  • Understanding the net nutrient balance in a farming system is crucial in assessing the system's sustainability. We quantified N, P and K balances under vegetable organic farming in a Eutric Haplud and in West Java, Indonesia in five planting seasons from 2005 to 2007. The ten treatments and three replications, arranged in a completely randomized block design, included single or combined sources of organic fertilizers: barnyard manure, compos ts or green manures. The organic matter rates were adjusted every planting season depending on the previous crop responses. The result sshowed that the application of ${\geq}20$ t $ha^{-1}$ barnyard manure per crop resulted in positive balances of N, P, and K, except in the second crops of 2006 where potassium balance were -25 to -11 kg $ha^{-1}$ under the treatments involving cattle barnyard manure, because of low K content of these treatments and high K uptake by Chinese cabbage. Application of 20 to 25 t $ha^{-1}$ of plant residue or 5 t $ha^{-1}$ of Tithonia compost also resulted in a negative K balance. Soil available P increased significantly under ${\geq}25$ t $ha^{-1}$ barnyard manure and that under chicken manure had the highest available P. Accordingly, chicken barnyard manure gave the highest crop yield because of relatively higher N, P, and K contents. Plant residues gave the lowest yield due to the lowest nutrient content among all sources. Reducing the use of barnyard manure to 12.5 t $ha^{-1}$ and substituting it with Tithonia compost, Tithonia green manure or vegetable plant residue compost gave insignificantly different yield compared to the application of 25 t $ha^{-1}$ barnyard manure singly. In the long run, application of 25 t ha-1 cattle, goat, and horse manure or about 20 t $ha^{-1}$ chicken manure is recommendable for sustaining the fertility of this Andisol for vegetable production.

  • The structure of concanavalin A (ConA) has been studied intensively owing to its specific interactions with carbohydrates and its heterometal ($Ca^{2+}$ and $Mn^{2+}$) coordination. Most structures from X-ray crystallography have shown ConA as a dimer or tetramer, because the complex formation requires specific crystallization conditions. Here, we reported the monomeric structure of ConA with a resolution of $1.6{\AA}$, which revealed that metal coordination could trigger sugar-binding ability. The calcium coordination residue, Asn14, changed the orientation of carbohydrate-binding residues and biophysical details, including structural information, providing valuable clues for the development and application of detection kits using ConA.

  • Because of the phenological variation of vegetation growth in temperate region, it is often difficult to accurately assess the surface conditions of agricultural croplands, grasslands, and disturbed forests by multi-spectral remote sensor data. In particular, the spectral similarity between soil and dry vegetation has been a primary problem to correctly appraise the surface conditions during the non-growing seasons in temperature region. This study analyzes the spectral characteristics of the mixture of dry vegetation and soil. The reflectance spectra were obtained from laboratory spectroradiometer measurement (GER-2600) and from EO-1 Hyperion image data. The reflectance spectra of several samples having different level of dry vegetation fractions show similar pattern from both lab measurement and hyperspectral image. Red-edge near 700nm and shortwave IR near 2,200nm are more sensitive to the fraction of dry vegetation. The use of hyperspectral data would allow us for better separation between bare soils and other surfaces covered by dry vegetation during the leaf-off season.

  • A pepper strain of Cucumber mosaic virus (Pf-CMV) induces a mild chlorotic spot symptom in zucchini squash at 9 days post-inoculation (dpi), wile Fny strain of CMV causes severe mosaic and stunting symptom at 4 dpi in this host. Pseudorecombinants were constructed between the two strains, and assessments of symptom severity were indicated that both RNA2 and RNA3 were responsible for both mildness and the slow appearance of symptom elicited by Pf-CMV in zucchini squash. With various RNA2 and RNA3 chimeras between two strains of CMV, the genetic symptom determinants of phenotype of Pf-CMV were mapped to Tyr residue at positions amino acid 267 in 2a protein and at positions amino acid 168 in 3a movement protein (MP). Chimeras changed the sequences (both changed Tyr to lie) in the codons of both amino acid 168 of 3a MP and amino acid 267 of 2a protein were resulted in the high RNA accumulation, severity of symptom, and the rapid systemic spread, suggesting that 2a replicase as well as MP is involved in virus movement. The RNA accumulation pattern of all pseudorecombinants and chimeras are identical in protoplast of zucchini squash, indicating the virus movement is responsible for the phenotypes of two CMV strains rather than virus replication.

