Protein Patterns on a Vaginal Mucus during Spontaneous and Estrus Synchronization using CIDR in Korean Native Cattle (Hanwoo)

  • The aim of the present recent study was to compare the protein patterns in the vaginal mucus of Hanwoo cattles during spontaneous and CIDR induced-estrus. Ten cattles, who had been observed in estrus, received no treatment and served as the group of cattles with normal spontaneous estrus. Thirteen cattles in the CIDR received an CIDR insert on day 14 were removed and cattles were injected GnRH on day 15. Vaginal mucus samples were collected from all cattles at the same time the single AI in cattles with spontaneous estrus and the AI in cattles with induced estrus. Spontaneous and CIDR-induced estrus vaginal mucus samples were analyzed on two different array surfaces: cation-exchange (CM10), anion-exchange (Q10). In addition, using the NaCl solution by which the proteins combined after washing are 0.5, 1 and 2 M, it was fractionated and a protein was collected successively. The results are summarized as follows: 1) Ionic surfaces chemistries (Q10 and CM10) gave the best results in terms of detectable protein peaks, with more than 100 protein peaks in the two fractions and under each condition. 2) Protein mass spectrometer using 11 different proteins in protein identification of 7 were able to determine the protein. List of identified proteins as follows; Ribosome-binding protein 1, GRIP 1-associated protein 1, Katanin p60 ATPase-containing subunit A-like 1, Protein FAM44A, DUF729 domain-containing protein 1, Prolactin precursor, Dihydrofolate erductase. Conclusively, on the basis of this study, protein expression in the vaginal mucus could be used as an indicator for time of estrus manifestation in order to increase conception rates by applying AI at an optional time.

Factors Affecting on Production of Dairy Cattle Embryos (젖소 수정란의 생산에 영향을 미치는 요인에 관한 연구)

  • The present study was carried out to investigate effects of gonadotropin, age of donor, day of estrus cycle gonadotropin injection started and season on embryo production after superovulation in dairy cattles. Embryo collection records were obtained from 177 embryo donor collections from 98 Holstein cows aged from 3 to 9 years during 4 years(1993~1996) at National Livestock Re-search Institute. Superovulation was induced by injections of 3 gonadotropins(FSH-P, FOLLTROPHIN-v* or SUPER-OV*) beginning on days 9 to 14 of the estrus cycle. Em-bryos were collected from donors using a nonsurgical technique on days 6 to 8 after insemi-nation. The results were as follows ;Number of total and freezable embryos per donor cow was affected by gonadotropin(P<0.01). The more number of total and freezable embryos wereobtained by use of FOLLTROPHIN-V (13.2, 7.4) or FSH-P (11.0, 5.7) than SUPER-OV* (5.0, 2.4). Age of donor, the day that gonadotropin was started or season didn't affect total or freezable embryos(P >0.05).

Comparison of Embryo Production Performance and Conception Rate after Embryo Transfer between Mongolian Cattle and Korean Native Cattle

  • This study is to compare the effect of estrus synchronization and embryo transfer between Korean and Mongolian cattle. Embryos were collected from 9 donors housed in Asan city in South Chungcheong Province, South Korea. Embryos were collected 9 donors from Khushaat sum, Selenge province and Bayanchandmani sum, Tov province in Mongolia. Follicle Stimulating Hormone (FSH), Controlled Internal Drug Release (CIDR) and Prostaglandin (PG) were used for superovulation. Subsequently, Artificial Insemination (AI) was done for donor cow and embryo was collected after 7 and 8 days. Collected embryos were compared between Mongolian and Korean cattle. Finally, good quality and fresh embryos were transferred to 50 and 22 recipients of cows in Korea and Mongolia respectively. The findings show that Korean native cattle each donor cow produced on an average 16.9 embryos and, 10.9 embryos were found transferable. But in case of Mongolia the average production of embryos per donor cow was 8.6 embryos and, 6.2 embryos were found transferable. Embryo collection after 7 and 8 days was not difference in embryo production in Korea. But, in Mongolia embryo production after 8 days was found more efficient than the 7 days. Korean native recipient's cows (74.6%) and Mongolian recipient's cows (71.0%) respectively were found transferable ovarian stage. The result suggested that efficiency of embryo production from the superovulation method treated of Korean cow were higher than the Mongolian cow. The pregnancy rate of Korea native cattle was 72%, which was about 10% higher than that of Mongolia cattle.

