The antibacterial mechanism of enterobacter Salmonella typhimurium was studied. The rfa (Waa) gene cluster of S. typhimurium encodes the core oligosaccharide biosynthesis of lipopolysaccharide (LPS). Among the rfa gene cluster, we recently cloned the rfaE gene, which is involved in ADP-L-glycero-D-manno-heptose biosynthesis. The rfaE mutant synthesizes heptose-deficient LPS, which consists of only lipid A and 3-deoxy-D-manno-octulosonic acid (KDO), thus making an incomplete LPS and a rough phenotype mutant. S. typhimurium deep-rough mutants with the heptose region of the inner core show a reduced growth rate, sensitivity to high temperature, and hypersensitivity to hydrophobic antibiotics such as baicalin isolated from the medicinal herb of Scutellaria baicalensis Georgi. Thus, in this study, the cloned rfaE gene was added to the S. typhimurium rfaE mutant strain SL1102 (rfaE543), which makes heptose-deficient LPS and has a deep-rough phenotype. The complementation created a smooth phenotype in the SL1102 strain. The sensitivity of SL1102 to bacteriophages was also recovered to that of wild-type strain, indicating that LPS is used as the receptor for bacteriophage infection. The permeability barrier of SL1102 to hydrophobic antibiotics such as novobiocin and baicalin was restored to that of the wild-type, suggesting that antibiotic resistance of the wild-type strain is highly correlated with their LPS. Through an agar diffusion assay, the growth-inhibition activity of baicalin was fully observed in the mutant SL1102 strain. However, only a half of the inhibitory activity was detected in the rfaE complemented SL1102 strain. Furthermore, the LPS produced by the rfaE-complemented SL1102 strain was indistinguishable from LPS biosynthesis of smooth strains.

High temperature electrical conductivity of Aluminum Nitride (AlN) ceramics sintered with $Y_2O_3$ as a sintering aid has been investigated with respect to various sintering conditions and MgO-dopant. When magnesium oxide is added as a dopant, liquid glass-film and crystalline phases such as spinel, perovskite are formed as second phases, which affects their electrical properties. According to high temperature impedance analysis, MgO doping leads to reduction of activation energy and electrical resistivity due to AlN grains. On the other hand, the activation energy and electrical resistivity due to grain boundary were increased by MgO doping. This is a result of the formation of liquid glass film in the grain boundary, which contains Mg ions, or the elevation of schottky barrier due to the precipitation of Mg in the grain boundary. For the annealed sample of MgO doped AlN, the electrical resistivity and activation energy were increased further compared to MgO doped AlN, which results from diffusion of Mg in the grains from grain boundary as shown in the microstructure.

Corrosion is the degradation of metals by reaction with the environment. It is difficult to completely remove. Corrosion proceeds rapidly after the protective barrier is destroyed, and several reactions occur that alter the composition and properties of the metal surface and local environments, such as diffusion of metal cations into the matrix, the formation of oxides, and local pH changes. The study of corrosion of steel and iron is of theoretical and practical interest and is receiving considerable attention. Acid solutions, which are widely used in industrial pickling, acid descaling, cleaning and acidification of oil wells, require the use of corrosion inhibitors to suppress corrosion attacks on metallic materials. Physical removal of rust requires expensive special equipment, and chemical removal of it can cause corrosion or shorten the life of the metal. In this study, an eco-friendly rust cleaner was developed using cosmetics and food materials by applying the concept of perm reducing agent and chelate, and applied to remove rust from industrial and hot water pipes and various industrial devices. As a result, it was found that rust cleaners remove rust more effectively and safely compared to conventional treatment methods. At the same time, the rust removal efficiency was 1.75 to 2.5 times better for industrial piping and 1.56 to 2.2 times better for boiler hot water than conventional methods.
Bentonite is widely recognized and utilized as a buffer material in high-level radioactive waste repositories, mainly due to its favorable characteristics such as swelling capability and low permeability. Bentonite buffers play an important role in ensuring the safe disposal of radioactive waste by providing a low permeability barrier and effectively preventing the migration of radionuclides into the surrounding rock. However, the long-term performance of bentonite buffers still remains a subject of ongoing research, and one of the main concerns is the erosion of the buffer induced by swelling and groundwater flow. The erosion of the bentonite buffer can significantly impact repository safety by compromising the integrity of buffer and leading to the formation of colloids that may facilitate the transport of radionuclides through groundwater, consequently elevating the risk of radionuclide migration. Therefore, it is very important to numerically quantify the erosion of bentonite buffer to evaluate the long-term performance of bentonite buffer, which is crucial for the safety assessment of high-level radioactive waste disposal. In this technical note, Two-region model is introduced, a proposed model to simulate the erosion behavior of bentonite based on a dynamic bentonite diffusion model, and quantitative evaluation is conducted for the bentonite buffer erosion with this model.

