Facile synthesis of nanostructured n-type SiGe alloys with enhanced thermoelectric performance using rapid solidification employing melt spinning followed by spark plasma sintering

  • SiGe alloy is widely used thermoelectric materials for high temperature thermoelectric generator applications. However, its high thermoelectric performance has been thus far realized only in alloys synthesized employing mechanical alloying techniques, which are time-consuming and employ several materials processing steps. In the current study, for the first time, we report an enhanced thermoelectric figure-of-merit (ZT) ~ 1.1 at $900^{\circ}C$ in ntype $Si_{80}Ge_{20}$ nano-alloys, synthesized using a facile and up-scalable methodology consisting of rapid solidification at high optimized cooling rate ${\sim}3.4{\times}10^7K/s$, employing melt spinning followed by spark plasma sintering of the resulting nano-crystalline melt-spun ribbons. This enhancement in ZT > 20% over its bulk counterpart, owes its origin to the nano-crystalline microstructure formed at high cooling rates, which results in crystallite size ~7 nm leading to high density of grain boundaries, which scatter heat-carrying phonons. This abundant scattering resulted in a very low thermal conductivity ${\sim}2.1Wm^{-1}K^{-1}$, which corresponds to ~50% reduction over its bulk counterpart and is amongst the lowest reported thus far in n-type SiGe alloys. The synthesized samples were characterized using X-ray diffraction, scanning electron microscopy and transmission electron microscopy, based on which the enhancement in their thermoelectric performance has been discussed.

The Danger-Associated Peptide PEP1 Directs Cellular Reprogramming in the Arabidopsis Root Vascular System

  • When perceiving microbe-associated molecular patterns (MAMPs) or plant-derived damage-associated molecular patterns (DAMPs), plants alter their root growth and development by displaying a reduction in the root length and the formation of root hairs and lateral roots. The exogenous application of a MAMP peptide, flg22, was shown to affect root growth by suppressing meristem activity. In addition to MAMPs, the DAMP peptide PEP1 suppresses root growth while also promoting root hair formation. However, the question of whether and how these elicitor peptides affect the development of the vascular system in the root has not been explored. The cellular receptors of PEP1, PEPR1 and PEPR2 are highly expressed in the root vascular system, while the receptors of flg22 (FLS2) and elf18 (EFR) are not. Consistent with the expression patterns of PEP1 receptors, we found that exogenously applied PEP1 has a strong impact on the division of stele cells, leading to a reduction of these cells. We also observed the alteration in the number and organization of cells that differentiate into xylem vessels. These PEP1-mediated developmental changes appear to be linked to the blockage of symplastic connections triggered by PEP1. PEP1 dramatically disrupts the symplastic movement of free green fluorescence protein (GFP) from phloem sieve elements to neighboring cells in the root meristem, leading to the deposition of a high level of callose between cells. Taken together, our first survey of PEP1-mediated vascular tissue development provides new insights into the PEP1 function as a regulator of cellular reprogramming in the Arabidopsis root vascular system.

High-performance computing for SARS-CoV-2 RNAs clustering: a data science-based genomics approach

  • Nowadays, Genomic data constitutes one of the fastest growing datasets in the world. As of 2025, it is supposed to become the fourth largest source of Big Data, and thus mandating adequate high-performance computing (HPC) platform for processing. With the latest unprecedented and unpredictable mutations in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the research community is in crucial need for ICT tools to process SARS-CoV-2 RNA data, e.g., by classifying it (i.e., clustering) and thus assisting in tracking virus mutations and predict future ones. In this paper, we are presenting an HPC-based SARS-CoV-2 RNAs clustering tool. We are adopting a data science approach, from data collection, through analysis, to visualization. In the analysis step, we present how our clustering approach leverages on HPC and the longest common subsequence (LCS) algorithm. The approach uses the Hadoop MapReduce programming paradigm and adapts the LCS algorithm in order to efficiently compute the length of the LCS for each pair of SARS-CoV-2 RNA sequences. The latter are extracted from the U.S. National Center for Biotechnology Information (NCBI) Virus repository. The computed LCS lengths are used to measure the dissimilarities between RNA sequences in order to work out existing clusters. In addition to that, we present a comparative study of the LCS algorithm performance based on variable workloads and different numbers of Hadoop worker nodes.