A Method of White Noise Reduction for Recognizing Cattle's Gulp Downing Sounds

  • In this paper, we proposed a method to measure the feed intake of cattle using the cattle's gulp downing sounds. To measure the sound of cattle's gulp downing, the recording is performed through a wearable device attached to the cattle's neck. A lot of noises are recorded according to the ranching environment. This paper proposed a method for spectralizing raw gulping sound data containing white noise and removing white noise through the signal transformation using a filter. This allows the feed intake to be measured. Through the proposed white noise reduction method, it was possible to extract only the cattle's gulp downing sound, and through this, the number of cattle's gulp downing could be measured. The proposed method in this paper makes it possible to measure cattle's feed intake easily, so that estrus prediction, health care for cattle, and feed management can be done efficiently.

Industrial Application of Embryo Transfer in Korea (수정란이식의 산업화 방안)

  • Animal in dustry in Korea urgently needs the domestic introduction and the industrial a, pp.ication of embryo transfer technique. Namely, this technique can be utilized effectively, as means of the improvement of livestocks, as means of the increase of meat production, as means of substitute for the livestock import, and dissemination of new breed. However, as this technique avaliable in our country is remaining initial stage, we can not make use of the technique industrially unless we make much improvement as follows; induction of superovulation, non-surgical recovery of embryos, synchronization between the estrus such cycles of donor and recipient, non-surgical transfer of embryos, etc. Simultaneously, the basic studies such as harvesting oocytes from ovary, in vitro culture of oocytes, in vitro capacitation of spermatozoa, cloning by culture of blastomeres and transfer of nuclei, sexing embryo, etc. should not be neglected in order to make the technique of embryo transfer more simple and convenient. For the success of these studies, universities, national and public institutes, large scale cattle farms, and small scale cattle farms should cooperate each other. For instance, universities undertake basic researches, and the national and public institutes a, pp.y the results of the researches to animal industry along with cooperation by large scale cattle farms. By the help of the cooperative organizations, the technique relevant to our environment and farm condition may be able to be finalized, and to be a, pp.ied to samll scale cattle farm. Consequently, being served to stimulate animal productivity, this technique can be contributed to the development of livestock industry in Korea.

Effect of human chorionic gonadotrophin injection after artificial insemination on pregnancy establishment in dairy cattle

  • The objective of this study was to evaluate the effect of treating dairy cattle with exogenous human chorionic gonadotrophin (hCG), five (5) days post artificial insemination (AI) on serum progesterone (P4) concentration and pregnancy rate. In this experiment, five days after AI, cows were assigned randomly to two groups namely: a) treated group (67) which were administrered with 1500 IU hCG (Chorulon) and b) control group (61), which received no treatment. On day 5, 10, 15 and 20 after the artificial insemination, blood samples from a total of 8 cows (4 from each group) were collected and were analyzed for serum P4 concentration. Cows were detected for estrus according to standing heat by visual observation. Cows that were detected still in estrus after days 18-24 were re-inseminated and recorded as not pregnant (open). Pregnancy diagnosis was conducted by ultrasonographic examination and transrectal palpation of the uterus on approximately 60 days in cows that observed to be not in estrus. The conception rate in hCG treated and control groups were 52.5 and 36.1%, respectively. The results proved that there were no significant differences in conception rate between two groups (p=0.0568). However, pregnancy rates were reduced by hCG treatment. Average serum P4 concentrations did not differ between Hcg-treated and control groups on day 5 (0.377 versus 0.375 ng/ml). On day 20 serum P4 concentrations were greater in the treated group compared with the control group (3.085 versus 2.010 ng/ml). The treatment with hCG seemed to increase P4 level compared with the control. In conclusion, the results of this study showed that 1500 IU of hCG administered on 5 day post AI increased conception rate in dairy cows. This was supported by the results on serum P4 concentration which was greater in hCG treated group.

Reproductive Performance of Holstein Cattle Treated with Progesterone and Combination of Progesterone and Estradiol (인공수정 후 외인성 Progesterone과 Estradiol 병용 투여 및 Progesterone 단독 투여가 젖소의 번식 성적에 미치는 영향)

  • This study was carried out to investigate the effects of hormones (progesterone, and combination of progesterone and estradiol) on the reproductive performance of dairy cattle. The intravaginal CIDR was inserted in the vagina of cow on day 14 post-insemination to stimulate progesterone secretion with and without estradiol. The CIDR was removed on the day of next expected estrus. The cows for control group were not treated with CIDR or hormones. Conception rate, estrus interval, estrus intensity and serum progesterone were measured. Conception rates in control, CIDR without estradiol and CIDR treated cows were 15.4, 38.9 and 60%, respectively. Estrus interval in cows treated with CIDR was 26.5 days compared with 37.1 and 48.5 days in control and CIDR without estradiol treated cows. Estrus signs were more intense in cows treated with CIDR compared with control and CIDR without estradiol treated cows. Pregnant cows treated with CIDR showed higher serum progesterone concentration than pregnant and non-pregnant cows in control group. Particularly, serum progesterone was significantly higher in CIDR treated pregnant cows compared to non-pregnant cows at days 1, 2 and 6 of gestation. It may be concluded from present results that CIDR treatment is better than CIDR without estradiol to improve conception rate in dairy cattle.

