Meloidogyne incognita 알을 제어하는 Paecilomyces lilacinus의 현미경적 관찰 (Microscopic observation of Paecilomyces lilacinus that have control over Meloidogyne incognita eggs)

  • M. incognita are plant-parasite nematode that cause severe damage to the crops. P. lilacinus are renowned for inhibitation of development of M. incognita's egg. We make a study for enzymatic examining the cause closely that P. lilacinus suppress development of M. incognita's egg by parasiting. The research result is explained the place below. 1. The egg that is exposed to co-enzymes which are cultured in the broth culture starts to change the membrane of egg from 3days. And in 10days, that membrane completely disappear. These are observed through light microscope. Therefore, we know that M. incongnita are controlled by extracellular lytic enzymes that are produced by P. lilacinus. 2. Through scanning electron microscope, we can find that the egg that is attacked by P. lilacinus loses it's membrane gradually, and that loss of the membrane causes transform, which suppresses the development of egg.

The Expression of Egg Plant Flavonoid 3',5'-Hydroxylase Gene in Tobacco Plants (Nicotiana tabacum cv. Xanthi)

  • The anthocyanin gene encoding flavonoid 3',5'-hydroxylase(F3,5H) was normally expressed in Nicotiana tobacco (Xanthi) plants cocultivated with Agrobacterium tumefaciens LBA4404 carrying egg plant flavonoid 3',5'-hydroxylase cDNA. Northern blot analysis showed the normal expression of F3', 5'H gene from transgenic plants. Here we found the phenotypic differences between transgenic plants and wild-type plants. The petal shape of transgenic plants showed more round shape and around petal tube area was compared to that of wild-type tobacco plants. And the petal color of transgenic plants was much lighter than that of wild-type tobacco plants.

The Effect of Mustard Meal in Laying Hen Diets

  • A total of 252, 50 week-old Isa-brown laying hens were randomly allotted to 7 groups of 3 replicates. Mustard meal (MM) which is a by-product from mustard processing plant, was dried under the sun or in a gas heated pan. It contained on DM basis 30-32% CP, 19-22% EE and 12-13% CF. The meal from either drying method was incorporated into the diets at 0, 10, 20 and 30% which was equivalent to the substitution levels for soybean meal at 0, 31, 63 and 94%, respectively. All birds were individually kept in battery cages where feed and water were freely accessed throughout 84 days experimental period. It was found that egg production, feed intake, body weight gain and egg weight significantly decreased with the increased MM level. The inclusion of 20% MM did not show a significant difference in egg production and quality from the control, but produced 6-8% lower egg production. Feed intake was linearly decreased with the MM levels, except the 10% sun dried MM group. Fat deposition of the birds fed MM diets significantly decreased, while kidney weight increased when compared with the control group. However, the weight of thyroid glands and spleen trended to be heavier in the MM groups, but this was not significantly different among dietary treatments. It was concluded, MM from both drying methods could be incorporated in laying hen diets at the level of 10% without any adverse effect.

First Report on Isolation of Salmonella Enteritidis from Eggs at Grocery Stores in Korea

  • Salmonella Enteritidis is responsible for causing foodborne diseases upon consumption of egg products. While cases of S. Enteritidis isolation from eggs have been reported in other countries, no such cases have previously been reported in Korea. In this study, we report the first isolation and identification of S. Enteritidis from domestically distributed eggs in Korea. Eggs were collected from eight countrywide grocery stores during different seasons between 2011 and 2012. Egg contents and washing solution of egg shells were incubated in buffered peptone water, and the enriched broth was further enriched in tetrathionate broth and Rappaport-Vassiliadis. The secondary enriched broth was streaked on xylose lysine desoxycholate agar. The suspected colonies were confirmed to S. Enteritidis by a biochemical test, serotyping, and PCR test. Genetic relatedness among the isolates was analyzed using Diversilab Salmonella kit. Three strains of S. Enteritidis were isolated from egg contents and egg shells collected from grocery stores of the Eumseong-city in the fall of 2011. All three stains showed resistance to chloramphenicol, streptomycin, nalidixic acid, and ampicillin by the disk diffusion method. In addition, the isolates showed more than 99% DNA homology, indicating that they were presumably identical strains. Therefore, there is a requirement to monitor and control against S. Enteritidis from eggs in Korea.

