  • Though a micro pump is a crucial element in miniaturized bio-fluidic systems or drug delivery systems, most of the conventional micro pumps still have some limitations to miniaturize their controller system and to obtain the sufficient back pressure which can rise over the inner pressure of human body or experimental animals. In this paper, to overcome these limitation, a new PZT disc and its controller were designed and fabricated to get the sufficient flowrate and the back pressure with guaranteeing embeddability of the controller into pumping body. The amplitudes of the disc deflections were as large as 40 ${\mu}m$ at 200 V - 100 Hz condition. As results of experiments, the flow rate and the back pressure increase when the frequency increases. The obtainable maximum flow rate and back pressure are 5.2 ml/min at 95 Hz and 13.14 kPa at 90 Hz respectively.

  • Wound healing is a complex and dynamic process, making the accurate and timely assessment of skin wounds a crucial aspect of effective wound care management, especially for chronic wounds. Unlike conventional wound dressings that simply cover the wound area once some form of medicine is administered onto the wound, recent studies have introduced versatile approaches to smart wound dressings capable of interacting with wound fluids to monitor physicochemical and pathological parameters to determine the wound healing status. Such electrochemical wound dressings can be integrated with on-demand, closed-loop drug delivery or stimulation systems and ultimately expanded into an ideal technological platform for the prevention, treatment, and management of skin wounds or illnesses. This article briefly reviews the wound healing mechanism and recent strategies for effective wound care management. Specifically, this review discusses the following aspects of smart wound dressings: sensor-integrated smart bandages to detect wound biomarkers, smart bandages developed to accelerate wound healing, and wireless, closed-loop automatic (on-demand) wound healing systems. This review concludes by providing future perspectives on effective wound care management.

Clinical and microbiological effects of adjunctive local delivery of minocycline (Periocline®) in patients receiving supportive periodontal therapy: a pilot study

  • Purpose: This study aimed to evaluate the clinical and microbiological efficacy of adjunctive local delivery of minocycline (Periocline®) in patients receiving supportive periodontal therapy (SPT) after initial treatment. Methods: The participants were 16 men and 8 women (age, 20-65 years) who had at least 15 natural teeth, underwent SPT for more than 1 year due to chronic periodontitis, had 4 or more periodontal pocket sites deeper than 5 mm, and showed >25% gingival bleeding on probing (BoP). They were randomly assigned to the test and control groups. In the test group, mechanical debridement and local antibiotic delivery were performed for all periodontal sulci/pockets; in the control group, mechanical debridement and saline irrigation were performed. In patients who underwent SPT for more than 1 year, clinical and microbiological examinations were performed at baseline and 1 and 3 months after SPT. The clinical examination included an assessment of the periodontal pocket depth, clinical attachment level, plaque index, and BoP. Microbial tests were performed using real-time polymerase chain reaction; the relative ratios of Porphyromonas gingivalis and Fusobacterium nucleatum were determined. Results: Both groups showed significant improvements in clinical parameters at 1 and 3 months from baseline; there were no significant changes between months 1 and 3. Intergroup differences were insignificant. The microbiological analysis revealed no significant differences in P. gingivalis and F. nucleatum ratios across time points. While intergroup differences were insignificant, there was a tendency for the P. gingivalis and F. nucleatum ratios to decrease in the test group. Conclusions: Mechanical debridement in patients receiving maintenance therapy resulted in clinically significant improvement; the effectiveness of additional local delivery of antibiotics was not significant. The ratios of P. gingivalis and F. nucleatum showed a tendency to decrease in the test group, although it was not significant.

생체 적합한 나노입자와 계면화학 (Surface Chemistry in Biocompatible Nanocolloidal Particles)

  • 콜로이드와 계면화학은 표면적과 표면에너지의 학문이다. 계면상의 위치에 따라서 분자밀도, 분자간의 상호작용력, 분자 배향성 그리고 반응성이 달라진다는 것은 흥미있는 주제가 되고 있다. 이러한 계면에너지가 중요하게 작용하는 시스템으로서 회합체, 에멀젼, 입자분산, 거품, 2차원적 표면이나 필름을 들 수 있다. 특히 나노 입자에 관련된 생체 적합성 재료를 사용하여 약물 전달체와 화장품 나노 소재로 이용하는데 관심이 고조되고 있다. 나노 입자는 수 nm에서 수백 nm 크기를 갖는, 넓은 표면적을 가진 콜로이드 상의 불균일 분산 입자의 일종이다. 지금까지 나노 입자의 제조, 특성 규명, 나노입자를 이용한 약물 봉입에 관한 연구가 활발히 이루어져 약물 전달체로서의 가능성이 충분히 입증되었다. 또한 난용성분 가용화 나노소재, 피부 흡수 증진용 나노소재, 자외선 차단용 나노소재, 안정화용 나노소재, 서방형 나노소재 등의 화장품 연구에 생체적합 나노전달체를 이용한 예가 보고되었다. 나노/마이크로 입자 시스템은 제조방법과 형태에 따라 나노/마이크로 스피어, 나노/마이크로 캡슐, 나노/마이크로 에멀젼, 폴리머 마이셀, 리포좀 등으로 구분된다. 수용액상에서 자기 회합체를 구성하는 나노수준의 폴리머 마이셀입자, 고농도, 고활성 물질에 대하여 농도 및 활성을 일정하게 제어할 수 있는 나노/마이크로 캡슐, 단일 이중층 또는 다층(100~800 nm)을 형성하여 여러 생리 환성 물질의 전달체로 이용되는 리포솜(liposome)에 대하여 제조방법과 산업의 응용에 대해 소개하였다.

