Low Level of TERC Gene Amplification between Chronic Myeloid Leukaemia Patients Resistant and Respond to Imatinib Mesylate Treatment

  • The amplification of telomerase component (TERC) gene could play an important role in generation and treatment of haematological malignancies. This present study was aimed to investigate copy number amplification status of TERC gene in chronic myeloid leukaemia (CML) patients who were being treated with imatinib mesylate (IM). Genomic DNA was extracted from peripheral blood of CML-IM Resistant (n=63), CML-IM Respond (n=63) and healthy individuals (n=30). TERC gene copy number predicted (CNP) and copy number calculated (CNC) were determined based on $Taqman^{(R)}$ Copy Number Assay. Fluorescence in situ hybridization (FISH) analysis was performed to confirm the normal signal pattern in C4 (calibrator) for TERC gene. Nine of CML patients showed TERC gene amplification (CNP=3), others had 2 CNP. A total of 17 CML patients expressed CNC>2.31 and the rest had 2.31>CNC>1.5. TERC gene CNP value in healthy individuals was 2 and their CNC value showed in range 1.59-2.31. The average CNC TERC gene copy number was 2.07, 1.99 and 1.94 in CML-IM Resistant patients, CML-IM Respond and healthy groups, respectively. No significant difference of TERC gene amplification observed between CML-IM Resistant and CML-IM Respond patients. Low levels of TERC gene amplification might not have a huge impact in haematological disorders especially in terms of resistance towards IM treatment.

Physical Properties and Cytotoxicity of Dental Pit and Fissure Sealants Containing Cerium Oxide Nano Particles(CNPs) (세륨옥사이드나노입자(Cerium Oxide Nano Particles; CNPs)를 첨가한 치면열구전색재의 물리적 특성 및 세포독성)

  • In this study cerium nano particles(CNPs) with 0-4.0 wt% was incorporated to the conventional dental pit and fissure sealant(ConciseTM) to produce new pit and fissure sealant the physical properties and cytotoxicity. The physical properties were measured for polymerizing depth the degree of water absorption and solubility. The cytotoxicity of cell viability was analyzed by MTT assay using immortalized human oral keratinocyte(IHOK). As a result of this preceding study the polymerizing depth was decreased by the increasing of the amount of CNPs. The solubility degree of the sealant added CNPs with 2.0 wt% showed was the lower and the water absorption showed no significantly difference with the control groups(p>0.05). The cytotoxicity test results showed high survival rates in all experimental groups. Therefore, pit and fissure sealant by the addition of CNPs excellent cell viability be produced without weaken the physical property of the cell viability fissure sealant containing CNPs does not weaken physical properties and has no cytotoxic effects biocompatibility. Considering its properties effect of CNPs, further studies are required for distribution technology application.

Increased Gene Expression in Cultured BEAS-2B Cells Treated with Metal Oxide Nanoparticles

  • Recent publications showed that metal nanoparticles which are made from $TiO_2,\;CeO_2,\;Al_2O_3,\;CuCl_2,\;AgNO_3$ and $ZnO_2$ induced oxidative stress and pro-inflammatory effects in cultured cells and the responses seemed to be common toxic pathway of metal nanoparticles to the ultimate toxicity in animals as well as cellular level. In this study, we compared the gene expression induced by two different types of metal oxide nanoparticles, titanium dioxide nanoparticles (TNP) and cerium dioxide nanoparticles (CNP) using microarray analysis. About 50 genes including interleukin 6, interleukin 1, platelet-derived growth factor $\beta$, and leukemia inhibitory factor were induced in cultured BEAS2B cells treated with TNP 40 ppm. When we compared the induction levels of genes in TNP-treated cells to those in CNP-treated cells, the induction levels were very correlated in various gene categories (r=0.645). This may suggest a possible common toxic mechanism of metal oxide nanoparticles.

