Antibacterial Activity of Zein Hydrolysate with Pepsin (Pepsin에 의한 Zein 가수분해물의 항균활성)

  • A study was carried out to produce antimicrobial peptides from zein treated with pretenses of six kinds. Among the pretenses of six kinds, zein hydrolysate treated with pepsin showed the highest antimicrobial activity. The zein hydrolysate with pepsin was fractionated with membrane filter (30,000 10,000 and 3,000 molecular weight cut-off) and antimicrobial activity was measured for each fractions. Antimicrobial activity appeared greatly in the fraction below 3,000 (molecular weight cut-off) . The fraction was re-fractionated by HPLC and substances of two peaks collected as a sample to measure antimicrobial activity. All of both peaks showed the antimicrobial activity but 1st peak exhibited a consistently higher antimicrobial activity than 2nd peak. Minimum inhibitory concentrations (MIC) were between 2.5 and 3.0 mg/mL. The peptide was heat-stable since antimicrobial activity was maintained after treated with heat for 20 min at $121^{\circ}C$. N-terminal amino acid sequence of peptide fractionated by HPLC was leucine, glutamic acid, proline, phenylalanine, aspartic acid and argenine. These results indicated that peptide isolated from zein hydrosate with pepsin can use as a natural preservative ingredient in food industry.

Investigation of Fungi in Pesticide Fumigated Oak Wilt-Diseased Logs (훈증방제 처리한 참나무시들음병 감염목의 사상균 조사)

  • Korean oak wilt disease caused by Raffaelea quercus-mongolicae is vectored by the ambrosia beetle Platypus koryoensis. To prevent the spread of the disease, the beetle infested oak tree had been cut into logs, covered with plastic vinyl, fumigated with a pesticide, and stored for three years on the site where the tree was cut. This study was carried out to get information on the fungi colonizing the fumigated oak wood. Wood disk samples collected from the fumigated oak logs at two locations in the Taejo Mountain, Cheonan city, were used for fungal isolation. A total of 99 filamentous fungal isolates were obtained from the wood disk samples. Hypocrea spp., Trichoderma spp. and Penicillium spp. were identified based on morphological characteristics and nucleotide sequence analysis of translation elongation factor 1-alpha gene and ITS rDNA region. Trichoderma was the major fungal group. R. quercus-mongolicae, and P. koryoensis were not detected from the fumigated oak wood. Our work provided evidence that after three years of storage, the fumigated oak wilt-diseased logs should be no longer harmful source of oak wilt disease transmission.

Fast Scene Change Detection Using Macro Block Information and Spatio-temporal Histogram (매크로 블록 정보와 시공간 히스토그램을 이용한 빠른 장면전환검출)

  • Most of the previous works on scene change detection algorithm focus on the detection of abrupt rather than gradual changes. In general, gradual scene change detection algorithms require heavy computation. Some of those approaches don't consider the error factors such as flashlights, camera or object movements, and special effects. Many scenes change detection algorithms based on the histogram show better performances than other approaches, but they have computation load problem. In this paper, we proposed a scene change detection algorithm with fast and accurate performance using the vertical and horizontal blocked slice images and their macro block informations. We apply graph cut partitioning algorithm for clustering and partitioning of video sequence using generated spatio-temporal histogram. When making spatio-temporal histogram, we only use the central block on vertical and horizontal direction for performance improvement. To detect camera and object movement as well as various special effects accurately, we utilize the motion vector and type information of the macro block.

Occurrence of the Phytophthora Blight Caused by Phytophthora sansomeana in Atractylodes macrocephala Koidz. (Phytophthora sansomeana에 의한 큰꽃삽주 역병 발생 보고)

