To identify conserved structural or functional subunit(s) in the CP1 (editing) domains of class Ia tRNA synthetases, five available structures were compared and analyzed. Through sequence alignments of the CP1 domains, three conserved regions were found near the amino acid binding site in the editing domain. Structural overlapping of the three subunits clearly showed that there exist three common structural subunits in all of the five editing RS structures. The new alignment suggests a translocation movement of the CP1 domain caused by the binding with tRNA. Based on the experimental and modeling results, it is proposed that subunits 1 and 3 accommodate the incoming amino acid binding, while subunit 2 contributes to the interactions with the adenosine ring of the A76 to stabilize the overall tRNA binding.. Since these subunits are critical for the editing reaction, we expect that these key structures should be conserved through all class Ia editing RSs.

In this paper, several methods which guarantee the seamless editing when the splicing is applied to the MPEG-2 system streams are described. In the MPEG-2 system standard, it is recommended that the splicing technique is applied in the Transport Stream(TS) domain. However, in view of the video editing, the splicing at an arbitrary picture unit can not be guaranteed. The splicing in the PES domain is only considered in this paper because the PES is one of the MPEG-2 system streams and this result could be directly extended to the video splicing techniques in the TS or PS domain. Thus. the problems that might occur when different two PES streams are spliced and its effects to the video quality are compared and analyzed. Based on this analysis. several methods that can resolve these problems directly in the PES domain are proposed. Consequently, the computer simulation demonstrates that the splicing at a GOP modulo is guaranteed to have good video quality even without considering the state of the PES packet only if the PES_packet_length field of the PES packet header is used.
Wide-line 1H NMR intensity and differential scanning calorimetry measurements were carried out on the intrinsically disordered 73-residue full transactivation domain (TAD) of the p53 tumor suppressor protein and two peptides: one a wild type p53 TAD peptide with a helix pre-structuring property, and a mutant peptide with a disabled helix-forming propensity. Measurements were carried out in order to characterize their water and ion binding characteristics. By quantifying the number of hydrate water molecules, we provide a microscopic description for the interactions of water with a wild-type p53 TAD and two p53 TAD peptides. The results provide direct evidence that intrinsically disordered proteins (IDPs) and a less structured peptide not only have a higher hydration capacity than globular proteins, but are also able to bind a larger amount of charged solute ions.
As the full-motion videos in MPEG are widely available nowadays, an editor that could easily edit such kind of media data is required to develop various multimedia applications. In order to concatenate and apply a transition effect to two video streams encoded in MPEG, they should be decoded first since there are dependencies in the frames in MPEG-encoded video stream. Since this decode-edit-encode process requires a huge amount of computing/storage resources, a new editing scheme that could apply various transition effects to MPEG video streams directly while keeping the quality of video data is strongly required. This paper proposes a new editing scheme that could apply three transition effects (such as fade-in, fade-out, and dissolve) to MPEG video streams in a compressed domain. In the proposed scheme, an extension of previous method in which the frames are partially decompressed and transition effects are applied is adopted for I- and P-frames. In addition, a new processing scheme for B-frame that could apply the transition effects in DCT domain directly using an approximation of motion compensation based on the motion vector to reference frames. Since this processing scheme could apply the transition effects in a compressed domain directly, the editing process could be speed-up about $3{\sim}4$ times faster than previous decode-edit-encoding method while keeping the quality of video data as good as the source data. The proposed scheme could be used to build a software-only MPEG video editing system that helps to edit MPEG video data even on a low-cost desk-top computer.