Separation of cadmium and chromium heavy metals from industrial wastewater by using Ni-Zn nanoferrites

  • The potentials of NixZn1-xFe2O4 (x = 0.0, 0.2, 0.4, 0.6, 0.8 and 1.0) nanoadsorbents were investigated for removal of Cd and Cr from contaminated water from an electroplating industry in Himachal Pradesh, India. Optimal values were recorded under batch adsorption experiments performed to remove dissolved heavy metal ions from industrial wastewater. The specific surface area (SSA) of nanoadsorbents perceived to vary in a range 35.75-45.29 cm2/g and was calculated from the XRD data. The influence of two operating parameters, contact time and dopant (Ni) concentration was also investigated at pH ~7 with optimum dosage. Kinetic studies were conducted within a time range of 2-10 min with rapid adsorption of cadmium and chromium ions onto Ni0.2Zn0.8Fe2O4 nanoadsorbents. Pseudo-second-order kinetic model was observed to be well fitted with the adsorption data that confirmed the only existence of chemisorption throughout the adsorption process. The maximum adsorption efficiency values observed for Cd and Cr were 51.4 mg/g and 40.12 mg/g, respectively for different compositions of prepared series of nanoadsorbents. The removal percentage of Cd and Cr was found to vary in a range of 47.7%-95.25% and 21%-50% respectively. The prepared series of nanoferrite found to be suitable enough for adsorption of both heavy metal ions.

Influence of the CYP1A1 T3801C Polymorphism on Tobacco and Alcohol-Associated Head and Neck Cancer Susceptibility in Northeast India

  • Background: Tobacco and alcohol contain or may generate carcinogenic compounds related to cancers. CYP1A1 enzymes act upon these carcinogens before elimination from the body. The aim of this study was to investigate whether CYP1A1 T3801C polymorphism modulates the relationship between tobacco and alcohol-associated head and neck cancer (HNC) susceptibility among the northeast Indian population. Materials and Methods: One hundred and seventy histologically confirmed HNC cases and 230 controls were included within the study. The CYP1A1 T3801C polymorphism was determined using PCR-RFLP, and the results were confirmed by DNA sequencing. Logistic regression (LR) and multifactor dimensionality reduction (MDR) approaches were applied for statistical analysis. Results: The CYP1A1 CC genotype was significantly associated with HNC risk (P=0.045). A significantly increased risk of HNC (OR=6.09; P<0.0001) was observed in individuals with combined habits of smoking, alcohol drinking and tobacco-betel quid chewing. Further, gene-environment interactions revealed enhanced risks of HNC among smokers, alcohol drinkers and tobacco-betel quid chewers carrying CYP1A1 TC or CC genotypes. The highest risk of HNC was observed among smokers (OR=7.55; P=0.009) and chewers (OR=10.8; P<0.0001) carrying the CYP1A1 CC genotype. In MDR analysis, the best model for HNC risk was the three-factor model combination of smoking, tobacco-betel quid chewing and the CYP1A1 variant genotype (CVC=99/100; TBA=0.605; P<0.0001); whereas interaction entropy graphs showed synergistic interaction between tobacco habits and CYP1A1. Conclusions: Our results confirm that the CYP1A1 T3801C polymorphism modifies the risk of HNC and further demonstrated importance of gene-environment interaction.

Baleen Whale Sound Synthesis using a Modified Spectral Modeling (수정된 스펙트럴 모델링을 이용한 수염고래 소리 합성)

  • Spectral modeling synthesis (SMS) has been used as a powerful tool for musical sound modeling. This technique considers a sound as a combination of a deterministic plus a stochastic component. The deterministic component is represented by the series of sinusoids that are described by amplitude, frequency, and phase functions and the stochastic component is represented by a series of magnitude spectrum envelopes that functions as a time varying filter excited by white noise. These representations make it possible for a synthesized sound to attain all the perceptual characteristics of the original sound. However, sometimes considerable phase variations occur in the deterministic component by using the conventional SMS for the complex sound such as whale sounds when the partial frequencies in successive frames differ. This is because it utilizes the calculated phase to synthesize deterministic component of the sound. As a result, it does not provide a good spectrum matching between original and synthesized spectrum in higher frequency region. To overcome this problem, we propose a modified SMS that provides good spectrum matching of original and synthesized sound by calculating complex residual spectrum in frequency domain and utilizing original phase information to synthesize the deterministic component of the sound. Analysis and simulation results for synthesizing whale sounds suggest that the proposed method is comparable to the conventional SMS in both time and frequency domain. However, the proposed method outperforms the SMS in better spectrum matching.

