e-Pharmacophore modeling and in silico study of CD147 receptor against SARS-CoV-2 drugs

  • Coronavirus has left severe health impacts on the human population, globally. Still a significant number of cases are reported daily as no specific medications are available for its effective treatment. The presence of the CD147 receptor (human basigin) on the host cell facilitates the severe acute respiratory disease coronavirus 2 (SARS-CoV-2) infection. Therefore, the drugs that efficiently alter the formation of CD147 and spike protein complex could be the right drug candidate to inhibit the replication of SARS-CoV-2. Hence, an e-Pharmacophore model was developed based on the receptor-ligand cavity of CD147 protein which was further mapped against pre-existing drugs of coronavirus disease treatment. A total of seven drugs were found to be suited as pharmacophores out of 11 drugs screened which was further docked with CD147 protein using CDOCKER of Biovia discovery studio. The active site sphere of the prepared protein was 101.44, 87.84, and 97.17 along with the radius being 15.33 and the root-mean-square deviation value obtained was 0.73 Å. The protein minimization energy was calculated to be -30,328.81547 kcal/mol. The docking results showed ritonavir as the best fit as it demonstrated a higher CDOCKER energy (-57.30) with correspond to CDOCKER interaction energy (-53.38). However, authors further suggest in vitro studies to understand the potential activity of the ritonavir.

  • Effect of stripping solutions on the Duolite GT-73 chelating resin for ten elements, Ag(I), Al(III), Ca(II), Cd(II), Cu(II), Fe(II), Hg(II), Mn(II), Pb(II), and Zn(II), was investigated. Relation between affinities of the metal ions and solubility products of metal sulfides was studied. The smaller the solubility product of metal sulfideis, the larger the affinitie with the ionsis. The ions which have the solubility products larger than $10^{-23}$ could be effectively desorbed with nitric acid. Complexation with chloride ion enhanced the desorption efficiencies of the ions having moderately strong affinity with the resin. The ions which have very strong affinity by the chelating resin can be desorbed by complextion with thiourea and hydrochloric acid.

  • $Bi_2Te_3-Bi_2Se_3$ alloy which is typical n-type thermoelectric material were grown by traveling heater method (THM) technique. We investigate the effect of the composition of $100-x(Bi_2Te_3)-x(Bi_2Se_3)$ and doping of n-type dopants such as $SbI_3$ and $CdCl_2$. Maximum figure of merit of $Bi_2Te_3-Bi_2Se_3$ alloy was observed with $CdCl_2$ 0.1 wt% (Z: $2.73{\times}10^{-3}/K$) and $SbI_3$ 0.05 wt% (Z: $2.29{\times}10^{-3}/K$). Deviation along the length of $Bi_2Te_3-Bi_2Se_3$ ingot grown by THM method is low, which indicates that the ingot is very homogenized. Also we observed the close relationship of between anisotropy ratio and dopant in the $90(Bi_2Te_3)-10(Bi_2Se_3)$ alloys. And we confirmed the fact that anisotropy ratio exerts thermoelectric performance in $Bi_2Te_3$ based n-type thermoelectric material.

MCP-1 Derived from Stromal Keratocyte Induces Corneal Infiltration of CD4+ T Cells in Herpetic Stromal Keratitis

  • Herpetic stromal keratitis (HSK) is an inflammatory disorder induced by HSV-1 infection and characterized by T cell-dependent destruction of corneal tissues. It is not known what triggers $CD4^+$ T cell migration into the stroma of HSV-1-infected corneas. The keratocyte is a fibroblast-like cell that can function as an antigen-presenting cell in the mouse cornea by expressing MHC class II and costimulatory molecules after HSV-1 infection. We hypothesized that chemokines produced by stromal keratocytes are involved in $CD4^+$ T cell infiltration into the cornea. We found that keratocytes produce several cytokines and chemokines, including MCP-1, RANTES, and T cell activation (TCA)-3. HSV-1 infection increased the production of MCP-1 and RANTES by keratocytes, and these acted as chemoattractants for HSV-1-primed $CD4^+$ T cells expressing CCR2 and CCR5. Finally, when HSV-1-primed $CD4^+$ T cells were adoptively transferred into wild type and MCP-1-deficient mice that had been sublethally irradiated to minimize chemokine production from immune cells, infiltration of $CD4^+$ T cells was markedly reduced in the MCP-1-deficient mice, suggesting that it is the MCP-1 from HSV-1-infected keratocytes that attracts $CD4^+$ T cells into the cornea.

