쇄양(鎖陽)의 Diphenyl-picryl-hydrazyl (DPPH) 소거 활성 및 HepG2 세포에 대한 항산화 효과 (Antioxidant Effect of Cynomorii Herba on HepG2 Cells and Diphenyl-picryl-hydrazyl (DPPH) Radical Scavenging Activity)

  • The purpose of this study was to investigate the anti-oxidant effect of Cynomorium songaricum. The extract of Cynomorii Herba was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, HepG2 cell viability and $H_2O_2$-induced cytotoxicity by a modified MTT assay. DPPH radical scavenging activity was measured after 30 minutes. The extract was tested by 1. 5, 10, 50, 100 and 500 ${\mu}g/ml$ concentrations. HepG2 cell viability by a modified MTT assay was measured in the concentrations of 10, 50, 100, 250, 500 ug/ml for 24 h. The results showed that the extract scavenged DPPH radical up to 52.2% with 50 ug/ml concentration. The extract did not reduced the cell viability and $H_2O_2-induced$ cytotoxicity (69.4%) was blocked by the extract in the concentrations of 50, 100, 250 and 500 ${\mu}g/ml$. In conclusion, the extract of Cynomorii Herba has potent antioxidant activity.

톳 자숙액 에탄올 추출물의 항산화 활성 (Antioxidant Activity on Ethanol Extract from Boiled-water of Hizikia fusiformis)

  • Antioxidant activity of the ethanol extract from boiled-water of Hizikia fusiformis (EBH) were compared with those of BHA, L-ascorbic acid, gallic acid, caffeic acid and (-)-catechin. The free radical scavenging ability against DPPH (1,1-diphenyl-2-picrylhydrazyl), authentic peroxynitrite and reducing power were measured as indices of antioxidant activity. EBH showed the potent DPPH radical and peroxynitrite scavenging activities, showing 85.23 and 96.97% at final concentration of $1000{\mu}g/ml$, respectively. The reducing power increased with the increasing amount of EBH (final concentration of 1, 10, 100 and $1000{\mu}g/ml$). Total phenolic content of EBH was 588 mg (-)-catechin/g at the final concentration $1000{\mu}g/ml$. Total phenolic contents correlated with DPPH radical scavenging activity $(R^2=0.766)$ and reducing power $(R^2=0.944)$. These results suggested that EBH could be a natural antioxidative source containing antioxidative components.

흑삼의 항산화 활성 (Antioxidant Activity of Black Panax ginseng)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.

단백질 분해효소를 이용한 오계란 가수분해물의 항산화 활성 (Antioxidant Activity of Ogae Egg White Protein Hydrolysates using commercial Protease)

  • 식물 및 동물성 단백질 유래 펩타이드 형태의 단백질 가수 분해물들은 항산화, 고혈압 완화, 면역조절, 진통완화 및 항균작용 등 생리활성이 있는 것으로 알려져 왔다. 본 연구는 6가지 프로티아제를 이용하여 오계란 단백질 가수분해물을 생산하고, 생산된 펩타이드의 항산화 능력을 평가하였다. 그 결과 가수분해도의 최대값은 protamex(46.3%)이고, DPPH 라디칼 소거능 최대값은 bromelain(57.23%), 하이드록시 라디칼 소거능 최대값은 alcalase(30.21%), 슈퍼옥사이드 라디칼 소거능 최대값은 alcalase(58.07%), $Fe^{2+}$ 킬레이션 능력 최대값은 alcalase(72.06%)로 나타났다. 더 나아가 효소별 항산화 저해 능력 $IC_{50}$ 평가하였다. 그 결과 alcalase에 의한 최대값은 금속 킬레이션 능력($IC_{50}$, 1.24 mg/mL)이고, bromelain에 의한 최대값은 DPPH 소거능($IC_{50}$, 2.46 mg/mL)이고, flavourzyme에 의한 최대값은 금속 킬레이션 능력($IC_{50}$, 1.25 mg/mL)이고, neutrase에 의한 최대값은 DPPH 소거능($IC_{50}$, 3.64 mg/mL)이고, papain에 의한 최대값은 DPPH 소거능($IC_{50}$, 3.82 mg/mL)이고, protamex에 의한 최대값은 DPPH 소거능($IC_{50}$, 1.93 mg/mL)이었다. 따라서 protease를 이용하여 오계란 단백질에서 추출한 펩타이드는 항산화 기능성 식품소재로서 활용할 가치가 높을 것으로 기대한다.

