Isolation and Characterization of a Thermophilic Bacillus sp. producing a Thermostable $\alpha$-glucosidase (내열성$\alpha$-glucosidase를 생산하는 호열성 Bacillus sp. 균주의 분리 및 특성)

  • A thermophilic bacterium (strain DG0303) producing a thermostable $\alpha$-glucosidase was isolated from manure and identified as Bacillus sp. Strain DG0303 produced high level of $\alpha$-glucosidase compared with other thermophilic Bacillus strains. The cellular protein patterns were also compared with other Bacillus strains by sodium dodecyl sulfatepolyacrylamide gel electrophoresis(SDS-PAGE). On the basis of 16S rDNA analysis the Bacillus sp. DG0303 was found to be a member of Bacillus rDNA group 5. The optimum temperature for growth was 65$\circ$C and no growth was obtained at 40$\circ$C or 75$\circ$C. The optimum pH for growth was 5.5 to 8.5. $\alpha$-glucosidase activity was produced during growth and most activity was detected in the culture supernatant. The $\alpha$-glucosidase production was constitutive in the absence of carbohydrates. High level of enzyme activity was detected when the culture was grown on medium containing starch. Addition of glucose resulted in the repression of the $\alpha$-glucosidase production. The optimum pH and tempoerature for enzyme activity were pH 5.0 and 65$\circ$C, respectively. When analyzed by zymogram, the culture supernatant showed a single $\alpha$-glucosidase band with a molecular weight of approximately 60,000.

  • For selection of lactic acid bacteria for probiotics, we have examined isolated strains from Korean Dongchimi to assess the acid, bile, and pancreatic tolerance and the growth inhibition on the pathogens. Especially, a kind of isolated strains, FF-3 showed the highest resistancy to both of HCl and bile salt, as well as the highest inhibitory activities against Salmonella sp. and Escherichia coli. Further the bacteriocin of FF-3 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacterias. By using 16s rDNA sequencing method, FF-3 of the selected lactic acid bacteria were found to be identified as genus Lactobacillus.

Mariannaea samuelsii Isolated from a Bark Beetle-Infested Elm Tree in Korea

  • During an investigation of fungi from an elm tree infested with bark beetles in Korea, one isolate, DUCC401, was isolated from elm wood. Based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer and 28S rDNA (large subunit) sequences, the isolate, DUCC401, was identified as Mariannaea samuelsii. Mycelia of the fungus grew faster on malt extract agar than on potato dextrose agar and oatmeal agar media. Temperature and pH for optimal growth of fungal mycelia were 25oC and pH 7.0, respectively. The fungus demonstrated the capacity to degrade cellobiose, starch, and xylan. This is the first report on isolation of Mariannaea samuelsii in Korea.

Isolation and Identification of Acanthamoeba in a Contact Lens Storage Case

  • Acanthamoeba is a free-living amoeba that causes human infections, and recently the incidence of amoebic keratitis has increased among contact lens wearers. In order to investigate Acanthamoeba contamination of contact lens storage cases, a short survey was performed on 57 contact lens wearers, and Acanthamoeba was found in one contact lens storage case. To diagnose Acanthamoeba, the 18s small subunit ribosomal DNA (18s rDNA) gene was amplified by polymerase chain reaction (PCR), and subsequently, the isolate was identified as A. lugdunensis. This species was originally isolated from a freshwater pool in France, and was reported recently to be a cause of amoebic keratitis. This observation indicates the need for a large survey to investigate the extent of Acanthamoeba contamination, and suggests that contact lens wearers be aware of the importance of hygiene and of the implications of Acanthamoeba infection.

  • Aspergillus oryzae is a filamentous fungus classified in the group Aspergillaceae Ascomycetes. It is an important microorganism for industrial production of enzymes and fermented food productions. The genetic transformation system in A. oryzae was used to protoplast mediated transformation with PEG/$CaCl_2$. When the protoplast was used, the regeneration efficiency was decreased and then transformation frequence was also effected. In this study, fungal transformation was carried out by bypassing the protoplast isolation step, changing enzymes, such as hemicellulase and celluclast, and decreasing the culturing time for the increment of the transformation efficiency. 83 transformants/10ug of DNA with hemicellulase were obtained, compared with less than 10 transformants with novozyme234 and celluclast.

