Nutritional compositions and antioxidative activities of two blueberry varieties cultivated in South Korea (국내산 두 품종 블루베리의 영양성분 및 항산화 활성 비교)

  • The nutritional compositions and antioxidative activities of the Kimcheon lowbush and Pyungtaek highbush blueberries cultivated in South Korea were investigated. The approximate compositions, pH, and soluble-solid and mineral contents of P, Ca, Mg, Na, and Zn were determined. Both blueberry cultivars had mainly fructose and glucose as free sugars while disaccharides such as sucrose, maltose, and lactose were not detected in both. Oleic, linoleic, and linolenic fatty acids were the major fatty acids in both types of blueberries. The total of 16 free amino acids, including seven essential and nine non-essential amino acids, were higher in the Pyungtaek highbush blueberry. Among the amino acids, arginine was especially much higher in the Pyungtaek highbush blueberry. The anthocyanin, resveratrol, and polyphenol compounds, which are the important biologically active compounds in blueberries, were found. The anthocyanin contents of the Kimcheon lowbush and Pyungtaek highbush blueberries were 22.0 and 18.1 mg/100 g, respectively; the resveratrol contents by HPLC, 0.12 and 0.11 mg/100 g; and the total polyphenol contents, 141.3 and 138.4 mg/100 g. The electron-donating ability determined based on the DPPH radical scavenging activity was increased in a concentration-dependent manner, and it was higher than that of the Pyungtaek highbush blueberries, which implies that it is highly correlated with the higher amounts of total polyphenol, anthocyanin, and resveratrol in it. In conclusion, the two varieties of Korean blueberries can be suggested as potential sources of high-value-added functional foods.

Anti-inflammatory Activities Verification of Ambrosia trifida L. extract in RAW 264.7 Cells (RAW 264.7 세포에서의 단풍잎돼지풀 추출물의 항염증 활성 검증)

  • This study was performed to evaluate the anti-inflammatory activities of 70% ethanol extract from Ambrosia trifida L. (AT). The electron donating ability and ABTS+ radical scavenging ability of extract from AT was shown to be 84.1% and 92.5% at 1,000 ㎍/ml concentration. The astringent effect of extract from AT was shown to be 94.7% at 1,000 ㎍/ml. The anti- inflammatory activities of extract of AT were investigated using RAW 264.7 cells induced by lipopolysaccharide (LPS). The cell toxicity effect of AT extract on RAW 264.7 performed MTT assay. As a result of the measured cell toxicity effect, 90% or more was shown with cell viability at a 500 ㎍/ml concentration. In nitric oxide synthesis inhibition effect, it was shown that extract from AT concentration dependent inhibited nitric oxide production. The protein expression inhibitory effect of AT extract was measured by western blot at 25, 50, and 100 ㎍/ml concentration and the β-actin used as a positive control. Consequently, the inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 protein expression inhibitory effect was decreased by 8.6%, 25.1% at 100 ㎍/ml concentration. The phosphorylation of extracellular signal-regulated kinase 1/2, p38, c-Jun NH2-terminal kinase and Iκ-Bα protein expression inhibitory effect was a decreased dependent concentration. The mRNA expression inhibitory effect was measured by reverse transcription - polymerase chain reaction at 25, 50, and 100 ㎍/ml concentration and the glyceraldehyde-3-phosphate dehydrogenase used as a positive control. Consequently, the iNOS, COX-2, interleukin (IL)-1β, IL-6 and tumor necrosis factor-α mRNA expression inhibition effect was a decreased dependent concentration in an LPS-activated macrophage. In conclusion, AT extract may have some effects on inflammatory factors as potential anti-inflammatory agents and natural substance for cosmetics.

Antioxidant Activities of Phenolic Compounds from Medicinal Plants (Hibiscus esculentus, Cirsium japonicum, Zizania latifolia and Kalopanax pictus) (약용식물(오크라, 엉컹퀴, 엄나무, 줄풀) 유래 페놀성 물질의 항산화 활성)

