We investigated characteristics and self-incompatibility genotypes of 11 crabapple cultivars to introduce a new pollinizer of 'Fuji' apple tree in Korea. Flowering dates of eleven crabapples were two to seven days earlier than that of 'Fuji'. The rate of pollen germination in vitro was ranged from 85.6% to 98.0% except 'Virginia'. Controlled pollination treatment with each crabapples to 'Fuji' increased fruit set rate about 20.4% to 34.4%, the number of seed per fruit about 13.8% to 42.3% and fruit weight about 7.4% to 16.7% compared to open pollination. Tested crabapples were resistant to peach fruit moth, brown leaf spot and sooty blotch in general. A PCR amplification method using S-RNase primers was carry out in eleven crabapples. S-alleles, $S_3$, $S_5$, $S_9$, $S_{10}$, $S_{20}$, $S_{26} from six crabapples were determinated. Through sequencing analysis, $S_5$ ('Manchurian', 'Virginia') and $S_9$ ('Yantaishagou') showed 100% homologous to previous result. Based on our results, it was recommended that 'Manchurian', 'Hopa A', 'Hanyaehanakaidou', 'Spectabilis' could be promising pollinzers for 'Fuji' apple cultivar.
The objective of this research was to investigate the effects of planting date, root pruning and uprooting on growth, yield and budding ratio of 'Seolhyang' strawberry (Fragaria${\times}$ananassa Duch.). Planting dates of plantlets with 60 ~ 70 days old were September 7th, September 14th and September 21th. Root pruning rates were controlled to 25% (RP 25) or 50% (RP 50) before transplanting. In the uprooting treatments, the plantlets were pulled out on root media and were replanted into same bed at 10 days (UR 10) and 20 days (UR 20) after transplanting. The delayed planting dates of plantlet resulted in the suppressed growth of plant, reduced yield, and slight earlier appearance of flower buds. The RP 50 treatment showed the lowest value among root pruning treatments in growth and yield, but appearance of flower buds slightly fall behinds. The UR 20 treatment only made flower budding earlier by 6 days than non-treatment. All of root stress treatment was appeared to decrease vegetative growth and yield except the RP 25 treatment. Above results imply that strong root stress was required to emerge second flower bud earlier but which reduced overall growth and yield in 'Seolhyang' strawberry.
When the cattail pollen was identified by using fibrinolytic agents, we found that the fibrinolytic activity was controlled by an enzyme. Therefore, for determining the fibrinolytic activity of cattail pollen, the fibrinolytic enzyme in cattail pollen was purified by gel filtration using DEAE-cellulose, Sephadex G-150 and HPLC. Also, its purity was certified by polyacrylamide gel electrophoresis, and its physico-chemical properties, such as pH and temperature stabilities and effects of metal, inhibitors and substrates, were examined. The specific activity, purification fold, and molecular weight of the enzyme were 38U/mg, 86.4,and 75kDa, respectively. The optimum pH for the purified enzyme was at 4.0 and it was stable at pH 4.0-6.0. The optimum temperature was $55^{\circ}C$ and it was stable at $30-60^{\circ}C$. But the enzyme began to be inactivated at $70^{\circ}C$ and its activity was totally lost at temperatures above $80^{\circ}C$. As for substrate specificity, the enzyme was most effective in dissolving fibrin, followed by whole casein, ${\kappa}$-casein, ${\alpha}$-casein, ${\beta}$-casein, and BSA. With casein as the substrate, Km value was found to be 0.44mM and the enzyme showed a high affinity for casein. As for the metal ions affecting enzyme activity, $K^+$, $Na^+$, and $Mg^{2+}$ had no effect on enzyme reaction while $Zn^{2+}$ and $Fe^{2+}$ showed potent inhibitory activity. Judging from the fact that the purified enzyme was also strongly inhibited by PMSF, iodoacetic acid, and SDA, it assumed to be a serine protease.

This study was conducted to obtain the basic information on rice cold damage and relationship to be connected with cold tolerance. The 20 rice varieties were screened to cold tolerance under low water temperature condition. The water gradient were controlled to $17^{\circ}C$ at the inlet and $21^{\circ}C$ at outlet of screening field. In the relationship between cold water irrigation and growth of rice varieties, effect on medium maturing cultivars were higher than early maturing cultivars. In relationship of shorting of culm length and panicle exsertion, culm length and panicle exsertion showed positive correlation. Yield of cultivars and culm length and percent of fertile grain showed positive correlation. However, heading date presented negative correlation. In the early maturing cultivars, except 'Kumobyeo', all cultivars range of discoloration value (1-3) was resistant. In the medium maturing cultivars, 'Donghaebyeo' was resistant in both the seedling stage whereas 'Hwajinbyeo' was resistant only in tiller stage. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Unhulled rice yield was increased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment.
