Biological Activities of Magnolia denudata Desr. Flower Extracts (목련(Magnolia denudata Desr.) 꽃 추출물의 생리활성)

  • The antioxidant, antiproliferation, and nitrate synthesis inhibitory effects of Magnolia denudata extracts (ME) were evaluated. The ME was extracted with 70% (v/v) ethanol and fractionated with solvents of hexane, chloroform, ethyl acetate, n-buthanol and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid contents of 427.10 mg garlic acid eq/g and 356.05 mg catechin eq/g, respectively. The ethyl acetate fraction showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity with a 50% inhibition concentration ($IC_{50}$) of 0.20 mg/mL and total antioxidant activity was 0.90 mg AA eq/100 mg. From the results of cytotoxic effects of HCT116, NCL-H460, and HepG2 human cancer cells by MTT assay on the ME and its solvent fraction, chloroform fraction showed the highest cytotoxic effect ($IC_{50}$ value: 0.14, 0.37, and 0.41 mg/mL, respectively). Nitrate synthesis inhibitory effect of ME and its solvent fractions on nitric oxide synthase activity in LPS stimulated RAW 264.7 cells were decreased in dose-dependent manners, and $IC_{50}$ value of hexane and chloroform fractions were 0.39 and 0.49 mg/mL, respectively.

Antioxidant Activities of Extract with Water and Ethanol of Perilla frutescens var. acuta kudo Leaf (차조기(Perilla frutescens var. acuta kudo) 잎의 물과 에탄올 추출물의 항산화 활성)

  • This study was conducted to examine antioxidant activities of Perilla frutescens var. acuta leaf. For the this purpose, DPPH radical scavenging activity, lipid oxidation inhibition, SOD-like activity, and xanthine oxidase inhibitor activity of water extract, ethanol extracts (30, 50, 70, and 95%) and the fractions obtained from these extracts were determined. The electron donating abilities of the chloroform fraction obtained from the 70% and 95% ethanol extracts were 50%, and that of the ethyl acetate fraction for all of the extracts was above 75%. In particular, the electron donating ability of the ethyl acetate fraction of the 70% ethanol extract showed the greatest activity with 200.5 ppm of $RC_{50}$ value. The 70% ethanol extract was most effective to inhibit the automatic oxidation of linoleic acid at $40^{\circ}C$ storage. The highest inhibition effects appeared in the chloroform and ethyl acetate fractions of the water extract, and the 30, 50, and 70% ethanol extracts, and the highest lipid oxidation inhibiting effect of the 95% ethanol extract occurred in the hexane and acetate fractions. The SOD-like activity of the water extract was 30.3%, and the activities of the various concentration of ethanol extracts were 28-32% and the activity of the 70% ethanol extract was the highest. The SOD-like activity of the ethyl acetate fraction of the 70% ethanol extract was highest with 1,549.0 ppm of $RC_{50}$ value. Xanthine oxidase inhibition activity was greatest in the water extract and the activities of the ethanol extracts were 36-41.2%. The xanthine oxidase inhibition activity of the ethyl acetate fraction of the water extract was highest. In summary, we found that electron donating ability, lipid oxidation inhibition, and SOD-like activity of Perilla frutescens var. acuta leaf were greatest in the ethyl acetate fraction of the 70% ethanol extract, and xanthine oxidase inhibition activity was highest in the ethyl acetate fraction of the water extract.

Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants (유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색)

  • This study was designed to investigate antioxidative activity of substances isolated from 25 kinds of useful plants resources toward free radical and human low density lipoprotein(LDL). Methanol extracts of Oenothers odorate had the highest antioxidative activity similar with ${\alpha}$-tocopherol. Methanol extracts of Oenothers odorate was extracted again by the ethylacetate. The ethylacetate soluble acidic fraction obtained from methanol extract of Oenothers odorate showed highest activity toward human LDL. Each fraction was purified through Sepadex LH-20 chromatography by elution of chloroform-methanol mixture (90:10 v/v). Fraction, F-2 obtained from Oenothers odorate showed at highest levels of electron donating activity. Fraction, F-2 was identified as 3,4-dihydroxybenzoic acid and 3-hydroxycinnamic acid.

