Microbial Hazards and Microbe Reduction Technologies for Mushrooms (버섯의 미생물 위해성 및 저감화 처리기술 개발 현황)

  • Mushroom consumption is gradually growing annually worldwide for many centuries. Oyster mushrooms (Pleurotus ostreatus), button mushrooms (Agaricus bisporus), and enokitake (Flammulina filiformis) are mainly consumed in Korea. However, mushrooms can be contaminated with pathogenic microorganisms, such as Listeria monocytogenes, because antibacterial treatment during mushroom cultivation and processing is insufficient. Therefore, many cases of mushroom contamination-related foodborne illnesses and food recalls have been reported. Three representative treatments are used to prevent microbial contamination in mushrooms: chemical, physical, and combination treatments. Among the chemical treatments, chlorine compounds, peroxyacetic acid, and quaternary ammonium compounds are commercially used and ozone and electrolyzed water has recently been used. Additionally, physical treatments, including ultrasound, irradiation, and cold plasma, are being developed. Combination techniques include ultraviolet/chlorine compounds, ozone/organic acid, and ultrasound/organic acid. This review describes the domestically consumed mushroom types and their characteristics, and investigates the mushroom contamination levels. Additionally, effective antibacterial technologies for reducing microbial contamination in mushrooms are also discussed.

Climate Variability and Chum Salmon Production in the North Pacific (북태평양 기후변화와 연어 생산력 변동)

  • The relationship between North Pacific chum salmon (Oncorhynchus keta) population and climate variability was investigated in the North Pacific ecosystem. Time-series for the Aleutian Low Pressure, Southern Oscillation, Arctic Oscillation, and Pacific Decadal Oscillation (PDO) indices dating back to 1950 are compared with the chum salmon catch using a cross-correlation function (CCF) and cumulative sum (CuSum) of anomalies. The results of CCF and CuSum analyses indicated that there was a major change in climate during the mid 1970s, and that the chum salmon population responded to this climate event with a time-lag. The PDO and chum salmon returns showed a highly significant correlation with a time-lag of 3 years, while the AOI with a time-lag of $6{\sim}7$ years. The favorable environments for fry chum salmon might cause better growth in the coastal areas, but higher growth rate during the early stage does not seem to be related to the improved return rate of spawning adults. Rather, growth in the Okhotsk Sea or the Bering Sea during immature stages has a significant correlation with return rate, which implies the size-related mortality process. The development of a local climate index is necessary to elucidate the effect of climate variability on the marine ecosystem around the Korean Peninsula.

Production and Properties Chip Block Pallets from Teak Wood (Tectona grandis sp.) Biomass

  • Wood biomass, such as sawdust, particles, and chips from the wood industry, can be potentially used as a composite product. Chip block pallets (CBP) are composite products that can be produced from industrial wood waste and are in high demand in the logistics sector. Therefore, this study aimed to investigate the production of CBP from teak wood biomass with varying polyurethane contents. In addition, this study analyzed the optimum particle-size composition was determined. The CBP production of CBP be divided into two stages. The first stage evaluated the use of polyurethane adhesive content, whereas the second stage considered the effect of particle size composition. The 9 × 9 × 9 cm3 of CBP with 0.6 g/cm3 target density was fabricated using a cold press. The National Wooden Pallet and Container Association (NWPCA) standards were used to evaluate the density, moisture content, dimensional stability, water absorption, compressive strength (CS), and screw-holding strength (SHS) of our CBP products. The mechanical and physical properties of CBP products were investigated. As a result, the CBP sample prepared using 4-14 mesh particle size and 4.5% adhesive content showed the optimal strength values, such as CS of 14.67 MPa and SHS of 371.50 N. These findings demonstrate that the CBPs derived from teak wood waste closely resemble commercial chip blocks and have the potential to replace wood bearings as pallet pads.

