  • During the serial attempts to identify the chemical and biological characteristics of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima (Simaroubaceae), we find out the serious toxic effect on kidney by chloroform fraction of the methanolic extract of the herb drug. The toxicities were revealed as the increase of urea nitrogen amount in blood and lactate dehydrogenase and ${\gamma}-glutamyltransferase$ activities in urine and the decrease of the concentration of glutathione and both of protein bound and non-protein bound -SH in kidney tissue.

Effects of Fractions from Methanol Extract of Commelina communis on Blood Glucose Levels and Energy Metabolism in Streptozotocin-Induced Diabetic Rats (닭의장풀 분획물이 당뇨 유발 흰쥐의 혈당과 에너지원 조성에 미치는 영향)

  • Methanol extracts of Commelina communis, a Korean wild edible plant showing hypoglycemic effect on diebetic rats were fractionated systematically by hexane, chloroform, butanol, and water, and these four fractions were administered orally to streptozotocin-induced diabetic rats to evaluate the hypoglycemic effect. The butanol and water fractions of Commelina communis lowered blood glucose levels of diebetic rats at a dose of 500 mg/kg/day. Administration of each fraction except that of chloroform increased the plasma protein levels. Liver protein levels were significantly higher in the butanol- and water fraction-fed groups. The results suggest that the butanol fraction of Commelina communis has hypoglycemic effect on diabetic rats and the intake of Commelina communis may be beneficial for the management of diabetes mellitus.

  • Hemidesmus indicus var. indicus belonging to the family Asclepiadaceae was screened for its activity against Helicobacter pylori(Hp) human isolates. Flowering and vegetative period samples were analysed. Aqueous (hot and cold) and solvent extracts (acetone, chloroform and methanol) were screened. Among them chloroform extract was observed to recover bioactive principles with low MIC and MLC. MIC was $75\;{\mu}g$ in both seasons and MLC was $100\;{\mu}g$ for vegetative and $75\;{\mu}g$ for flowering periods respectively. Extracts from samples collected during flowering period was better than that of vegetative period.

Antimycobacterial and Antioxidant Flavones from Limnophila geoffrayi

  • The chloroform extract of the aerial part of Limnophila geoffrayi showed antimycobacterial and antioxidant activities. Bioassay-guided fractionation has led to the isolation of the flavones nevadensin (5,7-dihydroxy-6,8,4'-trimethoxyflavone, 1) and isothymusin (6,7-dimethoxy-5,8,4'-trihydroxyflavone, 2). Both compounds 1 and 2 exhibited inhibition activity against Mycobacterium tuberculosis, with equal MIC value of $200{\;}\mu\textrm{g}/mL$. Only compound 2 exhibited antioxidant activity against the radical scavenging ability of DPPH, with the $IC_{50}$ value of $7.7{\;}\mu\textrm{g}/mL$. The crude hexane, chloroform and methanol extracts as well as the pure compounds 1 and 2 did not exhibit mutagenic activity in the Bacillus subtilis recassay.

Antimutagenic Activity of Asterina pectinifera (별불가사리의 항돌연변이 활성)

  • The antimutagenic activities of the total extract and several fractions of starfish, Asterina pectinifera (Asteriidae), were investigated in vitro by SOS chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA100. When various fractions was tested, the chloroform and butanol fractions showed low induction factors, which means both fractions increased antigenotoxicity against the base substitution mutagen MNNG. Even though higher antigenotoxic effect of the chloroform fraction, no effective result of Ames test was found in revertant formation of S. typhimurium TA100. The most effective antigenotoxic and antimutagenic fraction was a butanol one: i.e., When 0.5 mg/tube of butanol fraction was applied, the induction factor was 0.68. As the concentration of the fraction was increased the formation of revertants of S. typhimurium TA100 by about 81%. There was no cytotoxic effect of butanol fraction against S.typhimurium TA100. This result might be useful for further study to search a possible anticancer agent from the starfish, Asterina pectinifera.

  • This study was designed to investigate the possible utilization of Zizyphus jujuba leaves as a source of functional ingredients. The physiological activity of different solvent fractions prepared from ethanol extract of Zizyphus jujuba leaves were analyzed. Xanthine oxidase inhibitory effect was very high in all fractions except chloroform fraction. The very high electron donating ability was observed in the ethylacetate fraction and the effect was similar to 0.1% tocopherol. Nitrite scavenging effect of all fractions was more than 40% even at low concentration of 1mg/ml and was increased with increasing concentration. Angiotensin I converting enzyme inhibitory activity was appeared in ethyl acetate and chloroform fractions only at high concentraton.