  • In this study, the antioxidant activity of water and ethanol extracts from Hibiscus esculentus, Cirsium japonicum, Zizania latifolia and Kalopanax pictus for functional food source were examined. The optimal conditions for phenolic compounds extraction from medicinal plants were at 50% ethanol with Hibiscus esculentus and Cirsium japonicum var. ussuriense, at 40% ethanol with Kalopanax pictus and at 60% ethanol with Zizania latifolia. The total phenolic contents from the extracts of medical plants were determined to be 2.72~34.15 mg/g in the water extracts and 2.83~34.23 mg/g in the ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 74% at the low concentration of $50{\mu}g/mL$. The ABTS radical-cation decolorization was above 88% at $100{\mu}g/mL$ concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) in the water and ethanol extracts of the Cirsium japonicum var. ussuriense extracts was $1.73{\pm}0.02PF$ and $1.76{\pm}0.01PF$ at $50{\mu}g/mL$ concentration respectively, and was higher than those of the other medicinal-plant extracts. The TBARs inhibition rates of all the medicinal-plant extracts, were above 80% at the $50{\mu}g/mL$ concentration except Hibiscus esculentus. These results confirmed that the various oriental medicinal plants (Hibiscus esculentus, Cirsium japonicum var. ussuriense, Kalopanax pictus and Zizania latifolia) that were included in this study are useful anti-oxidant and functional-food resources.

Anti-oxidative and Anti-cancer Activities of Extracting of Yacon (Yacon 추출물의 항산화 및 항암 활성)

  • This study investigated the anti-oxidative and anti-cancer activities by extracting of Yacon by fraction with organic solvents. The content of total phenol was the highest by 45.53% in ethyl acetate fraction and the results of electron donating abilities were 65.20% ($100\;{\mu}g/ml$), 91.81% ($500\;{\mu}g/ml$) and 95.06% ($1000\;{\mu}g/ml$) and those of nitrite scavenging abilities were 11.71% ($100\;{\mu}g/ml$), 36.81% ($500\;{\mu}g/ml$), 59.70% ($1000\;{\mu}g/ml$) in ethyl acetate fraction, which were higher concentration than the control group. Xanthine oxidase inhibitory activities were 23.74% ($100\;{\mu}g/ml$) and 43.41% ($500\;{\mu}g/ml$) in low concentration, which showed higher activities than the control group. After measuring the growth inhibitory activities for cancer cells, it has been found that hexane fraction had the highest growth inhibitory activities by 23.75% ($10\;{\mu}g/ml$), 34.67% ($50\;{\mu}g/ml$), 54.21% ($100\;{\mu}g/ml$) for SNU-1. Meanwhile, hexane fraction demonstrated high growth inhibitory activities by 41.38% ($10\;{\mu}g/ml$), 50.53% ($50\;{\mu}g/ml$), 60.91% ($100\;{\mu}g/ml$) and butanol fraction had 17.05% ($10\;{\mu}g/ml$), 43.87% ($50\;{\mu}g/ml$), 62.99% ($100\;{\mu}g/ml$) for HeLa. Therefore, it has been believed that there were peculiar properties of anti-oxidative and anti-cancer activities depending on each fraction. Furthermore, it has been expected that further studies on the development of functional food and concrete components depending on the components of each fraction to be conducted.

The Antioxidative Activity of Glutathione-Enriched Extract from Saccharomyces cerevisiae FF-8 in In Vitro Model System (In Vitro 과산화지질에 미치는 glutathione 고함유 효모 Saccharomyces cerevisiae FF-8의 항산화효과)

  • The Antioxidative accvities of the cell free extracts containing high glutathione by Saccharomyces cerevisiae FF-8 were tested in vitro experimental models : DPPH method for radical scavenging activity, ferric TBA method and ferric thiocyanate method using linoleic acid and tissue microsome for lipid peroxidation inhibitions. The concentration of intercellular glutathione by cultivating S. cerevisiae FF-8 in the YM optimal medium obtained $204\mug/ml$, which was increased by 2.76-fold from $74\mug/ml$ in the YM basal medium. A comparition between the YM basal medium and the YM optimal medium on antioxidative substance produced by S. cerevisiae FF-8 was investigated. In DPPH ($\alpha, \alpha-diphenyl-\beta-picrylhydrazyl$) method, the electron donating activity of the glutathione produced by S. cerevisiae FF-8 cultured in the YM optimal medium was as high as that of BHT ($ 0.05\%w/v $). The antioxidative a.tivity was measured by inhibition against lipid peroxidation of rat tissues' microsomes. The results of anti-oxidant activity of the cell free extracts by S. rerevisiae FF-8 cultured in the YM optimal medium was shown in the following order . $ liver 60.98\% > kidney 56.43\% > heart 52.91\% > brain 52.13\% > testis 45.57\% > spleen 42.95\% $. In antioxidative activities determined by ferric thiocyanate method and TBA methods against lipid peroxidation, the lipid peroxidation in the control mixture increased more rapidly than the typical peroxidation curve of linoleic acid from one day. The antioxidative activity of the cell free extracts by cultivating S. cerevisine FF-8 in the YM optimal medium were higher than that of the YM basal medium. These data indicate that the cell free extracts containing a high intercellular glutathione of S. cerevisiae FF-8 cultured in YM optimal medium showed strong antioxidative capacities by DPPH radical scavenging activity and ferric thiocyanate and TBARS measurements.