Of 125 isolates collected from 35 golf courses, sensitivity of 44 isolates of Pythium species to metalaxyl was determined on corn meal agar with various concentrations of metalaxyl (0.1, 1.0, 10.0, 50.0, 100.0, and $250.0{\mu}g\;a.\;i/ml$). The isolates were able to be categorized into the sensitive and resistant groups based on hyphal growth measured in terms of colony diameters on the medium with $1.0\;and\;10.0{\mu}g\;a.\;i./ml$. When compared with hyphal growth on the medium without metalaxyl, hyphal growth of the sensitive group which included 31 isolates was inhibited by $66{\sim}98%$ on the medium with $1.0{\mu}g\;a.\;i./ml$, whereas that of the resistant group which included 13 isolates grew well and the hyphal growth was inhibited only by $6{\sim}26%$. When $10.0{\mu}g\;a.\;i./ml$ of metalaxyl was included in the medium, hyphal growth of the sensitive and resistant groups was inhibited by $82{\sim}99%\;and\;27{\sim}47%$, respectively. Occurrence of metalaxyl-resistant isolates of Pythium spp. not only from turfgrasses on golf courses but also from other crops was observed for the first time in Korea. Metalaxyl-resistant isolates occurred most frequently in P. graminicola. Control effects of metalaxyl were determined by applying metalaxyl after and before inoculation of 4 and 3 isolates of sensitive and resistant isolates of P. graminicola, respectively, to creeping bentgrass in pots. The minimum concentration of metalaxyl to control metalaxyl-sensitive isolates was $6.25{\mu}g\;a.\;i./ml$, whereas the disease caused by the metalaxyl-resistant isolates could not be controlled with $12.50{\mu}g\;a.\;i./ml$ of metalaxyl. The disease was controlled more effectively by an application of metalaxyl prior to inoculation than after occurrence of the disease.
This study aimed to evaluate the fruit quality characteristics and incidence of flesh browning in response to air storage at $0{\pm}1^{\circ}C$ a fter controlled atmosphere s torage (CA condition: $O_2$$2.5{\pm}0.5$%, $CO_2$$1.5{\pm}0.5$%) at $0{\pm}1^{\circ}C$ in 'Fuji' apples (Malus domestica Borkh.). The storage system was performed as followed: air storage for one month, CA storage 4 months + air storage 3 months ( CA 4M + A ir 3M), CA storage 5 m onths + air storage 2 months ( CA 5M + Air 2M) a nd C A storage 6 m onths + air storage 1months (CA 6M + Air 1M), while the control fruits were stored at CA storage for 8 months right after harvest. The incidence of flesh browning ranged from 17.1% to 30.2% during CA storage but not detected under the treatments of CA 4M + Air 3M and CA 5M + Air 2M. The respiration rate was not affected by storage treatments for 6M while the respiration rate was lower in the treatments of CA 4M + Air 3M and CA 5M + Air 2M than the other storage treatments after 7 months. Ethylene production and internal ethylene concentration were lowest in rapid CA storage and increased with a decreasing CA storage duration. Therefore, the results indicate that CA 5M + Air 2M storage treatment should be recommended to maintain the fruit quality and reduce the risk development of flesh browning rather than typical CA storage in 'Fuji' apples.
This study was carried out to investigate the effect of irrigation method adopted for reducing nutrient solution drainage on the root zone environment, growth and yield of a tomato crop grown in a rockwool medium. The irrigation control methods used were large quantity irrigation at a long interval controlled by only an integrated solar radiation sensor (standard), medium quantity irrigation at a medium interval (zero drainage 1), and small quantity irrigation at a short interval (zero drainage 2) controlled by both an integrated solar radiation sensor and a zero drainage sensor. The amount of the nutrient solution supplied and the drain percentage per plant of the standard, zero drainage 1, and zero drainage 2 were 1.4, 0.9 and 0.8 L, and 23.8, 8.6 and 3.7%, respectively. The average, minimum, and maximum water contents and EC of the standard, zero drainage 1, and zero drainage 2 were 64.5~88% and $1.5{\sim}3.5dS{\cdot}m^{-2}$, 40.3~76.0% and $2.5{\sim}4.0dS{\cdot}m^{-2}$, and 56.3~69.0% and $2.7{\sim}3.7dS{\cdot}m^{-2}$, respectively. There was no difference in leaf width, number of leaves, and stem diameter among the treatments. However, plant height and leaf length decreased in the zero drainage 1 and 2 treatments as compared to the standard. The fruit marketable yield per 10a in the zero drainage 1 and 2 treatments was about 93 and 88%, respectively, of that in the standard treatment.

