Changes in Gut Microbial Community of Pig Feces in Response to Different Dietary Animal Protein Media

  • Beef, pork, chicken and milk are considered representative protein sources in the human diet. Since the digestion of protein is important, the role of intestinal microflora is also important. Despite this, the pure effects of meat and milk intake on the microbiome are yet to be fully elucidated. To evaluate the effect of beef, pork, chicken and milk on intestinal microflora, we observed changes in the microbiome in response to different types of dietary animal proteins in vitro. Feces were collected from five 6-week-old pigs. The suspensions were pooled and inoculated into four different media containing beef, pork, chicken, or skim milk powder in distilled water. Changes in microbial communities were analyzed using 16S rRNA sequencing. The feces alone had the highest microbial alpha diversity. Among the treatment groups, beef showed the highest microbial diversity, followed by pork, chicken, and milk. The three dominant phyla were Proteobacteria, Firmicutes, and Bacteroidetes in all the groups. The most abundant genera in beef, pork, and chicken were Rummeliibacillus, Clostridium, and Phascolarctobacterium, whereas milk was enriched with Streptococcus, Lactobacillus, and Enterococcus. Aerobic bacteria decreased while anaerobic and facultative anaerobic bacteria increased in protein-rich nutrients. Functional gene groups were found to be over-represented in protein-rich nutrients. Our results provide baseline information for understanding the roles of dietary animal proteins in reshaping the gut microbiome. Furthermore, growth-promotion by specific species/genus may be used as a cultivation tool for uncultured gut microorganisms.

Perchloroethylene과 Trichloroethylene의 혐기적 탈염소화 및 미생물 군집 분석 (Analysis of Microbial Community During the Anaerobic Dechlorination of Perchloroethylene and Trichloroethylene)

  • 울산, 여수 등 공단지역의 토양, 하천의 저니, 해양의 준설토 둥을 이용하여 난분해성 염소화합물인 PCE (perchloroethylene)및 TCE (trichloroethylene)의 혐기성 탈염소화에 관련하는 미생물을 탐색하고 이들의 탈염소화 효율을 조사하였다. 혐기성 상호대사에 의한 탈염소화 효율을 조사하기 위해 전자공여체로 acetate를 사용하여 혐기성 회분식 실험을 실시하였으며, 이와 병행하여 분자생물학적인 기법인 16S rDNA의 PCR-Double Gradient DGGE (DG-DGGE)를 이용하여 미생물의 군집을 분석하였다. 그 결과 울산 태화강 및 여수 하남천의 저니를 접종한 경우 PCE는 $70\%,\;65\%$, TCE는 $50\%,\;45\%$의 높은 탈염소화 효율을 나타내었다. 또한 16S rDNA의 PCR을 이용한 DG-DGGE로 미생물 군집을 분석한 결과, 탈염소화 효율이 높은 지역의 저니에는 Desulfovibrio sup.의 미생물이 주로 존재함을 확인하였다.

방향족 아미노산에 의한 김치 유산균 생장의 제어 (Control of Lactic Acid Bacterial Growth in Kimchi by Aromatic Amino Acids)

  • 아미노산이 김치유산균에 미치는 영향을 검토하였다. 발효온도 $15^{\circ}C$에서 형태가 다른 집락을 73개 분리하였다. 이 중에서 69.9%가 덱스트란 생성균주이었으며, Leuconostoc 속이 4.1%, Lactobacillus 속이 65.8%를 차지하였다. Tyrosine 500 ppm이 되도록 첨가한 배지에서 분리균주의 생장은 완전히 억제되었다. 그러나 실제로 이 아미노산을 첨가하여 만든 김치에서 분리된 총 집락수는 58개 이었고, 이 중에서 덱스트란 생성균주가 70.7%로 큰 변동이 없었으나 대신에 Leuconostoc 속이 41.4%로 증가하였고, Lactobacillus 속은 29.3%로 감소하였다. 각 속에서 우점종은 Leu. mesenteroides와 Lac. minor이었다. 따라서 실제 김치에서 tyrosine은 Lactobacillus 속의 생장을 억제하는 효과가 있었다.

농가 자가제조 액비의 발효과정 중 이화학성 및 미생물상 변화 (Changes in Chemical Properties and Microbial Population of Farm-Made Organic Liquid Fertilizer during Fermenting Process)

  • 본 실험에서는 유기재배 농가에서 사용되고 있는 액비의 특성을 과학적으로 구명하고자 발효기간 동안 액비의 화학성, 미생물상의 변화, 그리고 액비 처리가 무 생육에 미치는 효과를 조사하였다. 액비의 pH는 발효가 진행됨에 따라 7.2에서 4.3로 감소하였고 EC는 액비제조 직후 13.9 dS/m에서 계속 상승하여 70일에는 99.3 dS/m에 도달하였다. 액비내 서식하는 세균 밀도는 발효 과정 중 $8.2{\times}10^5$ cfu/ml에서 $3{\times}10^4$ cfu/ml로 감소하였으며, Bacillus 속은 제조 직후 $2.1{\times}10^2$ cfu/ml에서 $4.2{\times}10^3$ cfu/ml로 발효가 진행됨에 따라 증가하는 경향을 보였다. 액비로부터 분리한 세균은 지방산 분석을 통한 동정 결과, 발효가 진행될수록 Bacillus 유사속들이 액비내 우점하는 것을 알 수 있었으며 DGGE profile을 통해 발효과정 동안 액비내 미생물 군집의 변화를 확인하였다. 액비처리에 의한 무 유묘 생육을 살펴본 결과, 줄기보다는 뿌리 발달을 촉진하였다.