시간 분해 직렬 펨토초 결정학을 위한 3차원 프린팅 기반의 초고속 믹싱 및 인젝팅 시스템 (3D Printing-Based Ultrafast Mixing and Injecting Systems for Time-Resolved Serial Femtosecond Crystallography)

  • 매우 짧은 펄스 폭의 X선 자유전자 레이저(XFEL)를 이용한 시간 분해능 연속 펨토초 결정학(time-resolved serial femtosecond crystallography, TR-SFX)기법에서 반응 물질과 생체분자 결정 샘플간의 혼합률(mixing rate)과 결정 샘플과 X선 레이저 간의 충돌률(hit rate)은 생체분자의 시분해 구조 변화에 대한 정확한 이미지 획득 및 효율적인 샘플소비와 같은 TR-SFX의 분석 성능을 결정짓는 핵심인자이다. 본 연구에서는 극초단 내 일어나는 생체분자의 시분해 구조 변화 해석을 위해 초고속 믹싱 기능을 가짐과 동시에 공압 기반의 주문형 액적 젯팅이 가능한 두 가지 다른 방식의 샘플 전달시스템을 고안하였다. 한 방식은 이중 노즐을 통해 토출된 액적의 고속 충돌에 유발된 관성 믹싱을 기반으로 하고 있으며, 다른 방식은 마이크로믹서가 내장된 공압 젯팅을 기반으로 하고 있다. 먼저, 이중 노즐을 통해 토출된 액적의 충돌에 대한 동적 거동 및 액적 내부 관성 유동에 대한 믹싱에 대한 실험 및 수치해석적 연구를 수행하였다. 다음으로 마이크로믹서가 내장된 공압 젯팅 시스템의 성능을 유사한 방법을 통해 평가하였다. 본 연구에서 개발한 샘플 전달시스템은 질환을 유발하는 특정 단백질들의 기작을 규명하거나, 항체 의약품과 신약 후보 물질 탐색하는 데 있어 필수적인 3차원 생체 분자 구조분석 연구에 매우 유용하게 활용될 수 있을 것이다.

The Effect of Enhancers on the Penetration of Albuterol through Hairless Mouse Skin

  • Albuterol, a selective ${\beta}_2$-adrenergic receptor stimulant, has been introduced as a potent bronchodilator for patients with bronchial asthma, chronic obstructive bronchial disease, chronic bronchitis and pulmonary emphysema. The percutaneous permeation of albuterol sulfate was investigated in hairless mouse skin in vitro with and without pretreatment with enhancers. The enhancing effects of ethanol and various penetration enhancers such as terpenes, non-ionic surfactants, pyrrolidones, and fatty acids on the permeation of albuterol sulfate were evaluated using Franz diffusion cells. Among terpenes studied, 1,8-cineole was the most effective enhancer, which increased the permeability of albuterol sulfate approximately 33-fold compared with the control without enhancer pretrement, followed by d-limonene with enhancement ratio of 21.79. 2-Pyrrolidone-5-carboxylic acid increased the permeability of albuterol sulfate approximately 5.5-fold compared with the control. Other pyrrolidones tested showed only slight permeability enhancing effect with enhancement ratio less than 2.8. Nonionic surfactants showed moderate enhancing effects. Lauric acid increased the permeability of albuterol sulfate approximately 30-fold with decreasing the lag time from 2.85 to 0.64 hr. Oleic acid and linoleic acid showed enhancement ratio of 24.55 and 22.91, respectively. These findings would allow a more rational approach for designing formulations for the transdermal delivery of albuterol sulfate and similar drugs.