Study on the Content of ${NO_3}^-$ in Green Vegetable Juice by Different Sorts, Harvesting Time, Mixing Rate of Vegetable, Storage Condition and Manufacturers (채소 모재료의 종류, 수확시기별, 부위별 혼합비율, 저장조건 및 생산회사에 따른 녹즙의 ${NO_3}^-$ 함량차이)

  • After the consumption of green vegetable juice by Korean increase rapidly, the ${NO_3}^-$ intake through green vegetable juice have been ignored to consider for the calculation of daily ${NO_3}^-$ intake. It is necessary to collect the basic data on the ${NO_3}^-$ content in green vegetable juice by different sorts, harvesting time and mixing rate of vegetable, manufacturers and storage conditions for the next calculation of daily ${NO_3}^-$ intake for Korean. Followings are the research results from monitoring and laboratory experiment related with ${NO_3}^-$ and vitamine C in green vegetable juice. The ${NO_3}^-$ content of angelica plaant(tomorrow's leaf) and kale were higher in spring than those in summer and autumm. The highest value of ${NO_3}^-$ content in tomorrow's leaf and kale were 4.85 and 2.94 times higher compare to the lowest value. The average ${NO_3}^-$ content in the midribs of tomorrow's leaf and kale were 7.5 and 2.1 times higher than those in leafblades. It indicate the green vegetable juice made from leadblade of tomorrow's leaf and kale might be better compare to those from midrib in terms of ${NO_3}^-$ content. The content of ${NO_3}^-$ and vitamine C as affected by the timecourse after juice making were decreased rapidly compare to those by storage temperature in case of carrot, kale and cucumber juice. It show the positive comelation between the content of ${NO_3}^-$ and vitamine C in carrot, kale and cucumber juice regardless of room temperature(${NO_3}^-$) or cold temperature(${NO_3}^-$). The content of ${NO_3}^-$ and vitamine C of green vegetable juice by P company were the highest among the manufactuers. The lower content of ${NO_3}^-$ and vitamine C of green vegecable juice by TW company and GB company compare to P company is due to dilution with water to produce the juice. The content of ${NO_3}^-$ of green vegetable juice which were available in market showed 143ppm in carrot juice, 506ppm in tomorrow's leaf juice, 669ppm in wild water celery juice, 985ppm in kale juice, whereas the content of vitamine C were 43ppm in carrot juice, 289ppm in wild water celery juice, 353ppm in kale juice and 768ppm in tomorrow's leaf juice. It was calculated that people take 253mg by tomorrow's leaf juice, 335mg by wild water celery juice, 483mg by kale juice if they drink 500ml of green vegetable juice perday, and it suggest to excess 1.16, 1.53 and 2.21 times respectively only by green vegetable juice consumption.

The Actual Management State of Trial Contact Lenses and Lens Care Products in Local Optical Shops (안경원의 시험착용 콘택트렌즈 및 관리용품 관리 실태)

  • Purpose: In the present study, the actual management state of trial contact lenses and lens care products in local optical shops was surveyed and analyzed to reduce the risk of lens complication possibly induced by neglecting lens care. Methods: The feeling of contact lens wearers during the wear of trial contact lenses was surveyed. Futhermore, the actual management state of trial contact lenses such as cosmetic lens and RGP lens and lens care products was also investigated by surveying opticians who trade contact lenses in local optical shops. Results: It was found that consumers trusted the sanitary conditions of the lens since trial cosmetic contact lens and RGP lens were cleaned before and after trails by over 98% of opticians in local optical shops. For trial cosmetic lens, cleaning with normal saline, multipurpose solution for soft lens and combination of saline and multipurpose solution were 38.5%, 40.5% and 21%, respectively, before trials. After trials, cosmetic lenses were cleaned with normal saline, multipurpose solution for soft lens and a combination of saline and multipurpose solution were 13%, 75%, and 12%, respectively. On the other hand, cleaning with normal saline, multipurpose solution for RGP lens and combination of saline and multipurpose solution were 28.5%, 38.5% and 33%, respectively, before trying trial RGP lens. After trials, RGP lenses were cleaned with normal saline, multipurpose solution for RGP lens and a combination of saline and multipurpose solution were 2.5%, 70%, and 27.5%, respectively, indicating that relatively many opticians followed the lens cleaning regimen. In local optical shops, the cleaning trial cosmetic lens was mainly conducted at every 10 days or a month and the washing cycle of cosmetic lens case was in a month or 2~3 months. The cleaning interval of trial RGP lens was primarily in a month or 2~3 months. For those lens cases, more than 75% of opticians washed them with a surfactant and then rinsed with cold water. The storing periods of lens care products were primarily in a week for saline and in a month and 2~3 months indicating that storing period of lens care products was relatively well-kept in local optical shops. Conclusions: It is thought that the concern about any microbial infection is not that high since trial contact lenses and lens care products were generally well-managed by opticians in local optical shops from the results above. However, better public eye health and better public confidence in opticians may be possible if further strengthen in avoidance of lens cleaning with saline, keep of cleaning cycles within 2 weeks and rinsing of lens cases with hot water happens.

