  • This study aims to find the creative management system for the Company-Wide Creativity Development(CWCD). And this study consider managemant culture to find CWCD system because managemant culture have influence on the specific character of the personality and beheavior of the system member. To consider managemant culture, this study attempt to compare Korea managemant culture with America, Japanese managemant culture. Through this study we know that we must throw the authoritarianism, the egoism and try to resurrect Korean independence to find CWCD system in korea companies.

In Vitro Development of Porcine Parthenogenetic Embryos under the Oil-free Culture System

  • Optimization of the preimplantation mammalian embryo culture condition was widely focused on refining medium composition under the name of chemically defined media. However, recent research revealed that the alteration of physical environment can be a crucial factor to a successful embryo development. In this study, under the same embryo density, a novel culture device named oil-free micro tube culture (MTC) system was evaluated using porcine parthenogenetic embryos. The activated oocytes were placed into the 0.2 ml thin-wall flat cap PCR tube and cultured to the blastocyst stage. As a preliminary step, embryo density and culture medium volume were optimized under a standard drop culture system. The optimal embryo density range for in vitro culture was 0.5 embryos per ${\mu}l$ in $20\;{\mu}l$ drop (20.5%) and 1.0 embryos per ${\mu}l$ in $10\;{\mu}l$ drop (20.6%). Based on these results, we compared drop culture system and 'MTC' system in terms of the developmental rate to the blastocyst stage. In $20\;{\mu}l$ medium volume, the 'MTC' system showed similar blastocyst formation rate when compared with drop culture system (20.2% versus 20.5%, respectively) while the 'MTC' system showed lower blastocyst formation rate than drop culture system in $10\;{\mu}l$ one (12.7% versus 20.0%, respectively). Therefore the $20\;{\mu}l$ MTC system may be an alternative incubation system for short-distance embryo transport without carrying the $CO_2$ incubator and this provides novel embryo culture device to clinical veterinary embryologists.

  • This study conducted a fuzzy set qualitative comparative analysis study on culture and arts information portals to ensure practical and universal use and activation of culture and arts information portals. The capabilities and influence of affordances and system literacy according to user experience regarding accessibility to cultural arts information portals are as follows. First, physical affordances in the culture and arts information portal were key conditions for the culture and arts information portal user experience regarding system accessibility, system understandability, system usability, system usability, and system literacy. Second, the functional affordance of the user experience in the cultural arts information portal was relatively low and unimportant, and when the functional affordance was low, the user's system accessibility, system understanding, system usefulness, system usability, and system literacy were increased. Third, if the user's functional affordance and sensory affordance are low in the culture and arts information portal, the system accessibility, system understanding, system usability, and system literacy of the culture and arts information portal users are increased. Fourth, cultural arts information portals must build an innovative system with a systematic approach appropriate for the user base by linking system literacy capabilities with a focus on physical affordances.

Studies on the Air-Liquid Interface Culture as an Experimental Model for Physiology and Pharmacology of Tracheal Epithelial Cells (기관(氣管) 상피세포 생리 및 약리 실험모델로서의 공기-액체 접면 일차배양법 연구)

  • In this study, we intended to get a preliminary data for establishing rat tracheal surface epithelial(RTSE) cell culture system as an experimental model for physiology and pharmacology of tracheal epithelial cells. Primary culture on the membrane support and application of the air-liquid interface system at the level of cell layer were performed. The cell growth rate and mucin production rate were measured according to the days in culture. The results were as follows: this culture system was found to manifest mucocilliary differentiation of rat tracheal epithelial cells, the cells were confluent and the quantity of produced and released mucin was highest on culture day 9, the mucin was mainly released to the apical side and tbe free $^3{H}$-glucosamine which was not incorporated to process of synthesis of mucin was left on the basolateral side. Taken together, we suggest that air-liquid interface culture system can be used as a substitute for immersion culture system and as an experimental model for in vivo mucus-hypersecretory diseases.

Effect of Culture Conditions on Survival of Frozen-Thawed Blastocysts Fertilized In Vitro (소 체외수정란의 배양조건이 동결-융해 배반포의 생존에 미치는 영향)

  • This study was carried out to investigate the effect of co-culture system(bovine oviduct epithelial cells; BOEC) and defined culture system(modified TALP ; mTALP) on the development of IVM-IVF embryos, and survival of in vitro produced blastocysts after freezing and thawing. Occytes from the slaugheterhous ovaries were matured and fertilized using general protocol. The results obtained were as the following: 1. Survival rates of frozen-thawed blastocysts using 10% glycerol as cryoprotectant was higher in day 7 blastocysts than in Day 8 and 9 blastocysts from co-cultrue system, but survival rate of frozen-thawed blastocysts was higher in Day 10 blastocysts than in day 8 and 9 blastocysts from defined culture system. Regardless of their age, survival rate of frozen-thawed blastocysts was significantly higher (p<0.05) in co-culture system than in defined culture system. 2. The cell number of blastocysts was significanlty higher (p<0.05) in Day 7 blasotcysts than in Day 8 and 9 blastocysts from co-cultures, but the cell number of blsstocysts was significantly higher (p<0.05) in Day 10 blastocysts than in Day 8 and 9 blastocysts from defined culture system. Regardless of the culture system, blastocysts with higher cell number showed higher survival rates after freezing and thawing.