Pepper mild mottle virus에 대한 난황항체의 생산과 혈청학적 진단에의 활용 (Production of Egg Yolk Immunoglobulin and Its Application for Pepper mild mottle virus in Serological Tests)

  • 닭을 이용한 egg yolk immunoglobulin (IgY)의 생산과 이용 가능성을 조사하기 위하여 pepper mild mottle virus (PMMoV)로 닭을 면역하였다. Ring-test로 달걀의 난황에서 분리 정제한 IgY의 역가를 조사한 결과 IgY의 최고 역가는 1:2,560이었고 달걀 l 개에서 대략 $60{\sim}80 mg$의 IgY를 얻을 수 있었다. 최고역가를 갖는 IgY를 이용하여 Gelrite gel 이중확산법, 효소면역항체법 (ELISA), 조직자국면역반응법을 실시한 결과 간접면역효소항체법의 PMMoV 검출 감도는 1 ng/ml 이었고, 검정법의 바이러스 검출감도와 특이도는 IgG와 비슷한 경향을 보였다. 그러므로 한 마리의 닭으로부터 대량으로 생산 가능한 IgY는 식물바이러스에 대한 항체생산과 혈청학적 검정에 유용할 것으로 생각한다.

약용 식물 가공 부산물의 첨가가 산란계의 생산성 및 난질에 미치는 영향 (Effect of Dietary Medicinal Plant by-products on Egg Production and Egg Quality in Laying Hens)

  • 본 실험은 약용 식물 가공 부산물의 산란계에 대한 급여 효과를 시험하기 위해 수행되었다. 95주령의 산란계 560수를 7처리 4반복으로 반복당 20수씩 배치하여 12주간 사육하면서 동충하초, 인진쑥, 울금 가공 부산물을 각각 1.5%, 3.0% 를 사료에 혼합 급여하였다. 산란율은 전 기간에 걸쳐 처리 구간에 유의적인 차이를 보이지 않았으며, 사료 섭취량과 평균 난중도 영향을 받지 않았다. 난각 두께, 난각 강도, 난각 색, 난황색에 있어서도 역시 처리구간 유의차가 나타나지 않았으나, Haugh unit 에 있어서는 대조구에 비해 모든 약용식 물 처리구에서 유의적으로 높았다(p<0.05). 실험 결과를 종합할 때, 동충하초, 인진쑥, 울금 가공 부산물을 산란계 사료 내 $1.5{\sim}3.0%$ 첨가 급여시 사료 섭취량, 산란율에는 영향을 미치지 않았으며, Haugh unit 에 있어서 유의적으로 향상되는 결과를 나타내어 고품질 계란 생산 및 약용식물 가공 부산물의 사료 자원화 가능성을 보여주었다.

인삼포장에 발생하는 당근뿌리혹선층의 방제를 위한 길항식물의 탐색 (Screening and Utilization of Antagonistic Plants to Control Northern root-knot Nematode in Ginseng Fields)

  • This study was conducted to screen the antagonistic plants on northern root-knot nematode (Meloidogyne) and to utilize those in its control. Egg hatching of M. hapla was found to be inhibited by 17 plant methanol extracts, and 11 plant extracts among them were also toxic to M. hapla second stage juvenile. Egg hatching of iW. hapla was also found to be inhibited by squeezed extracts of Cassia tora and Zea mays, and they were also toxic to M. hapla second stage juvenile. Extracts of Achyranthes japonica, Melia axedrach and Acorus graminens were toxic to M. hapla second stage with a juvenile mortality above 70clc at the 10 folds diluted concent ration and A. graminens was toxic to tested juvenile mortality above 50% at the 100 folds diluted concentration. The toxicity was directly propotional to the diluted concentration of the plant extracts and to the exposure period. Punica granatum, Acorns graminens and Melia axedrach were effective in inhibiting root penetration of JW. hapla juveniles, among of them p. granatum is most effective Percent inhibition of penetration by second and third stage juveniles into tomato slants penetrating by it was 72.7 and 82.4%, respectively.