  • We isolated a cDNA clone that encodes a ribulose-1,5-bisphosphate carboxylase small subunit (rbcS) from spinach using a soybean rbcS cDNA as a probe. The small subunit consists of 180 amino acids including a transit peptide of 57 residues. Comparison of the amino acid sequence with those of other plant species shows a maximum of 70-80% identical residues. Southern blot analysis suggests the existence of multiple rbcS genes in the spinach genome. Northern blot analysis indicates that the rbcS gene is expressed predominantly in leaves and that the expression of the gene is induced by light.

  • Brain-derived neurotrophic factor (BDNF) is a small secretory protein and a member of the nerve growth factor (NGF) gene family. We cloned the flounder BDNF gene from a flounder brain cDNA library. The nucleotide sequence of the cloned gene showed an open reading frame (ORF) consisting of 810 bp, corresponding to 269 amino acid residues. The tissue distribution of flounder BDNF was determined by reverse transcription-polymerase chain reaction (RT-PCR) in brain, embryo, and muscle tissues. To express fBDNF using a eukaryotic expression system, we constructed the vector mpCTV-BDNF containing the fBDNF gene and transformed this vector into Chlorella ellipsoidea. Stable integration of introduced DNA was confirmed by PCR analysis of genomic DNA, and mRNA expression in C. ellipsoidae was confirmed by RT-PCR analysis.

Random Isolation of Cadmium-induced Gene by Reverse Transcriptase PCR in Schizosaccharomyces pombe

  • The polymerase chain reaction was used to study the genes inducible under stress from the heavy metal cadmium. Schizosaccharomyces pombe, grown in the presence or absence of sublethal concentration of cadmium, was isolated to purify the total RNAs. The Induced RNA Random Fishing (IRRF) method in which random oligonucleotides were used as primers was applied to the identification of cadmium-induced gene expressions. A PCR-DNA product of 400-bp was cloned and sequenced. Computer analysis showed that this DNA has no homology with any known DNA sequences in GenBank or EMBL databases. The induction of this gene was confirmed by Northern blot analysis of total RNAs isolated from both cadmium-treated and untreated yeast cells.

Molecular Cloning of Red Seabream, Pagrus major Somatolactin cDNA and Its Expression in Escherichia coli

  • Isolation, cloning and sequencing of red seabream (Pagrus major) somatolactin (rsbSL) cDNA from pituitary gland revealed an open reading frame of 693 bp coding for a pre-growth hormone of 231 amino acids with a 22 amino acid putative signal peptide. Deduced amino acid sequence showed that there was one possible N-glycosylation site at Asn$^{145}$ and seven Cys residues (Cys$_{29}$ , Cys$^{39}$ , Cys$^{66}$ , Cys$^{89}$ , Cys$^{205}$ , Cys$^{222}$ , Cys$^{230}$ ). Except Cys$^{66}$ , others may be involved in disulfide bond formation. The rsbSL presented a 93% amino acid sequence identity with the SL of gilthead seabream (Sparus aurata) and contained the conserved hormone domain region. Expression of rsbSL in E. coli (BL2l) cells and gel analysis revealed a higher molecular weight for rsbSL than expected theoretically, implying posttranslational modifications.

Isolation and Characterization of UV-Inducible Gene UV150 and UV200 in Eukaryotic Cells (진핵세포에서 DNA 상해에 반응하는 유전자 (UV150과 UV200) 기능연구 분리 및 특성 연구)

  • 본 연구는 DNA 상해유도기작을 규명하기 위하여 하등 진핵생물인 분열형 효모 Schizosaccharomyces pombe로부터 subtraction hybridization방법을 이용하여 자외선 유도 유전자인 UV150과 UV200을 분리하고 그 유전자 구조와 발현양상을 조사하였다. 분리한 유전자의 발현양상을 Northern hybridization 방법으로 살펴본 결과 자외선 조사 1시간 후부터 발현이 증가되었다. 또한 알킬화제인 Methyl Methanesulfonate (MMS) 처리에 의해서도 발현이 증가되었다. 이 결과 다른 UV-inducible유전자와는 다르게 분리한 UV150유전자는 UV에 UV200유전자는 MMS에 의하여 발현이 증가됨을 알 수 있었다. 유전자의 기능을 알기 위하여 URA4 유전자를 이용하여 null-mutant 세포주를 제조하여 그 특성을 살펴본 결과 분리한 UV150 유전자는 세포의 성장에 필수적인 유전자임을 알 수 있었다.
