Monitoring of pH and dissolved oxygen in microorganism fermentation processes using a 24-well microplate (24-웰 마이크로 플레이트를 이용한 미생물 발효공정에서 pH 및 용존산소의 모니터링)

  • In this study, the sensing membranes for detection of pH and dissolved oxygen(DO) were prepared by immobilizing 6-aminofluorescein or ruthenium complex onto the sol-gel matrixes of GPTMS, MTMS, and TEOS and then recoated with the mixture of hydrophobic sol-gel and graphite for light insulation. The pH and DO sensing membranes recoated with the light insulation layer showed a higher sensitivity than those without light insulation layer. The sensing membranes were immobilized on the wells of 24-well microplate and used to monitor the fluorescence intensity for pH and DO in E.coli JM109 and P.pastoris X-33 fermentation processes. The change of the fluorescence intensity in the DO sensing membrane agreed with the growth patterns of microorganisms, that the membranes are valuable to monitor the DO in fermentation processes. In the case of pH monitoring, the fluorescence intensity has showed good correlation to the off-line pH data, that the pH membranes are valuable to monitor pH values in fermentations.

A Study on the Kinetics and Mechanism of the Hydrolysis of Dihydro-1,4-oxathiin Derivatives (Dihydro-1,4-oxathiin 유도체의 가수분해 Mechanism과 반응속도론적 연구)

  • The kinetics of the hydrolysis of dihydro-1, 4-oxathiin derivatives were investigated by ultraviolet spectrophotometry in H2O at $25^{\circ}C.$ A rate equation which can be applied over a wide pH range was obtained. The substituent effects on the hydrolysis of dihydro-1, 4-oxathiin derivatives were studied and the rate of hydrolysis was shown to be accelerated by electron accepting groups. Final product of the hydrolysis was 2-(2-hydroxyethylthio)acetoacet-anilide enol form. On the basis of rate equations derived and judging from hydrolysis products obtained and from general base effect and substituent effects, plausible mechanism of the hydrolysis in various pH range have been proposed. Below pH 3.5, the hydrolysis was initiated by the protonation and followed by the addition of water to 2-carbon. Above pH 10.0, the hydrolysis was proceeded by the addition of hydroxide to 2-carbon. In the range of pH 4.0∼10.0, the addition of water to dihydro-1,4-oxathiin is rate controlling step.

  • In evaluation of blast-induced vibration, peak particle velocity (PPV) is generally calculated by using attenuation relation curve. Calculated velocity is compared with the value in legal requirements or the standards to determine the stability. Attenuation relation curve varies depending on frequency of test blasting, geological structure of the site and blasting condition, so it is difficult to predict accurately using such an equation. Since PPV is response value from the ground, direct evaluation of the structure is impractical. Because of such a limit, engineers tend to use the commercial numerical analysis program in evaluating the stability of the structure more accurately. However, when simulate the explosion process using existing numerical analysis program, it's never easy to accurately simulate the complex conditions (fracture, crushing, cracks and plastic deformation) around blasting hole. For simulating such a process, the range for modelling will be limited due to the maximum node count and it requires extended calculation time as well. Thus, this study is intended to simulate the elastic energy after fractured zone only, instead of simulating the complex conditions of the rock that results from the blast, and the analysis of response characteristics of the velocity depending on shape and size of the fractured zone was conducted. As a result, difference in velocity and attenuation character were calculated depending on fractured zone around the blast source appeared. Propagation of vibration tended to spread spherically as it is distanced farther from the blast source.

Analysis of Bioconversion Components of Fermentation Hwangryunhaedok-tang (발효 황련해독탕의 생물 전환 성분분석)

