The Selective Visualization of Lignin Peroxidase, Manganese Peroxidase and Laccase, Produced by White Rot Fungi on Solid Media

  • A visual method for the selective screen Eng of lignin degrading enzymes, produced by white rot fungi (WRF), was investigated by the addition of coloring additives to solid media. Of the additives used in the enzyme production media, guaiacol and RBBR could be used for the detection of lignin peroxidase (LiP), manganese peroxidase (MnP) and lactase. Syringaldazine and Acid Red 264 were able for the detection of both the MnP and lactase, and the LiP and laccase, respectively, and a combination of these two additives was able to detect each of the ligninases produced by the WRF on solid media.

  • A leak detection method for diagnosis of the leak position in a pipeline was developed using an estimation theory with the assumption that the measured flow rates and pressures are stochastic processes. A notch filter was designed using power spectral density analysis of measurements to reduce the effects of disturbances. The noise model dimension was determined by hypothesis testing and then recursive extended least square method was applied to estimate the leak position in real time. The proposed method was applied to an experimental system for evaluation of its performance.

  • We chose a fluorescent pentapeptide sensor (-CPGHE) containing a dansyl fluorophore as a model peptide and investigated whether the selectivity and sensitivity of the peptides for heavy and transition metal ions could be tuned by changing amino acid sequence. In this process, we developed a selective peptide sensor, Cp1-d (-HHPGE, $K_d\;=\;670\;nM$) for detection of $Zn^{2+}$ in 100% aqueous solution and a selective and sensitive peptide sensor, Cp1-e (-CCHPGE, $K_d\;=\;24\;nM$) for detection of $Cd^{2+}$ in 100% aqueous solution. Overall results indicate that the selectivity and sensitivity of the metallopeptide sensors to specific heavy and transition metal ions can be tuned by changing amino acid sequence.

  • In spite of the radical increase of chemical poisoning cases caused by prevalent of oranic chlorine like endosulfan, the study of detection of chemical poisoning has seldom been attempted; the study of poisoning in teeth, which is more secure physiochemically, and in all the oral tissues has been even less attempted. The author has administered endosulfan into the month of a rabbit, causing its death, and tried to detect endosulfan in the teeth, jaws, and oral tissues through forensic chemical and histopathological tests. The result obtained is as follows : 1. By thin layer chromatography, endosulfan was detected in the jews, dental hard tissues, dental pulp, blood, and the liver. 2. By gas chromatography, it was also detected in the jaws, dental hard tissues, dental pulp, blood, and the liver. 3. The congestion and hemorrhage in buccal mucosa, palatal mucosa, and dental pulp was detected.

  • In this paper, we have studied the in-situ end point detection (EPD) for direct chemical mechanical polishing (CMP) of shallow trench isolation (STI) structures without the reverse moat etch process. In this case, we applied a high selectivity $1n (HSS) that improves the silicon oxide removal rate and maximizes oxide to nitride selectivity Quite reproducible EPD results were obtained, and the wafer-to-wafer thickness variation was significantly reduced compared with the conventional predetermined polishing time method without EPD. Therefore, it is possible to achieve a global planarization without the complicated reverse moat etch process. As a result, the STI-CMP process can be simplified and improved using the new EPD method.

  • The electrochemical oxidation of ascorbic acid(AA), serotonin(SE) and epinephrine(EP) have been performed at poly N,N-dimethylaniline(PDMA) film coated diamond electrode. This cationic polymer film is electrochemically deposited on boron-doped diamond electrode surface. Unlike the bard electrode, the polymer film-coated diamond electrode can well separate the oxidation potential of AA by 200mV. Thus this electrode can be successfully used for the simultaneoud detection of both species. Increases in the concentration of AA do not affect the reponse of EP and SE.

The Chemical Kinetics for the Reaction of O(³P) with Ethylene

  • The kinetics of the reaction between O(3P) and C2H4 was investigated by measuring time-dependent concentrations of OH resulting from the reaction by using the LIF detection. Oxygen atoms were generated by titrating microwave discharged N2/He with NO to the chemiluminescent end point. The operating pressures in the flow reactor ranged from 5 to 15 torr and the mixtures consisted of He/O(3/P)/C2H4 in the approximate ratios from 100/1/0.1 to 100/1/1. The controlled residence time prior to the detection were estimated to be 0.8-17 ms at the reactor pressure of 7 torr. Experimentally determined profiles both in shape and magnitude were compared with the computed OH density for a specified set of experimental parameters, allowing us to arrive at a complete mechanism for the reaction of O(3P) with ethylene.

Electrochemical Determination of Chemical Oxygen Demand Based on Boron-Doped Diamond Electrode

  • A rapid and environment-friendly electrochemical sensor to determine the chemical oxygen demand (COD) has been developed. The boron-doped diamond (BDD) thin-film electrode is employed as the anode, which fully oxidizes organic pollutants and provides a current response in proportion to the COD values of the sample solution. The BDD-based amperometric COD sensor is optimized in terms of the applied potential and the solution pH. At the optimized conditions, the COD sensor exhibits a linear range of 0 to 80 mg/L and the detection limit of 1.1 mg/L. Using a set of model organic compounds, the electrochemical COD sensor is compared with the conventional dichromate COD method. The result shows an excellent correlation between the two methods.

Fast and Efficient Method for Fire Detection Using Image Processing

  • Conventional fire detection systems use physical sensors to detect fire. Chemical properties of particles in the air are acquired by sensors and are used by conventional fire detection systems to raise an alarm. However, this can also cause false alarms; for example, a person smoking in a room may trigger a typical fire alarm system. In order to manage false alarms of conventional fire detection systems, a computer vision-based fire detection algorithm is proposed in this paper. The proposed fire detection algorithm consists of two main parts: fire color modeling and motion detection. The algorithm can be used in parallel with conventional fire detection systems to reduce false alarms. It can also be deployed as a stand-alone system to detect fire by using video frames acquired through a video acquisition device. A novel fire color model is developed in CIE $L^*a^*b^*$ color space to identify fire pixels. The proposed fire color model is tested with ten diverse video sequences including different types of fire. The experimental results are quite encouraging in terms of correctly classifying fire pixels according to color information only. The overall fire detection system's performance is tested over a benchmark fire video database, and its performance is compared with the state-of-the-art fire detection method.
