Background: Cnidium officinale Makino have been used in traditional medicine in Northeast Asia. Although gamma-ray mutagenesis has been used to develop breeding resources with novel characteristics, research on the radiation sensitivity of C. officinale Makino is limited. Hence, the optimal gamma-ray dosage for mutation breeding in C. officinale Makino was investigated. Methods and Results: Seedstocks were exposed to doses of gamma rays (5 Gy - 50 Gy), and subsequently planted in a greenhouse. After 30 days of sowing, the survival rates and growth decreased rapidly at doses above 20 Gy, while all individuals died at 50 Gy. The median lethal dose (LD50) was 25.65 Gy, and the median reduction doses (RD50) for plant height, number of stems, and fresh weight were 12.81, 9.32, and 23.26 Gy, respectively. Post-irradiation levels of malondialdehyde (MDA), peroxidase (POD), and chlorophyll in the aerial parts of the plant were quantified using spectrophotometry. Relative to the controls, the levels of MDA and POD increased, while the level of chlorophyll decreased at doses ≥ 10 Gy, indicating cellular damage. Conclusions: A dose of 20 Gy was found to be optimal for mutation breeding in C. officinale Makino.

This experiment was conducted to determine the essential oil content and the aromatic constituents in the leaves of Saururus chinensis Baill, Angelica dahurica Fischer and Cnidium officinale Makino. Volatile aromatic compounds in three aromatic medicinal plants were extracted with steam distillation extraction method and identified by GC/MS. Major aromatic compounds in Saururus chinensis Baill were 1,6-octadien-3-ol, 1, 3-benzodioxole, myristicin, ${\alpha}-cadinol$ and patchouene. Major aromatic compounds in Angelica dahurica Fischer were terpinolene, 3-carene, ${\beta}-caryophyllene$, ${\beta}-cubebene$, butylated hydroxy toluene, caryophyllene oxide, piperonal, and in Cnidium officinale Makino were aristolene, benzocycloheptene, ylangene, valencene, ${\beta}-cedrene$, satene, and menthofuran. Essential oil content was highest in Saururus chinensis plant.
Background: Cnidium officinale is a medicinal crop sensitive to high temperature. It is necessary to develop environment control technology that can reduce environmental stresses such as high temperature. This study was conducted to develop technology for stable production of Cnidium officinale by reducing damage owing to high temperature by applying shading treatment of varying duration, and structure. Methods and Results: Black shading nets were used from May to September or November; shading structures such as pillar, flat roof, and tunnel type structures were installed. Environmental changes, rate of photosynthesis, and growth characteristics were investigated. The shading treatment reduced temperature by 3℃. The rate of photosynthesis and yield with shading treatment were higher by 134% and 127%, respectively, than those with full sunlight. The ratio of shading area ranged from 50% to 71% according to the type of shading structure. The effect of environmental control on growth varied depending on the type of shading structure. Conclusions: The shading treatment reduced damage owing to high temperature, shading rate of 55% - 75% was recorded between the period May - September, and the flat roof type shading structure was considered the most suitable among shading methods.
This study was conducted to improve the productivety of Ligusticum chuanxiong HORT. and Cnidium officinale MAKINO by establishing the effects of cultivating conditions. Growth characteristics of underground part of L. chuanxiong under the different mulching treatment showed the highest growth and yield in black vinyl mulching treatment. The highest dry weight of underground parts per plant in mulching treatment was 47.9g at white shading treatment while the lowest one was 16.5g at black shading treatment. Growth characteristics of underground parts C. officinale under the different mulching treatment were the higher in black vinyl mulching rather than in rice straw mulching and white vinyl mulching. Dry weight of underground parts per plant of C. officinale under the different shading treatments was the highest(37.8g) under the white shading while it was lowest(12. 2g) under the black shading.

