Protective effects of Dictamni Radicis Cortex water extract on acute pancreatitis (백선피(白鮮皮) 물 추출물의 급성 췌장염 보호 효과)

  • Objectives : Dictamni Radicis Cortex (DRC) has been used as an important traditional medicine for inflammation and fungal diseases. However, the protective effect of DRC water extract on acute pancreatitis (AP) has not been deeply reported. Therefore, we aimed to evaluate the protective effects of DRC water extract on cerulein-induced AP. Methods : AP was induced via intraperitoneal injection of supramaximal concentrations of stable cholecystokinin analogue cerulein ($50{\mu}g/kg$) every hour for 6 times. DRC water extract (0.05, 0.1, or 0.2 g/kg) or saline was administrated intraperitoneally 1 h before to the first injection of cerulein. The mice were sacrificed at 6 h after the final cerulein injection. Pancreas was rapidly removed for histochemical examination and myeloperoxidase (MPO) assay. In addition, polymerase chain reaction (PCR) was performed to examine mRNA levels of proinflammatory cytokines such as Interleukin $(IL)-1{\beta}$, IL-6 and Tumor necrosis factor $(TNF)-{\alpha}$. Results : Administration of DRC water extract significantly inhibited the pancreatic weight to body weight ratio, pancreas histological damages and increase of pancreatic MPO activity during cerulein-induced AP. In addition, increased pancreatic mRNA levels of $IL-1{\beta}$, IL-6 but not $TNF-{\alpha}$ were significantly inhibited by treatment of DRC water extract against cerulein-induced AP. Conclusions : In conclusion, we have revealed that pre-treatment of DRC water extract reduces the severity of cerulein-induced AP. Accordingly, our results could give a clinical basis that DRC could be used as a drug or agent to prevent AP.

Protective Effect of Glycyrrhiza New Domestic Variety on the Acute Pancreatitis (국내 감초 신품종의 급성 췌장염 보호 효과)

  • Glycyrrhiza korshinskyi Grig. (Gk), which is a new variety of Glycyrrhiza, was developed by the National Institute of Horticultural and Herbal Science in Korea Rural Development Administration. It is improved the limitations of traditional Glycrrhiza species such as G. uralensis Fisch., G. glabra L., and G. inflata Batal. and has been reported various pharmacological effects. The aim of this study is to investigate the protective effect of Gk on the acute pancreatitis, because which has not been revealed yet. AP was induced via intraperitoneal injection of cerulein (50 ㎍/kg) hourly for 7 times in C57BL/6 mice. Gk water extract (20 or 50 mg/kg) or saline was administrated via oral gavage 1 h before the first injection of cerulein. The mice were sacrificed at 5 h after the final injection of cerulein. The inflammatory cell infiltration in pancreatic tissue was decreased by the administration of Gk water extract compared to the AP group. Also, serum amylase and lipase levels were decreased by the administration of Gk water extract compared to the AP group. These results suggest that Gk have protective effect on the acute pancreatitis.

Protective Effects of Ethanol Extract from Aster Yomena on Acute Pancreatitis (쑥부쟁이 에탄올 추출물의 급성췌장염 보호 효과)

  • Aster Yomena (AY) has been used in traditional medicine to treat diseases such as obesity, hyperlipidemia, atherosclerosis, diabetes and osteoarthritis. However, protective effect of AY on acute pancreatitis (AP) has not been reported. The present study examined the anti-inflammatory effects of an ethanol extract of AY on cerulein-induced AP. AP was induced in mice by intraperitoneally injecting cerulein ($50{\mu}g/kg$) hourly for 6 times. 70% ethanol extract of AY (0.1, 0.2, and 0.5 g/kg) was orally administered for 1 week before acute pancreatitis induction. The mouse was killed at 6 hours after the final cerulein injection. The pancreas and lung were rapidly removed for histological examination and myeloperoxidase (MPO) assay. Blood samples were taken to determine serum amylase and lipase activity. In addition real-time reverse transcription-polymerase chain reaction (RT-PCR) was also performed to investigate mRNA expression of proinflammatory cytokines such as $TNF-{\alpha}$. $IL-1{\beta}$, and IL-6. Administration of AY significantly ameliorated pancreatic weight to body weight ratio, histological damages and MPO activity during AP. In addition, AY inhibited the serum amylase and lipase activity during AP. Also, mRNA expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 were inhibited by AY against AP. Our results revealed that pre-treatment of AY reduces the severity of cerulein-induced AP. Therefore, AY may have a protective effect drug against AP.

The Comparison Between Single And Yimyosan in Acute Pancreatitis : Atractylodes chinensis(AC) and Phellodendron amurense Ruprecht(PAR) (이묘산(二妙散)과 구성약재(構成藥材)의 효능(效能) 비교(급성(急性) 췌장염(膵臟炎)을 중심으로))

  • Objectives : Atractylodes chinensis(AC) and Phellodendron amurense Ruprecht(PAR) have been used as anti-inflammatory medicines. However, the effects of AC, PAR and Yimyosan on AP were not examined. To prove this, We decocted the dried prescription of AC and PAR with boiling distilled water and freeze-dried to be powdered. AC, PAR and Yimyosan was administrated intraperitoneally. Methods : 1h after administration, cerulein was injected hourly six times. 6hrs after last cerulein injection, mice were sacrificed, then the pancreas and blood were harvested. Serum amylase and lipase, neutrophil infiltration, pancreatic cytokines were used as the parameter of severity of AP. Results : As a result of assessment the parameters of AP, AC alone treatment did not inhibit the severity of AP, however PAR treatment inhibited the severity of AP significantly. Yimyosan also showed the protective effects against AP at lower doses, however AC alone plus PAR alone extract did not show the protective effects significantly. Conclusions : In conclusion, PAR extract has a protective effects on AP, and the effects could be increased by co-treatment with AC.