Production of Transgenic Cattle by Non-surgical Embryo Transfer (비외과적 수정란 이식에 의한 형질전환 소 생산 기술)

  • Recently, the transgenic animal production technique is very important for the production of bio-parmaceutical as animal bio-reactor system. However, the absence of survival evaluation in vitro produced transgenic embryos has been a problem of the low productivity of transgenic animal because of absent of pre-estimate of pregnancy after transgenic embryos transferred into recipient. Therefore, this study is conducted to improve efficiency of transgenic cattle production by improving the non-surgical embryo transfer (ET) method. Transgenic bovine embryos were produced by injection of feline immunodeficiency virus enhanced green fluorescent protein (FIV-EGFP) lentiviral vector into perivitelline space of in vitro matured MII stage oocytes, and then in vitro fertilization (IVF) was occured. Normal IVF and EGFP expressing blastocysts were transferred into recipients. Results indicated that 2 expanded blastocysts (34.7%) transferred group showed significantly (P<0.05) higher pregnancy rate than 1 expanded blastocyst (26.8%) transferred group. In case of parity of recipient, ET to heifer (34.9%) showed significantly (P<0.05) higher pregnancy rate than ET to multiparous recipient (21.2%). However, there are no significant differences of pregnancy rate between natural induced estrus and artificial induced estrus groups. Significantly (P<0.05) higher pregnancy rate was obtained from recipient group which have normal corpus luteum with crown group (34.8%) than normal corpus luteum without crown (13.6%). Additionally, treatment of $100{\mu}g$ Gn-RH injection to recipient group (38.6%) 1 day before ET significantly (P<0.05) increase pregnancy rate than non- Gn-RH injection to recipient group (38.6%). We also transferred 2 EGFP expressing expanded blastocysts to each 19 recipients, 7 recipients were pregnant and finally 5 EGFP transgenic cattle were produced under described ET condition. Therefore, our result suggested that transfer of 2 good-quality expanded blastocysts to $100{\mu}g$ of Gn-RH injected recipient which have normal corpus luteum with crown is feasible to produce transgenic cattle.

Relationship between Various Estrous Behavioral Signs and Ovulation Time in Dairy Cows (젖소의 다양한 발정 행동 징후와 배란 시간과의 관계)

  • The objective of this study was to investigate the relationship between various estrous behavior and ovulation time, and to determine which estrous behavior could predict ovulation time more accurately. In total, 37 ovulations and 28 estrous detection were observed in 51 Holstein-Friesian dairy cows. Various estrous behavior were observed during 72 h from two days after $PGF_2{\alpha}$ injection and their relation with the time of ovulation (ultrasound examinations at 4-h intervals) was investigated. In estrous periods, the rate of sniffing, chin resting, mounting, standing heat was 81%, 78%, 78% and 56%, respectively. Ovulation occurred $25.6{\pm}7.9h$ after onset of estrus (ranging between 7 and 37h) and $13.4{\pm}7.1h$ after end of estrus (ranging between 1 and 28h). Interval between onset of estrus and ovulation time was significantly (p<0.05) shorter for standing heat $(17.33{\pm}5.83h)$ than for mounting, sniffing and chin resting $(23.58{\pm}5.12h,\;24.25{\pm}6.09h,\;23.42{\pm}6.04h)$. In 88% of the animals that displayed mounting, ovulation occurred between $16{\sim}28h$ after onset of mounting. Onset of standing heat, sniffing and chin resting occurred between $10{\sim}22(81%)h,\;16{\sim}28(79%)h\;and\; 19{\sim}31(79%)h$ before ovulation respectively. Sniffing and chin resting were displayed during the non-estrous period and are therefore, not useful predictors of ovulation time. The standing heat and mounting can be a good predictor for time of ovulation but the disadvantage of using standing heat is that only a limited number of cows display standing heat. Thus, it is concluded that mounting behavior could be the best predictor for time of ovulation.