구아바 잎 추출물의 항산화 및 미백 활성 효과 (Antioxidant Activity and Whitening activity of Psidium guajava leaf extract)

  • 구아바 잎 추출물이 미백 기능성 화장품 소재로써의 활용 가능성을 확인하고자 하였다. DPPH radical 소거 활성, 세포 내 ROS 측정, B16F10 melanoma cell 에서의 세포 독성 및 자외선A에 대한 세포 보호효과, 시험관 내 tyrosinase 억제 효과, 멜라닌 생합성 억제 효과를 측정하였다. 구아바 잎 추출물의 높은 DPPH radical 소거 활성과, 세포 내 ROS 활성 억제 측정을 통해 항산화 효과를 확인하였다. B16F10 melanoma cell에서의 세포 생존율이 모든 농도에서 98% 이상의 생존율을 나타냈으며, UVA로부터의 세포 보호효과가 농도 의존적으로 높아지는 것을 확인하였다. 또한 10%의 시험관 내 tyrosinase 활성 억제 효과와 20%의 멜라닌 생합성 억제 효과를 확인되었다. 구아바 잎 추출물은 B16F10 melanoma cell에 대한 독성이 적고, 높은 항산화 활성과 tyrosinase 활성 억제 및 멜라닌 생합성 억제 효과를 통해 안전하고 우수한 미백 효과를 가진 미백 기능성 화장품 소재로써의 개발 가능성을 확인하였다.

Antioxidant activity and anti-inflammatory activity of ethanol extract and fractions of Doenjang in LPS-stimulated RAW 264.7 macrophages

  • BACKGROUND/OBJECTIVES: Fermentation can increase functional compounds in fermented soybean products, thereby improving antioxidant and/or anti-inflammatory activities. We investigated the changes in the contents of phenolics and isoflavones, antioxidant activity and anti-inflammatory activity of Doenjang during fermentation and aging. MATERIALS/METHODS: Doenjang was made by inoculating Aspergillus oryzae and Bacillus licheniformis in soybeans, fermenting and aging for 1, 3, 6, 8, and 12 months (D1, D3, D6, D8, and D12). Doenjang was extracted using ethanol, and sequentially fractioned by hexane, dichloromethane (DM), ethylacetate (EA), n-butanol, and water. The contents of total phenolics, flavonoids and isoflavones, 2,2-diphenyl-1 picryl hydrazyl (DPPH) radical scavenging activity, and ferric reducing antioxidant power (FRAP) were measured. Anti-inflammatory effects in terms of nitric oxide (NO), prostaglandin (PG) E2 and pro-inflammatory cytokine production and inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expressions were also measured using LPS-treated RAW 264.7 macrophages. RESULTS: Total phenolic and flavonoid contents showed a gradual increase during fermentation and 6 months of aging and were sustained thereafter. DPPH radical scavenging activity and FRAP were increased by fermentation. FRAP was further increased by aging, but DPPH radical scavenging activity was not. Total isoflavone and glycoside contents decreased during fermentation and the aging process, while aglycone content and its proportion increased up to 3 or 6 months of aging and then showed a slow decrease. DM and EA fractions of Doenjang showed much higher total phenolic and flavonoid contents, and DPPH radical scavenging activity than the others. At $100{\mu}g/mL$, DM and EA fractions of D12 showed strongly suppressed NO production to 55.6% and 52.5% of control, respectively, and PGE2 production to 25.0% and 28.3% of control with inhibition of iNOS or COX-2 protein expression in macrophages. CONCLUSIONS: Twelve month-aged Doenjang has potent antioxidant and anti-inflammatory activities with high levels of phenolics and isoflavone aglycones, and can be used as a beneficial food for human health.

복분자 에탄올 추출물의 항산화 및 항용혈 효능 (Antioxidant and Antihemolytic Activity of Ethanol Extracts of Rubus coreanus Miquel)

  • 천연에서 유래한 항산화 및 항용혈 소재개발을 위해 복분자 에탄올 추출물의 생리활성 효능 평가를 실시하였다. 복분자를 에탄올 추출한 후, 획득한 추출물을 섬유아세포주인 HS68에 처리하여 DPPH 라디칼 소거능과 SOD 유사 항산화 효능을 평가하였다. 또한 Sprague Dawley 렛트 적혈구 세포에 과산화수소로 산화를 유도한 후 추출물의 항용혈 효능을 살펴보았다. 그 결과 추출물은 농도 의존적으로 DPPH 라디칼 소거능과 SOD 유사 활성이 증가하는 것으로 나타났다. 렛트 적혈구를 이용한 항용혈 평가에서는 과산화수소를 단독으로 처리한 양성대조군보다 추출물을 처리한 군에서 항용혈율이 농도 의존적으로 유의하게 증가하였다. 이들 결과는 복분자 추출물이 항산화효능과 항용혈 약용 식물 소재로서의 가능성을 내는 것으로 사료된다.

