Antioxidant and Anti-adipogenic Effects of Fermented Rhus verniciflua (발효옻 추출물의 항산화 및 지방세포 분화 억제 효과)

  • This study investigates the antioxidant and anti-adipogeneic effects of fermented Rhus verniciflua., evaluating the total phenol, total flavonoids contents and antioxidant activity of the fermented Rhus verniciflua as well as assessing the lipid accumulation during adipogenesis of 3T3-L1 cells. Our results demonstrate that the total phenolic and flavonoids contents of fermented Rhus verniciflua were $29.2{\pm}0.12$ GAE mg/g and $20.4{\pm}1.52$ RE mg/g, respectively. The antioxidative activities of fermented Rhus verniciflua were significantly increased in a dose dependent manner on DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical scavenging, ABTS(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP(ferric reducing antioxidant power)activity, reducing power. In addition, the fermented Rhus verniciflua did not show any cytotoxicity up to 300 ug/mL. However, the anti-adipogenic effect of fermented Rhus verniciflua extract was barely detectable.

Investigation of Antioxidative and Tryosinase Inhibitory Activities of the Seed Extracts (종자 추출물의 항산화 활성 및 Tyrosinase 저해 활성 탐색)

  • Bioactive substances, antioxidant activities and tyrosinase inhibitory activities of seed extracts were evaluated to discover new functional materials, using 13 edible or medicinal plants. Total polyphenol and flavonoid contents were highest in Taxus cuspidata, with $57.51mg{\cdot}g^{-1}\;and\;7.98mg{\cdot}g^{-1}$, respectively. Seed extract of Vitis coignetiae${\times}$Vitis labruscana($RC_{30}=0.030mg{\cdot}ml^{-1}$) was found to be the most effective in DPPH radical scavenging test, and the highest ABTS radical scavenging activity was shown in Diospyros lotus($RC_{30}=0.044mg{\cdot}ml^{-1}$). Inhibitory effects on peroxidation of linoleic acid determined by ferric thiocyanate (FTC) method were similar to BHT in Diospyros kaki, Diospyros lotus, Magnolia officinalis, Styrax obassia, Vitis coignetiae, Vitis coignetiae${\times}$Vistis labruscana, Zizyphus jujuba, Zizyphus jujuba var. inermis. For the inhibition against mushroom tyrosinase, Diospyros kaki, Diospyros lotus, Poncirus tritoliata, Prunus serrulata var. spomtanea, Zizyphus jujuba and Zizyphus jujuba var. inermis exhibited inhibitory activity, and especially Diospyros lotus showed the strongest inhibition.

Cellular Protective Effects and Antioxidative Activity of Resveratrol (레스베라트롤의 세포보호 및 항산화 효과)

  • In this study, the cellular protective effect of resveratrol on oxidative damage and its antioxidative activity were investigated. The free radical-scavenging activity ($FSC_{50}$) of resveratrol was measured to be $103{\mu}M$. The reactive oxygen species-scavenging activity ($OSC_{50}$) of resveratrol on the ROS generated in a $Fe^{3+}-EDTA/H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. Resveratrol displayed $0.042{\mu}M$ ROS scavenging activity, which is 9.6-fold higher than that of L-ascorbic acid ($0.405{\mu}M$) and had a more prominent cellular protective effect than (+)-${\alpha}$-tocopherol. When HaCaT cells were exposed to $800mJ/cm^2$ of UVB or treated with $30{\mu}M$ rose bengal, resveratrol protected the cells against oxidative stress in a concentration-dependent manner; however, it was unable to protect the cells when the damage was induced by 10 mM $H_2O_2$. These results indicate that resveratrol could be employed to improve and prevent the skin aging through its antioxidative and cellular protective activities.