쌀겨와 밀기울의 토양 혐기발효 처리가 시설 재배지 토양의 미생물상에 미치는 영향 (Microbial Community Changes in the Soil of Plastic Film House as Affected by Anaerobic Fermentation of Rice Bran or Wheat Bran)

  • 본 연구는 밀기울과 쌀겨를 이용한 토양 혐기발효 처리가 토양 미생물상 변화에 미치는 영향을 구명하고자 수행하였다. 희석 평판법을 이용하여 토양 미생물상을 분석한 결과 쌀겨를 처리한 토양은 사상균이 크게 증가한 반면, 효모의 생장은 확인할 수 없었다. 반면 밀기울을 처리한 토양은 사상균의 생장이 크게 억제되었으며, 효모는 높은 밀도를 보였다. 인지질 지방산을 이용하여 토양 미생물상을 분석한 결과, 처리가 진행되는 20일 까지는 밀기울과 쌀겨를 처리구 모두 그람 음성균과 양성균의 변동이 미미하였으나, 처리가 종료되어 비닐을 제거한 이후에는 크게 상승하는 경향을 보였다. 각 처리별 미생물 군집구조를 분석한 결과, 밀기울과 쌀겨 처리는 미생물상에 큰 변화를 보였으며, 각각의 군집구조는 크게 달랐다.

Diversity and Antiaflatoxigenic Activities of Culturable Filamentous Fungi from Deep-Sea Sediments of the South Atlantic Ocean

  • Despite recent studies, relatively few are known about the diversity of fungal communities in the deep Atlantic Ocean. In this study, we investigated the diversity of fungal communities in 15 different deep-sea sediments from the South Atlantic Ocean with a culture-dependent approach followed by phylogenetic analysis of ITS sequences. A total of 29 fungal strains were isolated from the 15 deep-sea sediments. These strains belong to four fungal genera, including Aspergillus, Cladosporium, Penicillium, and Alternaria. Penicillium, accounting for 44.8% of the total fungal isolates, was a dominant genus. The antiaflatoxigenic activity of these deep-sea fungal isolates was studied. Surprisingly, most of the strains showed moderate to strong antiaflatoxigenic activity. Four isolates, belonging to species of Penicillium polonicum, Penicillium chrysogenum, Aspergillus versicolor, and Cladosporium cladosporioides, could completely inhibit not only the mycelial growth of Aspergillus parasiticus mutant strain NFRI-95, but also the aflatoxin production. To our knowledge, this is the first report to investigate the antiaflatoxigenic activity of culturable deep-sea fungi. Our results provide new insights into the community composition of fungi in the deep South Atlantic Ocean. The high proportion of strains that displayed antiaflatoxigenic activity demonstrates that deep-sea fungi from the Atlantic Ocean are valuable resources for mining bioactive compounds.

Short-Term Changes in Gut Microflora and Intestinal Epithelium in X-Ray Exposed Mice

  • Background: Gut microflora contributes to the nutritional metabolism of the host and to strengthen its immune system. However, if the intestinal barrier function of the living body is destroyed by radiation exposure, the intestinal bacteria harm the health of the host and cause sepsis. Therefore, this study aims to trace short-term radiation-induced changes in the mouse gut microflora-dominant bacterial genus, and analyze the degree of intestinal epithelial damage. Materials and Methods: Mice were irradiated with 0, 2, 4, 8 Gy X-rays, and the gut microflora and intestinal epithelial changes were analyzed 72 hours later. Five representative genera of Actinobacteria, Firmicutes, and Bacteroidetes were analyzed in fecal samples, and the intestine was pathologically analyzed by Hematoxylin-Eosin and Alcian blue staining. In addition, DNA fragmentation was evaluated by the TdT-mediated dUTP nick-end labeling (TUNEL) assay. Results and Discussion: The small intestine showed shortened villi and reduced number of goblet cells upon 8 Gy irradiation. The large intestine epithelium showed no significant morphological changes, but the number of goblet cells were reduced in a radiation dose-dependent manner. Moreover, the small intestinal epithelium of 8 Gy-irradiated mice showed significant DNA damaged, whereas the large intestine epithelium was damaged in a dose-dependent manner. Overall, the large intestine epithelium showed less recovery potential upon radiation exposure than the small intestinal epithelium. Analysis of the intestinal flora revealed fluctuations in lactic acid bacteria excretion after irradiation regardless of the morphological changes of intestinal epithelium. Altogether, it became clear that radiation exposure could cause an immediate change of their excretion. Conclusion: This study revealed changes in the intestinal epithelium and intestinal microbiota that may pave the way for the identification of novel biomarkers of radiation-induced gastrointestinal disorders and develop new therapeutic strategies to treat patients with acute radiation syndrome.