A Study on the Reactions of Diamines with 2,5-Dimethoxytetrahydrofuran and 1,3-Acetonedicarboxylic Acid (1,3-Acetonedicarboxylic acid와 2,5-dimethoxytetrahydrofuran에 의한 diamine들의 반응에 관한 연구)

  • In order to synthesize novel anticonvulsants, we researched that the reactions of diamines with 2,5-dimethoxytetrahydrofuran and 1,3-acetonedicarboxylic acid. The reaction of ethylenediamine with 2,5-dimethoxytetrahydrofuran and 1,3-acetonedicarboxylic acid afforded 8-(2-pyrrol-1-yl-ethyl)-8-aza-bicyclo[3,2,1]octan-3-one (yield; 5.0%) and 1,2-di-(8-aza-bicyclo[3,2,1]octan3-onyl)ethane (yield; 17.0%). In case of 1,3-diaminopropane, 8-(3-pyrrol-1-yl-propyl)-8-aza-bicyclo[3,2,1]octan-3-one(yield; 6.0%) and 1,3-di-(8-aza-bicyclo[3,2,1]octan-3-onyl)propane (yield; 21.0%) were obtained. In case of 1,8-diaminooctane, 8-(8-pyrrol-1-yl-octyl)-8-aza-bicyclo-[3,2,1]octan-3-one (yield; 2.6 %) and 1,8-di-(8-aza-bicyclo[3,2,1]octan-3-onyl)octane (yield; 24.9%) were obtained. In diaminobenzene reactions, synthetic yields of 8-aza-bicyclo-[3,2,1]octan-3-one derivatives were higher than those of pyrrole derivatives because re actions were done under room temperature. The longer the carbon chain of diaminoalkane is, the more reactive N atom is due to more electron donating effect, and the less steric hindrance around the carbon gave the higher chemical yields. The reaction of p-phenylenediamine as a diaminobenzene with 2,5-dimethoxyte-trahydrofuran and 1,3-acetonedicarboxylic acid produced p-dipyrrolylbenzene (yield; 4.0%), 8-(4-pyrrol-1-yl-phenyl)-8-aza-bicyclo[3,2,1]octan-3-one (yield; 12.0%), and 1,4-di-(8-aza-bicyclo[3,2,1]octan-3-onyl)benzene (yield; 59.0%). In case of m-phenylenediamine, 8-(3-pyrrol-1-yl-phenyl)-8-aza-bicyclo[3,2,1]octan-3-one(yield; 2.0%) and 1,3-di-(8-aza-bicyclo[3,2,1]octan-3-onyl)benzene (yield ; 28.0%) were obtained. But, synthesis of 1,2-di-(8-aza-bicyclo[3,2,l]octan-3-onyl)benzene by treatment of o-phenylenediamine was not successful, presumably due to the steric hindrance of 8-aza-bicyclo-[3,2,1]octan-3-one rings.

Biological Activity of Omija (Schizandra chinensis Baillon) Extracts (오미자(Schizandra chinensis Baillon) 추출물의 생리활성)

  • Extracts from Schizandra chinensis Baillon (Korean name: Omija) were tested for antioxidant and their inhibitory activities of ${\alpha}-amylase$ and ${\alpha}-glucosidase$. Total contents of phenolics were found as 4.35 mg/g (water extract)${\sim}$6.35 mg/g (60% ethanol extract). Electron donating ability (EDA), ABTS [2,2'-azinobis(3-ethyl-benzothiaznoline-6-sulfonic acid)] radical decolorization, antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARS) were measured for the antioxidative activity of the extracts from S. chinensis. The water extract were determined as 97.5% at 200 ${\mu}g/ml$ while the activity of 60% ethanol extract were 96.2% at 200 ${\mu}g/ml$ in EDA. The 60% ethanol extract showed higher antioxidant activity than water extract when evaluated by ABTS radical decolorization, antioxidant PF and TBARS. ${\alpha}-Amylase$ inhibitory activity of water extract was similar with that of 60% EtOH extract. ${\alpha}-glucosidase$ inhibitory activities of water extract (97.4%) was higher than that of 60% ethanol extract (84.5%) at 200 ${\mu}g$/ml. The water extract from S. chinensis did not show an antimicrobial activity against Helicobacter pylori, but the 60% ethanol extract showed high antimicrobial activities such as 23 ${\pm}$ 1.6 mm of clear zone in 200 ${\mu}g/ml$ of phenolics. The result suggest that the water and 60% ethanol extract from S. chinensis will be useful as natural antioxidants and functional foods.