  • Background: In September 2017, wilting and rhizome rot symptoms were observed on Atractylodes macrocephala Koidz. in Jecheon-si and Eumseong-gun. This study was carried out to isolate hitherto unidentified pathogenic fungi from A. macrocephala and to test the pathogenicity of isolated fungi against Atractylodes spp. genus such as A. macrocephala, A. japonica, and their interspecific hybrids. Methods and Results: The diseased plants were washed with running tap water, and the boundary between the healthy area and the diseased area was cut while the pathogens were isolated by growing cultures from the diseased areas on Phytophthora semi-selective medium. The internal transcribed spacer (ITS) region of the isolates was used in this study for identification. Test plants were cultivated in the glasshouse at 20℃ - 30℃ for 4 months and then used for pathogenicity test. The pots with plants inoculated with mycelial plugs and zoospores were placed at 25℃ for 48 h in a dew chamber where relative humidity was above 95%, and then moved into the glasshouse at 20℃ - 30℃. The presence or absence of pathogenicity of the strains was determined by evaluating the symptom of plant wilting. The inoculation test was performed in three replicates with a non-treated control. Conclusions: On the basis of results of ITS sequence analysis, the strains isolated from the diseased plants was identified as Phytophthora sansomeana. Biological assay using test plants confirmed the pathogenicity of P. sansomeana against Atractylodes macrocephala. This is the first report of rhizome rot in A. macrocephala caused by P. sansomeana.

Purification and Characterization Sucrose phosohorylase in Leuconostoc mesenteroides NRRL B-1149 (Leuconostoc mesenteroides NRRL B-1149의 Sucrose phosohorylase의 분리와 특성 연구)

  • Leuconostoc mesenteroides NRRL B-1149 produces various glucoseyltransferases for the synthesis of dextran, levan and glucose-1-phosphate using sucrose as a substrate. A sucrose phosphorylase (1149SPase) was purified from L. mesenteroides NRRL B-1149 culture by using hollow fiber filtration (30 kDa cut off), Toyopearl DEAE 650 M column chromatography and following two times of DEAE-Sepharose column chromatographies. The specific activity of the purified 1149SPase was 25.7 (U/mg) with $16\%$ yield. The 1149SPase showed a molecular size of 56 kDa on denatured $10\%$ SDS-PAGE. The N-terminal amino acid sequence of the enzyme was MEIQNKAM. The optimum pH and temperature of this enzyme were 6.2~6.5 and 37^{circ}C, respectively. It had an apparent K_{m} of 6.0 mM and K_{cat} of 1.62/s for sucrose. 1149SPase crystal was formed by hanging drop diffusion technique using 20 mM calcium chloride dihydrate, 100 mM sodium acetate trihydrate pH 4.6 and $30\%$ 2-methyl-2,4-pentanediol as vaporizing and reservation solution. The 1149SPase catalyzes transferring of glucose from isomaltose or sucrose to salicin and salicyl alcohol by disproportionation reaction or acceptor reaction and synthesized two acceptor products, respectively.

Realistic Building Modeling from Sequences of Digital Images

  • With the wide usage of LiDAR data and high-resolution satellite image, 3D modeling of buildings in urban areas has become an important research topic in the photogrammetry and computer vision field for many years. However the previous modeling has its limitations of merely texturing the image to the DSM surface of the study area and does not represent the relief of building surfaces. This study is focused on presenting a system of realistic 3D building modeling from consecutive stereo image sequences using digital camera. Generally when acquiring images through camera, various parameters such as zooming, focus, and attitude are necessary to extract accurate results, which in certain cases, some parameters have to be rectified. It is, however, not always possible or practical to precisely estimate or rectify the information of camera positions or attitudes. In this research, we constructed the collinearity condition of stereo images through extracting the distinctive points from stereo image sequence. In addition, we executed image matching with Graph Cut method, which has a very high accuracy. This system successfully performed the realistic modeling of building with a good visual quality. From the study, we concluded that 3D building modeling of city area could be acquired more realistically.

  • This thesis proposes a design algorithm of an efficient space response compactor for Built-In Self-Testing of VLSI circuits. The proposed design algorithm of space compactors can be applied independently from the structure of Circuit Cnder Test. There are high hardware overhead cost in conventional space response compactors and the fault coverage is reduced by aliasing which maps faulty circuit's response to fault-free one. However, the proposed method designs space response compactors with reduced hardware overheads and does not reduce the fault coverage comparing to conventional method. Also, the proposed method can be extended to general N -input logic gate and design the most efficient space response L'Ompactors according to the characteristies of output sequence from CUT. The prolxlsed design algorithm is implemented by C language on a SUN SPARC Workstation, and some experiment results of the simulation applied to ISCAS'85 benchmark circuits with pseudo random patterns generated bv LFSR( Linear Feedback Shift Register) show the efficiency and validity of the proposed design algorithm.