The Antioxidative Effect of Black Garlic Extract on Paraquat-induced Oxidative Stress in ICR Mice (Paraquat에 의해 유발된 산화적 스트레스에 의한 흑마늘 추출물의 항산화 효과)

  • We investigated the antioxidative effect of black garlic extract (BGE) on paraquat (PQ)- induced oxidative stress in mice. A DPPH radical scavenging activity assay showed that BGE had potent free radical scavenging activity, comparable to that exhibited by vitamin C. Mice were administered with either vitamin C or two levels of BGE by oral gavage for 10 days, with PQ being injected intraperitoneally on day five. It was found that BGE reduced the liver enzyme levels induced by PQ injection in mice. The concentrations of plasma and hepatic malonedialdehyde were both significantly reduced in the BGE groups compared with the levels in the PQ group, whereas the hepatic superoxide dismutase and catalase activities were significantly increased in the BGE groups compared with the PQ group. These findings suggest that BGE has potent antioxidative activities in vivo and thus could prevent the oxidative stress induced by PQ injection in mice by two mechanisms: the induction of antixoxidative enzyme systems and direct scavenging of reactive oxygen species.

  • Astragalus membranaceus has a long history of medicinal use in Chinese herbal medicine. It has been shown to have immunostimulant, tonic, antioxidant, antiperspirant, diuretic, anti-diabetic, expectorant properties, and a supplementary medicine during cancer therapy. In this study, we investigated the effect of anti-oxidation of Astragalus membranaceus root extract. The anti-oxidative activities of water, 80% methanol, and 100% methanol extracts from Astragalus membranaceus were analyzed by DPPH free radical scavenging activity, Superoxide dismutase-like activity, reducing power, and crude ash. The water extract demonstrated to be more effective than methanol extract for a DPPH radicals scavenging activities and reducing power. Superoxide dismutase-like activity showed higher efficiency in 80% methanol extract. Our results indicate that Astragalus membranaceus extracts could be used as a source of antioxidant ingredients in the food industry.

Comparison of Antioxidant Activities of Black Onion Extracts (추출용매에 따른 흑양파의 항산화 활성 비교)

  • The antioxidant activities of the ethanol, methanol, and water extracts of black onion were investigated. The highest extraction yield (28.56%) and total polyphenol content (13.5 mg/g) were found in the water extract. The water extract also showed the highest protective effect on the RAW 264.7 cell anti-inflammatory activity, and the water and ethanol extracts showed the highest reducing power. The water extract showed higher DPPH and ABTS radical scavenging activities (65.98 and 89.69%, respectively) than the other solvent extracts (ethanol, 47.77%; methanol, 66.12%). Hence, black onion can be used as a potent natural antioxidative source.

Effects of Abeliophyllum distichum Nakai Flower Extracts on Antioxidative Activities and Inhibition of DNA Damage

  • The present study attempts to evaluate antioxidant activities of extracts from Abeliophyllum distichum. Nakai flower. The samples were collected in Jangyyeon-myeon, Goesan-gun, Korea and extracted with either hot-water or ethyl acetate (EtOAC). In DPPH, hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 93.41%, 98.43%, and 7.38%, while those of hot-water extracts were 86.93%, 41.33% and 47.68% at 200 ${\mu}g/ml$, respectively. In ${\varphi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 82% and 17% at 200 ${\mu}g/ml$, respectively. Both extracts showed the protective effect of DNA migration by oxidative stress in intracellular DNA migration assay. Both extracts had no cytotoxity in NIH3T3 cells. Several polyphenolic compounds were identified such as 2-methoxy-benzoic acid, vanillic acid, phytol and pulegone by GC/MS. These results indicated that extracts of Abeliophyllum distichum Nakai flower showed antioxidant activities and protective activities against oxidative DNA damage and showed the possibility to be used as an effective natural antioxidants.

