The effects of purification using ${\alpha}-amlyase$ (Termamyl 120L) on physicochemical properties of ${\beta}-glucan$ from oat bran were studied. Four fractions were selected as fraction A ($55^{\circ}C$, 15%, pH 6), fraction B ($45^{\circ}C$, 15%, pH 6), fraction C ($50^{\circ}C$, 0%, pH 7), and fraction D ($50^{\circ}C$, 10%, pH 5) from the result of physiological test, and three consecutive subfractions were obtained by repeated ${\alpha}-amlyase$ treatments on the each fractions. The contents of ${\beta}-glucan$, protein, and ash after purification were in 81.4-88.2%, 4.1-6.3% and 2.6-6.2%, respectively. The apparent viscosities of purified ${\beta}-glucan$ aqueous solutions were similar to those of hydroxy methyl cellulose. Glucose was a major monosaccharide of ${\beta}-glucan$ extracts, and xylose and arabinose were also detected as minor constituents on TLC. The average molecular weight ranged $2.0{\times}10^6-5.1{\times}10^6$ and was decreased after purification. From the result of the differential scanning calorimetry, the melting point ranged $130-140^{\circ}C$ with purification step and thermal transition enthalpy was increased. The ratio of ${\beta}-(1{\rightarrow}3)\;to\;{\beta}-(1{\rightarrow}4) $ linkages were 1:2.22-1:2.52, and increased up to 1:5.50 after purification.

Three-dimensional registration is a process of matching data with or without a coordinate system to a reference coordinate system, which is used in various fields such as the absolute orientation of photogrammetry and data combining for producing precise road maps. Three-dimensional registration is divided into a method using points and a method using linear features. In the case of using points, it is difficult to find the same conjugate point when having different spatial resolutions. On the other hand, the use of linear feature has the advantage that the three-dimensional registration is possible by using not only the case where the spatial resolution is different but also the conjugate linear feature that is not the same starting point and ending point in point cloud type data. In this study, we proposed a method to determine the scale and the three-dimensional translation after determining the three-dimensional rotation angle between two data using quaternion to perform three-dimensional registration using linear features. For the verification of the proposed method, three-dimensional registration was performed using the linear features constructed an indoor and the linear features acquired through the terrestrial mobile mapping system in an outdoor environment. The experimental results showed that the mean square root error was 0.001054m and 0.000936m, respectively, when the scale was fixed and if not fixed, using indoor data. The results of the three-dimensional transformation in the 500m section using outdoor data showed that the mean square root error was 0.09412m when the six linear features were used, and the accuracy for producing precision maps was satisfied. In addition, in the experiment where the number of linear features was changed, it was found that nine linear features were sufficient for high-precision 3D transformation through almost no change in the root mean square error even when nine linear features or more linear features were used.

Forage grasses and legumes ar$\varepsilon$ the mam component of livestock diets in Canada. There are over 30 million ha of grassland in Canada and there is a large, undeveloped land base in fringe areas suitable for forage production. The short growing s season limits the grassland farming to the southern p parts of Canada. The win!er season is long and in most parts of Canada cold temperatures, fr$\varepsilon$ezmg, and thawing, and diseases exert sever$\varepsilon$ stress on overwintering forage plants. The development of persistent cultivars is essential for sustained production particularly in the fringe areas with short growmg s$\varepsilon$ason. The seasonality of dry matter production is a result of high growth rates in early summ$\varepsilon$r and low dry matter accumulation in late summer and fall. Innovative management practIces a and cultivars with improved regrowth capacity are n necessary to overcome such skewed production pattern and to extend effiectlVe grazmg season l Improved pasture production is an important part of reducing costs in livestock operations and remaining competitive. It is suggested that applying available technology would increase pasture productivity and reduce d$\varepsilon$pendence on stored feeds thus improving profitability of small producers in particeular. Reducing nutrient losses during harv$\varepsilon$stmg, s storage, and feeding is essential for improved production efficiency during confinement. The devclopment of low cost and labor saving methods of ensiling is critical for improved efficiency and profitability of forage based enterprises Livestock industries must respond to consumer preferences for low fat and cholesterol foods. Research and development of entire production systems is emphasized for dev$\varepsilon$loping viabl$\varepsilon$ enterprises. It is increasingly difficult to secure resources for r$\varepsilon$search, education, and extension, and alliane$\varepsilon$s and cooperation must expand among organizations with interests in forage based livestock systems.