  • The multiresidue method 4.1.2.2 in Korea Food Code was extended for the analysis of 24 unregistered pesticide residues. The method includes acetonitrile extraction, liquid-liquid partition, Florisil SPE clean-up and GC analysis. The limits of quantification (LOQ) range of the method was 0.02~0.05 mg/kg for orange, brown rice and banana. The linearity for targeted pesticides were $R^2$ > 0.99 at the level ranged from 0.05 to 5 mg/L. Recovery test was performed at two concentration levels of LOQ and 4~10 times of LOQ. Recoveries and relative standard deviations (RSDs) of target pesticides were acceptable, showing 70~120% range and less than 20%, respectively, except for ethiprole, picloram and sulcotrion. This method is effectively applicable to routine analysis of target pesticides in orange, brown rice and banana.

  • Chlorothalonil and imidacloprid sprayed onto the green peppers were degraded more faster at outdoor than in greenhouse. These results were affected by dew and photodegration, considering no rain during the experimental period. Chlorothalonil, esfenvalerate and imidacloprid in green pepper, green twist pepper and sweet pepper did not show any residual pattern, because green peppers are one of the continuous harvesting crops and pesticides could not be sprayed homogeneously on them. When green peppers were pickled with soy sauce and green twist peppers were fried with vegetable oil, the amounts of pesticides such as alpha-cypermethrin, bifenthrin, chlorfenapyr, esfenvalerate and imidacloprid were diminished to the levels of about $30{\sim}71$ and $20{\sim}41%$, respectively. Esfenvalerate and imidacloprid could not be detected in 2 month-old hot pepper paste. The removal rates of pesticide residues in leaves of green peppers were about $22{\sim}37%$ by washing, about $74{\sim}95%$ by parboiling, and about $17{\sim}55%$ by drying after parboiling.

  • BACKGROUND: This study was carried out to investigate pesticide residues from fifty streams in Korea. Water samples were collected at two times. Thee first sampling was performed from april to may, which was the season for start of pesticide application and the second sampling event was from august to september, which was a period for spraying pesticides multiple times. METHODS AND RESULTS: The 136 pesticide residues were analyzed by LC-MS/MS and GC/ECD. As a result, eleven of the pesticide residues were detected at the first sampling. Twenty eight of the pesticide residues were detected at the second sampling. Seven pesticides were frequently detected from more than 10 water samples. Ecological risk assessment (ERA) was carried out by using residual and toxicological data. Four scenarios were applied for the ERA. Scenario 1 and 2 were performed using LC50 values and mean and maximum concentrations. Scenarios 3 and 4 were conducted by NOEC values and mean and maximum concentrations. CONCLUSION: Frequently detected pesticide residues tended to coincide with the period of preventing pathogen and pest at paddy rice. As a result of ERA, five pesticides (butachlor, carbendazim, carbofuran, chlorantranilprole, and oxadiazon) were assessed to be risks at scenario 4. However, only oxadiazon was assessed to be a risk at scenario 3 for the first sampling. Oxadiazon was not assessed to be a risk at the second sampling. It seems to be temporary phenomenon at the first sampling, because usage of herbicides such as oxadiazon increased from April to march for preventing weeds at paddy fields. However, this study suggested that five pesticides which were assessed to be risks need to be monitored continuously for the residues.