Fractal Compression using Range Block Coherence (레인지 블록 유사성을 이용한 프랙탈 압축)

  • The fractal image compression is based on the self-similarity that some area in an image is very similar to others. This compression method offers high compression ratio and fast decompression, but it has very long encoding time. To cut-off the encoding time, most researches give a restriction on domain blocks to be compared with a range block or make an effective search sequence of the domain blocks for a range block. However, most of them take much encoding time yet. In this research, we propose an algorithm that greatly reduces the encoding time by considering the coherence between range blocks. This algorithm first classifies all range blocks into some groups using the coherence between range blocks, and then searches corresponding domain blocks only for the key block of each group. If this scheme is joined in a prior work of the other fractal compression algorithm, it will give a great effectiveness to encoding time.

Multiple Linkage Disequilibrium Mapping Methods to Validate Additive Quantitative Trait Loci in Korean Native Cattle (Hanwoo)

  • The efficiency of genome-wide association analysis (GWAS) depends on power of detection for quantitative trait loci (QTL) and precision for QTL mapping. In this study, three different strategies for GWAS were applied to detect QTL for carcass quality traits in the Korean cattle, Hanwoo; a linkage disequilibrium single locus regression method (LDRM), a combined linkage and linkage disequilibrium analysis (LDLA) and a $BayesC{\pi}$ approach. The phenotypes of 486 steers were collected for weaning weight (WWT), yearling weight (YWT), carcass weight (CWT), backfat thickness (BFT), longissimus dorsi muscle area, and marbling score (Marb). Also the genotype data for the steers and their sires were scored with the Illumina bovine 50K single nucleotide polymorphism (SNP) chips. For the two former GWAS methods, threshold values were set at false discovery rate <0.01 on a chromosome-wide level, while a cut-off threshold value was set in the latter model, such that the top five windows, each of which comprised 10 adjacent SNPs, were chosen with significant variation for the phenotype. Four major additive QTL from these three methods had high concordance found in 64.1 to 64.9Mb for Bos taurus autosome (BTA) 7 for WWT, 24.3 to 25.4Mb for BTA14 for CWT, 0.5 to 1.5Mb for BTA6 for BFT and 26.3 to 33.4Mb for BTA29 for BFT. Several candidate genes (i.e. glutamate receptor, ionotropic, ampa 1 [GRIA1], family with sequence similarity 110, member B [FAM110B], and thymocyte selection-associated high mobility group box [TOX]) may be identified close to these QTL. Our result suggests that the use of different linkage disequilibrium mapping approaches can provide more reliable chromosome regions to further pinpoint DNA makers or causative genes in these regions.

Expression and Characterization of Bovine DNA Methyltransferase I

  • In this study, bovine Dnmt1 cDNA was sequenced and detected Dnmt1 mRNA level in bovine tissues by northern blot, methylation pattern of genome by southern blot, specific localization of Dnmt1 in mouse and bovine preimplantation embryos by immunocytostaining and Dnmt1 protein level in ovary and testis by western blot. Bovine Dnmt1 cDNA sequence showed more homology with that of human than mouse and rat. The RNA level of Dnmt1 was 10 times higher expression in placenta than other tissues. This indicates that placenta was hypermethylated compared to others organs. The genomic DNA could not be cut by a specific restriction enzyme (HpaII) in placenta, lung and liver of bovine. It suggests that Dnmt1 in some somatic cells was already methylated. Dnmt1, which has the antibody epitope 1316~1616, was distributed in nucleus and cytoplasm including the stage of pronuclear stage and maturation of oocyte and gradually weaken to blastocyst stage compare to negative. In addition, Dnmt1 was strongly expressed in tetraploid embryo and cloned 8-cell than IVF 8-cell. An aberrant pattern of DNA methylation in cloned embryo may be abnormal development of fetus, embryonic lethality and placenta dysfunction. The somatic specific band (190kDa) was appeared in ovary and testis, but oocyte specific band (175kDa) was not. Further investigations are necessary to understand the complex links between the methyltransferases and the transcriptional activity of genes in the cloned bovine tissues.