This study was conducted to evaluate the bactericidal activity of weakly alkalic electrolyzed water (WEW) against Salmonella Typhimurium and Staphylococcus aureus on perilla leaves. The influences of organic matter, inform of bovine serum, and the ratio of WEW to perilla leaves on bactericidal activity of WEW were also examined. Treatment of these organisms with 25, 50, 75, and 100 ppm WEW was performed for 1 min, 3 min and 5 min, respectively. Higher bactericidal activity was observed after a treatment with 100 ppm WEW compared to a treatment with 25 ppm WEW by 0.7 $log_{10}CFU/g$. The bactericidal activity of WEW also decreased with increasing bovine serum concentration. At the ratio of 10:1 (WEW: perilla leave), levels of Salmonella Typhimurium and Staphylococcus aureus were only reduced by 0.57 and 0.79 $log_{10}CFU/g$, respectively. It is suggested that the removal of organic debris prior to application of sanitizers and treatment above the ratio of 25:1 (WEW: perilla leave) is needed in order to improve WEW activity.
The apoptogenic effect of p-coumaric acid, a phenolic acid found in various edible plants, on human acute leukemia Jurkat T cells was investigated. Exposure of Jurkat T cells to p-coumaric acid (50-$150{\mu}M$) caused cytotoxicity and TdT-mediated dUTP nick-end labeling (TUNEL)-positive apoptotic DNA fragmentation along with Bak activation, ${\Delta}{\psi}m$ loss, activation of caspase-9, -3, -7, and -8, and PARP degradation in a dose-dependent manner. However,these apoptotic events were completely abrogated in Jurkat T cells overexpressing Bcl-2.Under these conditions, necrosis was not accompanied. Pretreatment of the cells with the pan-caspase inhibitor (z-VAD-fmk) could prevent p-coumaric acid-induced sub-$G_1$ peak representing apoptotic cells, whereas it failed to block ${\Delta}{\psi}m$ loss, indicating that the activation of caspase cascade was prerequisite for p-coumaric acid-induced apoptosis as a downstream event of ${\Delta}{\psi}m$ loss. FADD- and caspase-8-positive wild-type Jurkat T cell clone A3, FADD-deficient Jurkat T cell clone I2.1, and caspase-8-deficient Jurkat T cell clone I9.2 exhibited similar susceptibilities to the cytotoxicity of p-coumaric acid, excluding an involvement of Fas/FasL system in triggering the apoptosis. The apoptogenic activity of p-coumaric acid is more potent in malignant Jurkat T cells than in normal human peripheral T cells. Together, these results demonstrated that p-coumaric acid-induced apoptogenic activity in Jurkat T cellswas mediated by Bak activation, ${\Delta}{\psi}m$ loss, and subsequent activation of multiple caspases such as caspase-9, -3, -7, and-8, and PARP degradation, which could be regulated by anti-apoptotic protein Bcl-2.
Polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dl-PCBs) are bioaccumulative chemicals that are considered to be toxic contaminants based on several epidemiological studies. These chemicals in colostrum were investigated and estimated for their residual consistencies by maternal characteristics like parity and maternal ages. Test subjects were healthy primipara and multipara mothers with a mean age of 31.5 (S.D=3.6) in 2007. Seven isomers of PCDDs, 10 of PCDFs, 4 of non-orthopolychlorinated biphenyls(non-ortho PCBs) and 8 of mono-orthochlorinated polychlorinated biphenyls (mono-ortho PCBs) were analyzed by HRGC/HRMS. From the analyzed data, the mean level of total WHO-TEQs was 9.41 pg TEQ/g lipid, which is significantly lower than the level found in individuals from other countries. The main contributors to the total WHO-TEQs with increasing percentages were 2,3,4,7,8-PeCDF, 1,2,3,7,8-PeCDD and 3,3',4,4',5-PCB (#126), and they accounted for more than 60% of the total WHO-TEQs. PCDFs concentrations and total WHO-TEQs were negatively associated with parity (p<0.05), and maternal age was positively associated with total WHO-TEQs (p<0.01). However, the associations with body mass index (BMI) and fish intake during pregnancy were not significant. These results were suggested that parity and maternal age are an important factor affecting the concentrations of PCDD/DFs and dl-PCBs in these specimens.
SH21B is a natural composition composed of seven herbs: Scutellaria baicalensis Georgi, Prunus armeniaca Maxim, Ephedra sinica Stapf, Acorus gramineus Soland, Typha orientalis Presl, Polygala tenuifolia Willd and Nelumbo nucifera Gaertner (Ratio 3:3:3:3:3:2:2). In our previous study, we reported that SH21B inhibited adipogenesis and fat accumulation in 3T3-L1 cells through modulation of various regulators in the adipogenesis pathway. The aim of this study was to analyze the transcriptome profiles for the anti-adipogenic effects of SH21B in 3T3-L1 cells. Total RNAs from SH21B-treated 3T3-L1 cells were reverse-transcribed into cDNAs and hybridized to Affymetrix Mouse Gene 1.0 ST array. From microarray analyses, we identified 2,568 genes of which expressions were changed more than two-fold by SH21B, and the clustering analyses of these genes resulted in 9 clusters. Three clusters among the 9 showed down-regulation by SH21B (cluster 4, cluster 6 and cluster 9), and two clusters showed up-regulation by SH21B (cluster 7 and cluster 8) during the adipogenesis of 3T3-L1 cells. It was found that many genes related to cell proliferation and adipogenesis were included in these clusters. Clusters 4, 6 and 9 included genes which were related with adipogenesis induction and cell cycle arrest. Clusters 7 and 8 included genes related to cell proliferation as well as adipogenesis inhibition. These results suggest that the mechanisms of the anti-adipogenic effects of SH21B may be the modulation of genes involved in cell proliferation and adipogenesis.
The purpose of this study is analyzing 'observation and recommendation letter by teacher', which is being submitted to screen and enhance the utilization of gifted students in accordance with recently introduced gifted students observation, recommendation and screening system. For the purpose, this study will provide with objective securing plan of 'observation and recommendation letter by teacher' by developing an optimum evaluation model. The research findings were as follows: First, the result of analysis on the mathematically gifted students behavior characteristic as appeared in 'observation and recommendation letter by teacher' suggested that the recommending teachers have the tendency of giving superficial statement instead of giving concrete case description. When it was analyzed for frequency by the 'observation and recommendation letter by teacher' analysis framework devised by the author, the teachers showed the tendency of concentrating on specific questions. Meanwhile, there was a tendency that teachers concentrate on specific gifted behavior characteristic or area for which concrete case had been suggested. The reason is believed that such part is easy to observe and state while others are not, or, teachers did not judge the other part as the characteristic of gifted students. Second, the gifted students behavior characteristics as appeared in 'observation and recommendation letter by teacher' were made into scores by Rubric model. When the interrater reliability was analyzed based on these scores, the correlation coefficient of 1st scoring was .641. After a discussion session was taken and 2nd scoring was done 3 weeks later, the correlation coefficient of 2nd scoring increased to .732. The reason is believed that; i) the severity among scorers was adjusted by the discussion session after the 1st scoring, ii) the scorers established detail judgment standard on various situations which can appear because of the descriptive nature, and, (iii) they found a consensus on scoring for a new situation appeared. It implies that thorough understanding and application of scorers on evaluation model is as important as the development of optimum model for the differentiation of mathematically gifted elementary students.