The effects of dielectric curing temperature and temperature/humidity treatment conditions on the interfacial adhesion energies between Ti diffusion barrier/polybenzoxazole (PBO) dielectric layers were systematically investigated for Cu redistribution layer applications of fan-out wafer level package. The initial interfacial adhesion energies were 16.63, 25.95, 16.58 J/m2 for PBO curing temperatures at 175, 200, and 225 ℃, respectively. X-ray photoelectron spectroscopy analysis showed that there exists a good correlation between the interfacial adhesion energy and the C-O peak area fractions at PBO delaminated surfaces. And the interfacial adhesion energies of samples cured at 200 ℃ decreased to 3.99 J/m2 after 500 h at 85 ℃/85 % relative humidity, possibly due to the weak boundary layer formation inside PBO near Ti/PBO interface.

Studies were conducted to select the candidate buffer material for a high-level waste (HLW) repository in Korea. This paper presents the hydraulic properties, the swelling properties, the thermal properties, and the mechanical properties as well as the radionuclide release-retarding capacity of Kyungju bentonite as part of those studies. Experimental results showed that the hydraulic conductivities of the compacted bentonite were very low and less than $10^{-11}$m/s. The values decreased with increasing the dry density of the compacted bentonite. The swelling pressures were in the range of 0.66 MPa to 14.4 ㎫ and they increased with increasing the dry density. The thermal conductivities were in the range of 0.80 ㎉/m $h^{\circ}C$ to 1.52 ㎉/m $h^{\circ}C$. The unconfined compressive strength, Young's modulus and Poison's ratio showed the range of 0.55 ㎫ to 8.83 ㎫, 59 ㎫ to 1275 ㎫, and 0.05 to 0.20, respectively, when the dry densities of the compacted bentonite were 1.4 Ms/㎥ to 1.8 Mg/㎥. The diffusion coefficients in the compacted bentonite were measured under an oxidizing condition. The values were $1.7{\times}10^{-10}$m^2$/s to 3.4{\times}10^{-10}$m^2$/s for electrically neutral tritium (H-3), 8.6{\times}10^{-14}$m^2$/s to 1.3{\times}10^{-12}$m^2$/s for cations (Cs, Sr, Ni), 1.2{\times}10^{-11}$m^2$/s to 9.5{\times}10^{-11}$m^2$/s for anions (I, Tc), and 3.0{\times}10^{-14} $m^2$/s to 1.8{\times}10^{-13}$m^2$/s $for actinides (U, Am), when tile dry densities were in the range of 1.2 Mg/㎥ to 1.8 Mg/㎥. The obtained results will be used in assessing the barrier properties of Kyungju bentonite as a buffer material of a repository in Korea.n Korea.

The purpose of this study was to evaluate the efficacy of blocking the oxygen in the air during the polymerization of sealant. All curing were performed with various light curing units under the application of oxygen gel barrier, stream of nitrogen and carbon dioxide gas for inhibition of oxygen diffusion into sealant surface. The results of present study can be summarized as follows : 1. The amount of eluted TEGDMA form the specimens cured with all the three different light units in the stream of $N_2$ and $CO_2$ gas and application of Oxygen gel barrier($DeOx^{(R)}$) were significantly lower than in the room-air atmosphere (Control) (p<0.05). 2. In the $DeOx^{(R)}$ application, the amount of eluted TEGDMA the specimen cured with PAC light for 10seconds was less than that cured in the stream of $N_2$ and $CO_2$ atmospheric conditions (p<0.05) 3. In the LED using 10 or 20sec irradiation times under the stream of $N_2$ and $CO_2$, the eluted TEGDMA showed to be no statistically significant difference (p>0.05). 4. The microhardness from the specimens cured with all the three different light units under each treated conditions were significantly higher than in the room-air atmosphere (p<0.05). 5. The surface treatment by $DeOx^{(R)}$, $N_2$ and $CO_2$ reduces the thickness of oxygen inhibited layer by sp proximately 49% of the untreated control value.

