Efficacy of Synthesized NO-releasing Nanoparticles on the Germination and Growth of Arabidopsis thaliana

  • Nitric oxide (NO) is a versatile signaling molecule, which is not only involved in plant growth and development but also regulates biological processes in response to biotic and abiotic stresses. Exogenous application of NO regulates the endogenous level of nitric oxide in response to stress conditions and therefore, NO donors are frequently used for stress alleviation. However, NO has very short half-life along with high reactivity. Therefore, conventional NO donors are often disadvantageous due to the relative instability of NO. On the contrary, development of NO releasing nanoparticles is a potential technique for enhancing the availability of NO in plants. Therefore, our aim was to synthesize such potential NO releasing nanoparticles which may be useful for application in agriculture. We have prepared Chitosan encapsulated S-nitrosoglutathione nanoparticles (GSNONP) and tried it with different concentrations for basic research in Arabidopsis thaliana. Our results suggest that lower concentration of this nanoparticle is highly effective for better growth of plants whereas higher concentration produces toxicity that leads to plant death. We observed better growth of Arabidopsis thaliana at 1µM concentration of the GSNONP compared to free GSNO.

Self-Assembled Nanoparticles of Bile Acid-Modified Glycol Chitosans and Their Applications for Cancer Therapy

  • This review explores recent works involving the use of the self-assembled nanoparticles of bile acid-modified glycol chitosans (BGCs) as a new drug carrier for cancer therapy. BGC nanoparticles were produced by chemically grafting different bile acids through the use of l-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC). The precise control of the size, structure, and hydrophobicity of the various BGC nanoparticles could be achieved by grafting different amounts of bile acids. The BGC nanoparticles so produced formed nanoparticles ranging in size from 210 to 850 nm in phosphate-buffered saline (PBS, pH=7.4), which exhibited substantially lower critical aggregation concentrations (0.038-0.260 mg/mL) than those of other low-molecular-weight surfactants, indicating that they possess high thermodynamic stability. The SOC nanoparticles could encapsulate small molecular peptides and hydrophobic anticancer drugs with a high loading efficiency and release them in a sustained manner. This review also highlights the biodistribution of the BGC nanoparticles, in order to demonstrate their accumulation in the tumor tissue, by utilizing the enhanced permeability and retention (EPR) effect. The different approaches used to optimize the delivery of drugs to treat cancer are also described in the last section.

  • 고분자 용액의 전기분무에서 금속 샘플 포집기들은 분무된 입자로부터 흔히 용매를 충분히 제거하거나 재분산이 가능한 분무 입자를 회수하는데 효과적이지 못하다. 단백질 약물의 캡슐화(encapsulation)를 위해 본 연구에서는 입자를 분산매(응고액제)의 층류 안으로 분무하는 새로운 전기분무시스템을 설계하였다. 캡슐화용 물질로 키토산과 폴리아크릴산이 사용되었다. 이 새로운 시스템을 사용하여 입자의 응집을 방지할 수 있었으며, 4~16 kV의 전압과 같은 유속에서 unimodal한 입도분포를 관찰할 수 있었다. 반면에 가한 전압이 평균 입자 크기에 미치는 영향은 크지 않았다.

Characterization of ginsenoside compound K loaded ionically cross-linked carboxymethyl chitosan-calcium nanoparticles and its cytotoxic potential against prostate cancer cells

  • Backgroud: Ginsenoside compound K (GK) is a major metabolite of protopanaxadiol-type ginsenosides and has remarkable anticancer activities in vitro and in vivo. This work used an ionic cross-linking method to entrap GK within O-carboxymethyl chitosan (OCMC) nanoparticles (Nps) to form GK-loaded OCMC Nps (GK-OCMC Nps), which enhance the aqueous solubility and stability of GK. Methods: The GK-OCMC Nps were characterized using several physicochemical techniques, including x-ray diffraction, transmission electron microscopy, zeta potential analysis, and particle size analysis via dynamic light scattering. GK was released from GK-OCMC Nps and was conducted using the dialysis bag diffusion method. The effects of GK and GK-OCMC Nps on PC3 cell viability were measured by using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay. Fluorescent technology based on Cy5.5-labeled probes was used to explore the cellular uptake of GK-OCMC Nps. Results: The GK-OCMC NPs had a suitable particle size and zeta potential; they were spherical with good dispersion. In vitro drug release from GK-OCMC NPs was pH dependent. Moreover, the in vitro cytotoxicity study and cellular uptake assays indicated that the GK-OCMC Nps significantly enhanced the cytotoxicity and cellular uptake of GK toward the PC3 cells. GK-OCMC Nps also significantly promoted the activities of both caspase-3 and caspase-9. Conclusion: GK-OCMC Nps are potential nanocarriers for delivering hydrophobic drugs, thereby enhancing water solubility and permeability and improving the antiproliferative effects of GK.

