한약이 난소제거 흰쥐와 SAM P6 생쥐의 골소주에 미치는 영향 (Effects of Herbal Medicine on Tibia Bone Changes in Ovariectomized Rats and SAM P6 Mice)

  • The effects of herbal medicine on travecular bone area were studied using ovariectomized rat as an animal model of Type I osteoporosis and SAM P6 as that of Type II. We counted red blood cells(RBC), hemoglobin(Hb), and hematocrit(Hct) using Couter`sR method. Each traditional boiling water extract of Achyrathis Radix, Psoraleae Fructus, Rehmanniae Radix Preparat, and Cornii Fructus and a systemic water extract of Astragali Ractiex was given 5g/kg/day, p.o., for 30 days in a group of 4-5 ovariectormized rats. One ml of blood was taken by tail vein at day 0, 7, 14, 21, and 30 days after administration of the extract. The traditional hot water extract of Cervi parvumn Corni (Cervi) was given the same dose as described above for 14 days in a group of 10 SAM P6 mice and systemic water extract of Astragali Radix was administered as the same dose as above for 30 days in 10 SAM P6 mice. Travecular bone area was measured 5 mcm decalcified and stained thin bone slice by image analysis using a digitalizer. In Type I, ovariectomized rats, administration of Astragali Radix, Rhemanniae Radix Preparat, and Corni Fructus decreased in RBC, Hb and Hct. In Type II, administration of Cervi increased in RBC and Hct and that of Astragali Radix was also elevated RBC. In Type I, any administration of herbal medicine used in this study did not elevate travecular bone area significantly except Corni Fructus showed a trend of increase in travecular bone area. However, Type II, Cervi and Astragali Radix increased in both mean and total travecular bone area. Thus, there are significant difference in response of herbal medicine in different types of osteoporosis.

산수유 에텔분획물의 약리작용에 관한 연구 (Pharmacological Studies on Ether Fraction of Corni Fructus)

  • The ether fraction obtained from dry fruit of Cornus officinalis was investigated for the anti-inflammatory action, acute toxicity and central nervous system activities. From several pharmacological examinations, it was found that the ether fraction suppressed considerably carrageenin edema at the dose of 600mg/kg p.o. in rats, inhibited the granuloma formation in rats as given 100mg/kg p.o. for 7 days and decreased the swelling of both of complete adjuvant injected and noninjected (contralateral) paws of the rats at the dose of 100mg/kg p.o. given for 14 days. The $LD_{50}$ of the fraction are estimated to be more than 2,000mg/kg p. o. and 642mg/kg i. p. in mice. And the fraction did not show any sedative, stimulative, analgesic and anticonvulsant action but exhibited hypothermic action. These results might be concluded that the ether fraction of Corni fructus showed anti-inflammatory action in both of acute and chronic type models without any considerable central nervous depressant activity and exhibited very weak acute toxicity in mice.

  • Free radicals cause many diseases and aging. In our body coexist useful and harmful bacteria. Some useful intestinal bacteria such as L. plantarum, B. adolescentis, B .bifidum, B. infantis, C. butyricum were presented positive growth in culture medium bearing natural products. L. plantarum was more effectively grown by mixed natural products of CORNI FRUCTUS+THEAE FOLIUM+COPTIDIS RHIZOMA+PAEONIA JAPONICA, B. adolescentis was BEIAMCANDA CHINENSIS+ANGELICAE GIGANTIS RADIX+EPIMEDII HERBA+PAEONIA JAPONICA, B. bifidum was NELUMBO NUCLFERA GAERTNER+THEAE FOLIUM+BEIAMCANDA CHINENSIS+CORNI FRUCTUS, B. infantis was THEAE FOLIUM+CHELIDONIUM MAJUS L.+ASTRAGALI RADIX+EPIMEDII HERBA, C. butyricum was EPIMEDII HERBA+PUERARIAE RADIX+NELUMBO NUCLFERA GAERTNER+PAEONIA JAPONICA,etc. These natural products will support an important role in bio-industry.

