Antioxidant Activities and Inhibitory Effect on Oxidative DNA Damage of extracts from Abeliophylli distichi Folium (미선나무 잎 추출물의 항산화 및 산화적 DNA 손상억제 활성)

  • Objective : In this study, we demonstrate the protective effect on oxidative DNA damage of leaf extracts from Abeliophylli distichi Folium via its antioxidant activity for the establishment of new value for the herbal medicine. Methods : Abeliophylli distichi Folium leaves were extracted with hot-water and ethylacetate (EtOAC). The 1,1-diphenyl-2- picrylhydrazyl (DPPH) radical and hydroxyl scavenging assay and $Fe^{2+}$ chelating assay were performed for antioxidative effect and ${\varphi}$X-174 RF I DNA cleavage assay and intracellular DNA damage assay were used for inhibitory effect of intracellular DNA damage. Results : In DPPH, Hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 94.72%, 62.88%, 41.13%, and hot-water extracts were 88.86%, 56.7%, 37.4% at 200 ${\mu}g/m{\ell}$, respectively. Also, those extracts showed protective effect of DNA damage against the oxidative stress. Conclusion : These results indicated that the leaf extracts of Abeliophylli distichi Folium can be used as a natural antioxidants, which effectively inhibits the oxidative DNA damage.

Cosmetic Effect of Angelica gigas Nakai Root Extracts (참당귀(Angelica gigas Nakai) 뿌리 추출물의 화장품소재 특성)

  • Root extracts of Angelica gigas Nakai were tested to see the possibility for functional cosmetic agents. From ethanol extraction method, 97% of decursin and decursinol angelate was obtained, and concentration ratio of decursin to decursinol angelate was about 3:2. To test possibility as a functional cosmetic agent, DPPH free radical scavenging assay, UVA/B absorption, tyrosinase inhibition assay, melanogenesis inhibition assay, elastase inhibition assay and MTT assay were done. Root extracts of Angelica gigas Nakai showed $45.2{\pm}3.9%$ tyrosinase inhibition of tyrosine, and $24.2{\pm}12.0%$ melanin inhibition at $15{\mu}g/ml$ extract concentration, so that it indicated good whitening effect. DPPH free radical scavenging activity was $40.9{\pm}9.1%$ at $240{\mu}g/ml$ concentration, which is relatively good. Anti-wrinkle effect was poor such that it was $12.7{\pm}6.8%$ at $100{\mu}g/ml$. UVA/B absorption was also negligible. From the research, root extracts of Angelica gigas Nakai showed good potential as a whitening agent.

Antioxidant Effect of Cynomorii Herba on HepG2 Cells and Diphenyl-picryl-hydrazyl (DPPH) Radical Scavenging Activity (쇄양(鎖陽)의 Diphenyl-picryl-hydrazyl (DPPH) 소거 활성 및 HepG2 세포에 대한 항산화 효과)

  • The purpose of this study was to investigate the anti-oxidant effect of Cynomorium songaricum. The extract of Cynomorii Herba was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, HepG2 cell viability and $H_2O_2$-induced cytotoxicity by a modified MTT assay. DPPH radical scavenging activity was measured after 30 minutes. The extract was tested by 1. 5, 10, 50, 100 and 500 ${\mu}g/ml$ concentrations. HepG2 cell viability by a modified MTT assay was measured in the concentrations of 10, 50, 100, 250, 500 ug/ml for 24 h. The results showed that the extract scavenged DPPH radical up to 52.2% with 50 ug/ml concentration. The extract did not reduced the cell viability and $H_2O_2-induced$ cytotoxicity (69.4%) was blocked by the extract in the concentrations of 50, 100, 250 and 500 ${\mu}g/ml$. In conclusion, the extract of Cynomorii Herba has potent antioxidant activity.

  • Objective : This study was conducted to investigate the antioxidant activity and inhibitory effect on oxidative DNA damage of Nelumbinis Semen Extracts Methods : Nelumbins semen were extracted with hot-water and ethylacetate (EtOAC). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay were performed for antioxidative effect and ${\phi}X$-174 RF I DNA cleavage assay and intracellular DNA damage assay were used for inhibitory effect on intracellular DNA damage. Results : In DPPH, Hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity of EtOAC extracts were 96.22%, 53.53%, 64.72%, while those of hot-water extracts were 20.86%, 10.72%, 29.74% at $200{\mu}g/m{\ell}$, respectively. In ${\phi}X$-174 RF I plasmid DNA cleavage assay, the protective effects of EtOAC and hot-water extracts against oxidative DNA damage were 76% and 6% at $200{\mu}g/m{\ell}$, respectively. Conclusion : These results indicated that the seed extracts of Nelumbo nucifera can be used as a natural antioxidants, which effectively inhibits the oxidative DNA damage.

