Estrogen Receptor-α Mediates the Effects of Estradiol on Telomerase Activity in Human Mesenchymal Stem Cells

  • Sex steroid hormone receptors play a central role in modulating telomerase activity, especially in cancer cells. However, information on the regulation of steroid hormone receptors and their distinct functions on telomerase activity within the mesenchymal stem cell are largely unavailable due to low telomerase activity in the cell. In this study, the effects of estrogen ($E_2$) treatment and function of estrogen receptor alpha ($ER{\alpha}$) and estrogen receptor beta ($ER{\beta}$) on telomerase activity were investigated in human mesenchymal stem cells (hMSCs). Telomerase activity and mRNA expression of the catalytic subunit of telomerase (hTERT) were upregulated by treatment of the cells with $E_2$. The protein concentration of $ER{\alpha}$ was also increased by $E_2$ treatment, and enhancement of $ER{\alpha}$ accumulation in the nucleus was clearly detected with immunocytochemistry. When $ER{\alpha}$ expression was reduced by siRNA transfection into hMSCs, the effect of $E_2$ on the induction of hTERT expression and telomerase activity was diminished. In contrast, the transient overexpression of $ER{\alpha}$ increased the effect of $E_2$ on the expression of hTERT mRNA. These findings indicate that the activation of hTERT expression and telomerase activity by $E_2$ in hMSCs depends on $ER{\alpha}$, but not on $ER{\beta}$.

Dehydration effects on spectroscopic properties of $Er^{+3}$ doped phosphate laser Glass ($Er^{+3}$ 첨가된 인산염 레이저 유리의 탈 수산기 영향에 관한 분광학적 특성)

  • Phosphate glass samples with various $Cr_2O_3$ and $Er_2O_3$ contents based upon $55P_2O_5\cdot24BaO\cdot10K_2O\cdot4Al_2O_3\cdot6Yb_2O_3$ were prepared. The prepared glass compositions are dehydrated using gas bubble flow method in open system and investigated the effects of the eliminating of OH groups from the glass melts with bubbling time. It was found that the probability of $Er^{+3}$ fluorescence quenching by OH groups oscillations linear depends upon the OH groups absorption coefficients in the maximum of the stretch vibrations band at $3500cm^{-1}$ while $Er^{+3}$ concentration range is between $1.6\times10^{19}$ and $21.2\times10^{19}$ ion/$cm^3$.

  • Electro-Rheological(ER) fluid are suspensions which show an abrupt increase in rheological properties under electric fields. ER effects arise from electrostatic forces between the starch particles dispersed in the electrically insulating silicone oil, induced when an electric field is applied. Yield stress of the fluids were measured on the couette cell type rheometer as a function of electric fields. This paper presents performance analyses of four types of the two parallel-plate. Which have different electrode length and width but same electrode area. On the basis of the pressure drop and flow rate analysis. Four types of the two parallel-plate are designed and manufactured. Using ER fluid, it is possible to directly interface between electric signals and fluid power without moving parts.

  • This paper presents some inherent characteristics of a semi-active variable damper featuring electro-rheological (ER) fluid. The damping force of the damper can be selectively adjusted or controlled by employing electric field to the ER fluid domain. This is possible owing to the pressure drop across the piston occured by field-dependent variable yield stress of the ER fluid. This is fundamentally different than the performance of a conventional adjustable viscous damper. To demonstrate the effectiveness and superiority over the conventional one, the proposed damper is incorporated with a suspension system. A quarter car model with the suspension system is formulated and represented by a state equation. By choosing numerical values based on realistic package size, power requirements and suitable ER properties, the performance characteristics of the suspension system are obtained and evaluated in both frequency and time domains. The effects of constant electric field and on-off controlled electric field which relates to the damping force are also examined.

  • This paper addresses the lubrication analysis of a short squeeze film damper operating with electro-rheological (ER) fluids which have large and reversible changes in yield shear stresses with respect to an applied electric field. The ER fluids are assumed to be modeled as Bingham fluids. The governing lubrication equation for the ER short squeeze film damper is developed on the basis of a Bingham fluid model, and the equation is subsequently solved in order to investigate the effects of the ER fluids on the damping capability of the damper. It is shown that a substantial increase in damping (both direct and cross coupled) is accomplished by increasing the yield shear stress of the ER fluids. This significant improvement of the damping capability suggests that the ER short squeeze film damper could be very effective for reducing the vibration and controlling the critical speeds of a rotor system.