The effects of elevated atmospheric $CO_2$ on photosynthesis and growth of Chinese cabbage (Brassica campestris subsp. napus var. pekinensis) were investigated to predict productivity in highland cropping in an environment where $CO_2$ levels are increasing. Vegetative growth, based on fresh weight of the aerial part, and leaf characteristics (number, area, length, and width) of Chinese cabbage grown for 5 weeks, increased significantly under elevated $CO_2$ ($800{\mu}mol{\cdot}mol^{-1}$) compared to ambient $CO_2$ ($400{\mu}mol{\cdot}mol^{-1}$). The photosynthetic rate (A), stomatal conductance ($g_s$), and water use efficiency (WUE) increased, although the transpiration rate (E) decreased, under elevated atmospheric $CO_2$. The photosynthetic light-response parameters, the maximum photosynthetic rate ($A_{max}$) and apparent quantum yield (${\varphi}$), were higher at elevated $CO_2$ than at ambient $CO_2$, while the light compensation point ($Q_{comp}$) was lower at elevated $CO_2$. In particular, the maximum photosynthetic rate ($A_{max}$) was higher at elevated $CO_2$ by 2.2-fold than at ambient $CO_2$. However, the photosynthetic $CO_2$-response parameters such as light respiration rate ($R_p$), maximum Rubisco carboxylation efficiency ($V_{cmax}$), and $CO_2$ compensation point (CCP) were less responsive to elevated $CO_2$ relative to the light-response parameters. The photochemical efficiency parameters ($F_v/F_m$, $F_v/F_o$) of PSII were not significantly affected by elevated $CO_2$, suggesting that elevated atmospheric $CO_2$ will not reduce the photosynthetic efficiency of Chinese cabbage in highland cropping. The optimal temperature for photosynthesis shifted significantly by about $2^{\circ}C$ under elevated $CO_2$. Above the optimal temperature, the photosynthetic rate (A) decreased and the dark respiration rate ($R_d$) increased as the temperature increased. These findings indicate that future increases in $CO_2$ will favor the growth of Chinese cabbage on highland cropping, and its productivity will increase due to the increase in photosynthetic affinity for light rather than $CO_2$.

Rotavirus is a major cause of acute gastroenteritis in young children in developed and developing countries. The use of probiotics for the treatment of gastrointestinal diseases is both safe and easily accessible. In this study, we evaluated the anti-rotaviral activities of probiotic mixtures in a Sprague-Dawley rat. 24 litters with their dams were randomly assigned to four groups; placebo, phosphate buffered saline (PBS), and two probiotic mixture (PRO-1 and PRO-2) groups. All rats were inoculated with rotavirus at dose of 8 log plaque forming units per rat at 5 days old. Animals in the PRO-1 and PRO-2 groups were orally administered probiotic mixtures 1 or 2, respectively, at a dose of 8 log colony forming units daily during 4 days. For control purposes, placebo and PBS groups were orally administered the same amount of placebo (containing maltose and polydextrose) or PBS once daily for 4 days, respectively. Antiviral analysis was performed by real-time quantitative PCR (RT-qPCR) and observing intestinal villi. As a result, weights of small intestines were greater in the PRO-1, PRO-2 groups than in control groups. Villi were short and villous epithelial necrosis was exhibited in control groups, but these morphological changes were not observed in PRO-1, PRO-2 treated rats. RT-qPCR analysis showed that VP7 gene level of rotavirus in fecal samples and small intestinal epithelial cells were lower in the PRO-1 and PRO-2 groups. These findings suggest that probiotic mixtures may be useful probiotics for the treatment of or as alternative therapies for rotaviral gastroenteritis.