Quality Characteristics and Antioxidant Activity of Chrysanthemum indicum L., C. boreale M., and C. zawadskii K. Flowers Candies (감국, 산국 및 구절초꽃 캔디의 항산화활성과 품질특성)

  • In order to investigate the quality characteristics of candies prepared with Compositae species flower extracts, the samples of Chrysanthemum indicum Linne (CI), C. boreale Makino (CB), and C. zawadskii var. latilobum (Maxim.) Kitam (CZ) were extracted with hot water and concentrated, and then added with various concentrations of 5$\sim$20% for candy preparation. L-value of candy color was decreased, but a- and b-value were increased with increasing amount of added extracts. pH was decreased, but total acidity was increased with increasing amount of added extracts. Moisture content of CI, CB, and CZ candies were 2.61$\sim$4.11, 2.46$\sim$4.57, and 2.27$\sim$4.43%, respectively. Hardness of candies were decreased with increasing amount of added extracts. CZ candy of 20% added extract showed the highest total polyphenol, flavonoid content, electron donating ability and anol, flavonaid content at 2.71 mg/g, 0.86 mg/g, 76.24%, and 3.17 mg AA eq/g, respectively. In sensory evaluation, the CI and CZ candies of 10% added extract obtained higher sensory scores.

Quality Characteristic of Glasswort (Salicornia herbacea L.) Fermented by Bacillus subtilis (Bacillus subtilis를 이용하여 발효시킨 퉁퉁마디(Salicornia herbacea L.)의 품질 특성)

  • In this study, we investigated the quality of glasswort (Salicornia herbacea L.) fermented by Bacillus subtilis at $37^{\circ}C$ for 48 hours. We determined the changes in temperature, the contents of moisture, crude protein, crude fat, crude ash, carbohydrate, solid content, pH, Brix, salinity, free amino acid, Hunter's color value, electron donating ability (EDA) activities and angiotensin converting enzyme (ACE) inhibition. The contents of crude protein, crude fat, crude ash and carbohydrate after fermentation to mixing ratio of glasswort were $10.45{\sim}30.18%$, $11.69{\sim}19.26%$, $17.65{\sim}25.56%$ and $21.01{\sim}59.10%$ (dry basis), respectively. The solid content, pH, sugar and salinity of fermented glasswort were $39.56{\sim}52.25%$, $6.01{\sim}6.71$, $1.2{\sim}1.7$ Brix and $0.3{\sim}0.6%$, respectively. Total amino acid level of the fermented glasswort was $447.57{\sim}668.89\;mg$/100 g, and was the highest in glasswort mixed with rice bran at a ratio of 1 (w) : 1 (w). Moreover, aspartic acid, asparagine, glutamic acid, alanine, valine, $\beta$-aminoisobutyric acid, lysine and arginine were the main free amino acids. EDA activities and ACE inhibition after fermentation were $46.66{\pm}1.21{\sim}50.87{\pm}1.84%$ and $96.77{\pm}1.23{\sim}97.56{\pm}1.23%$, respectively.

Optimization of Microwave Extraction Conditions for Antioxidant Phenolic Compounds from Ligustrum lucidum Aiton Using Response Surface Methodology (반응표면분석법을 이용한 여정자의 페놀계 항산화 성분에 대한 마이크로웨이브 추출조건 최적화)

  • Response surface methodology (RSM) was applied to optimize the microwave-assisted extraction (MAE) conditions for electron-donating ability, total phenol content, and total flavonoid content of Ligustrum lucidum Aiton. Ligustrum lucidum Aiton from different regions was tested, and Ligustrum lucidum Aiton from Haenam was chosen due to its higher total phenolic content, total flavonoid content, DPPH radical scavenging activity and ABTS radical scavenging activity compared to the other samples. Central composite design was used to optimize extraction of Ligustrum lucidum Aiton from Haenam as well as determine the effects of extraction temperature ($X_1$) and extraction time ($X_2$) on dependent variables ($Y_n$). Determination coefficients ($R^2$) of the regression equations for dependent variables ranged from 0.8858 to 0.9517. The optimum points were $131.68^{\circ}C$ for extraction temperature and 5.49 min for extraction time. Predicted values of the optimized conditions were acceptable when compared to experimental values.