  • Wood-culture is new paradigm which is substituted for the cement culture, and the study of wood-culture should keep pace with not only the study of a engineering science but also a sociological study to form a theory system. The purpose of this study is to suggest the method of sociological approach of the wood culture which have not yet tried at home and abroad; in view of structural-functionalism, systemical-functionalism, conflict theory, social-change theory by analyzing cases in Korea. In view of structural-functionalism, social system consists of cognition system, institution system, life system and technology system. These sub-systems fulfill the function of what-should-be, reasonability, reality and development. In view of systemical-functionalism, according to the acknowledgment and spreading proccess of the wood usefulness, wood culture system consists of the individual system, organizational system and the social system. In view of conflict theory, the society which has the wood culture can be classified into three types. The one is society which the traditional wood culture is declining and the cement culture is spreading. The other one is society which the cement culture is declining and the traditional wood culture is spreading. Another is Finally the society which the two type balance with. In view of social-change theory, the society can be classified into three types. The one is society which the wood culture is proceeding to the cement culture. The other is society which the cement culture is proceeding to the wood culture. Another is society which the wood culture changes itself. Finally, from a view of changing level, the society can be classified into three types: the micro-change, the middle-level-change, and the macro-change. It's need to study wood culture systemically in view of engineering and sociological science. And then it will be possible to make 'wood culture theory system'.

Development of the Three-Dimensional Perfusion Culture Technology for the Salivary Ductal Cells (타액선 도관세포의 관류 배양 기술 개발)

  • Background and objectives: Salivary hypofunction is one of the common side effects after radioiodine therapy, and its pathophysiology is salivary ductal stenosis resulting from ductal cell injury. This study aimed to develop the functional culture environment of human parotid gland ductal cells in in vitro three-dimensional perfusion culture system. Materials and Methods: We compared plastic dish culture method and three-dimensional culture system containing Matrigel and nanofiber. Morphogenesis of reconstituted salivary structures was assessed by histomorphometry. Functional characteristics were assessed by immunohistochemistry and reverse transcription polymerase chain reaction (aquaporin 5, CK7, CK18, connexin 43, and p21). In addition, we designed the media perfusion culture system and identified higher rate of cell proliferation and expression of connexin 43 in perfusion system comparing to dish. Results: Human parotid ductal cells were well proliferated with the ductal cell characters under environment with Matrigel. In the presence of Matrigel, aquaporin 5, CK18 and connexin 43 were more expressed than 2D dish and 3D nanofiber setting. In the media perfusion culture system, ductal cells in 3D culture media showed higher cells count and connexin 43 expression compared to 2D dish. Conclusion: This in vitro ductal cell perfusion culture system using Matrigel could be used to study for radioiodine induced sialadenitis model in vivo.

Cultural Implications of Science and Technology (과학기술의 문화적 함의)

  • Though science and technology can understood as a part of cultural system, the relationship of science/technology with culture is often viewed as contradictory or mutually exclusive by many scholars. Therefore, in the first section of the present paper, the nature of science and technology as a cultural form is examined. Here, science is viewed as a form of symbolic culture, while technology is regarded as a form of instrumental culture. In the second section, the contradictory relationship and/or tensions between science-technological system (instrumental culture) and cultural system (symbolic culture) within a society is discussed. In the final section of this paper, 'science culture' is conceptualized as a dynamic social process in which contradictory science sub-cultures of political system(state), economic system(cooperations), civil society (including academic community of scientists and other social organizations and movements), and cultural system are supposed to be coordinated. In conclusion, establishment of 'humanized science culture' and 'democratic science-culture movement' is proposed as an alternative way of resolving contradictory relationship between cultural system and science-technological system in the modern world.

Growth Rate of Entomopathogenic Fungi in Mass Culture System (곤충병원성 진균의 대량 배양체계에서의 성장율)

  • To develope a microbial pesticide for the control of agricultural and forestal pests in Korea, the mass culture system of entomopathogenic fungi was studied. Previously, we have developed the mass culture system which was adaptable for the culture of Beauveria bassiana. In this study, we determined the efficacy of this mass culture system for other entomopathogenic fungi, B. bassiana, Beauveria brongniartii, Metarhizium anisopliae, and Verticillium lecanni. To determine the efficacy of mass culture system, we examined the growth rate of entomopathogenic fungi in this system which was composed of 1st liquid media for growth of blastospore and 2nd pellet media for growth of conidia. As the result, we obtained that the blastopore numbers increased 103-104 times in liquid media at 72 hrs post inoculation. The results showed that this mass culture system for the growth of entomopathogenic fungi was effective.

  • The possibility of application of membrane type immobilized adsorbent to the fed-batch or perfusion culture system with anchorage-independent cells as well as batch system was investigated. The improvement in cell density and cell viability due to the combination of immobilized adsorbent with each culture system was evaluated for the investigation, and the optimum culture system employing immobilized adsorbent system was suggested based on the results. It was observed that the system with immobilized adsorbent showed better cell growth and cell viability than that without immobilized adsorbent in every operation system of batch, fed-batch, and perfusion. In case of batch system, 200% improvement of maximum cell density was observed in the system where ammonium chloride was added on purpose. And 50% improvement of maximum cell density was observed in the fed-batch system where ammonium ion accumulates significantly, while small increase in maximum cell density was observed in the perfusion system where dilution of waste byproducts exists. Especially, the fed-batch system showed the most significant improvement on cell growth because both compensation of nutrient and removal of ammonium ion occurred simultaneously in the system. Therefore a combined system of immobilized adsorbent and fed-batch operation could be suggested as an optimum system with in situ removal of ammonium ion.