  • Hwangryunhaedok-tang (HRT) is a traditional herbal medicine, which has been known as a useful prescription for anti-biotic, anti-inflammatory, anti-oxidative and immunosuppressive activity. In this study, the variation in the amount of eight bioactive components of Hwangryunhaedok-tang (HRT) and its fermentation HRT with Lactobacillus casei KFRI 127, Lactobacillus curvatus KFRI 166 and Lactobacillus confuses KFRI 227 was investigated via high-performance liquid chromatography coupled with diode array detection (HPLC-DAD). Simultaneous qualitative and quantitative analysis of eight bioactive components; geniposide, genipin, baicalin, wogonoside, palmatine, berberine, baicalein and wogonin was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. All calibration curve of standard components showed good linearity ($r^2$ >0.979). As a result, the geniposide amount was $15.52{\pm}0.19{\mu}/mg$ that as a main components in HRT. The wogonoside was decreased by 29.28~58.35% with Lactobacillus casei KFRI 127 and L. confuses KFRI 227 ($3.17{\pm}0.31{\mu}g/mg$ and $3.55{\pm}0.13{\mu}g/mg$) compared with the original HRT ($5.02{\pm}0.14{\mu}g/mg$). Otherwise wogonin was increased by 16.28~41.86% with Lactobacillus casei KFRI 127 and L. confuses KFRI 227 ($0.61{\pm}0.01{\mu}g/mg$ and $0.50{\pm}0.02{\mu}g/mg$) compared with the original HRT($0.43{\pm}0.00{\mu}g/mg$). HRT fermented with L. casei KFRI 127 and L. confuses KFRI 227 were evaluated as creating the changes in wogonoside to that aglycon wogonine. In the fermented HRT using Lactobacillus acidophilus KFRI 166, the genipin was only detected, among 3 species of fermentation strains. Thus, these results considered that the strains 166 were exhibited the remarkable changes in genipin.

Use of Oil Red O Staining Method in Non-Comedogenic Test for Cosmetics (화장품의 면포 비유발 평가에서 오일 레드 오 염색법의 응용)

  • It has been reported that certain ingredients added to cosmetics clog the skin pores and this can cause outbreaks of comedones which are the primary sign of acne leading to inflammatory acne. This research aims to establish objective evaluation criteria for non-comedogenic cosmetics suitable for acne prone skin. The research has been carried out to examine non-comedogenic test performed in foreign clinical institutions and to establish the evaluation method for detecting comedones outbreaks through repetitive closed back-patch test, Also, usability evaluation on face skin is performed additionally to the same subjects. The analysis of the comedones collected through repetitive closed back-patch test confirmed that the test products, moisturizer and sunscreen product, did not cause comedones. These results had no correlation with the analysis result of the comedones collected from face skin or visual evaluation of acne by Global Acne Grading System (GAGS) in face usability test. Additionally, Oil red O staining was performed on the collected comedones specimen for easy distinction of comedones from hair follicle in image analysis. The analysis result of stained specimen showed higher precision than that of non-stained specimen. This study established a new version of non-comedogenic test for cosmetics, whose objectivity and reliability were improved by inclusion of comedones staining step.

  • Next generation wireless communication systems require RF transceivers that enable multiband/multimode operations. Polar transmitters are known as good candidates for high data rate systems such as EDGE (Enhanced Data Rates for GSM Evolution), WCDMA (Wideband Code Division Multiple Access), and WLAN (Wireless Local Area Network) because they can obtain high efficiency by using efficient switched-mode RF power amplifiers. In this paper, we investigate the performance of a simple peak windowing scheme for the OFDM (Orthogonal frequency Division Multiplexing) polar transmitter, which requires no change of a receiver structure or no additional information transmission. The approach we employed is to apply the peak windowing scheme to the amplitude modulated signals of the polar transmitter to reduce the PAPR (Peak-to-Average Power Ratio). The BER (Bit Error Rate) and EVM (Error Vector Magnitude) performances are measured for various window types and lengths. The simulation results demonstrate that the proposed algorithm mitigates out-of-band distortion introduced by clipping along with PAPR reduction.

  • Local anesthesia procedure in pediatric dentistry using needle-tipped syringes is stressful and painful for the child patients The Syrijet Mark II (Mizzy Inc. USA) is a jet injection instrument, which uses high pressure to propel fluids into soft tissue without the use of a hollow neeldle, so it seems to be able to reduce the injection phobia of the child patients. The authors compared the Syrijet with the conventional syringe by assessing the pain level after local anesthetic procedures using CAS(color analogue scale) which was developed from VAS(visual analogue scale). The result showed that the pain was reduced by the use of syrijet. The advantages of syrijet were no use of neeldes, the reuse of the anesthetic cartridge, and the safety to nerves and vessels. The disadvantages were the large size, the high price, and the need of skill to use.

