To investgate growth effect of tomato by Bacillus sp. WRD-1 isolated from soil, the Bacillus sp. WRD-1 cultures were treated into tomato cultivated soil with different dilutions (1:100, 1:300, and 1:500) and autoclaved Bacillus cultures as control. Growth and yeild of tomato enhanced in treatments of the Bacillus cultures compared to control. The populations of native bacteria and actinomyces were increased twice in field treated with Bacillus sp. WRD-1 cultures, but the number of mold was decreased. Since the Bacillus sp. WRD-1 promoted growth of tomato and affected population dynamics of microorganism in field, this strain is prominent candidate as a microbial biocide to improve soil potential.
The present work presents a novel approach for the dynamic quantification of respiration rates on a small scale by using lysine-producing Corynebacterium glutamicum ATCC 21253. Cells sampeld from batch cultures at different times were incubated ina 12-ml scale bioreactor equipped with a membrane mass spectrometer. Under dynamic conditions, gas exchange across the gas-liquid phase, specific respiration rates, and RQ values were precisely measured. For this purpose, suitable mass balances were formulated. The transport coefficients for $O_2$ and $CO_2$, crucial for calculating the respiration activity, were determined as $k_La_{O2}=9.18h^{-1}$ and $k_La_{CO2}=5.10h^{-1}$ at 400 rpm. The application of the proposed method to batch cultures of C. glutamicum ATCC 21253 revealed the maximum specific respiration rates of $q_{O2}=8.4\;mmol\;g^{-1}h^{-1}\;and\;q_{CO2}=8.7\;mmol\;g^{-1}h^{-1}$ in the middle of the exponential growth phase after 5 h of cultivation. When the cells changed from growth to lysine production due to the depletion of the essential amino acids theonine, methionine, and leucine, $q_{O2}\;and\;q_{CO2}$ decreased significantly and RQ increased. The respiration data exhibited an excellent agreement with previous cultivations of the strain [13]. This confirms the potential of the developed approach to realistically reflect the metabolic activities of cells at their point of sampling. The short-term influence of added threonine, methionine, and leucine was highest during the shift from growth to lysine production, where $q_{O2}\;and\;q_{CO2}$ increased 50% within one minute after the pulse addition of these compounds. Non-growing, yet lysine-producing cells taken from the end of the batch cultivation revealed no metabolic stimulation with the addition of threonine, methionine, and leucine.
Two malonate-specific enzymes, malonyl-CoA synthetase and malonamidase, were found in free-living cultures of Rhizobium japonicum, Rhizobium meliloti, and Rhizobium trifolii, that infect plant roots where contain a high concentration of malonate. Malonyl-CoA synthetase catalyzes the formation of malonyl-CoA, AMP, and PPi directly from malonate, coenzyme A, and ATP in the presence of $Mg^{2+}$ Malonamidase is a novel enzyme that catalyzes hydrolysis and malonyl transfer of malonamate, and forms malonohydroxamate from malonate and hydroxylamine. Both enzymes are highly specific for malonate. These results show that Rhizobia have enzymes able to metabolize malonate and suggest that malonate may be used in symbiotic carbon and nitrogen metabolism.

A bench-scale feasibility study was conducted with solid farming feed to evaluate a treatment process for microbiological removal of nitrogen (N) and phosphorus (P). Strains, Bacillus sp. CK-10 and Bacillus sp. CK-13, were originally isolated from water samples of shrimp farming pond. Simultaneous removal of N/P in marine media was monitored in the co-cultures, CK-10 and CK-13. As the results, $400\;{\mu}M\;NH^{+}_4$ and $400\;{\mu}M\;NO^{-}_2$ were eliminated within 12 hours and $NO^{-}_3$ within 36 hours, and $500\;{\mu}M\;PO^{-3}_4$ was completely disappeared within 36 hours from the media. Cultures of CK-10 and CK-13 were applied for removal of N/P leached from shrimp farming fred. HPAEC-PAD system was used to analyze sugars in farming feed, resulting in resolution of various sugars including glucose, galactose, galatosamine, mannose, and fucose. $0.2\%$ (w/v) Pulp densities of the farming feed contained approximately $33.3\;{\mu}M\;NH^{+}_4,\;12.9\;{\mu}M\;NO^{-}_2.\;81.5\;{\mu}M\;NO^{-}_3\;and\;248\;{\mu}M\;PO^{-3}_4$ which could dissolved within 72 hours of leaching in aqueous solution followed by bacterial removal. Complete bacterial removal of N/P was achieved within 84 hours at $0.2\%$ of the feed in co-cultures, whereas single cultures removed to incompletion of N/P during the incubation period. This work demonstrated that test cultures, CK-10 and CK-13 showed effective removal of N/P derived from shrimp farming feed.

