In Korean ginseng, Panax ginseng C.A Meyer, it was difficult to isolate chloroplast DNA with classical methods, because of the high polysaccharide content of ginseng chloroplast The simple and efficient method of chloroplast DNA isolation from ginseng leaves has been developed by motificalion of recently advanced methods. Also, it can be successfully applied to ctDNA isolation of Chinese cabbage, radish, petunia tobacco as well as ginseng. Isolated chloroplast DNA from ginseng was digested with various restriction endonucleases. It was estimated that the molecular weight of Korean ginseng chloroplast DNA was about 142 kb. There was no difference in restriction endonuclease digestion patterns between two variants of Korean ginseng, which are Jakyung-Jong (violet-stem variant) and Hwang- sook-Jong (yellow-berry variant).

Along the developmental processes in higher plants, chloroplast follows a major route of development which is proplastid - etioplast - chloroplast. Development of chloroplast can be determined according to the expressions of the genes coded in the chloroplast DNA as well as in the chromosomal DNA. Most of the processes occuring in proplastid and etioplast seems to be coded in the chromosomal DNA, which the development of chloroplast from etioplast upon the exposure of plants to light is determined by harmonious expressions of the genes in the chloroplast and the chromosome.

A cDNA clone encoding chloroplast triosephosphate isomerase (TPI-cp) was isolated from strawberry fruit cDNA library. Sequence analyses indicated that the cDNA contains an open reading frame of 314 amino acids (33.5 kDa) composed of a transit peptide (59 amino acids) in amino terminal region and mature protein (255 amino acids). The existence of transit peptide in the deduced amino acid sequence implies that it encodes a chloroplast isoform. The protein sequence is more similar to other plant chloroplast isoforms than cytosolic isoforms. RNA blot analysis indicated that its expression is ubiquitous in examined five tissues, flowers, leaves, petioles, roots and fruits, and shows differential pattern according to fruit ripening. Genomic DNA blot analysis showed that TPI-cp is encoded by multiple genes in strawberry. Through sequence comparison and phylogenetic tree construction, TPI-cp is distinctively grouped into dicot and chloroplast isoforms.

Plant chloroplast DNA exists as an unique circular structure in which large single copy(LSC) region and small single copy (SSC) region are separated by large inverted repeat sequences (IRS). It has been known that the unique existence of inverted repeat sequences in chloroplast DNA has no relation with the stability of the chloroplast DNA, but causes the inversion between inverted repeat its biological significance has not been understood so far. In rice, several gene clusters have been cloned and sequenced which contain ribulose-5-biophosphate car-boxylase large subunit (rbcL). Especially, one rbcL gene is linked with rp12 gene which is located in the IRS region in one of the gene clusters. By comparison of nucleotide sequence, the two genes are found to be linked through 151 bp repeat sequence which is homologous to the rp123 gene in IRS region. The repeat sequence is found to be located 3' downstream of rfcL gene and near psbA gene in LSC region. The existence of these repeat sequences and the presence of gene clusters caused by the gene rearrangement thorough the repeat sequence provide a possible which is found to be dispersed chloroplast DNA provide the model system to explaine the heterogeneity of the chloroplast DNA in rice in term of gene rearrangement.
Previously we reported the migration and rearrangement of a chloroplast gene cluster into mitochondria. The exact genomic locations of the clusters, modes of the gene rearrangement and mechanisms of the interorganellar migration of the clusters have yet to be understood. The detailed analysis needs to include a larger region of DNA surrounding each cluster. To study DNA rearrangement and migration in more detail a cosmid library was constructed using the total rice genomic DNA including nuclear, chloroplast and mitochondrial DNA. From this cosmid library, a sub-library was obtained by selecting the clones hybridized to various regions of chloroplast DNA. According to the hybridization pattern 136 clones from the sub-library were classified into 29 groups. Detailed analysis of these clones revealed that in addition to authentic chloroplast DNA, the clones contain its homologs resulted from rearrangement and mutation. We analyzed two clones in detail, which contain different rp12 homologs resulted from rearrangement and/or migration, respectively.
There is a considerable difference in morphological traits between Bokbunja cultivated in Korea (KCB) and Korea native Rubus coreanus, contrary to the conviction that the cultivated Bokbunja is the domestication of R. coreanus. To infer the phylogenetic relationship of KCB with other Rubus species, we compared the chloroplast DNA spacers of KCB with those of several Rubus species including black raspberry, R. occidentalis. The three chloroplast DNA spacers, atpB~rbcL, trnL~trnF, and trnT~trnL, were amplified using the specific primer pairs and converted to Single Strand Conformational Polymorphism (SSCP) markers. The SSCP makers of the chloroplast DNA spacers showed a considerable variation both within and among Rubus species. In the phylogenetic tree generated by the SSCP markers, KCB accessions were located in the same clade with R. occidentalis, but R. coreanus accessions in the different clade. Also, in the phylogenetic tree by the nucleotide sequences of the chloroplast DNA spacer trnL~trnF, KCB located in the same clade with R. occidentalis but not with R. coreanus. These results suggest that the three KCB accessions share higher similarity with R. occidentalis than with R. coreanus in the three chloroplast DNA spacers.