Antioxidant Activities of Fractions from Sedum sarmentosum

  • This study was conducted to evaluate the antioxidant activity of each fraction from Sedum sarmentosum. Antioxidant activity of each fraction was measured using the DPPH radical assay, the ferric thiocyanate (FTC) method, and thiobarbituric acid (TBA) method. The antioxidant activities were then compared with that of BHT(synthetic antioxidant). The ethyl acetate and butanol fractions were found to have significant DPPH radical scavenging activity, with scavenging potencies showing 90.61 % and 87.02%, respectively. Total phenolic compound contents, determined according to the Folin-Denis method, were found to be in the order of ethyl acetate>butanol>ethanol>chloroform>aqueous fraction. From the results, we have been able to establish a positive correlation between the antioxidant activity and the total phenolic compound content of the sample. The antioxidant activity in a linoleic acid system was measured using the ferric thiocyanate (FTC) method and thiobarbituric acid (TBA) method. The ethyl acetate fraction had the highest antioxidant activity among the tested fractions. On the basis of these results, the ethyl acetate fraction provided equivalent or higher antioxidant activity as compared to BHT. These results suggest that Sedum sarmentosum is a potentially useful antioxidant for foods, cosmetics, and medicine.

Cytotoxic Activity of Methanol Fractions and Solvent Extracts from Houttuynia cordata $T_{HUNS}$ (IX) on Various Cancer Cells (어성초 용매추출물과 메탄올 분획물의 암세포주에 대한 세포독성)

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata T/sub HUNB/ extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 ㎍/㎖ concentration of methanol extract (63.81 %) of Houttuynia cordata T/sub HUNB/ was shown significantly antitoxic activity on A549 cell lines. The order of cytotoxicity fractions of methanol from Houttuynia cordata T/sub HUNB/ extracts against cancer cell lines in vitro is as follows : hexane fraction layer > chloroform fraction layer > ethyl acetate fraction layer > buthanol fraction layer > water fraction layer. These results suggest that the hexane fraction of methanol extract from Houttuynia cordata T/sub HUNB/ extract may be a valuable choice for the development of antitumor agents.

Anti-inflammatory Activity of Opuntia ficus-indica (손바닥선인장의 항염증 활성)

  • Cactus (Opuntia ficus-indica var saboten Makino) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. We previously found that the ethanol extract of cactus stem showed anti-inflanunatory action. This investigation was designed to isolate the active fraction of anti-inflanimatory action from cactus stem by solvent extraction and colunm chromatography. Carrageenan-induced paw edema in rats and acetic acid-induced writhing test in mice were used as animal models to search anti-inflammatory and analgesic activities. respectively. The ethanol extract of cactus stem was consecutively extracted with hexane, ethyl acetate, and n-butanol. The hexane fraction was the most effective in carrageenan-induced paw edema, and then was separated in colunm chromatography of silica gel by the elution with hexane/ethyl acetate mixture. The most effective fraction 1 was separated in a second colunm chromatography by eluting with hexane/diethyl ether mixture. The most effective fraction 1-5 was obtained, and separated in a third column chromatography by eluting with hexane/chloroform mixture. It produced the most effective fraction 1-5-1. Moreover, fraction 1-5-1 showed an inhibitory effect on acetic acid-induced writhing in the doses of 30mg/kg and 60mg/kg,p.o.,indicating that it also contained analgesic activity.