  • Steady flow of an ER (electro-rheological) fluid in a two-dimensional electrode channel is studied by using FEM. Hydrodynamic interactions between the particles and the fluid are calculated by solving the Navier-Stokes equation combined with the equation of motion for each particle, where the multi-body electrostatic interaction is described by using point-dipole model. Motion of the particles in the ER fluid is elucidated in conjunction with the mechanisms of the flow resistance and the increase of viscosity. The ER effects have been studied by varying the Mason number and volume fraction of particles. These parameters have an influence on the formation of the chains resulting in the changes of the fluid velocity and the effective viscosity of ER fluids.

Annealing effects of ZnO:Er films on UV emission (ZnO:Er막의 UV 발광에 미치는 열처리 효과)

  • Er-doped ZnO(ZnO:Er) films were deposited onto MgO wafers by ultrasonic spray pyrolysis at 550 $^{\circ}C$ varying the concentration of Er in the deposition source from 0.5 wt% to 3.0 wt%. Annealing of the films in a vacuum was carried out to increase the intensity of ultraviolet(UV) emission from the films. The annealing temperature was between 600$^{\circ}C$ and 800$^{\circ}C$. The crystallographic properties and surface morphology of the films were investigated by X-ray diffraction(XRD)and scanning electron microscope(SEM), respectively. The properties of photoluminescence(PL) for the films were investigated by the dependence of PL spectra on the annealing temperature. X-ray photoelectron spectroscopy(XPS) was conducted to find the composition change in the films by the annealing.

Molecular Cloning and Characterization of the Estrogen Receptor from the Slender Bitterling (Acheilognathus yamatsutae)

  • Objectives: In order to identify the possibility of slender bitterling (SB) (Acheilognathus yamatsutae) being used as a test species for estrogenic endocrine disrupting chemicals (EEDCs), we carried out the cloning and sequence characterization of the estrogen receptor (ER). Methods: The ER from a slender bitterling was obtained by reverse transcriptase-polymerase chain reaction (RT-PCR), 5'- and 3'-rapid amplification of cDNA ends (5'-RACE and 3'-RACE) and T-vector cloning. The expression of ER mRNA was also analyzed in six tissues (brain, liver, kidney, gill, gonad, and intestines) by real-time PCR. Results: We obtained an ER from the slender bitterling. The SB ER cDNA was 2189 base pairs (bp) in length and contained a 1707 bp open reading frame that encoded 568 amino acid residues. The SB ER amino acid sequence clustered in a monophyletic group with the $ER{\alpha}$ of other fish, and was more closely related to zebrafish $ER{\alpha}$(88% identity) than to the $ER{\alpha}$ of other fish. The SB ER cDNA was divided into A/B, C, D, E and F domains. The SB ER has conserved important sequences for ER functions, such as the DNA binding domain (D domain), which are consistent with those of other teleosts. Conclusions: The ER of the slender bitterling could provide basic information in toxicological studies of EEDCs in the slender bitterling.

  • The Egr-1 is an immediate early response gene encoding a transcription factor that functions in the regulation of cell growth, differentiation, and apoptosis. Estrogen has diverse physiological effects, including cellular proliferation and neuroprotection against brain injury. There are two types of estrogen receptors (ERs), $ER{\alpha}$ and $ER{\beta}$. $ER{\alpha}$-induced Egr-1 expression has been extensively studied; however, the role of $ER{\beta}$ is yet not known. In the present study, we investigated whether or not $ER{\beta}$ induces Egr-1 expression in C6 rat glioma cells, which express $ER{\beta}$ but not $ER{\alpha}$. Our results show that $ER{\beta}$ promoted up-regulation of Egr-1 expression via a non-genomic mechanism involving the Raf/MEK1/Erk/Elk-1 signaling cascade.

A Fundamental Study on Bingham Characteristics of Electro-Rheological Fluids for Control System Application (제어 시스템 적용을 위한 ER유체의 빙햄 특성에 관한 기초적 연구)

  • This paper describes the properties of temperature-viscosity characteristics of hydrous and anhydrous electro-rheological fluids containing starch and titanium particle in silicone oil ER effects arise from electrostatic forces between the starch particles and titanium particles dispersed to the electrically insulating silicone oil induced when electric field is applied ER fluids under electric field control have been found to provide resonable estimates of ER fluid viscosity variation characteristics. Yield shear stress of the ER fluids were measured the couette cell type rheometer as a function of electrlc fields. The outer cup is connected to positive electrode(+) and bob becomes ground(-). The electrie field is applied by high voltage DC power supply. In this experiment shear rates were increased from 0 to 200 $s^{-1}$ in 2 minutes. The ER fluid's viscosity change is very small and stable at the temperature range of $40^{\circ}C$ to $60^{\circ}C$. Therefore, applications of a new ER fluid to control systems application are suitable.
