Plant Growth Promotion Activity of Endophytic Fungi Isolated from the Roots of Calystegia soldanella (갯메꽃 뿌리로부터 분리된 내생진균의 식물생장촉진활성)

  • Eight endophytic fungal strains were isolated from the roots of Calystegia soldanella from the western coast of South Korea. The culture filtrate of the eight endophytic fungi were applied to waito-c rice seedlings in order to verify potential plant growth promotion activities. The results of bioassay indicated that the Cs-9-7 fungal strain possessed the highest plant growth promotion activity. Fungal culture filtrates were analyzed to verify secondary metabolites using gas chromatography and mass spectroscopy with selected ion monitoring (GC/MS-SIM). The culture filtrate of the Cs-9-7 fungal strain was confirmed to contain gibberellins GA3 (1.229 ng/mL), GA4 (3.535 ng/mL), GA7 (1.408 ng/mL) and GA12 (0.378 ng/mL). Polymerase chain reactions (PCR) were performed so as to determine the internal transcribed spacer (ITS) regions for the identification of isolated strains with universal primers ITS-1 and ITS-4. The Cs-9-7 fungal strain, isolated from the root of C. soldanella, has been named Aspergillus tubingensis Cs-9-7.

Synecology and Habitat Environment of Coastal Sand Dune Vegetation in Uido (Island), Korea (우이도 해안사구식생의 군락생태와 입지환경)

  • The aim of this study is to provide a fundamental data which can be applied to management conservation, and restoration of coastal sand dune vegetation by determining the classification and distribution of community, and ecological characteristics of the habitat. This research was performed by the Braun-Blanquet's method. The coastal sand dune vegetation of Uido (I.) was composed with 9 communities as follows : Vitex rotundifolia community, Zoysia macrostachya community, Carex kobomugi community, Iachaemum anthephoroides community, Imperata cylindrica var. koenigii community, Carex pumila community, Calystegia soldanella community Messerschmidia sibirica community and Pinus thunbergii community, the evergreen needle-leaved forest. The constancy degree showed high in Calystegia soldanella (77%), Vitex rotundifolia (74%), Carex kobomugi (66%), Zoysia macrostachya (50%) and Imperata cylindrica var. koenigii (47%). However the highest constancy degree Calystegia soldanella has, it turned out to have low net contribution degree (NCD) in each community. In terms of the distribution and growth feature of the composition species in the coastal sand dune, Vitex rotundifolia, Carex kobomuri and Zoysia macrostachya were widely distributed from unstable sand dune to stable one but Iachaemum anthephoroides and Imperata cylindrica var. koenigii were mainly found at the stable sand dune. Carex kobomugi was especially dominant at the unstable sand dune where the sand continued to be deposited. On the other hand, Carer pumila and Messerschmidia sibirica showed regional distributions around fresh water.

Comparison of the Total Phenolic and Flavonoid Contents and Antioxidant Activities of Four Kinds of Sand Dune Plants Living in Taean, Korea (태안에서 서식하는 사구식물 4종의 폴리페놀, 플라보노이드 함량과 항산화 활성 비교)

  • This study attempted to investigate the antioxidant properties of four different species of sand dune plants (Calystegia soldanella, Messerschmidia sibirica, Vitex rotundifolia and Rosa rugosa). In order to validate the antioxidant activity of these plants, we first determined the total amount of flavonoid versus phenolic contents (TFC/TPC) and extracted crude flavonoids for measuring antioxidant activities that were determined by DPPH, ABTS and FRAP assays with radical scavenging effects. We found that highest amounts of TPC were detected in R. rugose with values of 110.20 mg/g (leaves) and 65.71 mg/g (stems), while the highest amounts of TFC in V. rotundifolia with values of 38.07 mg/g (leaves) and 6.55 mg/g (stems). We further examined how closely related the amounts of TFC/TPC in antioxidant activities and found that R. rugose has the highest activities of radical scavenging with values of $63.4{\mu}g/ml$ and $51.2{\mu}g/ml$ determined by DPPH and ABTS assays compared with the value of $21.2{\mu}g/ml$ by FRAP assay. It is of note that there is a statistically significant correlation between the resulting antioxidant activities and the total ratio of TFC and TPC, suggesting that the different amounts of TFC/TPC may directly contribute to the various antioxidant activities.