Studies on Antioxidant, Anti-inflammation and Whitening Activities of Hordeum vulgare L. Extracts and Their Fractions (청보리 추출물과 분획물의 항산화, 항염 및 미백활성 연구)

  • This study was carried out to evaluate the antioxidant, anti-inflammation, and whitening effect of Hordeum vulgare L. extracts and their fractions. Total polyphenol and flavonoid contents in fractions were varied from 13.58 to 40.06 mg GAE/g and 7.67 ~ 13.67 mg CE/g, respectively. Among the three fractions(chloroform, hexane, and water), $400{\mu}g/mL$ of the chroloform fraction showed similar antioxidant activity to ascorbic acid ($30{\mu}M$) against the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The chloroform and hexane fractions inhibited the NO production of RAW 264.7 cells similar to quercetin ($15{\mu}M$) and the chloroform fraction of $100{\mu}g/mL$ significantly reduced IL-6, iNOS and COX2 gene expression. Additionally, the chloroform fraction inhibited ${\beta}$-hexosaminidase degranulation, IL-4, and IL-13 gene expression in RBL-2H3 cells. All of the fractions inhibited tyrosinase activity in a concentration-dependent manner, and the hexane fraction at $50{\mu}g/mL$ and the chloroform fraction at $100{\mu}g/mL$ significantly inhibited melanin production of B16F10 cells. These results indicated that H. vulgare L. can be used as an effective cosmetic ingredient having anti-inflammation and whitening activity.

Anti-Inflammatory Action of the Fractions of Platycodi radix (길경(Platycodi radix) 분획물의 항염증 활성)

  • The extract of Platycodi radix has long been used as an anti-inflammatory traditional medicine in Korea for the treatment of bronchitis, asthma and acute respiratory disease. In this study, the anti-inflammatory actions of the methanol extract of Platycodi radix administered via oral administration were evaluated. The results revealed that the extracts exhibited potent anti-inflammatory action. Of the hexane, chloroform, ethyl acetate, butanol and water fractions, the butanol fraction had the most potent anti-inflammatory action. Additioinally the sapogenin obtained through acid hydrolysis of the butanol fraction did not show anti-inflammatory action. These findings confirm the value of Platycodi radix as a traditional Korean medicine.

Flavonoid Fraction Purified from Rhus verniciflua Stokes Actively Inhibits Cell Growth Via Induction of Apoptosis in Mouse Tumorigenic Hepatocytes

  • Dietary flavonoids are currently receiving considerable attention in developing novel cancer-preventive approaches because of their potential capacities to actively induce apoptosis of cancer cells. In our previous report, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein and named RCMF (RVS chloroform-methanol fraction), was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) that is traditionally used as food additive and herbal medicine. In this study, we evaluated the effects of the RCMF on cell proliferation and apoptosis using SV40-transformed tumorigenic hepatocytes, BNL SV A.8. Tritium uptake assay showing the proliferative capacity of the cells was strongly suppressed in the presence of RCMF. This anti-proliferative effect was further confirmed through trypan blue exclusion. RCMF-mediated suppression of cell growth was verified to be apoptotic, based on the increase in DNA fragmentation, low fluorescence intensity in nuclei after propidium iodide staining, and the appearance of DNA laddering. Collectively, this study demonstrated that RCMF can be approached as a potential agent that is capable of significantly inhibiting cell growth of hepatic cancer cells.

Protective Effects of Alnus japonica Steude on Hepatic Injury Induced by Carbon Tetrachloride in Rats (사염화탄소로 유발된 흰쥐의 간손상에 대한 오리나무 분획물의 간 보호효과)

  • This study was done to investigate the protective effect of Alnus japonica Steude on hepatotoxicity in carbon let-rachloride $(CCl_4)$ intoxicated rats. Alnus japonica Steud was extracted with methanol and fractionated with hexane, chloroform, ethylacetate, butanol and water. Rats were treated with those orally once a day for 6 days. The activities of aminotransferase and ${\gamma}-glutamyltranspeptidase$ and contents of cholesterol, TG and hepatic lipid peroxide in butanol fraction pretreated rats were significantly decreased compared to the only $CCl_4$ treated rats but the content of glutathione was significantly increased compared to the only $CCl_4$ treated rats. Also activities of hepatic superoside dismutase, catalase, glutathione perpxidase in butanol fraction pretreated rats were signigicantly decreased compared to the only $CCl_4$ treated rats. These result indicated that butanol fraction of Alnus japonica Steude showed hepatoprotective effect in carbon tetrachloride intoxicated rats.