An investigation on fermentative profile, microbial numbers, bacterial community diversity and their predicted metabolic characteristics of Sudangrass (Sorghum sudanense Stapf.) silages

  • Objective: This study aimed to investigate the fermentation profiles, bacterial community and predicted metabolic characteristics of Sudangrass (Sorghum sudanense Stapf.) during ensiling. Methods: First-cutting Sudangrass was harvested at the vegetative stage and ensiled in laboratory-scale silos (1 L capacity). Triplicate silos were sampled after 1, 3, 7, 15, 30, and 60 days of ensiling, respectively. The bacterial communities on day 3 and 60 were assessed through high-throughput sequencing technology, and 16S rRNA-gene predicted functional profiles were analyzed according to the Kyoto encyclopedia of genes and genomes using Tax4Fun. Results: The Sudangrass silages showed good fermentation quality, indicated by higher lactic acid contents, and lower pH, butyric acid and ammonia nitrogen contents. The dominant genus Lactococcus on day 3 was replaced by Lactobacillus on day 60. The metabolism of amino acid, energy, cofactors and vitamins was restricted, and metabolism of nucleotide and carbohydrate was promoted after ensiling. The 1-phosphofructokinase and pyruvate kinase of bacterial community seemed to play important roles in stimulating the lactic acid fermentation, and the promotion of arginine deiminase could help lactic acid bacteria to tolerate the acidic environment. Conclusion: High-throughput sequencing technology combined with 16S rRNA gene-predicted functional analyses revealed the differences during the early and late stages of Sudangrass ensiling not only for distinct bacterial community but also for specific functional metabolites. The results could provide a comprehensive insight into bacterial community and metabolic characteristics to further improve the silage quality.

농업지역 지하수의 수리지화학 및 미생물 군집 구조 분석 (Hydrogeochemistry and Microbial Community Structure of Groundwater in an Agricultural Area)

  • This study evaluated the potential threat of agricultural and human activities to groundwater in the Noseong stream watershed, a typical agricultural area, through hydrogeochemical characteristics and microbial community analyses. The groundwater in the study area was Ca-SO4 and Ca-HCO3 types alluvial aquifer mainly used for agricultural and living purposes, and contained high levels of NO3- and Cl- ions generated from anthropogenic sources such as fertilizer, livestock wastewater, and domestic sewage. Proteobacteria was most abundant in all samples with an average of 46.1% while Actinobacteria, Bacteroidetes, and Cyanobacteria were dominant on an occasional basis. The prevalence of aerobic bacteria such as the genus Mycobacterium, Flavobacterium, and Sphingomonas suggests that groundwater was well connected with the surface layer. The potential pathogen Mycobacterium was detected in most samples, and other pathogenic bacteria were also widely distributed, indicating the vulnerability to contamination. Therefore, an integrated management system is required to secure the sustainable use of groundwater in agricultural areas with high groundwater dependence.

The impact of different diets and genders on fecal microbiota in Hanwoo cattle

  • Bovine fecal microbiota is important for host health and its composition can be affected by various factors, such as diet, age, species, breed, regions, and environments. The objective of this study was to evaluate the impact of diet and gender on fecal microbiota in Korean native Hanwoo cattle. The 16S rRNA gene amplicon sequencing of fecal microbiota was conducted from 44 Hanwoo cattle divided into four groups: (1) 11 heifers fed an oat hay plus total mixed ration (TMR) diet for breeding (HOTB), (2) 11 heifers fed an early fattening TMR diet (HEFT), (3) 11 steers fed the early fattening TMR diet (SEFT), and (4) 11 steers fed the late fattening TMR diet (SLFT). Firmicutes and Bacteroidota were the first and second most dominant phyla in all the samples, respectively. The Firmicutes/Bacteroidota (F/B) ratio associated with feed efficiency was significantly greater in the SLFT group than in the other groups. At the genus level, Romboutsia, Paeniclostridium, and Turicibacter were the most abundant in the SLFT while Akkermansia, Bacteroides, and Monoglobus were the most abundant in the HOTB group. Although the same early fattening TMR diet was fed to Hanwoo heifers and steers, Marvinbryantia and Coprococcus were the most abundant in the HEFT group while Alistipes and Ruminococcus were the most abundant in the SEFT group. Shannon and Simpson diversity indices were significantly lower in the SLFT group than in the other groups. Distribution of fecal microbiota and functional genetic profiles were significantly different among the four treatment groups. The present study demonstrates that different diets and genders can affect fecal microbiota and the F/B ratio may be associated with feed efficiency in Hanwoo cattle. Our results may help develop strategies to improve gut health and productivity through manipulation of fecal microbiota using the appropriate diet considering Hanwoo cattle gender.
