Acetylcholinesterase Inhibition and Behavioral Changes of Crucian carp (Carassius auratus) Exposed to the Waterborne Parathion (Parathion에 노출된 붕어(Carassius auratus)의 Acetylcholinesterase 억제와 행동변화)

  • The investigation of the Acetylcholinesterase (AChE) activity in tissues (brain, eye, muscle and serum) of crucian carp (Carassius auratus) exposed to the waterborne parathion was carried out for application as biomarker of organophosphate pesticides. The AChE activities were significantly inhibited in the experimental organs of C. auratus treated ${\geq}63{\mu}g/L$ of concentrations of parathion. The AChE activity of C. auratus was significantly reduced in response to brain (79.1~92.4%), eye (76.0~91.5%), muscle (89.7~97.6%) and serum (68.9~78.0%) after 30 days exposure. No significant mortality occurred during the experiment duration but behavioral changes occurred in the carp after exposure to the parathion were erratic swimming and convulsions. The anomaly in the carp exposed to parathion were observed in the form of scoliosis. The use of AChE activity and other adverse responses of the carp might be use as a reliable monitoring tool to detect parathion in aquatic ecosystem which might produce significant population changes.

  • Carp, Cypinus carpio, has been esteemed as one of the most tasty fresh water fishes in Korea. But little study on its food has been reported. The composition of fatty acids, amino acids, nucleotides and their related compounds as chemical component of carp muscle were analyzed by gas-liauid chromatography, amino acid autoanalyzer and high speed liauid chromatography. The fatty acids of the carp muscle lipid consisted of the large amount of $C_{18}\;:\;2,$ $C_{18}\;:\;1,$ $C_{18}\;:\;3,$ $C_{16}\;:\;0,$ $C_{16}\;:\;1,$ acids, small amount of $C_{16}\;:\;2,$ $C_{18}\;:\;0,$ $C_{18}\;:\;4,$ $C_{14}\;:\;0,$ $C_{14}\;:\;1,$ $C_{12}\;:\;0,$ acids, and $C_{13}\;:\;1,$ $C_{13}\;:\;0,$ $C_{20}\;:\;1,$ $C_{17}\;:\;0,$ $C_{15}\;:\;1,$ $C_{17}\;:\;1,$ $C_{22}\;:\;1$ acids were smaller. In fresh carp, the results showed that inosine $17.25{\mu}mole/g$, hypoxanthine $10.06{\mu}mole/g$ were dominant and the content of GMP, IMP, AMP, UMP, CMP were 1.56, 1.36, 0.92, 0.49, $0.34\:{\mu}mole/g$ on dry base, respectively. Histidine and lysine were dominant amino acids in carp extract, having 49.4% (214.6mg%), 33.9% (147.1mg%) of total free amino acid contents, respectively, but the content of glycine, serine, alanine and glutamic acid were low, and arginine, aspartic acid, threonine, proline, valine, methionine, isoleucine, phenylalanine were detected in trace amount. The content of amino acid composition showed that glutamic acid, aspartic acid, lysine, leucine, alanine, valine and arginine were the most abundant amino acids, while such amino acids as glycine, isoleucine, phenylalanine, threonine, histidine, methionine, serine and tyrosine were low, and proline was detected in trace amount.

  • Bioaccumulation status and distribution characteristics of PCDDs, PCDFs and DL-PCBs in cruian carp collected from the four representative sites of major river systems in Korea were investigated. The recovery rates of PCDDs and PCDFs ranged from 50.6% to 88.3%, and those of DL-PCBs ranged from 52.3% to 93.2%. The mean, median and concentration range of ${\sum}$dioxins, which represents the total concentratons of PCDDs, PCDFs and DL-PCBs accumulated in the muscle of crucian carp, were 0.39, 0.14 and 0.047-1.0 pg TEQ/g wet wt., respectively. DL-PCBs were detected above the detection limit from all the samples, whereas PCDDs and PCDFs were detected from limited crucian samples. The relative contribution of DL-PCBs to ${\sum}$dioxins was remarkably larger than those for PCDDs and PCDFs. The percent contribution was 83.6% for DL-PCBs, and followed by 12.7% and 3.7% for PCDFs and PCDDs, respectively.