Improvement of antithrombotic activity of red ginseng extract by nanoencapsulation using chitosan and antithrombotic cross-linkers: polyglutamic acid and fucoidan

  • Background: Red ginseng (RG) extract, especially ginsenoside Rg1 and Rb1 fractions has been reported to have antithrombotic activities. However, gastric instability and low intestinal permeability are considered to be obstacles to its oral administration. We hypothesized that stability, permeability, and activities of RG might be improved by encapsulation within nanoparticles (NPs) prepared with antithrombotic coating materials. Methods: RG-loaded chitosan (CS) NPs (PF-NPs) were prepared by complex ionic gelation with the antithrombotic wall materials, polyglutamic acid (PGA), and fucoidan (Fu). The concentrations of PGA (mg/mL, X1) and Fu (mg/mL, X2) were optimized for the smallest particle size by response surface methodology. Antithrombotic activities of RG and PF-NPs were analyzed using ex vivo and in vivo antiplatelet activities, in vivo carrageenan-induced mouse tail, and arteriovenous shunt rat thrombosis models. Results: In accordance with a quadratic regression model, the smallest PF-NPs (286 ± 36.6 nm) were fabricated at 0.628 mg/mL PGA and 0.081 mg/mL Fu. The inhibitory activities of RG on ex vivo and in vivo platelet aggregation and thrombosis in in vivo arteriovenous shunt significantly (p < 0.05) increased to approximately 66.82%, 35.42%, and 38.95%, respectively, by encapsulation within PF-NPs. For an in vivo carrageenan-induced mouse tail thrombosis model, though RG had a weaker inhibitory effect, PF-NPs reduced thrombus significantly due to the presence of PGA and Fu. Conclusion: PF-NPs contributed to improve the activities of RG not only by nanoencapsulation but also by antithrombotic coating materials. Therefore, PG-NPs can be suggested as an efficient delivery system for oral administration of RG.

The Power of Being Small: Nanosized Products for Agriculture

  • Certain agrochemicals may be tuned for increased effectiveness when downsized to nanoparticles (NPs), where one dimension is less than 100 nm. The NPs may function as fertilizers, pesticides and products to improve plant health through seed priming, growth promotion, and induction of systemic tolerance to stress. Formulations will allow targeted applications with timed release, reducing waste and pollution when compared to treatments with bulk-size products. The NPs may be a single component, such as nano-ZnO as a fertilizer, or be composites of compatible materials, for example where N, P, and K plus micronutrients are available. The active materials could be loaded into porous carriers or tethered to base nanostructures. Coatings could include such natural products alginate, chitosan, zein, or silica. Certain NPs are taken up and transported in the plant's phloem and xylem so systemic effects are feasible. Timed and targeted release of the active product could be achieved in response to changes in pH or availability of ligands within the plant or the rhizosphere. Global research has revealed the many potentials offered by NP formulations to aid sustainability in agriculture. Current work will provide information needed by regulatory agencies to assess their safety in the agricultural setting.

Development of New Materials of Ginseng by Nanoparticles

  • For centuries, Panax ginseng Meyer (Korean ginseng) has been widely used as a medicinal herb in Korea, China, and Japan. Ginsenosides are a class of triterpene saponins and recognized as the bioactive components in Korean ginseng. Ginsenosides, which can be classified broadly as protopanaxadiols (PPD), protopanaxatriols (PPT), and oleanolic acids, have been shown to flaunt a vast array of pharmacological activities such as immune-modulatory, anti-inflammatory, anti-tumor, anti-diabetic, and antioxidant effects. In recent years, a number of ginseng and ginsenoside researches have increasingly gained wide attention owing to its unique pharmacological properties. Although good efficacies of ginsenosides have been reported, lack of target specific delivery into tumor sites, low solubility, and low bioavailability due to modifications in gastro-intestinal environments limit their biomedical application in clinical trials. As a result to this major challenge, nanotechnology and drug delivery techniques play a significant role to solve this problematic issue. Thus, we reported the preparation of poly-ethylene glycol (PEG) and glycol chitosan (GC) functionalized to ginsenoside (Compound K and PPD) conjugates via hydrolysable ester bonds with improved aqueous solubility and pH-dependent drug release. In vitro cytotoxicity assays revealed that PEG-CK, and PPD-CK conjugates exhibited lower cytotoxicity compared to bare CK and PPD in HT29 cells. However, GC-CK conjugates exhibited higher and similar cytotoxicity in HT29 and HepG2 cells. Furthermore, GC-CK-treated RAW264.7 cells did not exhibit significant cell death at higher concentration of treatment which supports the biocompatibility of the polymer conjugates. They also inhibited nitric oxide production in lipopolysaccharide (LPS)-induced RAW64.7 cells. In addition to polymer-ginsenoside conjugates, silver (AgNps) and gold nanoparticles (AuNps) have been successfully synthesized by green chemistry using different m. The biosynthesized nanoparticles demonstrated antimicrobial efficacy, anticancer, anti-inflammatory, antioxidant activity, biofilm inhibition, and anticoagulant effect. Special interest on the effective delivery methods of ginsenoside to treatment sites is the focus of metal nanoparticle research.In short, nano-sizing of ginsenoside results in an increased water solubility and bioavailability. The use of nano-sized ginsenoside and P. ginseng mediated metallic nanoparticles is expected to be effective on medical platform against various diseases in the future.

