오리나무 수피엑스의 위염 및 위궤양에 대한 효과 (The Effect of Alnus japonica Cortex Extract on Gastric Lesion and Ulcer of Rats)

  • Alnus japonica cortex has been used as antidiarrhea, antihemorrhage and the remedy of indigestion. This study was performed to investigate the effectiveness of the methanol extracts of the Alnus japonica cortex on the gastric lesion and ulcer. The methanol extract was fractionated with hexane, chloroform and butanol, followed by bioassay on antigastritic and antiulcer activity. The methanol extract showed low acute toxicity with minimum lethal dose of more than 5000 mg/kg, p.o. in mice. The chloroform and the butanol fraction reduced gastric lesion in HCI. ethanol induced gastritic model. On gastric secretion in pylorus ligated rat, the hexane and chloroform fraction decreased the volume and acidity. The butanol fraction had significant inhibitory effects on aspirin and Shay's ulcer. The butanol fraction showed a tendency to inhibit the decrease of mucin secretion due to ingestion of absolute ethanol.

홍경천의 면역활성이 증진된 용매별 분획 추출물 (Improvement of Immune Activities of Rhodiola sachalinensis A. Bor. by Serial Solvent Fractionization)

  • This study was performed to compare the effects of immuno-modulating activities of Rhodiola sachalinensis A. Bor. fractionized by consecutive solvent separation. The Cytotoxicity of all fractionized extracts on human kidney cell (HEK293) was lower than crude extracts. Generally, the butanol and chloroform extracts showed less cytotoxicity on about 10.07% and 9.67% than the crude extracts. For human immune B and T cell growth, chloroform fraction showed the highest cell growth compared to the control. The secretion of cytokines (IL-6, $TNF-\alpha$) on human B and T cells were increased by adding chloroform extracts. Also, NK cell growth was significantly improved up to nearly 30% by adding the supernatant of B cell medium grown with the chloroform fraction. It was also found that chloroform fraction could yield higher nitric oxide production from macrophage than untreated control cells. Differentiation of HL-60 cells was increased up to 131.9% after treatment with chloroform fraction extracts, compared to the control. These results indicate that the chloroform fraction of R. sachalinensis have high immune activation activity than others fractions and the crude extracts, implying that this chloroform fractions could be used a new functional material.

감초 추출물로부터 항균성 물질의 분리 및 동정 (Isolation and Identification of Antimicrobial Compounds from Licorice Extracts)

  • 식품부패미생물에 대한 감초추출물의 항균력을 확인하고 유효성분을 분리한 후 그 구조를 확인 하였다. 감초추출물은 대표적인 항균제인 메타중아황산칼륨과 myconazole과 비슷한 항균 활성을 나타내었다. 용매분획을 실시한 결과 chloroform fraction이 가장 항균활성이 높은 것으로 나타났다. 이에 chloroform분획을 silica gel column chromatography를 통해 분리하여 항균활성이 가장 큰 분획물인 KC를 분리하였다. 이 화합물은 ESI-MS 와 $^1H$-NMR, $^{13}C$-NMR 결과로부터 활성물질의 구조를 동정하였으며 그 결과 KC는 (R)-glabridin인 것으로 확인하였다.

Monascus purpureus P-57 변이주가 생산하는 홍국색소의 항산화효과 (Antioxidant Activity of Monascus Pigment of Monascus purpureus P-57 Mutant)

  • Monascus purpureue KCCM 60016으로부터 색소생성 이 우수한 변이주 P-57이 생성한 홍국색소의 항산화 활성을 조사하였다. 홍국색소는 chloroform에 아주 잘 추출되며, 특히 hexane에서는 황색색소가 특이적으로 높게 추출되었다. DPPH radical소거 효과는 hexane분획, chloroform 분획, ethyl acetate분획, butanol 분획, water 분획의 순으로 높게 나타났다. Xanthine oxidase저해효과는 hexane분획, chloroform분획, ethyl acetate 분획의 순으로 높게 나타났으며, hexane 분획물의 경우 5 ppm에서 $41.2\%$, 50 ppm에서 $82.4\%$로 높은 저해율을 보였으며, 저해기작은 비경쟁적 저해였다.

지유 분획의 Candida albicans에 대한 항균효과 (Antifungal Effect of Sanguisorba officinalis L. fractions on Candida albicans)

  • We have studied the antifungal effect of 19 medicinal plants with paper disc diffusion method against candida albicans. As a result, Sanguisorba officinalis L., Cinnamomum cassia, Rheum coreanum, Perilla frutescens and Eugenia caryophyllata have been found to be effective against C. albicans. Among these, Sanguisorba officinalis L. was most effective at 24 hours and 48 hours. Its clear zone diameter was 17 mm for 24 hours and 16 mm for 48 hours. The antifungal activity of the solvent fraction of Sanguisorba officinalis L. by n-hexane, chloroform, ethyl acetate and n-butanol was the best for the chloroform fraction of 28 mm for 24 hours and 18 mm for 48 hours. The MIC concentration of the chloroform fraction was $80{\mu}g/50{\mu}l$ at 24 hours and $240{\mu}g/50{\mu}l$ at 48 hours.