Background: Evaluation of transpiration is required for agricultural and environmental management applications, as crop yields and plant growth are primarily water limited. This study aimed to determine the transpiration and carbon accumulation of Cnidium officinale. Methods and Results: The transpiration of C. officinale was evaluated using weighing lysimeter. The relationship between transpiration and factors such as solar radiation, air temperature, and leaf area was assessed. Transpiration increased as the leaf area increased with the growth stage. Furthermore, daily transpiration per unit leaf area was 0.69 ± 0.16 g·cm-2·day-1 and there were no significant differences in daily transpiration during the cultivation period. The maximum transpiration was 620.6 g m-2·h-1 and diurnal changes in transpiration were highly correlated with solar radiation although the maximum transpiration was observed at the air temperatures of 20℃ - 26℃. The ratio of carbon accumulation to transpiration was 0.12%. Conclusions: Our results indicated that the transpiration of C. officinale is primarily regulated by solar radiation energy on clear days and that 97% of the water is discharged through transpiration for heat dissipation. Therefore, weighing lysimeters can measure transpiration accurately and may be useful in interpreting plant growth.
Field experiment for two years was conducted at Ulleung island to determine the optimum planting date and density of Cnidium officinale Makino. Early planting(Mar. 20) showed the highest yield by 2,880kg per ha as dry wt. Planting time after Mar. 20 was decreased. Late planting had higher occurrence of diseases and insect damage. Early planting had higher extract contents than late planting without violation of the Korean crude drug regulation such as ash contents. Although plants grew better in lower density, they had fewer growing points per plant which actually determine the yield components. Planting density did not affected extract contents in Cnidium officinale Makino. High density planting($35\times15cm$)yielded 33% than those of lower planting density($45\times25cm$).

The activity of phase II enzyme in the liver of mice was examined following the application of Cnidium officinale Makino aqua-acupuncture solution (COMAS). The application of COMAS $CV_{12}$ to mice for 7 days increased quinone reductase(QR) activity. The glutathione S-transferase(GST) activity was also increased with COMAS. A greater increase was observed in the glutathione(GSH) levels than in the QR and GST activities.

Evapotranspiration (ET) information is needed for many applications in agricultural and environmental resource management because crop yields, plant growth and physiological characteristics are primarily water limited. This study was conducted to evaluate the diurnal change of ET using electronic weighing lysimeter and to determine whether the ET of Cnidium officinale Makino could be manipulated through meteorological factors such as solar radiation, windy conditions and air temperature etc., Pot has a diameter of 35 cm and an height of 38 cm. A disturbed soil of sandy loam (coarse, mixed, mesic family of Dystric Fluventic Eutroudepts) within lysimeter has a mass of approximately 40.0 kg. In 2017, 10-minute recordings of data were used for measuring actual ET, and also evaluating a relationship between ET and meteorological factors during from 2 Aug. to 6 Aug. The maximum daily ET of Cnidium officinale was $44.04{\pm}3.949g$ per hour in lysimetric measurements. Diurnal changes of ET was highly correlated with solar radiation ($r^2=0.7778$) and followed by wind speed ($r^2=0.6400$). But on the other hand, air temperature was not consistent with ET ($r^2=0.2260$). This results imply that ET of Cnidium officinaele seems to be mainly governed by radiation energy in clear days, and approximately 40% of solar radiation is likely to be converted into ET. Therefore, weighing lysimeter can be used to accurately estimate actual ET and is expected to attract a great deal of attention to reliable application of water management in agriculture.
Mycoplasma-like organisms (MLO) were identified from three plants. The disease symptoms show stunting, yellowing and witche's broom on Cnidium officinale Makino collected from Ulreung Island, Bupleurum falcatum L. collected from Jangsu, and Plantago asiatica L. collected from Daegwanreung. The particles of MLO were observed in the phloem tissues by a Hitachi Hu-11E electron microscope. C. officinale infected with MLO was frequently observed in Ulreung Island.

This study was conducted to develop a somatic embryogenesis protocol for the Cnidium officinale Makino difficult to seed propagation. The immature flowers were used as explants. The concentration of a 2,4-D 1.0mg/L was found to be optimal concentration for induction of embryogenic callus and somatic embryos. Addition of 0.3mg/L, 0.5mg/L and 1.0mg/L to the embryo germination medium promoted somatic embryo germination. Among four concentrations, GA3 1.0mg/L were superior to others. Shoots were transferred to hormone-free MS medium after 2 months of culture in the dark. We obtained an optimized protocol for the regeneration of C. officinale.
