Isolation of Eudesmanolides Derivatives from the Flower of Chrysanthemum coronarium L. (쑥갓 꽃에서 Eudesmanolide 유도체 분리)

  • Two sesquiterpene lactones were isolated from the flowers of Chrysanthemum coronarium L. by silica gel chromatography and recrystalization. Their structures were determined using various spectroscopic data, and were identified as eudesmanolide derivatives douglanine and reynosin, respectively. This is reported for the first time in this plant. The cytotoxic activity of two compounds showed strong activities against human cancer cell lines such as A549, PC-3 and HCT 116.

Plant Regeneration Depending on Explant Type in Chrysanthemum coronarium L.

  • Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.

  • Three terpenoids were isolated from the leaves of Chrysanthemum coronarium L. by silica gel chromatography and recrystalization. The structure of compounds 1, 2, and 3, using various spectroscopic data, were identified as to be terpenoid derivatives of dihydrotulipinolide, loliolide, and $5{\alpha},8{\alpha}-peroxyergosterol$, respectively. The presence of these 3 is the first time reported in this plant. The cytotoxic activity of $5{\alpha},8{\alpha}-peroxyergosterol$ showed strong activities against human cancer cell lines such as A549, PC-3, and HCT 116.

Effects of Chrysanthemum coronarium L. on the Thermotropic Behavior of DPPC Liposomal Membrane

  • To understand the effects of the fraction from Chrysanthemum coronarium L. (CC), we prepared five different types of samples, denoted here as CCMM, CCMH, CCMEA, CCMB and CCMA. We studied the effects of these samples on the phase transition of liposomal membranes by high-sensitivity differential scanning calorimetry (nano-DSC). We used dipalmitoylphosphatidylcholine (DPPC) bilayers which make most stable liposomes among the other phosphatidylcholines. When the samples were added to the bilayers, the phase transition temperatures of DPPC liposomes incorporated with CCMH and CCMEA were decreased by 1.5 and 2^{\circ}C$, while the other three fractions showed less tendencies. The CCMH and CCMEA fractions markedly affected the thermotropic properties of DPPC liposomes, broadened and shifted the thermograms of DSC. It also significantly reduced the size of cooperative unit of the transition. In all cases, there was no change in enthalpy of transition within the concentration range of the CC fractions studied. We concluded that the incorporation of the CCMH and CCMEA into DPPC liposomes was preferentially located in the hydrophobic core of DPPC bilayers compared to the other three fractions CCMM, CCMB and CCMA. These results suggest that certain substances in CCMH and CCMEA fractions might have biologically significant effects on the fluidity of biological membrane.

  • Chrysanthemum coronarium L. contains various antioxidants such as beta-carotene, vitamins A and C, and polyphenols, and is known to have anti-inflammatory effects. Under the assumption that the PDRN contained in the extract can mediate the anti-inflammatory response, the mouse macrophages, RAW264.7 cells, were stimulated with LPS to induce the conversion to inflammatory cells, and then the addition of PDRN extracted from the extract was effective in inhibiting inflammation. It was analyzed whether there was The gene expression of IL-1β and TNF-α was used as an anti-inflammatory index, and the relative expression levels of each gene were confirmed by RT-PCR. As a result, RT-PCR confirmed the effect of PDRN-induced inhibition of inflammation in both IL-1β and TNF-α genes. Therefore, based on this study, it is considered to be a precedent data that can be developed as a treatment for inflammatory diseases, and it will be helpful in research and development of a treatment that can improve the anti-inflammatory mechanism.

Antioxidant Activities of Chrysanthemum coronarium L. Fractions on the Liposomal Phospholipid Membrane (인지질막 liposome에 미치는 쑥갓 분획물의 항산화 효과)

  • This experiment was designed to investigate the antioxidant effects of Chrysanthemum coronarium L. (CC) fractions on the liposomal phospholipid membranes. The sample CC was extracted and fractionated into five different types, methanol (CCMM), hexane (CCMH), ethylacetate (CCMEA), butanol (CCMB), and aqueous (CCMA) fractions. The antioxidant activities of CC fractions in oxidized dilinoleoylphosphatidylcholine (DLPC) liposomes were examined by spectrophotometry measuring conjugated dienes. The oxidation indices of five CC fractions exhibited weaker antioxidant activities than that of BHT in oxidized DLPC liposomes, however, showing much similar antioxidant activities of $\alpha$-tocopherol in the oxidized DLPC liposomes, which is known as a potent antioxidant. Among CC fractions, CCMM and CCMA in oxidized DLPC liposomes showed rather effective than $\alpha$-tocopherol after 2 h. These results strongly indicate that bioactive substances in CC fractions have a kind of function as potent antioxidants against biomembrane oxidation.

Quality Characteristics of Sulgidduk with Added Ssukgat (Chrysanthemum coronarium L. var. spatiosum) Powder (쑥갓가루를 첨가한 설기떡의 품질 특성)

  • This study was performed to investigate the quality characteristics of Sulgidduk added ssukgat. Sulgidduk was prepared with freeze-dried ssukgat powder, rice flour, sugar, and salt. The Sulgidduk was made with various amounts(0, 1, 3, 5, 7%) of added ssukgat powder. The moisture content, Hunter's color value, dietary fiber, texture profile analysis, and sensory characteristics of ssukgat Sulgidduk were examined. The addition of ssukgat powder has a tendency to decrease the water content of Sulgidduk. As the amount of ssukgat powder increased, the lightness (L) and redness (a) decreased, and the yellowness (b) increased. In texture profile analysis, the hardness decreased with increasing ssukgat powder content. Chewiness, cohesiveness and springiness did not show significant differences with different amounts of ssukgat powder. The results of sensory evaluation showed that the sweetness, softness, and overall acceptability scores were the highest with Sulgidduk containing 3% ssukgat powder. In conclusion, these results show that the Sulgidduk with 3% of ssukgat powder contained dietary fiber is the best.

The Effect of Daylength, Shading and Irrigation on the Flowering of Chrysanthemum coronarium L. (쑥갓의 화아분화에 미치는 일장, 차광 및 관수 처리의 영향)

  • In order to study flowering of Chrysanthemum coronarium, several investigations on the daylength, shading, irrigation and plant growth regulator treatment were carried out. As daylength was treated for 12, 14 and 16hrs, flowering was accelerated and plant height was increased. Leaf number, length and width were decreased by the increased daylength. Sensitivity to daylength and flower development were accompanied by the plant growth, especially the effect of long day treatment was appeared remarkably when the number of leaves was more than 8 leaves. The minimum days of long day treatment for flowering was more than 10 days. Flowering was delayed by 65% shading treatment, over irrigation.