Dietary Supplementation of Resveratrol and Methoxylated Resveratrol Affects on Chicken Thigh Meat Quality (Resveratrol과 Methoxylated Resveratrol이 닭 다리육의 저장 품질에 미치는 영향)

  • This study was carried out to determine the effect of dietary supplementation of resveratrol and methoxylated resveratrol extracted from branch of Morus alba L. on the quality of chicken thigh meat during cold storage. For 35 days, 1-day-old 320 broiler chicks (Ross) were divided into 8 groups and supplemented the diet; basal diet only (BD), BD with antibiotics (AB), vitamin E 20 IU (VE 20), vitamin E 200 IU (VE 200), resveratrol 20 ppm (RV 20), resveratrol 200 ppm (RV 200), methoxylated resveratrol 20 ppm (MR 20), and methoxylated resveratrol 200 ppm (MR 200). After slaughtering the broilers, thighs were collected and analyzed the quality change of the meat during storage at $4^{\circ}C$ for 5 days. The meat quality factors such as pH, color, water holding capacity, and sensory characteristics of thigh meat were determined on the experiment day 1, 3, and 5. AB, VE, and MR increased pH value of chicken thigh compare to BD (p<0.05). Lightness ($L^*$) showed no significant difference during storage day 1 and 5. VE 200 and MR 20 stabilized the redness ($a^*$) of chicken thigh as the value of day 1. Water holding capacity of chicken thigh from VE 20, RV 200, MR 20, and MR 200 on storage day 3 was higher than that of BD (p<0.05). In sensory evaluation, the panelist discriminated the tenderness and gave the higher score on the chicken from AB, VE20, RV 20, and MR 20 compare to BD (p<0.05). These results suggest that the dietary resveratrol and methoxylated resveratrol could be used as chicken meat quality enhancer in broiler industry.

The Chemical Composition of Abies koreana Wilson Wood (구상나무(Abies koreana Wilson)재(材)의 화학적(化学的) 조성(組成))

  • The Chemical components of Abies koreana Wilson grown in Korea were analized. The results obtained were summarized as follows. 1. The ash content is ca 0.56% on the average, and of the range is 0.33 to 0.76%. 2. The cold water extractive content is ca 8.76% on the average and of the range is 5.55 to 12.5%. 3. The hot water extractive content is ca 10.16% on the average and of the range is 4.80 to 13.65%. 4. Basic extractive content is ca 14.60% on the average and of the range is 5.51 to 25.44%. 5. The alcohol benzol soluble fraction is ca 4. 23% and of the range is 2.94 to 5.44%. 6. The holocellulose content is ca 76.49% on the average and of the range is 73.68% to 79.10%. 7. The cellulose content is ca 56.30% on the average and of the range is 46.02% to 61. 33%. The cellulose contains 78.54% ${\alpha}$-cellulose, 7.66% ${\beta}$-cellulose and 14.04% ${\gamma}$-cellulose respectively. 8. The Klason lignin content is ca 25.03% on the average and of the range is 22.5 to 27.0%. In conclusion, ash content is comparable to the other needle leaf trees. It has 76.49% in holocellulose content which is comparable value to the Pinus densiflora's. It has lower value than the Pinus densiffora in pentosan and lignin content. Having not resin cannal in xylem and long tracheid, this wood could be usable industrial material.

