxybenzoate (A) and N-Alkyl-3,4-dihydroxybenzamide (B) derivatives were analyzed and discussed. The statistical results of HQSAR model for the activities showed the best predictability of the activities based on the cross-validated $r^2_{cv}\;(q^2=0.674),$ non-cross-validated, conventional coefficient $(r^2_{ncv}=0.936).$ The melanogenesis inhibitory activities and hydrolytic reactivity of (A) were slightly higher than that of (B) (A>B) and the activities depends upon the $R_1-substituents\;(R_1>R_2).$ It has been found using frontier molecular orbital (FMO) theory that the hydrolysis reactions of (A) and (B) proceeded to an orbital-controlled reactions, while the nucleophillc addition-elimination reactions $(Ad_{N-E})$ between LUMO energy of (A) and (B) and HOMO energy of water molecule are occurred.

  • Iron alkynyl(amino)carbene complexes, (${\beta}$-aminovinyl)carbene complexes, ${\eta}^3$-(2-(alkylcarbonyl)vinyl)carbene complexes, and a 3-aminoallenylidene complex were formed in the reactions of iron alkynyl(ethoxy)carbene complexes with amines. The ratio of the products, which were formed by substitution reaction, the Michael addition of amines, rearrangements after the addition reaction, and the addition followed by the elimination of the ethoxy group, respectively, was dependent on reaction temperature, the substituent of the alkynyl moiety, and employed amines.

Substituent Effect on Fragmentations and Ion-Molecule Reactions of Ionized Alkyn Alcohols

  • The fragmentation patterns and ion-molecule reactions of two alkyn alcohols, 2-propyn-1-ol (HC≡$CCH_2$OH) and 2-methyl-3-butyn-2-ol (HC≡CC($CH_3)_2$OH), were investigated using Fourier transform mass spectrometry (FTMS). The most abundant fragment ions formed from the molecular ions were [M-H]$^+$ for 2-propyn-1-ol and [M-$CH_3]^+$ for 2-methyl-3-butyn-2-ol. The dehydrated ion, [M-$H_2O]^+$ was formed only from 2-propyn-1-ol in which $\alpha$ -hydrogen atoms were available for $\alpha,\;\alpha$ -elimination reaction. The protonated molecules were dissociated into [M+H-$H_2O]^+$ and [M+H-$C_2H_2]^+$ through dehydration and deacetylenylation processes. The formations of [M+H-$H_2O]^+$ and [M+H-$C_2H_2]^+$ from 2-methyl-3-butyn-2-ol were more favorable than those from 2-propyn-1-ol due to stabilization by two methyl groups at $\alpha$ -carbon. Ion-neutral complexes formed at long ion trapping time gave dehydrated and/or deacetylenylated ion products by further dissociation.

Dynamics of OH Production in the Reaction of O(1D2) with Cyclopropane

  • The OH($X^2{\Pi}$, ${\upsilon}^{\prime\prime}=0,1$) internal state distribution following the reaction of electronically excited oxygen atom ($O(^1D_2)$) with cyclo-$C_3H_6$ has been measured using laser-induced fluorescence, and compared with that following the reaction of $O(^1D_2)$ with $C_3H_8$. The overall characteristics of the OH internal energy distributions for both reactions were qualitatively similar. The population propensity of the ${\Pi}(A^{\prime})$ ${\Lambda}$-doublet sub-level suggested that both reactions proceeded via an insertion/elimination mechanism. Bimodal rotational population distributions supported the existence of two parallel mechanisms for OH production, i.e., statistical insertion and nonstatistical insertion. However, detailed analysis revealed that, despite the higher exoergicity of the reaction, the rotational distribution of the OH following the reaction of $O(^1D_2)$ with $C_3H_8$ was significantly cooler than that with cyclo-$C_3H_6$, especially in the vibrational ground state. This observation was interpreted as the effect of the flexibility of the insertion complex and faster intramolecular vibrational relaxation (IVR).

  • Rate constants for the reaction of methyl chloroformate with substituted anilines, and for the halogen exchanges in phenyl chloroformate have been determined in acetone. Although the rate data can be interpreted equally well with the addition-elimination mechanism($S_AN$) involving an intermediate, results of MO and isotope effect studies strongly favor the synchronous ($S_N2$) mechanism for the reactions studied. It was concluded that for the fast reacting nucleophiles the transition state is of "late" type while for the slow reacting nucleophiles it is of "early" type.

  • The metallation of $\alpha$-methylpyridine 1(a∼f) with n-BuLi produced $\alpha-methylenylpyridinium$ salt 3(a∼f) by elimination of butane. The trapping reactions of 3(a∼f) with $Me_3SiCl\;and\;Me_2SiClCH(SiMe_3)CH_2tBu$ produced only 4(a∼f) and 5(a∼f). The $\alpha$-hydrogen atom of silylated methylene group in 4(a∼f) is more reactive than unreacted $CH_3$ of 4(a∼f) itself and 1(a∼f) toward n-BuLi at low temperature in pentane medium.

  • This review summarizes gender differences in pharmacokinetics, pharmacodynamics, and adverse drug reactions. Gender differences in pharmacokinetics are categorized by four major factors: absorption/bioavailability, distribution, metabolism, and elimination. There are sex-based differences in gastric emptying time, gastric alcohol dehydrogenase activity, apparent volume of distribution, ${\alpha}1$-acid glycoprotein level, phase I (CYP) and phase II metabolizing enzymes, glomerular filtration rate, and drug transporters. This review also reports gender differences in pharmacokinetics and pharmacodynamics of cardiovascular agents, central nervous system acting agents and antiviral agents. In addition, it has been reported that females experience more adverse reactions such as coughing, tachycardia, nausea, vomiting, rash, hypersensitivity, hepatotoxicity, and metabolic disorder after taking cardiovascular, central nervous system acting and antiviral agents. Therefore, in order to provide optimal drug dosage regimens both in male and female, gender differences in pharmacokinetics, pharmacodynamics, and adverse drug reactions must be considered.

Novel organic catalysts for nucleophilic fluorination including F-18 radiofluorination

  • To overcome the low reactivity and solubility of alkali metal fluorides (MFs), various types of phase transfer catalysts (PTCs) have been developed over the last decades. However, since the fluoride activated by such PTC sometimes has a strong basicity, it may cause various side reactions such as elimination reaction or hydroxylation reaction in the nucleophilic fluorination reaction. Also, they may cause separation problems in the compound purification process. In recent advanced study, various PTCs have been developed to solve these problem of conventional catalyst. In this review, we would like to introduce three kinds of novel multifunctional organic catalysts such as bis-tert-alcohol-functionalized crown-6-calix[4]arene (BACCA), easy separable pyrene-tagged ionic liquid (PIL) by reduced graphene oxide (rGO), and tri-tert-butanolamine organic catalyst.
