Effects of Banhaonpaetang(半夏溫肺湯) extract and its Constituent herbs on the Contraction of Isolated Guinea Pig Trachea Smooth Muscle (반하온폐탕(半夏溫肺湯) 및 그 구성약물(構成藥物)이 실험(實驗) 동물(動物)에 미치는 영향(影響))

  • This study has been carried out to investigate the effects of Banhaonpaetang extract and its constituent herbs on the contractile force of Isolated Guinea pig trachea smooth muscle and to elucidate its mechanism. The results were as follows; 1. Banhaonpaetang significanly inhibitd the contractile force of isolated guinea pig trachea smooth muscle pretreated by histamine. 2. Banhaonpaetang significantly inhibited the contractile force of isolated guinea pig trachea smooth muscle pretreated by acetylcholine. 3. Radix Asari(細辛), Flos Inulae(旋覆花), Pericarpium Citri Nobilis(陳皮) and Radix Ginseng(人蔘) extract significantly inhibited the contractile force of isolated guinea pig trachea smooth muscle pretreated by histamine. 4. Radix Asari(細辛), Cortex Cinnamomi(桂心), Flos Inulae(旋覆花), Pericarpium Citri Nobilis(陳皮) and Radix Ginseng(人蔘) extract significantly inhibited the contractile force of isolated guinea pig trachea smooth muscle pretreated by acetylcholine. 5. The effects of Banhaonpaetang extract on the changes of plasma ACTH in rats were shown to be insignificant. 6. The effects of Banhaonpaetang extract on the changes of serum electrolytes were shown to be significant in $K^+$ and $Ca^{2+}$.

  • This study was carried out to investigate the effect of samyootang and shinchulsan extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate it's mechanism The isolated were obtained as follows ; 1. The isolated trachea smooth muscle was suspended in the organ bath with oxygenated kreb's Henseleit bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recored with Isometric transducer (nacro F-60) The resting tension was approximately 0.5g 2. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine $10^{-4}M$ was significantly inhibited by samyootang and shinchulsan extract. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine $10^{-4}M$ was considerably inhibited by samyootang and shinchulsan extract. 4. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine $10^{-4}M$ was significantly inhibited by samyootang and shinchulsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine $10^{-4}M$ was significantly inhibited by samyootang and shinchulsan extract.

  • The effects of omission of buffers from Krebs-Ringer medium on contractile activity, membrane potentials and ATP content of electrically stimulated isolated rat atria were investigated. 1) Contractile status: A rapid and marked depression of the contractile activity of atria occurred when buffer-free medium was substituted for the normal Krebs-Ringer medium. 2) Electrical status: The omission of buffers from medium did not alter the resting or action potential magnitudes of atria. However, the action potential duration was on initial increase followed by a decrease in the buffer-free medium. 3) ATP concentration: The omission of buffers from medium resulted in a marked decrease in the ATP levels of atria. It has been also found in the present study that bicarbonate buffer plays an important role for the maintenance of the contractility and ATP levels of the heart. The contractile depression by the omission of buffers was not directly associated with electrical alterations in resting or action potentials of the heart. In the absence of bicarbonate-buffer, glucose no longer plays to maintain the contractile activity and the ATP levels of rat atria.

  • In order to study the effects of Yunpaejaesueum known clinically for their effects of treatment for cough and asthma, the study was carried out to investigate the effect of Yunpaejaesueum extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism. The results were obtained as follows: 1. The isolated trachea smooth muscle of guinea pig was suspended in the organ bath with oxygenated Kreb's Henseleite bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with isometric transducer(Nacro F-60). The resting tension was approximately 0.5g. 2. The isolated trachea smooth muscle of guinea pig was remakably relaxed by the administration of Yunpaejaesueum. 3. Yunpaejaesueum is significantly inhibited the contractile response of histimine 10-4 M in isolated guinea pig trachea smooth muscle. 4. Yunpaejaesueum is significantly inhibited the contractile response of acetylcholine 10-4 M in isolated guinea pig trachea smooth muscle. 5. Yunpaejaesueum is significantly inhibited the contractile response of 5-hydroxytryptamine 10-4 M in isolated guinea pig trachea smooth muscle. 6. Yunpaejaesueum is significantly inhibited the contractile response of prostaglandin F2a 10-7 M in isolated guinea pig trachea smooth muscle.

The results were obtained as follows: 1. The contractile response of the trachea smooth muscle of the isolated guinea pig by prostaglandin $F2\;{\alpha}$ 10-7 M was significantly inhibited by Shihngigiltang extract.