Anti-invasion Effects of Calystegia soldanella Solvent Extracts and Partitioned Fractions on PMA-stimulated Fibrosarcoma Cells (갯메꽃 추출물과 유기용매 분획물의 암전이 억제 효과)

  • Calystegia soldanella is distributed in coastal sand dunes and has high environmental adaptability; it is also known to be effective for anti-oxidant, anti-pyretic, anti-septic, and diuretic action. This study investigated the effect of crude extracts and organic solvent fractions of C. soldanella on MMP-2 and MMP-9 expression, MMP activity, and cell mobility in phorbol-12-myristate-13-acetate (PMA)-induced fibrosarcoma HT-1080 cells. C. soldanella was twice extracted, once with methylene chloride (MC) and once with methanol (MeOH). After the MC and MeOH extracts were combined, their suppressive effects on MMP-2 and MMP-9 expression, MMP enzymatic activity, and gene and protein expression were measured by gelatin zymography, enzyme-linked immunosorbent assay, reverse-transcription polymerase chain reaction, and western blot method. Cell mobility for the HT-1080 cells was observed by wound healing assay. The combined crude extracts showed a significant suppressive effects on MMP-2 and MMP-9 expression. To explore active inhibitory elements, the combined extracts were fractionated according to polarity into with n-hexane, 85% aqueous methanol, n-butanol, and water. Across these four solvent fractions, MMP-2 and MMP-9 activity and cell mobility in the HT-1080 cells were all strongly inhibited by the n-hexane fraction. These results suggest that C. soldanella extract and organic solvent fractions could be used as potent MMP inhibitors for effective anti-cancer treatments to suppress cancer invasion and metastasis.

Effect of Halophyte (Spartina anglica and Calystegia soldanella) Extracts on Skin Moisturizing and Barrier Function in HaCaT Cells (염생식물인 갯끈풀과 갯메꽃 추출물의 HaCaT 세포에서 피부 보습 및 피부 장벽 기능에 미치는 영향)

  • As aging progresses, reactive oxygen species (ROS) reduces skin moisturization and collapses skin barrier function. In this study, we evaluated the efficacy of skin moisturizing and skin barrier function enhancement by extracts from halophytes using HaCaT cells. Spartina anglica (S. anglica; SAE) and Calystegia soldanella (C. soldanella; CSE), a kind of halophytes, were collected from Dongmak beach in Incheon, and extracted with 70% ethanol. At the first, we evaluated the cytotoxicity of extracts in HaCaT cell using WST-8 Kit. As a result, the other experiment was conducted by setting the concentration at which the cell viability was 90% or more. SAE and CSE showed high radical scavenging activity through ABTS assay. Expression levels of genes related to skin moisturizing and skin barrier functions, were analyzed by real-time qPCR. As a result, it showed that the expression of aquaporin 3, hyaluronan synthase 2, and transglutaminase 1 was increased by SAE treatment but not changed by CSE. Activation of extracellular signal regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 mitogen activated protein kinase was induced by SAE. These results suggest that SAE can be used as functional materials for cosmetics for skin moisturizing and barrier function enhancement.