Antioxidative Properties of Extract/Fractions of Suaeda asparagoides and Salicornia herbacea Extracts(I) (함초(나문재 및 퉁퉁마디) 추출물의 항산화 작용에 관한 연구(I))

  • In this study, the antioxidative effects of Sueada asparagoides and Salicornia herbacea extracts were investigated. The free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$) of extract/fractions of Sueada asparagoides was in the order: 100 % ethanol extract(329.33 ${\mu}g/mL$) < 50 % ethanol extract(40.73) < ethylacetate fraction(13.87) < deglycosylated aglycone fraction (7.80). In case of Salicornia herbacea, the free radical scavenging activities of ethylacetate fraction and aglycone fraction were 13.87 and 7.80 ${\mu}g/mL$, respectively. Reactive oxygen species(ROS) scavenging activities($OSC_{50}$) of Sueada asparagoides and Solicornia herbacea extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity of Sueada asparagoides extracts was 50 % ethanol extract($OSC_{50}$, $0.99{\mu}g/mL$) < ethylacetate fraction (0.05) < aglycone fraction (0.03). Aglycone fraction showed the most prominent scavenging activity. In case of Salicornia herbacea, the ROS scavenging activities of ethylacetate fraction and aglycone fraction were 0.10 and 0.20 ${\mu}g/mL$, respectively. The protective effects of extract/fractions of Sueada asparagoides and Salicornia herbacea on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract(100%) of Sueada asparagoides diminished photohemolysis in a concentration dependent manner($1{\sim}100{\mu}g/mL$). Particularly deglycosylated aglycone fraction exhibited the most prominent celluar protective effect($\tau_{50}$, 310 min at 50 ${\mu}g/mL$). In case of Salicornia herbacea, ethylacetate fraction exhibited more potent protective effect. These results indicate that extract/fractions of Sueada asparagoides can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS.

Antioxidative and antimicrobial activities of Oenothera biennis extracted by different methods (추출방법을 달리한 달맞이꽃 추출물의 항산화 및 항균 활성)

  • A effect of extraction methods, including stirrer extraction method (SE), ultrasonification extraction method (USE), reflux extraction method (RE), autoclave extraction (AE) and low temperature high pressure extraction (LE) method on the antioxidative and antimicrobial activities of ethanol extracts of Oenothera biennis was investigated. The extraction yield (46.33%), total polyphenol (463.05 mg GAE/g) and flavonoid (71.71 mg RHE/g) content of Oenothera biennis extract obtained by RE were higher than those from other extraction methods. The antimicrobial activity of Oenothera biennis extract was only observed against Bacillus cereus among other tested organisms (Bacillus cereus, Staphylococcus aureus, Escherichia coli and Salmonella Typhimurium). Oenothera biennis obtained by RE showed the best DPPH radical scavenging ability (74.40%), ABTS radical scavenging ability (65.29%), reducing power (1.370 ($OD_{700}$)) and ferrous ion chelating ability (90.14%) compared with other tested extraction methods tested. The RE method was the most efficient method for extracting crude antioxidant and antimicrobial substances from Oenothera biennis. These results suggested that Oenothera biennis obtained by RE could be used as a bioactive and functional material in the food industry.

Antioxidant and Anti-Adipogenic Effects of Ethanolic Extracts from Ixeris dentata Nakai (씀바귀 에탄올 추출물의 항산화 및 지방세포 분화억제 효과)

  • The aim of this study was to evaluate the total phenol, total flavonoids contents and antioxidant activity of 80% ethanolic extract from Ixeris dentata Nakai(IDE) as well as to assess the lipid accumulation during adipogenesis of 3T3-L1 cells. The results demonstrated that the total phenolic and flavonoids contents of the IDE were $4.01{\pm}0.63$ GAE mg/g and $0.05{\pm}0.01$ RE mg/g, respectively. The antioxidative activities of the IDE were significantly increased in a dose dependent manner on DPPH(1,1-Diphenyl-2-picrylhydrazyl) radical scavenging, ABTS(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt) radical scavenging, reducing power value. During adipocyte differentiation, IDE significantly inhibited lipid accumulation compared with the control cells. These results indicated that the anti-adipogenesis effect of Ixeris dentata Nakai could be attributed to phenolic compounds that may potentially inhibit ROS(reactive oxygen species) production.