Physicochemical and Sensory Properties of Yogurt Supplemented with Corni fructus during Storage

  • This study was carried out to determine a possibility of adding Corni fructus extract (CFE) into yogurt for improving the neutraceutical properties of yogurt and the effects of adding CFE (2~6%, v/v) on the physicochemical and sensory properties of the products during a 15-day storage period at $4^{\circ}C$. Incorporation of CFE into the yogurt samples resulted in a significant pH reduction and a significant increase in titratable acidity. When evaluating the color of the yogurt, the $L^*$-values were not significantly influenced by CFE supplementation; however, the $a^*$- and $b^*$-values significantly increased with the addition of CFE during storage. The power law and Casson models were applied to assess the flow behavior of CFE-added yogurt samples. The magnitudes of apparent viscosity (${\eta}_{a,100}$), consistency index (K), and yield stress (${\sigma}_{oc}$) for 4~6% CFE yogurt samples were significantly greater than those for the control, indicating that CFE can be used as a thickening agent for yogurt. The sensory test revealed that addition of CFE (2~4%) to yogurt did not significantly affect the overall scores, but the overall preference score for 6% CFE yogurt was significantly decreased. Based on the data obtained from the present study, we concluded that the concentrations (2~4%) of CFE could be used to produce a CFE-added yogurt without the significantly adverse effects on the physicochemical and sensory properties.

산수유 퓨레로 제조한 젤리의 품질 특성 (Quality Characteristics of Jelly Using Fresh Puree of Sansuyu (Corni Fructus))

  • 본 연구는 산수유 퓨레를 첨가하여 젤리를 제조하였으며, 젤화제 종류 및 함량, 가열시간에 따른 젤리의 이화학적 품질 특성을 조사하였다. 산수유 퓨레는 수분 함량 89.39%, pH 2.92, 당도 $10.28^{\circ}Brix$, 총폴리페놀 함량 1,791.71 mg GAE/100 g이었다. 산수유 퓨레 첨가량이 증가할수록 pH는 3.04~2.97로 소폭 감소하였으나 부착성은 큰 폭으로 감소하였다. 또한, 가열시간이 증가할수록 붉은색이 불안정하여 황색으로 변하였으며, 5분 이상 가열 시에는 젤리가 되지 않았다. 산수유 젤리의 pH는 3.04~2.97, 당도는 $64.31{\sim}66.96^{\circ}Brix$ 범위였다. 산수유 퓨레 10%, 한천 1.5%, 카라기난 1.0% 첨가한 젤리의 기호도가 5.15로 가장 높게 평가 되었다. 산수유 과육을 이용함으로써 과육에 포함된 다양한 생리활성 물질을 효과적으로 섭취할 수 있는 젤리 제조가 가능할 것으로 본다.

음경해면체에 미치는 산수유(山茱萸) 물 추출물의 이완효과 (Relaxation Effects of Corni Fructus Water Extract on Corpus Cavernosum)

  • To investigate the effect of Corni Fructus(CF) water extracts on the relaxation of the corpus cavernosum, organ bath studies, histochemical and immunohistochemical methods were used and obtained the following results. In the organ bath study, the maximal contraction of the corpus cavernosum tissue by PE showed a significant relaxation effect at $3.0mg/m{\ell}$ of CF water extract. When ${\text\tiny{L}}$-NNA was pretreated corpuscular relaxation effect of CF water extract was significantly inhibited compared without ${\text\tiny{L}}$-NNA pretreatment. In $Ca^{2+}$-free solution, the increase of contraction due to $Ca^{2+}$ influx significantly inhibited in the pretreatment compared with no pretreatment of CF water extract. Histochemical and immunohistochemical studies showed that the ratio of smooth muscle to collagen fiber was increased in the CF group compared to the PE group in the corpus cavernosum, and the eNOS positive reaction increased and the PDE5 positive reaction decreased. These results suggest that CF extract has increased NO production through activation of eNOS and inhibited the action of PDE5 to block the extracellular $Ca^{2+}$ influx, thereby relaxing the smooth muscle of the corpus cavernosum.