Lab scale experiments were conducted in order to assess the applicability of $CaCO_{3}$-alginate beads as neutralizer for the high cell density cultivation and prepare the direct vat inoculation cultures of isolated sourdough lactic acid bacteria. With increasing the amount of bead and decreasing the diameter of bead in acidic solution, the neutralizing effect of $CaCO_{3}$-alginate bead became higher. In batch process with $CaCO_{3}$-alginate beads, Lactobacillus amylovorus DU-21 isolated from sourdough showed the highest viable cell counts and optical density in MRS broth. The values of viable cell counts and optical density were 9.996 log CFU/mL and 3.97, respectively. Experiments on the conditions which increase viability during lyophilization were carried out and the following results were obtained; 15% glycerol revealed the high cryoprotective effect on the concentrated cultures during lyophilization among the two cryoprotective agents. Consequently, $CaCO_{3}$-alginate beads and 15% glycerol were found to be useful not only to cultivate Lactobacillus amylovorus DU-21 but also to preserve strain.

The influences of inoculum size, pH, and medium composition on mycelial growth and exopolysaccharides (EPS) production were investigated in shake flasks and in a bioreactor. The optimum inoculum size for both mycelial growth and EPS production was identified to be 10% (v/v) in shake flask cultures. The optimal initial pH for mycelial growth and EPS production in shake flask cultures were found to be 5.0 and 7.0, respectively. However, the optimal pH was 5.0 for both mycelial growth and EPS production in bioreactor cultures where the pH was regulated. The optimal mass ratio of the two major carbon sources, glucose to dextrin, was 1:4. The optimal mass ratio of the two major nitrogen sources, yeast extract to soy tone peptone, was 2:1. When 500 mg $1^{-1}$ of $MnSO_4-5H_2O$ was added to the bioreactor culture, both mycelial growth and EPS production were enhanced by approximately 10%. Under the optimized conditions, a mycelial biomass of 9.85 g $1^{-1}$ and an EPS concentration of 4.92 g $1^{-1}$ were obtained in 4 days.
To investigate the effect of growth regulators and antioxidant in anther floating cultures of rice, anthers were cultured in liquid media supplemented with different growth regulators, and the effect of antioxidant mixture : (Sigma Chemical Co.) on callus induction and plant regeneration were investigated. N6 medium with 1 mg/L 2,4-D and 1 mg/L kinetin was the best for rice anther floating cultures, which showed 48.5% of callus induction and 6.8% of green plant regeneration. The callus induction was not affected by antioxidant mixture in liquid medium, and antioxidant mixture (250 mg/L) was effective for the reduction of brownish callus and improvement of plant regeneration Antioxidant mixture showed better effectiveness when it was supplemented to both media for callus induction and plant regeneration.

The study was carried out to establish an improved protocol for shoot organogenesis and plant regeneration from leaf explant cultures of Scrophularia buergeriana M. with the treatment of ethylene inhibitors [silver nitrate (AgNO$_3$), aminoethox-yvinylglycine (AVG), Cobalt chloride (CoCl$_2$)]. The regenerated shoots obtained from leaf explant cultures on MS medium containing 2 mg/l BAP, The additions of AgNO$_3$. AVG and CoC1$_2$ substantially improved the shoot regeneration frequency, at the optimal concentration of 7 mg/L, 7 mg/L, and 3 mg/L respectively, The regenerated shoots could be easily rooted with 0.1 mg/L IBA treatment. The noted plants were hardened and transferred to vermiculite with a 85% survival rate where they grew normally.

In a search for bacteria capable of degrading styrene better than previously isolated strains, bacterium IS-3 was isolated from activated sludge and found to be most closely related to Pseudomonas sp. Styrene degradation by this strain was tested in liquid cultures and polyurethane-packed biofilters. In liquid cultures, the rate of styrene degradation by this bacterium increased from 24.93 to $76.53\;{\mu}mol\;g^{-1}\;DCW\;H^{-1}$ for an initial mass range from 8.7 to $34.8{\mu}mol$. The maximum styrene elimination capacity was 580-635 $g/m^{3}\cdot$h at a space velocity (SV) of 50-200/h. The critical elimination capacities guaranteeing $95\%$ removal of the input styrene were determined to be 635, 170, and 38 $g/m^{3}\cdot$h, respectively, at SVs of 50, 100, and 200/h. Kinetic analysis revealed that the maximum styrene elimination velocity ($V_{m}$) for this biofilter was 1,000 g/m$\cdot$h, and the saturation constant ($K_{m}$) was 454 ppmv. Together, these results suggest that a polyurethane biofilter containing Pseudomonas sp. IS-3 could have potential practical applications for the effective removal of styrene gas.