The epidermal growth factor (EGF) has a intrinsic function of inducing growth and proliferation of cells through interacting with cell membrane receptors in human epidermis and dermis layer. These functions of EGF are used as a main ingredient for wound healing medicines and anti-aging cosmetics. As a cosmetic ingredient, the EGF has a problem in exhibiting its natural efficacy due to the lack of the ability to penetrate through the stratum corneum, which is known as the skin barrier. In this study, a recombinant human epidermal growth factor ($MTD_{151}-EGF$) fused with the macromolecule transduction domain $(MTD)_{151}$ with the skin penetration ability was developed to improve the skin penetration efficiency of the EGF. Expression of $MTD_{151}-EGF$ was performed in E. coli transformed with a vector encoding the $MTD_{151}-EGF$ gene and then purified. The purified $MTD_{151}-EGF$ was evaluated using cell proliferation assay, cytotoxicity test and skin penetration test by franz diffusion cell assay and artificial skin. Cell proliferation activity of $MTD_{151}-EGF$ purified to high purity of 99% or above was equivalent to the EGF or better, and cytotoxicity was not observed. In addition, the $MTD_{151}-EGF$ showed an excellent penetration efficiency compared to the EGF in the skin penetration test with EGF and $MTD_{151}-EGF$ labeled by FITC in an artificial skin penetration model. Based on the quantitative analysis of the penetrating substance using franz diffusion cell assay, the amount of penetration was about 16 times more than that of EGF. These results can be regarded as an effective alternative to improve the existing physical transdermal penetration method related to the use of various active ingredients for cosmetics.
The roles of RNA- and protein-synthesis and $H^{+}$ excretion in 1AA ($10\;\mu\textrm{M}$)-induced elongation were investigated using abraded hypocotyl segments of sunflower (Helianthus annuus L.). The response of elongation initiated about 13 min after IAA treatment. Removal of cuticle, acting as diffusion barrier for inhibitors, by mechanical abrasion of hypocotyl segments enhanced the effect of inhibitors markedly, but the degree of abrasion for the saturated effect of inhibition was different among inhibitors. The elongation induced by 1M was completely inhibited when cycloheximide ($10\;\mu\textrm{M}$) was applied to abraded hypocotyl segments as shortly as 4 min before the onset of the growth response (= 10 min after administration of IAA). Cordycepin ($200\;\mu\textrm{M}$) prevented completely 1AA-induced elongation when applied as shortly as 19 min before the onset of the growth response (=5 min before administration of 1AA). Vanadate (1 mM) inhibited both lAA-induced elongation and medium acidification via lAA-induced $H^{+}$ excretion to apoplast. Cycloheximide and cordycepin also prevented lAA-induced $H^{+}$ excretion strongly. However, inhibition by cycloheximide of lAA-induced elongation was not alleviated by acidifying the cell wall to pH 4.5. The results indicate that, a few minutes before the initiation of growih, protein synthesis is demanded for the initiation of 1AA-induced elongation and the $H^{+}$ excretion to cell wall, and that the H+ excretion, even though it may be necessary for elongation, does not seem to bring about acid growth simply through acidifying cell wall.l wall.
Numerous factors concern with the absorption of substances through the membrane of the gastrointestinal tract. To simplify the experimental condition, present work has been restricted to observe the disappearance rate of substance from the intestinal loop which was made in the jejunum, 70 cm apart from the pylorus of the adult rabbit. The purpose of the study is to clarify the absorption of urea through the jejunal wall is solely attributable to the concentration difference between the luminal fluid and plasma, and to observe the effect of adding red pepper upon the rate of absorption. The rabbits were anesthetized with nembutal, 35mg/kg I.V. Jejunal loop was made by ligating at 2 spots, 70 cm and 80cm apart from the pylorus. After rinsing with normal saline solution through the polyethylene tubing inserted from the end of the loop, 8 ml of test solution was placed through the same tubing. The test solution contained 200 mg% of urea and 150mg% of polyethylene glycol(M.W. 4,000) in normal saline solution. Right after placing the test solution the first specimen was taken through the tubing, and successive samplings were performed at 5, 10, 20, and 30 minutes. Logarithm of the difference of urea concentration between the luminal fluid and plasma was plotted against time elapsed after the onset of the experiment. If straight line is revealed, it would verify the nature of transport mechanism as diffusion, obeying the Fick's principle. The concentration of polyethylene glycol (PEG) was also measured in order to examine the change in the volume. PEG was used as the marker substance because it is not absorbable in the intestinal tract. Consequently the concentration of PEG relates inversely to the volume of the loop. Instantaneous concentration of urea in the loop times the volume will give the amount of urea remaining in the luminal fluid. The change in the amount of any substance is directly relate to the volume of the compartment and differs from the change in the concentration which is independent of the volume. After completion of the experiment without red pepper, it was added in the test solution and was centrifuged after thorough mixing. Supernatant of the mixture was placed in the loop and similar sampling were performed with the same time intervals that of previous run in order to observe the effects of the red pepper on the passive transport of the water soluble small substance, urea. The results obtained were as follows: 1. Logarithm of the concentration difference of urea between the luminal fluid and plasma was diminished exponentially as time elapsed. The decay constant in the experiment without red pepper was 0.0563/min. By adding red pepper in the test solution as much as the concentration rose to 4,000 mg% and 8,000 mg%, the decay constants were lowered to 0.0493/min and to 0.0506/min, respectively. The time interval by which the concentration difference dropped to one half of the initial value was prolonged. Without red pepper the half concentration time was 13.30 minutes, and by adding extract of red pepper, 15.31 minutes and 15.71 minutes were revealed. 2. The profile of the diminishing rate of tile amount of urea was quite different from that of the concentration because of the change in the volume of the loop during the observed period. 3. By adding the extract of red pepper, it slowed down the rate of absorption of urea in the intestinal loop, suggesting an increase in the diffusional barrier. 4. Larger dosage of red pepper brought an increase in the secretion of intestinal fluid with concomitant expansion of the luminal volume, and the retardation of the absorption of urea was noticed. This effect was largely dependent on the sensitivity of the individual animal to the red pepper, extract. The amount of urea remained after 10 minutes interval was 55.5% of the initial amount in the experiment without red pepper. On the other hand it was not consistent after administration of red pepper, showing 50.6% and 66.5% of the initial figures by adding 400 mg and 800 mg of red pepper in the test solution, respectively. It was postulated that symptom of diarrhea often encountered by taking a hot (red pepper) food might be attributable to the increase of secretion and the retardation of absorption in the intestinal tract.