Study of Serum Constituents in Several Species of Cultured Fish (주요 양식 어류의 혈액 성분에 관한 연구)

  • This study was performed to obtain the basic data on the serum components of several marine fish species commonly cultured in Korea. Blood samples taken from five species of fish were analyzed for various components of serum, total protein (TP), albumin (ALB), triglyceride (TG), cholesterol (CHOL), glucose (GLC), sodium (Na). Potassium (K), chloride (Cl), Phosphorus (P), lipase (LIPA), alanine aminotransferase (ALT) and aspartate aminotransferase (AST). The fish used were coho salmon(Oncorhynchus kisutch), rock fish (Sebastes schlegeli), sea bass (Lateolabrax japonicus), olive flounder ( Paralichthys olivaceus) and parrot fish (Oplegnathus fasciatuss) reared at the Chungmu Experimental Fish Culture Satation of KORDI. TP concentration of warm-water species (2.9-5.1 g/dl) was higher than that of cold-water species, and ALB concentration was ranged at the level of 1.2-1.9 g/dl. Coho salmon showed the highest ration of A/G(1.1), and the other species were about 0.5-0.6. The concentrations of TG and CHOL, components of lipids, varied with the different species. The concentration of TG was high, but CHOL concentration was low in olive flounder, while the reversed results were shown by sea bass. The sum of these two components was the highest with 600mg/d1 in olive flounder, and about 400mg/d1 for sea bass and rock fish, and 300mg/d1 for parrot lish and coho salmon. Concentration ot GLC in coho salmon and rock fish ranged from 61 to 76mg/d1 which were about lour times higher than that of flounder. The highest lipase activity was observed in coho salmon, while it was nearly nil in flounder. The reversed tendency was found for TG concentration. Mineral concentrations of Na, Cl and K were 160-204 mmol/l, 137-183mmo1/1 and 0.5-3.1 mmol/l, respectively, but no significant difference between the species was observed. However, the concentrations of P were high in relatively active species such as coho salmon and rockfish. AST activity in all species examined was higher than that of ALT with being highest in coho salmon. The highest ALT activity was found in olive flounder.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. Optimal Enrichment Temperature, Time and Materials for L-type Rotifer (Brachionus plicatilis) Cultured at a Low Temperature (저온 배양한 L-type 로티퍼(Brachionus plicatilis)의 적정 영양강화 수온, 시간 및 영양강화제 종류)

  • This study was undertaken to improve the survival and early life growth rates of cold-water fish by culturing rotifer (Brachionus plicatilis) with low-temperature tolerance. The enrichment experiment was carried out at different temperatures and over different time intervals. Cultivation of the rotifer at low temperatures was repeated, with the selected and cultured as the water temperature was gradually lowered from $20^{\circ}C$ to $10^{\circ}C$. Enrichment of the rotifer was completed using A, S, SCV and SCP. Enrichment was carried out after 6, 12 and 24 hours at three different temperatures (10, 15 and $20^{\circ}C$). In the growth experiments, the rotifer increased to approximately triple their original size, from $350{\pm}7.9ind./ml$ to $1,064{\pm}5.7ind./ml$ at $10^{\circ}C$ over 50 days. The fatty acid composition of the four enrichment materials was species-specific, with the highest ratios belonging to eicosapentaenoic acid (EPA, C20:5n-3) and docosahezaenoic acid (DHA, C22:6n-3) in SCP. The fatty acid composition of the rotifers was affected by the enrichment materials. The EPA (% of total fatty acid) was more than 2 % in SCP, which showed a higher ratio than the other enrichment materials. DHA was higher in S reaching 12.40 % at $15^{\circ}C$ for 24 hours. The highest levels of EPA (3.09 %) and DHA (11.65 %) were obtained after the rotifers were enriched with S at $20^{\circ}C$ for 12hours.