Effects of Different UV-B Levels on Growth, Antioxidant Contents and Activities of Related Enzymes in Cucumber(Cucumis sativus L.). (UV-B 강도 변화가 오이의 생장 및 항산화 물질 함량과 관련 효소의 활성에 미치는 영향)

  • To investigate the effects of different UV-B levels on growth and biochemical defense response in plants, cucumber plants were subjected to three levels of biologically effective ultraviolet-B $(UV-B_{BE})$ radiation [daily dose: 0.03 (No), 6.40 (Low) and $11.30\;(High)\;kJ{\cdot}m^{-2}$, $UV-B_{BE}$] in the growth chambers for 3 weeks during the early growth period. Enhanced UV-B radiation drastically decreased both dry weight and leaf area of cucumber. With increasing UV-B intensity, chlorophyll content was decreased, however the level of malondialdehyde was highly increased linearly. Total contents of ascorbic acid and glutathione were tended to increase by UV-B, while the ratios of dehydroascorbate/ascorbate and oxidized glutathione/reduced glutathione were significantly increased with increasing UV-B intensity in cucumber. All the enzyme activities investigated (superoxide dismutase, ascorbate peroxidase, dehydroascorbate reductase, guaiacol peroxidase etc.) in cucumber were increased by the UV-B enhancement. These results suggested that enhanced UV-B irradiation caused photooxidative stress in cucumber plant and resulted in significant reduction in plant growth. Biochemical protection responses might be activated to prevent the leaves from damaging effects of oxidative stress generated by UV-B irradiation.

Manual Liquid Based Cytology in Primary Screening for Cervical Cancer - a Cost Effective Preposition for Scarce Resource Settings

  • Conventional pap smear (CPS) examination has been the mainstay for early detection of cervical cancer. However, its widespread use has not been possible due to the inherent limitations, like presence of obscuring blood and inflammation, reducing its sensitivity considerably. Automated methods in use in developed countries may not be affordable in the developing countries due to paucity of resources. On the other hand, manual liquid based cytology (MLBC) is a technique that is cost effective and improves detection of precursor lesions and specimen adequacy. Therefore the aim of the study was to compare the utility of MLBC with that of CPS in cervical cancer screening. A prospective study of 100 cases through MLBC and CPS was conducted from October 2009 to July 2010, in a Medical College in India, by two independent pathologists and correlated with histopathology (22 cases). Morphological features as seen through MLBC and CPS were compared. Subsequently, all the cases were grouped based on cytological diagnosis according to two methods into 10 groups and a subjective comparison was made. In order to compare the validity of MLBC with CPS in case of major diagnoses, sensitivity and specificity of the two methods were estimated considering histological examination as the gold standard. Increased detection rate with MLBC was 150%. The concordance rate by LBC/histopathology v/s CPS/histopathology was also improved (86% vs 77%) The percentage agreement by the two methods was 68%. MLBC was more sensitive in diagnosis of LSIL and more specific in the diagnosis of inflammation. Thus, MLBC was found to be better than CPS in diagnosis of precursor lesions. It provided better morphology with increased detection of abnormalities and preservation of specimen for cell block and ancillary studies like immunocytochemistry and HPV detection. Therefore, it can be used as alternative strategy for cervical cancer prevention in limited resource settings.

Response of Antioxidative Enzymes of Two Rice Cultivars to Ozone Exposure and Nutrient Supply