  • This study was carried out to investigate the pesticide residue pattern among different leafy vegetables applied with foliar spraying under greenhouse and to check extrapolating from some residue trial data to other minor crops. Leafy vegetables used in this study were: Mustard greens (Brassica juncea L.), Kale (Brassica oleracea L.), Dacheongchae (a kind of pak-choi (Brassica rapa subsp. chinensis L.)), Leaf broccoli (Brassica oleracea var alboglabra), Perilla leaf (Perilla frutescens (L.) Britton var. Frutescens), Leaf lettuce (Lactuca sativa L.), Swiss chard (Beta vulgaris L. subsp. vulgaris) and Red leaf chicory (Cichorium intybus L. var. foliosum Hegi). These are cultivated all year under indoor or outdoor and cut the leaf from plant continuously during harvest time. The amounts of pesticide deposit in/on the continuous harvesting leafy vegetables were affected by the ratios of leaf area to weight. Ratio of perilla leaf was the largest among crops as 58 $cm^2/g$. The residue levels of 7 pesticides in/on perilla leaf were the highest than those of other crops through the statistical analysis from zero day to fifth day after last application. The representative crop in 8 crops was perilla leaf selected based on the amounts of daily consumption and the high residues. This study suggest that the continuous harvesting leafy greens should be separated from the one time harvesting leafy vegetables for the pesticide recommendations because of different harvesting habits and pre-harvest intervals.

  • Recent outbreak of new diseases and pests which were introduced from abroad, seriously hampered both quality and safety of paprika fruits. This study has been carried out to aid an establishment of guideline for safe use of pesticides and reduction of their residues on paprika. Systemic fungicides boscalid and pyraclostrobin of either mixed (a.i.; 13.6+6.8%) or single (a.i.; 47 and 18.8%, respectively) water dispersible granule formulation(WG) products were sprayed with recommended or double dosage on paprika grown in green house at March and June. To draw pre-harvest residue limit, residues of each fungicide were analyzed from fruits collected eight times from 18 to 1 day pre-harvest. The biological half-lives of both boscalid and pyraclostrobin in mixed formulation in March and June were slightly shorter than those of single formulation which ranged from 14.4 to 20.1 days. Residue levels of both fungicides of single formulation in fruits in June were about one lower compared to those in March. However, application of double dosage frequently exceeded MRLs from fruits grown both seasons. These results showed that residue levels on fruits persisted longer period of time, more than two weeks, and so the case applied in winter season. The dissipation of fungicides on leaves and fruits was compared. The distribution of both fungicides in leaves was 20-200 times higher than that of fruits and persisted up to 18 days of pre-harvest period at the concentration of 10-40 ${\mu}g\;g^{-1}$. This study indicated that the mixed formulation product exhibited low residues in fruits, but high and long enough to pathogen growth in leaves.

  • Soybean can produce high-N residue due to N-fixation, so soybean rotation may increase yield of subsequent corn and reduce N fertilizer on the corn fairly. To find out the contribution of nitrogen to subsequent corn following soybean cultivation, soil nitrate, corn yield, and nitrogen uptake were measured for three continuous corn cropping years after soybean rotation. Three N rates of 0, 80, and 160 kg/ha were applied to three continuous corn following soybean cropping. At 6-leaf stage, soil nitrate amount at the soil depth of 0-30cm ranged from 60 to 80 kgN/ha higher in the first corn cropping year than that in the second and third corn cropping years. Judging from corn N status such as SPAD value, N concentration of ear-leaf and stover at silking stage, N contribution of previous soybean to corn in the first corn year was N fertilizer of approximately 80 kg N/ha. Stover N uptake at silking stage increased from 47 to 52 kg N/ha at the 0, and 80 kg N/ha of N rates in the first corn cropping year compared with those in the second and third corn cropping years. Corn grain yield at the 0 kg N/ha of N rate was 6-7 ton/ha higher in the first corn cropping year than that in the second and third corn cropping years, respectively. When compared the first corn year following soybean cropping with the second and third corn cropping years, N uptake of grain and stover at harvest with low N rates such as 0 and 80 kg N/ha increased from 45 to 67kg N/ha, from 35 to 60 kg N/ha, respectively. N uptake of whole plant by soybean rotation increased from 93 to 118 kg N/ha in the first year compared with that in the second and third corn cropping years. However, the N contribution by soybean cropping was small in the second and third continuous corn cropping years. Therefore, it was concluded that the nitrogen fertilizer of 80-100 kg N/ha in the first corn cropping year could be saved by soybean rotation and annual alternative corn-soybean rotation could be the best rotation system.