Effect of additives on the hydrothermal synthesis of manganese ferrite nanoparticles

  • Superparamagnetic iron oxide nanoparticles (Nps), composed of magnetite, $Fe_3O_4$, or maghemite, ${\gamma}-Fe_2O_3$, core and biocompatible polymer shell, such as dextran or chitozan, have recently found wide applications in magnetic resonance imaging, contrast enhancement and hyperthermia therapy. For different diagnostic and therapeutic applications, current attempt is focusing on the synthesis and biomedical applications of various ferrite Nps, such as $CoFe_2O_4$ and $MnFe_2O_4$, differing from iron oxide Nps in charge, surface chemistry and magnetic properties. This study is focused on the synthesis of manganese ferrite, $MnFe_2O_4$, Nps by most commonly used chemical way pursuing better control of their size, purity and magnetic properties. Co-precipitation syntheses were performed using aqueous alkaline solutions of Mn(II) and Fe(III) salts and NaOH within a wide pH range using various hydrothermal treatment regimes. Different additives, such as citric acid, cysteine, glicine, polyetylene glycol, triethanolamine, chitosan, etc., were tested on purpose to obtain good yield of pure phase and monodispersed Nps with average size of ${\leq}20nm$. Transmission electron microscopy (TEM), X-ray diffraction, energy dispersive X-ray spectroscopy (EDX), $M\ddot{o}ssbauer$ spectroscopy down to cryogenic temperatures, magnetic measurements and inductively coupled plasma mass spectrometry were employed in this study.

단백질 약물 전달을 위한 마이크로/나노 입자의 전기분무 제조법 (Electrospraying of Micro/Nano Particles for Protein Drug Delivery)

  • 전기분무에서의 전기유체 역학적 힘에 의한 표면 에너지의 조절은 간단한 입자 크기 조절, 단분산성, 높은 회수율, 그리고 약한 가공조건과 같은 이점을 제공할 수 있다. 이러한 이점은 단백질 약물전달체 제조에 적절할 것으로 예상되어, 본 연구에서 전기분무법을 이용하여 단백질 약물의 나노포집을 시도하였다. 모델 단백질인 알부민을 단축 혹은 동축 전기분무로 가공하였고 키토산, 폴리카플로락톤(PCL), 폴리 (에틸렌 글리콜) (PEG) 등이 포집물질로 사용되었다. 효율을 최대로 높이기 위해 분무액의 전기전도도, 유속, 전기포텐셜 구배의 거리 등과 같은 가공변수들이 조사되었다. 키토산 시스템에서 입자크기에 대한 공정 변수의 영향은 유속이 늦어질수록, 노즐과 집적부 사이의 거리가 가까울수록 입자 크기가 감소하는 것을 알 수 있었다. PCL 시스템에서는 단축 전기분무의 경우 유속이 늦어질수록, 동축 전기분무의 경우 내부와 외부 물질의 유속비가 클수록 입자 크기가 증가하는 것을 확인할 수 있었다. 전기분무 노즐에서 생성된 초기 입자들은 좁은 입자 크기 분포를 보였으나, 그것들이 집적부에 도달했을 때 입자들이 응집되는 경향이 있었다. 이러한 전기분무법에서 PCL, PEG, 키토산을 사용한 알부민의 효과적인 나노 포집은 12 kV 이상에서 성공적으로 이루어졌다.

Preparation of Flame Retardant and Antibacterial Wood with Composite Membrane Coating

  • A novel flame retardant and antibacterial composite membrane coating for wood surfaces was prepared by adding POSS-based phosphorous nitrogen flame retardant (later referred to as NH2-POSS) and silver nanoparticles (Ag NPs) to chitosan (CS). The effects of NH2-POSS content (mass fractions of CS 0%, 0.5%, 1%, 3%, 5%, and 7%) on the structure and properties of the composite membrane coating on wood were investigated. The composite film was prepared by the method of blending and ducting. Contact angle, tensile property and antibacterial effects of the composite film were measured, and infrared spectroscopy was used. The results show that the addition of NH2-POSS can not only improve the toughness of the membrane, but also the flame retardancy of the membrane, which improves the application of the membrane in wood products. However, with the addition of NH2-POSS, the transparency of the composite membrane was weakened. The inhibitory effect of the composite membrane on the growth of Escherichia coli was enhanced with the increase in Ag NPs. This research provides a foundation for the application of functional wood.