산수유의 혈관이완효과 기전에 대한 연구 (Mechanism of Corni Fructus Induced Vasorelaxation in Rabbit Carotid Artery)

  • This study is conducted to investigate vasorelaxant effect of Corni Fructus(CF) on rabbit carotid artery. To determine vasorelaxant effect of CF on rabbit carotid artery, arterial sections with intact or removed endothelium were used in this organ bath study. After being contracted by phenylephrine(PE), arterial sections were treated with CF extract in a dose-dependent manner. To identity its mechanism, the contracted arterial sections by PE were pretreated with indomethacin(IM), tetraethylammonium chloride(TEA), Nω-nitro-L-arginine(L-NNA) or methylene blue(MB) and 1.0 ㎎/㎖ CF extract. We also studied to confirm the effect on influx of extracellular calcium chloride(Ca2+) of the CF extract in rabbit carotid artery. To measure the cytotoxicity of the CF extract, cell viability of human umbilical vein endothelial cell(HUVEC) was measured by MTT assay. Generation of nitric oxide(NO) was also measured by Griess reagent. The arterial sections with intact endothelium were relaxed significantly by CF extract, but this effect was inhibited in the arterial sections with damaged endothelium. The vasorelaxant effect was inhibited significantly when arterial sections were pretreated with IM, TEA, L-NNA, MB. In Ca2+-free krebs solution, increasing of arterial contraction by Ca2+ was also inhibited by CF significantly. The treatment of CF extract increased NO concentration in HUVEC. This study suggested that the vasorelaxant effect of CF extract would be related with endothelium derived relaxing factor(EDRF) such as NO, prostacyclin(PGI2), endothelium derived hyperpolarization factor(EDHF).

산수유의 유리자유기에 의한 간손상 보호효과 (Study on the Protective Effect of Corni Fructus against Free Radical Mediated Liver Damage)

  • Carbon tetrachloride ($CCl_4$)-induced liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Alternations in the activity of cytochrome P450 enzymes affect the susceptibility to hepatic injury from $CCl_4$. In this study, we evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), against an experimental model of hepatotoxicity induced by $CCl_4$. The CF exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Sprague-Dawley (SD) rats, as measured by GOT, GPT, ALP and histological observation. The CF also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. In addition, the expression of CYP2E1, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, was significantly decreased in the liver of CF treated SD rats. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and regulation of CYP2E1 expression.

산수유로부터 추출한 ursolic acid가 과산화수소로 손상된 HEI-OC1 청각세포보호에 미치는 영향 (Protective Effect of Ursolic Acid from Corni fructus on the Hydrogen Peroxide-induced Damage of HEI-OC1 Auditory Cells)

  • The fruits of Cornus officinalis have been used in traditional Oriental medicine for treatment of inner ear diseases, such as tinnitus and hearing loss. In the present study, we showed that the ursolic acid obtained from Corni fructus protected HEI-OC1 auditory cells from hydrogen peroxide cytotoxicity in a dose-dependent fashion. In addition, to investigate the protection mechanism of ursolic acid on hydrogen peroxide cytotoxicity toward HEI-OC1, we measured the effects of ursolic acid on lipid peroxidation and activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) in hydrogen peroxide treated cells. Ursolic acid (0.05 - 2 ${\mu}g/ml$) had protective effect against the hydrogen peroxide-induced HEI-OC1 cell damage and reduced lipid peroxidation in a dose-dependent manner. Pre-treatment with ursolic acid significantly attenuated the decrease in activities of CAT and GPX, but SOD activity was not affected by the ursolic acid or hydrogen peroxide. These results indicate that ursolic acid protects hydrogen peroxide-induced HEI-OC1 cell damage through inhibition of lipid peroxidation and induce the antioxidant enzymes CAT and GPX.