Antioxidant Activity of Solvent Fraction from Black Garlic (흑마늘 용매 분획물의 항산화 활성)

  • To confirm antioxidant activity of black garlic, methanol extract of black garlic was fractioned by hexane, chloroform, ethyl acetate, buthanol and water. Antioxidant activities of solvent fractions were assayed in 100, 250, 500 and $1,000\;{\mu}g/mL$ concentrations. The contents of total phenol and flavonoids were significantly higher 5.5~11.6 times in chloroform, ethyl acetate and hexane fraction than other fractions. Antioxidant activities of solvent fractions were increased by higher sample concentrations and their activities were significantly higher in chloroform and ethyl acetate fractions than others. DPPH radical scavenging activity was over 50% in $1,000\;{\mu}g/mL$ concentration, except butanol and water fraction. In the same concentration, reducing power was also significantly lower in butanol and water fraction. ABTS radical scavenging activity was higher in hexane, chloroform and ethyl acetate fractions and was over 70% at $1,000\;{\mu}g/mL$ concentration. In $1,000\;{\mu}g/mL$ concentration, the range of hydroxy radical scavenging activity was 50.27~81.02% and SOD-like ability was 26.73~47.64%. Antioxidant activity in linoleic acid reaction system was significantly higher when storage time was longer and sample concentration was higher in non-polar solvent fractions. Nitrite scavenging activity was relatively higher than antioxidant activity and the activity in $100\;{\mu}g/mL$ concentration was over than 50%, except butanol fraction.

A Study on the Antioxidative and Antimicrobial Activities of the Chopi(Zanthoxylum Pipperitum DC.) Solvent Extracts (초피 용매 추출물의 항산화성 및 항균성에 관한 연구)

  • The antioxidative and antimicrobial activity were carried on the Chopi(Zanthoxylum pipperitum) extracts by six kinds of solvents in order to find out new natural food additives. Six solvents were used methanol(MeOH), n-hexan(hexane), chloroform$(CHCl_3)$, ethylacetate(EtOAc), buthanol(BuOH), and water(water) and methanol extract(MEex), hexan extract(HEex), chloroform extract(CHex), ethylacetate extract(EAex), and buthanol extract(BUex), water extract(WAex). The antioxdative activities of them extracts were determined by peroxide value(POV), conjugated diene value(CDV) of corn germ oil storaged for 30 days at $60{\pm}2^{\circ}C$. These extracts were added as 0.02, 0.1, 0.2% of each extracts and compared with ${\alpha}-tocopherol$, and BHT. The antioxidative activities of 0.02% extract were as follows in decreasing order BUex > WAex, BHT, MEex > HEex, EAex, CHex > TOC, and control. While, BUex and CHex among these extracts were shown to be had antimicrobial effects on the microorganism such as Escheria coli, Salmonella typhimurium, Psedomonas aeruginosa, Staphylococcus aureus, and Listeria monocytogenes. Finally to find out the preference of chopi we had made Yugwanzageon (a kind of shallow fried meat ball)by adding chopi powder. The result were similar to control(not added chopi powder) in case of 0.1% chopi adding Yugwanzageon.(p<0.05) Therefore it was thought to be possible that chopi powder was used Yugwanzageon preperation.

Purification of Cucurbitacins D, E, and I from Ecballium Elaterium (L.) A. Rich Fruits and Study of Their Cytotoxic Effects on the AGS Cell Line

  • Background: The plant Ecballium elaterium (L.) A. Rich, belongs to the Cucurbitaceae family which occupies an important position in traditional medicine prescriptions. It has been reported that a freeze-dried aqueous extract of E. elaterium fruits has cytotoxic effects on the AGS human stomach adenocarcinoma cell line. We here focused on anticancer effects of the main chemicals purified from E. elaterium fruits. Materials and Methods: We isolated cucurbitacins D, E, and I from chloroform, and ethyl acetate fractions of a methanolic extract of E. elaterium fruits and assessed their cytotoxic effects on the AGS cell line by MTT assay. The methanolic extract was fractionated to petroleum ether, chloroform, and ethyl acetate fractions. The compounds isolated by column chromatography were identified by NMR spectroscopy. Results: After 24 h of incubation with AGS cells, the IC50 values were 0.3, 0.1, and $0.5{\mu}g/ml$ for cucurbitacins D, E, and I respectively. Conclusions: This finding suggests that because of its cucurbitacins, E. elaterium fruit may have some cytotoxic effects on gastric cancer cells. Also, compared with D and I, cucurbitacin E showed greater potency in this regard.

Antioxidative Activity of Ethanol Extract from Lotus (Nelumbo nucifera) Leaf (연잎 에탄올 추출물의 항산화 효과)

  • This study was investigated on the antioxidative activity of $70\%$ ethanol extracts from lotus (Nelumbo nucifera) leaf. The extraction yields of $70\%$ ethanol extract was $8.16\%$. The extract was further fractionated subsequently by hexane, chloroform, ethyl acetate, butanol and water. Antioxidative activity was examined by electron donating ability (EDA) using DPPH, TBA value, acid value (AV), and peroxide value (POV), in comparison with commercial antioxidants. Butanol fraction showed the highest extraction yields but ethyl acetate fraction showed the highest phenol content. The ethyl acetate fraction showed the highest EDA. The antioxidative activity of ethyl acetate fraction determined by AV and POV in accelerated oxidation condition of lard was similar to that of BHA.