Effects of Soil Textures on Infectivity of Root-Knot Nematodes on Carrot

  • This study was conducted to examine infectivity (penetration and gall and egg-mass formations) of the root-knot nematodes, Meloidogyne incognita and M. hapla, on carrots grown in soil conditions of 5 different soil textures consisting of bed-soil (b) and sand (s) mixtures (b-s mixtures) at the ratios of 10:0, 7:3, 5:5, 3:7, and 0:10. For M. incognita, the nematode penetration rates in b-s of 0:10 (100% sand) were significantly higher than in the other b-s mixtures, more greatly at 2 and 5 days after inoculation than at 10 DAI, while no significant differences in the penetration rates were mostly shown for M. hapla at the above DAI. However, for both nematodes, gall and egg-mass formations were remarkably increased in the b-s mixture of 0:10, compared to the other b-s mixtures, which is coincided with the general aspects of severe nematode infestations in sandy soils. This suggests the increased gall and egg-mass formations of M. incognita should be derived from the increased penetration rates in the sandy soil conditions, which provide a sufficient aeration due to coarse soil nature for the nematodes, leading to their mobility increased for the enhanced root penetration. For M. hapla, it is suggested that the sandy soil conditions affect positively on the healthy plant growth with little accumulation of the inhibitory materials and sufficient aeration, enhancing the nematode growth and feeding activities. All of these aspects provide information reliable for the development screening techniques efficient for the evaluation of the nematode resistance in the breeding programs.

Identification of 2-methylbutyric Acid as a Nematicidal Metabolite, and Biocontrol and Biofertilization Potentials of Bacillus pumilus L1

  • The present study described the isolation of 2-methylbutyric acid (2-MBA) produced from Bacillus pumilus L1, to subsequently investigate its nematicidal activity for the control of the root-knot nematode. The results showed that 2-MBA could be purified by chromatographic techniques and was identified using nuclear magnetic resonance and liquid chromatography-mass spectrometry. Crude extract and partially purified compounds had a significant effect on the inhibition of egg hatchability and second-stage juvenile (J2) mortality. A dose-dependent effect of 2-MBA was observed for J2 mortality and egg hatchability. Egg hatchability was 69.2%, 59.9%, 32.7%, and 0.0% at 125, 250, 500, and $1000{\mu}g\;mL^{-1}$ of 2-MBA after 4 d of incubation, respectively. Meanwhile, J2 mortality was in the range of 24.4%-100.0% after 2 d of incubation, depending on the concentrations of 2-MBA used. A pot experiment also demonstrated that treatment of B. pumilus L1 culture caused a significant reduction in the number of galls, egg masses, and J2 population than that of the tap water (TW) control. However, as the B. pumilus L1 culture concentration was decreased, the efficacy of nematode control by treatment of B. pumilus L1 culture was reduced compared to that of TW. B. pumilus L1 inoculation at different concentrations also promoted cucumber plant growth. Therefore, our study demonstrated the potential of 2-MBA from B. pumilus L1 as a biocontrol agent against the root-knot nematode and a plant growth promoter for cucumber plants.

EFFECT OF DIETARY LIPIDS ON LIVER, SERUM AND EGG YOLK CHOLESTEROL CONTENTS OF LAYING HENS

  • The effect of dietary lipid factors (plant and animal oil, cholesterol and ${\beta}$-sitosterol) on the liver, serum, and egg yolk cholesterol levels of the laying hen was studied. Single Comb White Leghorn laying hens, at 28 weeks of age, were fed two basal diets containing 8.0% soybean oil or 8.0% fish oil, with or without supplemental cholesterol (1.0%), ${\beta}$-sitosterol (2.0%) or combinations of both. Restricting caloric intake resulted in significantly (p<.05) decreased egg production and the total amount of cholesterol excreted via the egg was significantly (p<.05) different among treatment groups. Cholesterol supplementation to the two basal diets resulted in a significant elevation of liver, serum and egg yolk cholesterol levels. The addition of ${\beta}$-sitosterol lowered the cholesterol levels in liver and serum, while increased in the egg yolk (SO + ST, FO + ST). The anticholesterogenic effect of dietary ${\beta}$-sitosterol was not clearly exhibited in this study.