This study was carried out to examine the base temperature to flowering and the average days to flowering by accumulated hours of low temperature ($5.0^{\circ}C$) or changing temperature after bud breaking. Over-all, the prediction of flowering time in the commercial apple cultivars ('Fuji' and 'Tsugaru') and apple cultivars ('Chukwang', 'Gamhong', 'Hongan', 'Honggeum', 'Hongro', 'Hongso', 'Hwahong', 'Summer dream', 'Sunhong') bred in Korea at the Gunwi region for 4 years (from 2009 to 2012) was investigated. Also, this study estimated the flowering time when the air temperature of Gunwi region rises at $5.0^{\circ}C$ was investigated using the same data. The range of accumulated hours of low temperature (chilling requirement) was from 0 hour to 1,671 hours, and the range of high temperature (heat requirements) to flowering after low temperature treatment was from $5.0^{\circ}C$ to $29.0^{\circ}C$. The treatments of changing temperature after bud breaking were classified as constant temperature treatment (control) and $5.0{\sim}10.0^{\circ}C$ elevation or descent treatments. The results show that the average days to flowering was longer with shorter accumulated hours of low temperature, and the average days from bud breaking to flowering of 0 hour treatment was longer about 2~4 weeks than that of 1,335~1,503 hours treatments. In comparing to apple cultivars, the all cultivars were not flowered under $10.0^{\circ}C$ after bud breaking, and the cultivars with low chilling requirements needed low heat requirements for flowering. The average days to flowering of treatments that the air temperature after bud breaking was controlled about $15.0^{\circ}C$ was shorter about 1~3 weeks than that of treatments was controlled about $10.0^{\circ}C$. In the treatment of changing temperature after bud breaking, the average days from bud breaking to flowering of temperature elevation treatment was shorter than that of constant temperature treatment. By use of these results, the base temperature to flowering of main apple cultivars in Korea was seemed to $10.0^{\circ}C$, and if the air temperature of Gunwi region rises about $5.0^{\circ}C$ than that of current, the flowering time was estimated to be delayed by 1 week.
This study was conducted to determine the optimum nutrient solution, pH, irrigation interval, light intensity and planting density to growth of common ice plant (Mesembryanthemum crystallinum L.) in a closed-type plant production system. Three-band radiation type fluorescent lamps with a 12-h photoperiod were used. Nutrient film technique systems with three layers were used for the plant growth system. Environmental conditions, such as air temperature, relative humidity and $CO_2$ concentration were controlled by an ON/OFF operation. Treatments were comparison of the nutrient solution of Horticultural Experiment Station in Japan (NHES) and the nutrient solution of Jeju National University (NJNU), pH 6.0 and 7.0, irrigation interval 5 min and 10 min, light intensity 90 and $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and within-row spacing 10 cm, 15 cm, 20 cm and 25 cm with between-row spacing 15 cm. Optimum macronutrients were composed N 7.65, P 0.65, K 4.0, Ca 1.6 and Mg $1.0mM{\cdot}L^{-1}$. There were no significant interactions between pH 6.0 and 7.0 about shoot fresh weight and shoot dry weight of common ice plant. Irrigation interval 5 min and 10 min was also the same result. Shoot fresh weight and shoot dry weight were highest at $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Shoot fresh weight and shoot dry weight were decreased according to increasing the planting density. From the above results, we concluded that optimum nutrient solution, optimum levels of pH, irrigation interval, light intensity and planting density were 6.0-7.0 and 10 min, $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and $15{\times}15cm$, respectively for growth of common ice plant in a closed-type plant production system.
The objective of this study was to investigate the effects of light intensity and electrical conductivity (EC) of nutrient solution during short day treatment in an ebb and flow systems on the growth and nutrient uptake of potted Kalanchoe blossfeldiana 'Rako' and the nutrient accumulation of growing medium. Nutrient concentrations in the growing medium were also analyzed to investigate the accumulation rates of macro-nutrients such as T-N, P, K, Ca, and Mg, respectively. To achieve the objectives, plants were fed with a nutrient solution with 1.2, 1.8, or $2.4dS{\cdot}m^{-1}$ under three daily photosynthetic photon flux (PPF) of 4.26, 5.51, or $9.75mol{\cdot}m^{-2}{\cdot}d^{-1}$. Both light intensity and EC of nutrient solution significantly influenced the crop growth. The elevation of PPFs resulted in the increase of plant growth. For each light condition, plant growth, such as dry and fresh weight and leaf area, was the highest when the electrical conductivity of nutrient solution was controlled to $2.4dS{\cdot}m^{-1}$. However, growth was acceptable in the EC ranges from 1.8 to $2.4dS{\cdot}m^{-1}$. Both light intensity and EC of nutrient solution significantly influenced the uptake of nutrients in the solution tanks and the accumulation of nutrients in the growing medium. As the EC of nutrient solution was elevated, the absorption rates of $NO_3^-$, $PO_4^{-3}$, $K^+$, and $Mg^{2+}$ by crops and accumulation of those in growing medium increased, but the light intensity did not significantly influence the absorption rates. Based on the above results, the regression models were suggested for anticipating the macro-nutrient accumulations in growing medium.