Cytotoxic Effects of Chloroform Extracts and Fraction from Cornis fructus on Cancer Cell Lines

  • Cornis fructus were extracted by successive extractions and then fractionated with chloroform extract to get active fractions. This study was performed to determine the cytotoxic effect of chloroform extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. All extracts did not exhibit cytotoxicity in HIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. (omitted)

  • Yields of various solvent extracts for Zanthoxylum schinifolium and Z. piperitum leaves were higher in ethanol extract layer. Ethanol Extract in sancho and chopi leaves was fractionated with different solvents, such as hexane, chloroform, ethyl acetate, butanol and water, yields of hexane fraction was higher. In the solvent extracts using methanol, ethanol, hexane, chloroform, and water, ethanol extract showed the most effective antimicrobial activities. Ethanol extract in sancho and chopi leaves was fractionated, the most antimicrobial activities of sancho leaf were ethyl acetate fraction, and chopi leaf were chloroform fraction.

췌장 콜레스테롤 에스터레이즈 저해제인 초두구 추출물의 혈중 콜레스테롤 저해효과 (Plasma Cholesterol-Lowering Effects of Alpiniae katsumadaii Extract as an Inhibiter of Pancreatic Cholesterol Esterase Activity)

  • 췌장에서 분비되는 cholesterol esterase(CEase)와 소장의 acyl coenzyme A: cholesterol acyltransferase(ACAT)는 흡수된 콜레스테롤을 다시 ester화하는데 관여한다. 한방재료 중 건위, 진토 및 수렴지사작용 등을 가지는 초두구(Alpiniae Katusumadaii Semen)의 에탄올 추출물이 in vitro에서 cholesterol esterase활성에 대하여 강한 저해작용을 보였으며 에탄올추출물의 용매분획 중 클로로포름분획이 다른 분획들, 즉 에틸 아세테이트분획, 부탄올분획 및 물 분획보다 강하였다. 초두구의 클로로포름분획이 고 콜레스테롤식이를 섭취시킨 rat에서 혈중 콜레스테롤치를 저하시키는 효과가 있는지 측정한 실험에서 chloroform분획을 100 mg/kg 및 150 mg/kg 섭취시켰을 경우 각각 총cholesterol 함량이 11.8% 및 20.8% 감소하였으며 HDL-cholesterol 함량 및 동맥경화지수는 정상군에 가깝게 회복된 것으로 나타났다. 또한 초두구 chloroform분획의 독성검사 결과 mouse에서 $LD_{50}$은 1,625mg/kg으로 산출되었다.

Sarcoma-180 세포를 이용한 in vivo에서 감잎의 항암효과 (Antitumor Effect of Persimmon Leaves in vivo using Sarcoma-180 Cells)

  • 감잎의 시료 중 항돌연변이 효과와 암세포 증식 억제효과가 컸던 감잎의 헥산, 클로로포름 획분 및 감잎 탄닌의 항암효과를 Sarcoma-180 종양세포를 이용하여 살펴 보았다. Sarcoma-180 종양세포에 대한 시료의 세포 독성작용을 관찰한 후 세포에 큰 영향을 주지 않는 범위내에서 고형암 성장저지 효과 및 수명 연장 효과를 살펴 본 결과 감잎의 헥산 획분이 59.6%로 가장 높은 고형암 성장 저지율을 나타냈으며 수명 연장 효과도 대조군의 경우 17.4일인데 반해 헥산 획분을 투여한 경우 생존일수가 23.6일로 가장 큰 수명 연장 효과가 관찰되었다. 면역 관련 장기 중 하나인 비장의 체중에 대한 중량 변화는 대조군의 경우 0.6%인데 반해 감잎의 헥산 및 클로로포름 획분 그리고 감잎 탄닌의 경우 0.7%, 0.8%, 0.8%로 약간의 증가를 나타내었다.

측백나무 열매 추출물의 항균활성 (Antimicrobial Activities of Organic Extracts from Fruit of Thuja orientalis L.)

  • This study was carried out to investigate the antimicrobial activities of organic extracts obtained from the fruit of Thuja orientalis L. The native fruits in Korea were collected and extracted by 80% ethanol, and the extract was sequentially fractionated with n-hexane, chloroform, ethylacetate, and butanol. The fraction yields of n-hexane, chloroform, ethylacetate, butanol and water of ethanol extract were 10.15%, 10.05%, 1.45%, 45.35% and 27.55%, respectively. n-Hexane-soluble fraction showed the highest antibacterial activity against gram positive bacteria, while the chloroform, ethylacetate, butanol and aqueous fractions did not show any antibacterial activity. Minimum inhibitory concentration (MICs) on Staphyloycoccus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction were $100\;{\mu}g$, $500\;{\mu}g$ and $50\;{\mu}g$/disc, respectively. The antibacterial activity was not destroyed by heating at 80, 100, $120^{\circ}C$ for 30 min and was not affected by pH. In the inhibitory test against the Staphylococcus aureus, Bacillus subtilis and Sateptococcus pneumoniae, n-hexane-soluble fraction showed potent growth inhibition at the concentration of 0.1 and $0.5\;{\mu}g/mL$ for 12~24 hours and n-hexane-soluble fraction did not show any mutagenic activity.