Antilipid Peroxidative Effect of Houttuynia cordata (즙채의 과산화지질 생성 저해효과)

  • Houttuynia cordata (Saururaceae) has pungent smell and taste. It has been regarded as detoxicant, antipyretic, antiinflammantory and diuretic agents. In order to evaluate antioxidative and antilipidperoxidative efficacies, its fractions ($H_2O$, 20% MeOH, 40% MeOH, 60% MeOH, 170% MeOH) were measured by DPPH method and TBARS assay on rat liver homogenate. It was revealed that 60% MeOH fractions had potential antioxidative activity and inhibited lipid peroxidation significantly: In active fractions, we isolated rutin, hyperin, quercitrin and quercetin.

Antioxidative Effects of Purple Sweet Potato Extracts (자색고구마 추출물의 항산화 효과)

  • The colored sweet potato, particularly purple sweet potato, has been well known to contain anthocyanins abundantly. This study was conducted to examine the antioxidant properties of purple sweet potato. The chopped purple sweet potato was extracted 2 times with water or acetone for 18 hours at $28^{\circ}C$. The antioxidative potential of each solvent extract was assessed by DPPH free radical scavenging activity assay, FRAP assay, and total phenolic contents. The results showed that both extracts had not only high DPPH free radical scavenging activity but had high level of total phenolic compounds. Furthermore, both solvent extracts were found to have antioxidative effects in human colon cancer cells (HCT 116, HT 29) in DCFDA assay. The notable antioxidant activity of purple sweet potato suggests its significant health benefit and deserves further study to develop into functional food ingredient.

Measurement of the Anti-oxidative Properties of Extract from Medicinal Plants Using an On-line HPLC-DPPH Assay (HPLC와 DPPH radical 소거능 측정 방법의 결합에 의한 약용 식물 추출물의 항산화 활성 비교)

  • Natural anti-oxidative compounds have important disease prevention and food preservation properties, in addition to anti-bacterial, anti-inflammation, anti-cancer, and skin whitening effects. High-performance liquid chromatography (HPLC), with an ultra vilolet (UV) detector coupled to a reverse phase C18 column and an online measurement system for 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radicals, was used to search for potent antioxidative compounds in crude extracts. The online HPLC-DPPH assay was then applied to confirm antioxidative compounds in water extracts from Radix of Pueraria lobata, Rhizoma of Zingiber officinale, Fructus of Chaenomeles sinensis, Cortex of Ulmus pumila, and Radix of Astragalus membranaceus. To determine the yields of the extracts, the Brix% of each extract solution was measured using a refractometer. When the relative DPPH radical scavenging ability values of the water extracts were compared with those of a positive control (ascorbic acid), the water extracts of P. lobata, C. sinensis, and U. pumila were 7.77%, 4.71%, and 4.19%, respectively. The results suggest that this method provides a useful assay for rapid measurement of DPPH radical scavenging abilities and conformation of antioxidative compounds in natural products. Moreover, it can reduce the time spent on the separation of active compounds from natural materials, such as medicinal plants, in addition to the use of reagents for separation.

Properties of Fucoidan as Raw Materials of Water-holding Cream and Cosmetics (후코이단의 보습제 및 화장품재료로서 특성)

  • Properties of fucoidan used for functional cosmetics agents were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls. To test possibility of fucoidan as a cosmetics material, water-holding property measurement, DPPH free radical scavenging assay and MTT assay were done. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease. Fucoidan showed high stability from MTT assay and good anti-oxidation property from DPPH assay. To evaluate the effect of water-holding property and anti-alergy property of fucoidan on the atopic dermatitis(AD), 46 AD patients were treated with fucoidan cream. After 6 weeks treatment, Investigation Global Assessment(IGA) scores decreased from 3.04 to 2.15, that is fucoidan cream had a 39.8% benefit effect on atopic dermatitis.

Antioxidant potentials of Hypericum hookerianum (Family: Hypericaceae) on CCl4 induced hepatotoxicity in rats

  • Free radicals are known to play important role in pathophysiology of hepatic disorders and antioxidants are employed along with other chemotherapeutic agents in treatment of such diseases. In search of natural antioxidant, successive extracts of Hypericum (H.) hookerianum (Family: Hypericaceae) were evaluated by in vitro and in vivo methods. Extracts of aerial parts of H. hookerianum were subjected for 1,1-diphenyl 2-picryl hydrazyl radical scavenging activity (DPPH assay), nitric oxide radicals scavenging assay and thiobarbituric acid reactive substances (TBARS) assay. Methanolic extract was found to be more active than other extracts in DPPH and in vitro TBARS assay with $IC_{50}$ at 5.82 ${\pm}$ 1.33 ${\mu}g/ml$ and 49.78 ${\pm}$ 3.79 ${\mu}g/ml$ respectively. While petroleum ether extract showed more potentials in scavenging the nitric oxide radicals with $IC_{50}$ 220.97 ${\pm}$ 2.69 ${\mu}g/ml$. The administration of $CCl_{4}$ to the control animals caused decrease in the level of catalase and superoxide dismutase, together with significant increase in the level of TBARS in liver and kidney. Reversal of these changes towards normal group was observed by administration of H. hookerianum methanolic extract at 50 and 100 mg/kg body weight, while other extracts were found to be less active.