Nutritional and Digestive Properties of Israeli Carp Cyprinus carpio as Affected by Size, Part and Commercial Value (향어(Cyprinus carpio)의 크기별, 부위별 및 상품성별 영양 및 소화 특성 비교)

  • This study was conducted to compare sensory and nutritional and digestive properties of the Israeli carp Cyprinus carpio muscle (ICM) by size [normal large (NLF) and small (NSF) fish], part [ventral (VM) and dorsal (DM) muscle], and commercial value [NLF, recessive (RF) or deformed (DF) fish]. In NLF, the major amino acids were lysine, aspartic acid, and glutamic acid, whereas aspartic acid and glutamic acid were predominant in NSF, DM, VM, DF, and RF. Taurine content increased with fish size, and was higher in DM than in VM, and higher in NLF than in DF but lower than that in RF. Regardless of ICM size, part, and commercial value, potassium and iron were expected in the health function, but calcium and zinc were not expected. The major fatty acids present in ICM were 16:0, 18:1n-9, and 18:2n-6, regardless of size, part, and commercial value. The digestion rate of ICM by digestive organ was lower in NLF than in NSF, and lower in VM with a high lipid content than in DM, and lower in DF than in RF.

  • To obtain basic data on food components of triploid fish, we undertook the analysis of free amino acid, nucleotide, total creatinine, betaine, trimethylamine oxide(TMAO) and their related compounds in diploid and triploid carps(Cyprinus carpio). The contents of total free amino acid and its related compounds in belly and dorsal muscles of triploid carp were 346.1mg/100g and 333.4mg/100g. Histidine occupied $45.1\%\;and\;46.9\%$ in belly and dorsal muscles, and followed by taurine, lysine and glycine in order. As for the compositions of nucleotide and its related compounds in those muscles of triploid carp, IMP were revealed $73.9\%\;and\;65.8\%$ of total nucleotide and its related compounds. The major component of the other organic base in those muscles of triploid carp was total creatinine, but betaine and TMAO were poor. The contents of taste compounds such as free amino acid, nucleotide, total creatinine, betaine, TMAO and their related compounds were less in triploid carp than in diploid carp of nonspawning season, while were more in triploid carp than in diploid carp of spawning season. Total amino acid contents were more in diploid carp of nonspawning season than in triploid carp, but mineral contents were more in triploid carp than in diploid carp of spawning season. Therefore, it is believed that triploid carp is very worthy for a tasty and nutritional food source.

First report on Myxobous artus infection in leather carp (Cyprinus carpio nudus) in Korea (향어(Cyprinus carpio nudus)의 Myxobolus artus 국내 첫 감염사례 보고)

  • Ellipsoidal-shaped spores with two polar capsules were detected in leather carp (Cyprinus carpio nudus) muscle. 18S rDNA gene analysis of the spore showed a 99.58% match to Myxobolus artus, a myxozoan parasite. As a result of phylogenetic analysis using the Bayesian inference model and maximum likelihood model among other Myxobolus species, the isolate in the present study belonged to the M. artus cluster. This is the first case report of M. artus infection detected in domestic aquaculture organisms in Korea.