We performed phylogenetic analyses of a total of 21 acessions covering 5 species in the Korean Trigonotis and one outgroup species using nuclear ribosomal ITS and chloroplast rbcL, matK, ndhF sequences. Outgroup were chosen from the closely related genus Lithospermum zollingeri. Both parsimony and Bayesian Inference methods were used to reconstruct the evolutionary history of the group. The evidence collected indicated that phylogenetic relationships among Korean Trigonotis species are unresolved based on nuclear marker (ITS), as the same as based on separated chloroplast sequences. While the phylogenetic relationships of Korean Trigonotis species almost clearly were resolved in combined chloroplast sequences. Thus, the members of Trigonotis coreana can be distinguished to the members of Trigonotis peduncularis in combined cpDNA sequences and Trigonotis nakaii was treated as a synonymed to Trigonotis radicans var. sericea. In addition, the MP and BI analysis showed Trigonotis icumae as sister of the remained Korean Trigonotis species based on combined molecular markers (BI: PP = 1).
한국에 분포하는 두릅나무과 식물 대부분은 중요한 약용 식물로 경제적인 가치가 크다. 본 연구는 분자적 방법인 엽록체 DNA 바코드 염기서열 분석을 통해 한국에 자생하고 있는 두릅나무과 식물 14종 전체의 속 및 종간 유연관계를 파악해 보고 이를 구별할 수 있는 마커를 개발하기 위해 수행되었다. 국제 생물 DNA 바코드 컨소시엄(CBOL, the Consortium for the Barcode of Life)이 DNA barcoding marker로 제안한 엽록체 DNA 7영역의 염기서열을 분석한 결과, psbA-trnH영역에서 가장 많은 삽입, 결실 및 염기치환이 나타났으며 조사된 한국의 두릅나무과 식물 14종 모두 구분 될 수 있었다. 또한 각각의 영역에서 특정 속과 종만이 지니는 특이적인 염기서열을 찾을 수 있었다. 인삼의 경우 중국삼과 한국삼의 염기서열에는 차이가 전혀 없었다. 7영역을 모두 유합하여 작성한 계통수에서는 통탈목이 특이성을 나타내며 가장 기부에 분계조를 형성하였다. 두릅나무속과 인삼속은 자매군을 형성하였고, 오갈피속 5 종 역시 서로 높은 유연관계를 나타내었다. 결론적으로 한국에 자생하는 14종의 두릅나무과 식물들이 모두 엽록체 DNA 바코드 마커 개발을 통해 동정이 가능함을 확인하였다.
In order to acquire information on chloroplast DNA markers to evaluate the genetic diversity of evergreen broad leaved plants, we investigated the intraspecific variation of cpDNA in eight non-coding regions of nine species commonly distributed in East Asia. Although no variations were detected in psbA-trnH, rpoB-trnC, rpl16 and atpB-rbcL regions, a relatively large amount of intraspecific variations was detected in the psbC-trnS, rps16 and trnL-F regions. These results suggested that these three cpDNA markers are suitable to assess genetic diversity of the species investigated in this study. In contrast, intraspecific variations were detected in seven taxa except Hedera rhombea and Neolitsea aciculata. Neolitsea sericea and the taxa of Quercus had many polymorphic sites.
Objectives : The application of polymerase chain reaction (PCR) for the discrimination of the herbal medicine Leonuri Herba (Leonurus japonicus) was evaluated by the comparison of the DNA sequence with Lamiaceae herbal medicine. Method : Genetic analysis showed that phylogenetic tree and comparing sequences through the DNA analysis of rbcL (ribulose-1, 5-bisphosphatecarboxylase) region and trnL-F (tRNA-Leu, trnL-trnF intergeni cspacer, and tRNA-Phe) region of chloroplast DNA from six Lamiaceae sold in market. And we developed IMCF and IMCR primers in order to distinction Leonuri Herba in six Lamiaceae using rbcL and trnL-F sequences. Results : Genetic analysis showed that six Lamiaceae showed individual group on phylogenetic tree. PCR amplification product of Leonuri Herba and another five Lamiaceae were developed for amplification of a 281 bp sequence and the specific PCR amplification of a 460 bp sequence that was exclusive to Leonuri Herba was designed using IMCF and IMCR primers. Conclusion : PCR analysis based on the chloroplast DNA sequences allows the discrimination of Leonuri Herba-based medicine.