The distribution of Jeju coastal sand dune plants and its restoration implications (제주 해안사구 식물 분포와 복원을 위한 의미)

  • The coastal dune ecosystem is one of the ecosystems under the most development pressure in Korea. Therefore, it is necessary to study the ecological location and related ecological phenomena of coastal dune plants, but related studies are lacking. Through this study, we intend to conduct research on the structure and restoration of dune plants, focusing on the coastal dunes in Jeju Island, which are affected by artificial development pressure and the continuous increase in tourists among many coastal dunes in Korea. Ecosystems of coastal sand dunes for vegetation survey in Jeju Island are selected based on naturalness and preservation. In this study, 23 major coastal dunes on Jeju Island including Udo were selected. In the coastal dunes of Jeju Island, a whole species survey and quadrat survey were carried out. The vegetation survey at study sites were conducted on May to September 2022, when the vegetation is clearly visible. At the survey site, the dune area was identified at the beginning and the plant species were recorded until no more new species appeared. Vegetation survey in the field was performed by 103 quadrat establishments and was conducted using Braun-Blanquet method. A total of 277 species appeared, and the most common species were Vitex rotundifolia and Calystegia soldanella. The frequency of both Vitex rotundifolia and Calystegia soldanella was approximately over 90%. The proportion of woody and herbaceous in all emerging species was 7.2% and 92.8%, respectively. The total number of species found in the quadrat survey was 98. As a result of classifying plant communities based on species dominance in the quadrats, it was analyzed into 30 plant communities. The plant communities that appeared with a frequency of 2 or more on the main island of Jeju were Vitex rotundifolia, Imperata cylindrica var. koenigii, Ischaemum antephoroides, Wedelia prostrata, Elymus mollis, Calystegia soldanella, Artemisia scoparia, and Tetragonia tetragonoides. The DCCA(detrended canonical correspondence analysis) based on the vegetation and environment factor matrix showed that the height and covers of the dominant plant species explain significantly the variation and distribution of coastal sand dune species on Jeju island. Thus, we may propose a plan to restore the coastal dunes of Jeju island as helping colonization and establishment of mainly sand dune native perennials and trees, preserving native plant communities that are declining and preserving present tree strips of Pinus thunbergii, Litsea japonica, Pittosporum tobira and Vitex rotundifolia.

Physiological Responses of Calystegia soldanella under Drought Stress

  • This study was conducted to determine the extent of drought resistance based on physiological responses of Calystegia soldanella under water deficit. In order to investigate the changes of plant growth, stomatal density, photosynthesis, chlorophyll fluorescence, the contents of chlorophyll and carotenoid, osmolality, total ion contents, the contents of carbohydrate and proline, C. soldanella was grown under well watered and drought stressed conditions for 12 days. In this study, water-deficit resulted in remarkable growth inhibition of C. soldanella. The effect of water-deficit on plant growth was associated with low osmotic potential of soil. On day 12 after drought treatment, dry weight, relative water contents, number and area of leaves and stem length were lower than those of control. The stomatal conductance and net photosynthetic rate were significantly reduced in water stressed plant to regulate inner water contents and $CO_2$ exchange through the stomatal pore. Chlorophyll fluorescence and chlorophyll contents were not different in comparison with the control, indicating that the efficiency of photosystem II was not affected by drought stress. This results could be explained that water-deficit in C. soldanella limits the photosynthetic rate and reduces the plant's ability to convert energy to biomass. A significant increase in total ion contents and osmolality was observed on day 7 and day 12. Accumulation of proline in leaves is associated with the osmotic adjustment in C. soldanella to soil water-deficit. Consequently, this increase in osmolality in water stressed plant can be a result in the increase of ion contents and proline.

Isolation and Characterization of Bacteria Associated with Two Sand Dune Plant Species, Calystegia soldanella and Elymus mollis