Effects of Phosphodiesterase 5 Inhibition with Sildenafil on Atrial Contractile and Secretory Function

  • Selective inhibition of phosphodiesterase (PDE) 5 opened a new therapeutic approach for cardiovascular disorders. Therefore, the effect of PDE5 inhibition on the cardiac function should thoroughly be defined. The purpose of the present study was to define the effects of sildenafil, a selective inhibitor of PDE5, on the atrial cGMP efflux, atrial dynamics, and the release of atrial natriuretic peptide (ANP). By perfusing rabbit left atria to allow atrial pacing, changes in atrial stroke volume and pulse pressure, transmural extracellular fluid translocation, cGMP efflux, and ANP secretion were measured. SIN-I, an NO donor and soluble (s) guanylyl cyclase (GC) activator, and C-type natriuretic peptide (CNP), an activator of particulate (p) GC activator, were used. Sildenafil increased basal levels of cGMP efflux slightly but not significantly. Sildenafil in a therapeutic dose increased atrial dynamics (for atrial stroke volume, $2.84{\pm}1.71%$, n=12, vs $-0.71{\pm}0.86%$, n=21; p<0.05) and decreased ANP release ($-9.02{\pm}3.36%$, n=14, vs $1.35{\pm}3.25%$, n=23; p < 0.05), however, it had no effect on the SIN-1- or CNP-induced increase of cGMP levels. Furthermore, sildenafil in a therapeutic dose accentuated SIN-1-induced, but not CNP-induced, decrease of atrial pulse pressure and ANP release. These data indicate that PDE5 inhibition with sildenafil has a minor effect on cGMP levels, but has a distinct effect on pGC-cGMP- and sGC-cGMP-induced contractile and secretory function.

Analgesic Effects of Transcranial Direct Current Stimulation on Central Neuropathic Pain in Spinal Cord Contusive Rat Model

  • The aim is to investigate the analgesic effect of transcranial direct current stimulation(tDCS) on central neuropathic pain(CNP) in spinal cord contusive rat model. Twenty Sprague-Dawley rats($250{\pm}50$ g, male) were used. Thoracic spinal cord(T10) was contused using New York University(NYU) spinal cord impactor. The animals were randomly assigned to two groups; GroupI: Non-treatment after SCI induction(n=10), GroupII: application of tDCS(0.1 mA, 20 min/time, 2 times/day, 5 days/6week) after SCI induction(n=10). Assess the effect of tDCS using the Basso Beattie Bresnahan(BBB) locomotor rating scales, Touch $test^{TM}$ sensory evaluator(TTSE), Plantar test$^{\circledR}$after contusion at the $2^{nd}$, $3^{rd}$, $4^{th}$, $5^{th}$, $6^{th}$ week and the immunohistochemistric response of c-fos in the thalamus, cerebral cortex after contusion at the $3^{rd}$, $6^{th}$ week after SCI. The scores of BBB scales were significantly different from $3^{rd}$week. TTSE were different significantly over time, but there were no differences at each evaluation times on between-measure time effects. Plantar test were different significantly over time and there were difference at the $4^{th}$, $6^{th}$ week after SCI on between-measure time effects. Also, immunohistochemistric response of c-fos was reduced significantly from $3^{rd}$, $6^{th}$ week after SCI in tDCS group compared with control group in thalamus and cortex. These results identified that tDCS of non-invasive therapeutic method may have beneficial analgesic effect on CNP after SCI with behavioral test and immunohistochemical test.

  • Achondroplasia is autosomal dominant genetic disease and fibroblast growth factor receptor 3 (FGFR3) is currently known to be the only gene that causes achondroplasia. Gain-of function mutation in fibroblast-growth-factor-receptor 3 (FGFR3) causes the disease and C-type natriuretic peptide (CNP) antagonizes FGFR3 downstream signaling by inhibiting the pathway of mitogen-activated protein kinase (MAPK). As FGFR3-related skeletal dysplasias are caused by growth attenuation of the cartilage, chondrocytes appear to be unique in their response to FGFR3 activation. However, the full spectrum of molecular events by which FGFR3 mediates its signaling is just beginning to emerge. This article summaries the mechanisms of FGFR3 function in skeletal dysplasias, the extraordinary cellular manifestations of FGFR3 signaling in chondrocytes, and finally, the progress toward therapy for ACH.