Study of Efficacy of Foeniculi Fructus and Lonicerae Flos Extract on Acute Pancreatitis (소회향과 금은화 추출물의 급성췌장염에 대한 예방효과)

  • Objectives : This study was undertaken to compare the effect of Foeniculi Fructus and Lonicerae Flos extract on the cerulein-induced acute pancreatitis in rats. Methods : Acute pancreatitis was induced by intraperitoneal injection of cerulein. Foeniculi Fructus extract (FE; 300 mg/kg) and Lonicerae Flos extract (LE; 300 mg/kg) were injected 2hr before induction of acute pancreatitis. Rats were sacrificed 6hr after first injection of cerulein. The severity of pancreatitis was assessed by measuring pancreatic weight/body weight ratio, neutrophil, lymphocyte, serum amylase activity, platelet activating factor (PAF) activity, tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) activity, interleukin 6 (IL-6) activity and by histological assessments of inflammatory cell infiltration. Results : 1. The pancreatic weight/body weight ratios of FE and LE group compared with the control group were decreased significantly. 2. The neutrophil content ratio of FE and LE group compared with the control group were decreased. 3. The lymphocyte content ratio of FE and LE group compared with the control group were increased significantly. 4. The activities of serum amylase of FE and LE group compared with the control group were decreased significantly. 5. The activities of serum PAF of FE and LE group compared with the control group were decreased significantly. 6. The activities of TNF-${\alpha}$ of FE and LE group compared with the control group were decreased significantly. 7. The activities of IL-6 of FE and LE group compared with the control group were decreased significantly. 8. The pancreas injected with FE and LE showed reduction of swelling of acinar cells, inflammation and vacuolization than the control group on light microscopic observation. Conclusions : These results suggest that Foeniculi Fructus and Lonicerae Flos extract have an effect to suppress inflammation on cerulein-induced acute pancreatitis in rats. But there are no significant differences between Foeniculi Fructus extract and Lonicerae Flos extract.

Protective Effects of Individual Extract and Mixture of Forsythia Fructus and Corydalis Tuber on Chronic Pancreatitis in Mice (마우스 만성 췌장염 모델에서 연교(連翹)와 현호색(玄胡索) 단일 추출물 및 추출 혼합물의 보호 효과)

  • Objectives : Forsythiae Fructus has been reported to have anti-inflammatory effects in many diseases, and Corydalis Tuber has been used as a pain suppressor in Eastern Asia. However, the protective effects of individual water extract of Forsythiae Fructus (FF) and Corydalis Tuber (CT) and the mixture of FF and CT (FC) on chronic pancreatitis (CP) were not well-investigated. Therefore, we investigated the protective effects of FF, CT, and FC on CP in mice. Methods : To induce CP, cerulein was injected 6 times a day, 4 times a week for 3 weeks. 1 h before the every cerulein injection, 200 mg/kg of FF, CT, or FC was intraperitoneally injected to mice. Histological analysis of pancreas was examined by hematoxylin and eosin stain and collagen deposition was examined by Masson's trichrome stain. Fibrogenic parameters such as α-smooth muscle actin (α-SMA), extracellular matrix (ECM) deposition, and fibrotic cytokines such as transforming growth factor (TGF)-β1 and platelet-derived growth factor (PDGF) were also analyzed by immunofluorescence stain and real-time PCR. Results : Histological damages in pancreas were inhibited by pre-treatment of FF or FC but not CT. α-SMA and ECM in pancreas were inhibited by pre-treatment of CT or FC but not FF. Moreover, the expression of TGF-β1 and PDGF in pancreas were inhibited by FF, CT or FC. Conclusions : Our results suggest that FC have protective effect on CP in mice through inhibition of α-SMA, ECM, TGF-β1 and PDGF in pancreas, and these findings could suggest new clinical strategy for CP.

The inhibitory effects of Nardostachys jatamansi on alcoholic chronic pancreatitis

  • Nardostachys jatamansi (NJ) belonging to the Valerianaceae family has been used as a remedy for gastrointestinal inflammatory diseases for decades. However, the potential for NJ to ameliorate alcoholic chronic pancreatitis (ACP) is unknown. The aim of this study was to examine the inhibitory effects of NJ on ACP. C57black/6 mice received ethanol injections intraperitoneally for 3 weeks against a background of cerulein-induced acute pancreatitis. During ACP, NJ was ad libitum administrated orally with water. After 3 weeks of treatment, the pancreas was harvested for histological examination. NJ treatment increased the pancreatic acinar cell survival (confirmed by amylase level testing) and reduced collagen deposition and pancreatic stellate cell (PSC) activation. In addition, NJ treatment reduced the activation but not death of PSC. In conclusion, our results suggest that NJ attenuated ACP through the inhibition of PSC activation.