Hot Water Extract of Leather Carp (Cyprinus carpio nudus) Improves Exercise Performance in Mice

  • The hot water extract of leather carp (Cyprinus carpio nudus) has been used as a nourishing tonic soup and as an aid for recovery from physical fatigue. In this study, we investigated the effect of leather carp extract on exercise performance in mice. Swimming endurance and forelimb grip strength were assessed following oral administration of the extract (once per day for 7 days) at a dose of $0.5 mg/10{\mu}L/g$ body weight. After 7 days, mice given the leather carp extract had significantly greater swimming endurance [$105{\pm}18s$ (P<0.05); 52% longer than day 0] and forelimb grip strength [$1.18{\pm}0.05$ Newton (P<0.01); 17% greater than day 0]. The extract increased muscle mass, but had little effect on body weight. Following the swimming exercise, blood glucose, glutathione peroxidase, and superoxide dismutase levels in extract-fed mice were significantly higher (145%, 131%, and 106%, respectively) than in the saline control group. Blood levels of high-density lipoprotein cholesterol were also significantly increased (128%) in mice given the extract compared to the controls. These results suggest that leather carp extract can improve physical exercise performance and prevent oxidative stress caused by exhaustive workouts.

  • The sensor to determine ATPase activities was consisted of an immobilized enzyme membrane(purine nucloside phosphoryrase and xanthine oxidase) and an oxygen electrode. The proposed sensor was used for the determination of $K^+-, Ca^{2}+- and Mg^{2+}-$ATPase activities in several fish muscle proteins such as Thunnus albacares(Yellowfin tuna), Tetrapturus audax(Striped marlin), Prognichthys agoo(Japanese flyingfish), and Cypvinus carpio(Carp). $K^+-, Ca^{2}-$ATPase activities measured by the proposed sensor system were in good agreement with the results obtained by a conventional colorimetric assay. One cycle of assay could be completed within 3mlnutes.

  • The present study was undertaken to investigate and compare the effect and mode of action of tachykinins on isolated strip preparations of the intestinal smooth muscle from the nile tilapia, Oreochromis niloticus and the Israel carp, Cyprinus carpio. Both of neurokinin 1(NK-1) receptor agonist, substance P(SP) and neurokinin 2(NK-2) receptor agonist, neurokinin A(NKA) caused concentration-dependent contractions of intestinal smooth muscle in the nile tilapia and the israel carp. The efficiency and potency of these agonists varied between two fish species. In the nile tilapia intestine, the efficiency and potency of SP were greater than those of NKA. However, the efficiency and potency of SP were similar to those of NKA. In the nile tilapia intestine and the israel carp intestine, the contractile responses of SP and NKA were noncompetitively antagonized by NK-1 receptor antagonist, L-732, 138 but unaffected by NK-2 receptor antagonist, MDL 29913. In addition, SP-induced contractions in the both of preparation were significantly inhibited by muscarinic antagonist, atropine($5{\times}10^{-7}$M) and ganglionic inhibitor, tetrodotoxin($2{\times}10^{-7}$M) but NKA-induced contractions were unaffected by those. These results indicate that two tachykinin agonists, SP and NKA predominately modulate the mechanical activity of isolated preparation from the nile tilapia and the israle carp directly through the activation of NK-1 receptor on the intestinal smooth muscle cells, but in the case of SP action, the indirect action through activation of cholinergic nerve terminals seems to be also implicated.

Preparation and Sensory Properties of Semi-dried Israeli Carp Cyprinus carpio (향어(Cyprinus carpio)를 활용한 반건제품의 제조 및 관능 특성)

  • The aims of this study were to optimize the processing procedure of high-quality semi-alternating temperature dried Israeli carp Cyprinus carpio (SAD-IC) and to investigate the sensory properties of the product. Based on the differences in moisture content of the dorsal and ventral muscles, high quality SAD-IC was prepared by alternating the drying temperature between 4 h at 35℃, and 2 h at 5℃, three times before final drying for 2 h at 35℃. The surface in SAD-IC produced under the optimal alternating-temperature drying process had a markedly superior browning index value and softer texture compared to products produced using constant-temperature drying. SAD-IC produced from dorsal and ventral muscle had significantly superior taste values than the raw material. These results suggest that SAD-IC has the potential to be industrialized.