글루코스를 함유한 음이온 하이드로젤의 pH 감응성 동적 팽윤거동 (pH-Sensitive Dynamic Swelling Behavior of Glucose-containing Anionic Hydrogels)

  • 음이온 하이드로젤은 그들이 가지고 있는 pH 감응성 팽윤거동 때문에 단백질 약물의 경구투여용 전달물질로써 많은 주목을 받고 있다. 본 연구에서는 음이온 하이드로젤의 pH 변화에 따른 용매의 침투 메커니즘을 규명하기 위하여 methacrylic acid와 2-methacryloxyehtyl glucoside를 공중합하여 P(MAA-co-MEG) 하이드로젤을 합성한 후 pH 변화에 따른 하이드로젤의 동적 팽윤거동을 관찰하였다. 용매의 침투 메커니즘이 Fickian 또는 non-Fickian 인지를 설명할 수 있는 특성지수 n을 $M_t/M_{\infty}=kt^n$ 관계식으로부터 계산하였다. 하이드로젤에 대한 용매의 침투 메커니즘은 주위 pH의 영향을 많이 받았으며, 젤의 $pK_a$ 보다 높은 pH인 7.0에서는 침투 메커니즘이 상대적으로 고분자사슬의 이완에 의한 지배를 많이 받는다는 것을 알 수 있었다. 한편, pH 7.0에서 고분자 이완에 의한 용매의 침투 메커니즘은 하이드로젤에 존재하는 카르복실산의 이온화에 기인한 것임을 ATR-FTIR 분광분석을 이용하여 확인하였다.

Glutamate Receptor Abnormalities in Schizophrenia: Implications for Innovative Treatments

  • Schizophrenia is a devastating psychiatric illness that afflicts 1% of the population worldwide, resulting in substantial impact to patients, their families, and health care delivery systems. For many years, schizophrenia has been felt to be associated with dysregulated dopaminergic neurotransmission as a key feature of the pathophysiology of the illness. Although numerous studies point to dopaminergic abnormalities in schizophrenia, dopamine dysfunction cannot completely account for all of the symptoms seen in schizophrenia, and dopamine-based treatments are often inadequate and can be associated with serious side effects. More recently, converging lines of evidence have suggested that there are abnormalities of glutamate transmission in schizophrenia. Glutamatergic neurotransmission involves numerous molecules that facilitate glutamate release, receptor activation, glutamate reuptake, and other synaptic activities. Evidence for glutamatergic abnormalities in schizophrenia primarily has implicated the NMDA and AMPA subtypes of the glutamate receptor. The expression of these receptors and other molecules associated with glutamate neurotransmission has been systematically studied in the brain in schizophrenia. These studies have generally revealed region- and molecule-specifi c changes in glutamate receptor transcript and protein expression in this illness. Given that glutamatergic neurotransmission has been implicated in the pathophysiology of schizophrenia, recent drug development efforts have targeted the glutamate system. Much effort to date has focused on modulation of the NMDA receptor, although more recently other glutamate receptors and transporters have been the targets of drug development. These efforts have been promising thus far, and ongoing efforts to develop additional drugs that modulate glutamatergic neurotransmission are underway that may hold the potential for novel classes of more effective treatments for this serious psychiatric illness.

Development of Estimation Methods of Skin Oxidation and Evaluation of Anti-Oxidative Effects of Genistein in Topical Formulations

  • The objective of the present study was to establish the method of measurement of hydrogen peroxide and to estimate the anti-oxidative effect of genistein in the skin. UVB induced skin oxidation and anti-oxidative effect of genistein formulations were evaluated by determining levels of hydrogen peroxide. The mechanism involved in the determination of hydrogen peroxide is based on a color reaction between ferric ion ($Fe^{3+}$) and xylenol orange, often called FOX assay and subsequent monitoring of absorbance values of the reactant at 540 nm. The reaction was to some extent pH-dependent and detection sensitivity was greatest at pH 1.75. Genistein liposomal gel demonstrated better anti-oxidative effect with regard to lowering hydrogen peroxide levels elevated by UVB irradiation compared to genistein-suspended gel. A linear relationship has been observed between anti-oxidative effect of genistein and drug deposition in the skin tissue. Genistein liposomal gel resulting in the localization of the drug in the deeper skin led to improved anti-oxidative effect compared to genistein gel. The suggested method for evaluation of oxidation of the skin can be used as a tool to screen effective anti-oxidative agents and their delivery systems acting on the skin.

  • 나노의약품(Nanomedicine)은 나노기술이 접목된 신기능성 의약품을 의미하며 빠르게 발전하고 있는 신생 분야이다. 아직 상용화된 나노의약품은 많지 않지만 최근 나노기술이 접목된 진단 및 치료제제가 속속 개발되면서 기존 의약품을 대체하는 추세에 있다. 나노의약품 연구는 지난 10여년 동안 미국과 유럽을 중심으로 활발히 진행되었으며, 그 결과로 2000년대에 접어서면서 나노의약품 관련 특허 및 논문의 수가 급속히 증가하고 있다. 본 총설에서는 탄소 나노소재, 고분자 약물전달 시스템, 진단영상 소재 등과 같은 대표적인 나노의약품의 연구개발 동향을 소개하고 있다.