Each year, significant amounts of Stereoscopic 3D(S3D) contents have been introduced. However, viewers who enjoy the contents readily experience a sense of fatigue on account of various factors. Consequently, many improvement studies have been conducted with the domain of disparity by, for example, simply controlling the disparity or optimizing the reaction speed of viewers' eyes to vergence. However, such studies are limited to the disparity domain and therefore are restricted to a limited number of applications. In this study, we attempted to transcend this limitation and analyzed how a reconstruction in color and brightness, as well as disparity and other important features, affects eyes in terms of vergence adaptation. As a result, we found that, the higher the color similarity is, the better it positively affects vergence adaptation during viewing. Based on this analysis, we propose in this paper a similar color extraction method between takes that are applicable to real-life situations. In an evaluation, the algorithm was applied to publicly available S3D contents and produced a converted color optimized image. The vergence adaptation time of this applied contents was significantly decreased. Also it was minimized through color reconstruction, thereby, being resulted in enhancing viewer concentration.
In this paper, We have studied for guaranteeing the clear display of MPEG-2 video sequence when conduct splicing of MPEG-2 system streams. we focus on the PES domain splicing considering video sequence. And we wish to make a base on the TS or PS domain splicing considering video sequence. For that, first, we compared and analyzed problems that is raised when different two PES streams are spliced and effects that affect the video sequence. And based on this analysis, we have searched for methods that resolve the cause of problems that can be happened in the display of video sequence directly in PES domain.
DNA methylation is a regulatory mechanism in epigenetics that is frequently altered during human carcinogenesis. To detect critical methylation events associated with gastric cancer (GC), we compared three DNA methylomes from gastric mucosa (GM), intestinal metaplasia (IM), and gastric tumor (GT) cells that were microscopically dissected from an intestinal-type early gastric cancer (EGC) using methylated DNA binding domain sequencing (MBD-seq) and reduced representation bisulfite sequencing (RRBS) analysis. In this study, we focused on differentially methylated promoters (DMPs) that could be directly associated with gene expression. We detected 2,761 and 677 DMPs between the GT and GM by MBD-seq and RRBS, respectively, and for a total of 3,035 DMPs. Then, 514 (17%) of all DMPs were detected in the IM genome, which is a precancer of GC, supporting that some DMPs might represent an early event in gastric carcinogenesis. A pathway analysis of all DMPs demonstrated that 59 G protein-coupled receptor (GPCR) genes linked to the hypermethylated DMPs were significantly enriched in a neuroactive ligand-receptor interaction pathway. Furthermore, among the 59 GPCRs, six GI hormone receptor genes (NPY1R, PPYR1, PTGDR, PTGER2, PTGER3, and SSTR2) that play an inhibitory role in the secretion of gastrin or gastric acid were selected and validated as potential biomarkers for the diagnosis or prognosis of GC patients in two cohorts. These data suggest that the loss of function of gastrointestinal (GI) hormone receptors by promoter methylation may lead to gastric carcinogenesis because gastrin and gastric acid have been known to play a role in cell differentiation and carcinogenesis in the GI tract.
As MPEG-2 is introduced, a new editing system that works at the compressed domain is required. It is efficient to paste the two different video sequences in the compressed domain, but after repeated pasting, the result sequence can violate MPEG-2 rate control algorithm. In other words, it causes the underflow of the buffer during the decoding time. The earlier study inserted an additional fade-in sequence but it is not natural when we make a program. In this paper, a nobel method using requantization of the underflow sequence is presented.
This paper is concerned with the program of the automatic mesh generation for 2-dimensional domain which contains the curved boundaries and holes. This program treats a new vertical - line drawing method. This method starts with 4-subdivisions of problem domain and the classification of the cross points of grid lines and boundaries. And the new node is generated by the vertical line to the line connecting the two intersections of a boundary and two grid lines in general. The boundaries are piecewise-curves composed of lines, circles, arcs, and free curves. The free curves are generated by B-Spline form. Although there were some bad elements for the complex boundary, it was possible to obtain the acceptable elements for the given boundaries. The results of automatic mesh generation can be verified directly by drawing on the computer monitor in executing the program. And it is possible to add the processes - that is, editing, hard copying, etc - using the script file in Auto-CAD.