Studies on the Physiological Properties of the Histamine Receptor of Ileal Smooth Muscle in Dog (개 회장 평활근에 있어서 Histamine Receptor의 생리적 특성에 관한 연구)

  • To validate the physiological properties of the histamine receptors of ileal smooth muscle in dog, the effects of adrenergic-, cholineric-, and H-receptor antagonists on the responses of ileal smooth muscle strips to histamine were investigated. The results were summarized as follows; 1. Histamine caused the contraction of ileal smooth muscle and the contractile responses were increased between the concentration of histamine $10^{-7}M$ and $10^{-5}M$ with dose-dependent manner in dog. 2. The shorter the treatment interval of histamine, the lower the contractile activity until the treatment interval extended to 40 minutes. 3. The contractile response induced by histamine was completely blocked by the pre treatment with a $H_1$-receptor blocker, chlorpheniramine and not by the pretreatment with a $H_2$-receptor blockers cimetidine. 4. The contractile response induced by histamine was not blocked by the pretreatment with a cholinergic receptor blocker, atropine. 5. The contractile response induced by histamine was not blocked by the pretreatment with an ${\alpha}$-adrenergic receptor blocker, phenoxybenzamine, or a ${\beta}$-adrenergic receptor blocker, propranolol. From these results, it was suggested that the contraction induced by histamine was elicited through $H_1$-receptor on the ileal smooth muscle in dog.

  • The effcets of adenosine 5'-triphosphate(ATP) were investigated on the uterine smooth muscle motility in the pig. The results were summarized as follows: 1. The effects of the porcine uterine smooth muscle and the contractile responses increased between the concentration of ATP $10^{-5}$ and $10^{-3}$ M with a dose-dependent manner. 2. The contractile response induced by ATP($10^{-4}$ M) was not blocked by pretreatment with cholinergic receptor blocker, atropine ($10^{-6}$ M) 3. The contractile response induced by ATP ($10^{-4}$ M) was not blocked by pretreatment with $\alpha$ -adrenergic receptor blocker, phentolamine(10$^{-6}$ M) and ${\beta}$-adrenergic blocker, propranolol ($10^{-6}$ M). 4. The contractile response induced by ATP($10^{-4}$ M) was not appeared in 4Ca^{++}$ -free medium. As the concentration of $Ca^{++}$ in $Ca^{++}$ -free medium was increased, the contractile response induced by ATP ($10^{-4}$ M) was enhenced but was completely inhibited by pretreatment with $Ca^{++}$ -channel blocker, papaverine($10^{-6}$ M) or verapamil($10^{-6}$ M). From these results, it was conclued that the effects of ATP were the contraction mediated by purinergic receptor in uterine smooth muscle of pig.

Change of Isometric Contractile Force and Muscle Activity Applying Heat according to the Time on Biceps Brachii Muscle

  • This study has investigated the effect of isometric contractile force and muscle activity applying sperficial heat according to the time from the biceps brachii muscle. In this study, 20 university students participants without musculoskeletal and neurological disorders. By applying a hot pack 5min, 10min, 20min and 30min respectively. After that measurement are skin temperature, contractile force and muscle activity. Skin temperature of the hot 5 min applied that rapidly changing. Increasing the time it takes to apply a variance has been reduced(p<.001). Isometric contractile force was not statistically significant but highest when applying the hot pack 5 minutes and lowest when applying the hot pack 30 minutes(p<.001). Muscle activity and median frequency was highest when applying the hot pack 5 minutes. To analyze the above results, it was found that isometric contractile force and muscle activity changed according to the applying time. These result lead us to the conclusion that this study will be more evidence for changes in muscle contraction to apply hot pack on clinic.

Pinacidil causes depresor action, catecholamine release and vasorelaxation in the normotensive rat

  • The present study was conducted to investigate the effects of pinacidil, a potassium channel opener, on arterial blood pressure, catecholamine release and vascular contractile responses in the normotensve rats and to establish the mechanism of action. Phenylephrine (an adrenergi $_1$-receptor agonist) and high potassium (a membrane- depolarizing agent) caused greatly contractile responses in the isolated aortic strips, respectively. These phenylephrine (10$\^$-5/ M)-induced contractile responses were dose-dependently depressed in the presence of pinacidil (25 ∼ 100 ${\mu}$M). (omitted)

  • In this study was proposed that a new estimating method for investigation of contractile state changes which generated from continuous isometric contraction of skeletal muscle. The physiological changes(EMG, ECG) and the psychological changes by CNS(central nervous system) were measured by experiments, while the muscle of subjects contracted continuously with isometric contraction in constant load. The psychological changes were represented as three-step-change named 'fatigue', 'pain' and 'sick(greatly pain)' from oral test, and the method which compared physiological change with psychological change on basis of these three steps was developed. The result of analyzing the physiological signals, EMG and ECG signal changes were observed at the vicinity of judging point in time of psychological changes. Namely, it is supposed that contractile states have three kind of states pattern (stable, fatigue, pain) instead of two states (stable, fatigue).