  • In-Ceram system is one of contemporary esthetic all ceramic restorations and has relatively high flexural strength. The purpose of this study was to evaluate the marginal fit of In-Ceram crown according to shoulder width and measuring position. In this study, twenty seven In-Ceram crowns devided into three groups, each prepared with shoulder width of 0.6mm, 0.9mm and 1.2mm. All specimens were cemented with glass ionomer cement. After cementation, specimens were measured marginal gap between the margin of the In-Ceram crown and the finishing line of resin master die by using a refrective microscope. Marginal gaps were recorded at the labial, lingual, mesial and distal measuring points on the resin master die. The results of this study were as follows: 1. Mean marginal fits of each group were $81.28{\mu}m$ in 0.6mm shoulder width, $70.78{\mu}m$ in 0.9mm shoulder width and $67.75{\mu}m$ in 1.2mm shoulder width. 2. There was significant difference between group 0.6mm and 0.9mm, 1.2mm.(p<0.05). 3. In comparison of marginal fit according to the measuring points, there was no significant difference.

Performance Evaluation of Long Span Bridge Columns Strengthened with High-Performance Glass Fiber (고성능 유리섬유로 보강된 해상장대교량 교각의 보강성능평가)

  • Researches and studies which have been conducted so far on external confinement of long span concrete columns have mainly concentrated on concentric loading. But, long span bridge concrete columns over the sea are mainly subjected to concentrated axial load, and at the same time lange amount of moment by eccentric load. This paper experimentally investigates the performance of externally confined high-strength concrete columns subjected to loading mechanism and evaluates the effectiveness of two confinement materials carbon fibre and high performance glass fibre. Twelve short columns with the same dimensions were cast and tested Six columns were reinforced with hoop bars, the remaining six columns were reinforced with spiral bars and wrapped with three layers of carbon failure and high performance glass FRP sheets. Test variables considered were the shape of internal reinforcement and strengthening materials according to loading location. The experimental results showed that eccentric load could obviously lower down the maximum failure load of FRP-confined concrete columns, compared with the columns under concentric load. And compared with the carbon FRP-confined reinforced concrete columns, high performance glass FRP-confined columns displayed a higher load capacity and ductility, when tested both concentrically and eccentrically.

A Study on the Career Decision-Making Patterns of Fisheries High School Students (수산고등학교(水産高等學校) 학생(學生)들의 직업결정형태(職業決定形態)에 관한 연구(硏究))

  • 1. Purpose of the study: This study was to investigate and analyse the career decision making patterns of students, which would enhance vocational guidance activities for the students. The specific purposes of the study are as follows. 1) To find how much the fisheries high school students are satisfied with the school. 2) To find out the factors which would influence the fisheries high school students in their career decision making. 3) To find out the person who plays the most important role in the career decision making of the students 4) To find out the patterns of behavior taken by the fisheries high school students in their career decision making. 2. Hypotheses: The following hypotheses were tested for this study. 1) There would be significant differences in the extent of school-satisfaction, depending on the class level and major. 2) There would be significant differences in the value criterion with which the fisheries high school students make decisions on their careers, depending on the grades, majors, and the levels of school-satisfaction. 3) There would be significant differences in the human factors which affect their decision-making on their career, depending on the class levels, majors, and the levels of school satisfaction. 4) There would be significant differences in the patterns of behavior they take when they make decisions on their careers depending on the grades, majors and the levels of school satisfaction. 3. Methods of the study: For the verification of the above hypotheses, a questionaire was developed by the researcher, and the questionaire was answered by the total 440 students from three fisheries high schools, located in the rural and urban areas, out of 8 fisheries high schools in Korea. The questionaire was composed of two major parts: school-satisfaction (10 items) and career decision making patterns (39 items). The career decision-making patterns include 16 value criterion items, 10 human category items, and 13 behavior category items. 4. Results of the study: 1) School satisfaction; There was no significant difference in school satisfaction depending on the class level and major. 2) Value criteria; a) Almost of the fisheries high school students think that the potential for advancement, job security, and salary are important factors for their job. b) The seafarer-students rated the salary, prestige of the occupation and leadership to be important than the nonseafarer-students. The nonseafarer-students rated the stabilization of their livelihood and opportunity to use their creative ideas than the seafarer-students. c) The students unsatisfied with their school emphasize the working environment, variety of work duties and leisure more than the satisfied ones. 3) Human factors: a) The fisheries high school students are affected most by their homeroom teachers, major teachers and their parents, but they are less influenced by their counselor. b) The seniors are more affected by their friends or classmates than the freshmen, and especially seafarer-students are more affected by major teachers. c) The persons, who are working in the occupation they are interested in, exert more influence on unsatisfied students than class-teachers do. 4) Behavior patterns; a) Most of them want test their abilities. b) The nonseafarer-students and seniors tend to depend on their own intuitions. c) The seafarer-students, the freshmen, and the satisfied with their school tend to discuss with other persons, and the unsatisfied tend to be influenced by the information that they collect for themselves.