  • The present series of investigations have been undertaken to know the effective removal method of some pesticides, heavy metals and detergent when the doses of oxidants and coagulant were changed in the treatment process of drinking water containing organic material. Three pesticides, IBP, diazinon and CNP, were removed by about 68 to 100% by treatment of oxidant, $Cl_2$. Especially, diazinon was completely removed by treatment of $Cl_2$ and other oxidants such as $Cl_2,\;KMnO_4\;and\;O_3$. However, butachlor removal measured only 20.3 to 26.7% due to treatment of all oxidant used. A detergent, sodium dodecylbenzenesulfonate, was effectively removed by treatment of $O_3$ but remained stable in the presence of other oxidants. The heavy metals of Cd, Pb, Cu and Zn in water were not affected by treatment of all oxidants used. By changing the concentration of humic matter and the dosage of coagulant, the removal of heavy metals was increased according to the increase of coagulant dosage, and decreased when increasing the concentration of humic matter, whereas the pesticides and detergent were not effectively removed.

  • The purpose of this study is to evaluate elite high school kayaker's iso-kinetic muscle function and provide physical training advices. Biodex system IV, Biodex stabilizer system, Inbody 3.0 and CNP-5403 are used to investigate 11 kayaker's muscle function(peak torque, average power and work fatigue), dynamic balance, body compositions, and flexibility. S4, S5, S8, S10, S11 are needed to train shoulder muscle strength and power. S5, S6, S10, S11 are needed to strengthen shoulder muscle endurance. S5, S7, S8, S10, S11 are needed to train knee muscle strength and power. S1, S4, S5, S10 are needed to strengthen knee muscle endurance. S3, S10, S11 are needed to train wrist muscle endurance. Based on these results we should establish an muscle function enhancing program with periodic inspection for elite kayakers. And standardized training program should be sublated.

  • In mathematics a prime number is the natural number that has no positive factors other than 1 and itself. As natural numbers greater than 1 can be factored characterized by prime numbers, identities of a culture could be understood if its cultural phenomena are analyzed through cultural prime numbers(CPN). It is not easy to resolve cultural phenomena into CPN and analyze them through CPN due to complexities of culture. Though it is difficult, however, it is not impossible. For CPN keeps relative independence in the context of history and thought. We call 2, 3 and 5 as CPN: 2 is representative of Yin and Yang theory, 3 of Three Principles theory, and 5 of Five Elements theory. We argue that the Ten Celestial Stems and the Twelve Earthly Branches, the core principles in the oriental tradition, could be factored by the CPN. Analyzing Sil-Hah Woo's arguments, we discuss that the CNP 3 achieved more qualitative valuation than the others in Korean culture.

Neurotoxiciligy. objectives of this study were to test the hypothesis that fumonisin $B_1$ ($FB_1$) alters sphinganine (Sa) levels and myelin synthesis in the central nervous system of developing rats. $FB_1$ (subcutaneous, 0. 4 or 0. 8 mg/kg/day) from postnatal days (PND) 3 to PND 12 resulted in a significantly increased in the brain of rats given 0. 8 mg $FB_1$/kg/day. To confirm the effect of limited nutrition on changes in the Sa levels and myelinogenesis, rats given 0.8mg $FB_1$/kg/day or treated by limited nutrition (temporary removal from dam during postnatal period) were compared to those in saline controls. Sa levels and Sa/So ratios were compared to those in saline in the 0.8 $FB_1$-treated, 3'-phosphohydrolase (CNP) activities were decreased significantly in both nutritionally limited and $FB_1$-exposed rats. These data indicate that sphingolipid metabolism in the central nervous system of develiping rats is vulnerable to $FB_1$ exposure. The hypomyelination associated with $FB_1$-treatment may be mediated by limited nutrition.