Comparative phenolic compound profiles and antioxidative activity of the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) according to cultivation years

  • Background: The study of phenolic compounds profiles and antioxidative activity in ginseng fruit, leaves, and roots with respect to cultivation years, and has been little reported to date. Hence, this study examined the phenolic compounds profiles and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free-radical-scavenging activities in the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) as a function of cultivation year. Methods: Profiling of 23 phenolic compounds in ginseng fruit, leaves, and roots was investigated using ultra-high performance liquid chromatography with the external calibration method. Antioxidative activity of ginseng fruit, leaves, and roots were evaluated using the method of DPPH free-radical-scavenging activity. Results: The total phenol content in ginseng fruit and leaves was higher than in ginseng roots (p < 0.05), and the phenol content in the ginseng samples was significantly correlated to the DPPH free-radical-scavenging activity ($r=0.928^{****}$). In particular, p-coumaric acid ($r=0.847^{****}$) and ferulic acid ($r=0.742^{****}$) greatly affected the DPPH activity. Among the 23 phenolic compounds studied, phenolic acids were more abundant in ginseng fruit, leaves, and roots than the flavonoids and other compounds (p < 0.05). In particular, chlorogenic acid, gentisic acid, p- and m-coumaric acid, and rutin were the major phenolic compounds in 3e6-yr-old ginseng fruit, leaves, and roots. Conclusion: This study provides basic information about the antioxidative activity and phenolic compounds profiles in fruit, leaves, and roots of Korean ginseng with cultivation years. This information is potentially useful to ginseng growers and industries involved in the production of high-quality and nutritional ginseng products.

Antioxidant Activity of Hawthorn Fruit in vitro

  • The antioxidant activity of hawthorn fruit (Crataegus pinnatifida Bunge var. typica Schneider) extracts was investigated by several in vitro antioxidants properties, including DPPH free radical scavenging activity, total phenolic and flavonoid contents, hydroxyl radical scavenging activity, reducing power activity, iron-chelating capacity and nitrite scavenging activity. Among the extracts in this study, the 70% EtOH extract showed higher antioxidant activity than the others. The $IC_{50}$ value of DPPH free radical scavenging activity was $99.26\;{\mu}g/mL$. Furthermore, the 70% EtOH extract also showed significantly high total phenolic and flavonoids contents and reducing power activity. However, the MeOH extract exhibited stronger effects on hydroxyl radical scavenging activity, iron-chelating capacity and nitrite scavenging activity. All the results implicated that, the hawthorn fruit may has the available potential to be utilize as a potential source of natural antioxidant.

민들레 뿌리를 첨가한 약주의 총 플라보노이드, 총 폴리페놀 함량과 전자공여능, 환원력 활성 조사 (Studies on Antioxidant Activity, Total Flavonoids and Polyphenols, and Reducing Power in Yakju with Different Ratios of Dandelion Root)

  • 민들레 뿌리를 이용한 기능성 약주 개발의 기초 자료로 활용하고자 민들레 뿌리를 각각 0, 5, 10, 15%로 첨가하여 민들레 약주(기본적특성: pH 4.0~4.2, 산도4.6~4.8%, 알코올14.92~15.78%, 민들레 뿌리 5% 첨가한 약주를 가장 선호함)의 총 플라보노이드와 총 폴리페놀 함량 및 DPPH 라디칼 소거 활성을 이용한 항산화력을 측정하였으며, 활성 산소종 및 유리기에 전자를 공여하는 환원력을 조사하였다. 총 플라보노이드와 총 폴리페놀 함량은 민들레 뿌리 분말 첨가 비율이 증가할수록 높아졌다. 또한, DPPH 라디칼 소거 활성과 환원력에서도 민들레 뿌리 가루의 첨가비율이 높을수록 증가하였다. 이상의 결과로 보아 민들레 뿌리 가루를 첨가한 약주는 높은 생리활성물질과 항산화 활성을 가지고 있어 기능성 약주로서 개발이 가능하리라 판단된다.