  • Ozone ($O_3$)-induced changes in chlorophyll content and specific activities of antioxidant enzymes, such as superoxide dismutase (SOD), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) were investigated in two rice cultivars (Oryza sativa L.) grown under variable nutrient treatments. For this study, two rice cultivars of Ilpumbyeo (IL) and Keumobyeo#l (KM), which were known as resistant and susceptible to $O_3$, respectively, were exposed to $O_3$at 0.15ppm for 30 days and investigated with 10 days interval. The available nutrient regimes were varied by doubling the supply of nitrogen (N), phosphorus (P) and potassium (K) Within a basic fertilizer status (N, P, K; 15, 12, 12kg/l0a$^{-1}$ ). In both cultivars and at all nutrient status, chlorophyll content in $O_3$-treated plants decreased with prolonged treatment period, although higher N, P and K supply with $O_3$ treatment alleviated the decrease in chlorophyll content. The activities of almost all enzymes investigated for this study were decreased during initial stages of $O_3$- exposure except GPX which maintained higher activity throughout the exposure period than the non-treated plant. However, the antioxidant enzymes in $O_3$-treated plants showed almost the same or higher activities on 30 days after $O_3$ - exposure. The most significant changes in activities were observed in GR of the $O_3$-treated leaves. With the prolonged treatment period, the activity of GR at 30 days was increased by 3-8 times compared to those in 10 days. Most of the investigated enzymes showed very similar tendency to $O_3$ treatment in all fertilizer status. There was no observed evidence for enhanced detoxification of $O_3$-derived activated oxygen species in plants grown under higher fertilizer status compared with that in plants grown under basic fertilizer status. The increase in the activities of SOD, APX and GR in rice leaves by relatively long-term treatment with $O_3$ at low concentration is considered to indicate that the plant became adapted to the $O_3$ stress and the protection system increased its capacity to scavenge toxic oxygen species. Our results in two rice cultivars indicated that there was little difference in the activities of antioxidant enzymes between IL and KM, which were known as resistant and susceptible cultivar to $O_3$

Metabolic engineering of Vit C: Biofortification of potato

  • Vitamin C (ascorbic acid) is an essential component for collagen biosynthesis and also for the proper functioning of the cardiovascular system in humans. Unlike most of the animals, humans lack the ability to synthesize ascorbic acid on their own due to a mutation in the gene encoding the last enzyme of ascorbate biosynthesis. As a result, vitamin C must be obtained from dietary sources like plants. In this study, we have developed two different kinds of transgenic potato plants (Solanumtuberosum L. cv. Taedong Valley) overexpressing strawberry GalUR and mouse GLoase gene under the control of CaMV 35S promoter with increased ascorbic acid levels. Integration of the these genes in the plant genome was confirmed by PCR and Southern blotting. Ascorbic acid(AsA) levels in transgenic tubers were determined by high-performance liquid chromatography(HPLC). The over-expression of these genes resulted in 2-4 folds increase in AsA intransgenic potato and the levels of AsA were positively correlated with increased geneactivity. The transgenic lines with enhanced vitamin C content showed enhanced tolerance to abiotic stresses induced by methyl viologen(MV), NaCl or mannitol as compared to untransformed control plants. The leaf disc senescence assay showed better tolerance in transgenic lines by retaining higher chlorophyll as compared to the untransformed control plants. Present study demonstrated that the over-expression of these gene enhanced the level of AsA in potato tubers and these transgenics performed better under different abiotic stresses as compared to untransformed control. We have also investigated the mechanism of the abiotic stress tolerance upon enhancing the level of the ascorbate in transgenic potato. The transgenic potato plants overexpressing GalUR gene with enhanced accumulation of ascorbate were investigated to analyze the antioxidants activity of enzymes involved in the ascorbate-glutathione cycle and their tolerance mechanism against different abiotic stresses under invitro conditions. Transformed potato tubers subjected to various abiotic stresses induced by methyl viologen, sodium chloride and zinc chloride showed significant increase in the activities of superoxide dismutase(SOD, EC 1.15.1.1), catalase, enzymes of ascorbate-glutathione cycle enzymes such as ascorbate peroxidase(APX, EC 1.11.1.11), dehydroascorbate reductase(DHAR, EC 1.8.5.1), and glutathione reductase(GR, EC 1.8.1.7) as well as the levels of ascorbate, GSH and proline when compared to the untransformed tubers. The increased enzyme activities correlated with their mRNA transcript accumulation in the stressed transgenic tubers. Pronounced differences in redox status were also observed in stressed transgenic potato tubers that showed more tolerance to abiotic stresses when compared to untransformed tubers. From the present study, it is evident that improved to lerance against abiotic stresses in transgenic tubers is due to the increased activity of enzymes involved in the antioxidant system together with enhanced ascorbate accumulated in transformed tubers when compared to untransformed tubers. At moment we also investigating the role of enhanced reduced glutathione level for the maintenance of the methylglyoxal level as it is evident that methylglyoxal is a potent cytotoxic compound produced under the abiotic stress and the maintenance of the methylglyoxal level is important to survive the plant under stress conditions.