  • Little is known about the bacterial communities associated with the plants inhabiting sand dune ecosystems. In this study, the bacterial populations associated with two major sand dune plant species, Calystegia soldanella (beach morning glory) and Elymus mollis (wild rye), growing along the costal areas in Tae-An, Chungnam Province, were analyzed using a culture-dependent approach. A total of 212 bacteria were isolated from the root and rhizosphere samples of the two plants, and subjected to further analysis. Based on the analysis of the 16S rDNA sequences, all the bacterial isolates were classified into six major phyla of the domain Bacteria. Significant differences were observed between the two plant species, and also between the rhizospheric and root endophytic communities. The isolates from the rhizosphere of the two plant species were assigned to 27 different established genera, and the root endophytic bacteria were assigned to 21. Members of the phylum Gammaproteobacteria, notably the Pseudomonas species, comprised the majority of both the rhizospheric and endophytic bacteria, followed by members of Bacteroidetes and Firmicutes in the rhizosphere and Alphaproteobacteria and Bacteroidetes in the root. A number of isolates were recognized as potentially novel bacterial taxa. Fifteen out of 27 bacterial genera were commonly found in the rhizosphere of both plants, which was comparable to 3 out of 21 common genera in the root, implying the host specificity for endophytic populations. This study of the diversity of culturable rhizospheric and endophytic bacteria has provided the basis for further investigation aimed at the selection of microbes for the facilitation of plant growth.

Changes of the Coastal Sand Dune Vegetation after the Construction of an Embankment in Anmado

  • This study examined the coastal sand dune vegetation before and after the construction of an embankment on Anmado Island in order to compare vegetation in relation to the development of islands. A total of 24 species distributed on the sand dunes. 18 species were found to be in common before and after the construction of the embankment, which included Vitex rotundifolia, Imperata cylindrica var. koenigii, Zoysia sinica, etc. The species which were not found in this survey included Rumex japonicus, Setaria viridis, Portulaca oleracea, Artemisia japonica, Poiygonum aviculare, etc, and new species included Arena fatua, Carex boottiana, Lycium chinense, Leonurus sibiricus, Torilis japonica, Solanum carolinense, etc. The washing away of sand brought about the changes in habitat and the increase in naturalized plants, which included Oenothera odorata, Lepidium apetalum, Bidens bipinnata, Erigeron canadensis, Datura stramonium, Xanthium strumarium, Arena fatua, Solanum carolinense etc. In addition, the disturbance to this habitat led to the changes in vegetation. The main plant communities in the surveyed site were classified as Vitex rotundifolia-Imperata cylindrica var. koenigil community, Zoysia sinica-Calystegia soldanella community and Messerschmidia sibirica community. The sand dune vegetation on Anmado Island changed with regard to the community and the composition of species after the construction of the embankment, due to the sand being severely eroded. While Vitex rotundifolia community and Commelina communis community were found before the construction of the embankment, they were replaced by Vitex rotundifolia-Imperata cylindrica var. koenigii community, Zoysia sinica-Calystegia soldanella community and Messerschmidia sibirica community, after the construction of the embankment.

A Herbological Study on the plants of Convolvulaceae in the Korean (한국산 메꽃과 식물에 관한 본초학적 연구)

  • Objective : For the purpose of developing Convoivulaceae plants recorded in Korean Herbology in Korea, the literatures of the successive generations have been thoroughly investigated to prepare this article. Methods : Books and research papers about Herbology which published at Korea and other countries were examined. Results : A list was made about Convoivulaceae plant which were cultivated or grew naturally in Korea, after investigated the data on domestic and foreign Convoivulaceae plants. Convoivulaceae(grew in Korea) were divided into 6 classes with 18 species. Out of those, 6 classes with 13 species were found serviceable which indicates approximately 72% of all. Out of all 18 species of Convoivulaceae family, Calystegia genus were found 6 species, which were shown the most. Among 4 genus of Calystegia species, medicinal plants were all one species each. Out of all serviceable parts in Convoivulaceae, Herb·Radix·Tuber·Semen·Flower·Stem and Folium parts took first place as 8 species. There are 2 species toxic, the survey said. 6 Classifying medicinal plants among the Convoivulaceae plants by part, whole Herbs are found to be 8 separate species each. When it comes to the nature of taste, 10 kinds of taste without action and 7 kinds of sweet taste are the most common. For channel tropism, kidney channel has the largest number of 7 types. Conclusion : A total of 6 genera and 18 species of Convoivulaceae was found in the Korean among which 6 genera and 13 species(approximately 72%) were medicinal plants. Two species toxic drugs were detected.