Distributions of Dissolved Pb and Cd in the Surface Water of East Sea, Korea (동해 표층수중 용존 Pb, Cd의 분포 특성)

  • The distributions of Pb and Cd concentrations in the surface seawater of the East Sea were investigated during the R/V Lavrentyev cruise (July 2009) in which four transects from Russia shore to South were conducted to collect 26 surface water samples. The total dissolved concentrations of Pb and Cd were measured using ICP-MS (Perkin Elmer, DRC-e). In the coastal area, their concentrations of Russia shore (Pb, 0.08; Cd, 0.10 nM) were comparable for Cd but on the other hand, 6 times lower for Pb than Korea shore (Pb, 0.49; Cd, 0.11 nM). In the subregion, their concentrations of Warm region (Pb, 0.22; Cd, 0.01 nM) were about 1.7 times higher for Pb but 0.4 lower for Cd than Cold region (Pb, 0.13; Cd, 0.14 nM). The distributions of Pb and Cd concentrations were divided by lowest level at $10^{\circ}C$ of water temperature. Below $10^{\circ}C$, Pb and Cd concentrations increased when surface water temperatures decreased. Above $10^{\circ}C$, their concentrations increased with temperature, which showed highest concentrations in the Ulleung basin, directly influenced by flux from East Korean Warm Current and neighboring countrys (Korea and Japan). Specially, in the case of Pb, the concentrations decrease remarkablely with temperatures decrease from D10 directly influenced by flux from East Korean Warm Current, which shows highest Pb level. By comparing with other sea areas (Western Mediterranean, East Pacific), Pb concentrations in the East Sea were a little higher. The influence of East Korean Warm Current and neighboring countrys (Korea and Japan) may be relatively important. Therefore, the distribution of Cd may primarily be influenced by mixing of different water masses while the distribution of Pb may mainly be influenced by flux from East Korean Warm Current and atmospheric inputs. River inputs and interaction with particulate materials may also some roles for the distribution of these elements.

The Clinical Significance of Cathepsin D and p53 Expression in Locally Advanced Rectal Cancer (국소진행된 직장암에서 Cathepsin D와 p53 발현의 임상적 의의)

  • Purpose: Cathepsin D(CD) is a lysosomal acid proteinase that is related to malignant progression, invasion, and a poor prognosis in several tumors. The aim of this study was to evaluate the prognostic clinical significance of CD and p53 expression in pretreatment biopsy specimens from patients with locally advanced rectal cancer who were treated with preoperative chemoradiation. Materials and Methods: Eighty-nine patients with locally advanced rectal cancer(cT3/T4 or N+) were included in this study. Preoperative chemoradiation consisted of a dose of 50.4 Gy of pelvic radiation and two concurrent cycles of administration of 5-fluorouracil and leucovorin. Surgery was performed six weeks after chemoradiation. CD and p53 expression in pretreatment formalin-fixed paraffin-embedded tumor biopsy specimens were assessed by immunohistochemical staining using a CD and p53 monoclonal antibodies. The threshold value for a positive stain in tumor tissue and stromal cells was 1+ intensity in 10% of the tumors or stromal cells, respectively. Results: Positive CD expression was found in 57(64%) of the tumors and 32(35%) of the stromal cell specimens. There was no association with CD expression of the tumor or stromal cells and patient characteristics. There was a correlation between tumor CD expression with stromal cell CD expression(p=0.01). Overexpression of p53 was not a significant prognostic factor. The 5-year overall survival(OS) and disease-free survival(DFS) rates were not different between tumor CD-negative and positive patient biopsy samples(69% vs. 65%, 60% vs. 61%, respectively). The 5-year OS rates in the tumor-negative/stromal cell-negative, tumor-negative/stromal cell-positive, tumor-positive/stromal cell-negative and tumor-positive/stromal cell-positive biopsy samples were 75%, 28%, 62%, and 73%, respectively. Stromal cell staining only without positive tumor staining demonstrated the worst overall survival prognosis for patients(p=0.013). Conclusion: Overexpression of p53 in rectal biopy tissue was not associated with prognostic significance. In the pretreatment biopsy specimens, an exclusive increase in CD expression in stromal cells without tumor expression was related to poor overall survival in patients with locally advanced rectal cancer treated with preoperative chemoradiation.

  • Objectives : The purpose of this study is to confirm the suppression effect of Asthma and Immune response improvement of Fasciculus Vascularis Luffae Herbal-acupuncture into Chok-samni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks Two experimental groups were treated with different concentrations(1%, 0.1%) of FVL-HA at Chok-samni(St36) for the later 8 weeks(3times/week). Results : 1. Lung weigh of the mice group treated with FVL-HA decreased significantly compared with that of control group. 2. Total cells of lung, total Leukocytes and Eosinophils in BALF of the mice group treated with FVL-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with FVL-HA in Photomicrographs decreased significantly compared with those of control group. 4. The concentration of IL-l3, IgE, IL-4 In serum and IL-4 in BALF of the mice group treated with FVL-HA decreased significantly compared with that of control group. 5. The number of Gr-1+/CB11b+, CD3-/CCR3+, CD4+, CD8+, CD3e+/CD69+, IgE+/B220+ cells in the lungs of the mice group treated with FVL-HA decreased significantly compared with those of control group. 6. The cytokine's manifestation of mRNA of the mice group treated with FVL-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : These result suggests that Fasciculus Vascularis Luffae Herbal-acupuncture Choksamni(St36) in C57BL/6mice may be an effective part to OVA-induced asthma in C57BL/6 mice.

Concentrations and Daily Intake of Arsenic and Heavy Metals in Polished Rice Around Abandoned Metal Mines in Korea (국내 휴·폐금속광산 주변 백미의 비소 및 중금속 함량과 일일 섭취량)

  • Rice is a staple food source in Asian countries. In paddy field, rice plant can take up toxic elements through its roots from contaminated soils, and its leaves and grain can absorb the toxic elements deposited on the soil surface. A totla of 40 soil and polished rice samples were collected around four abandoned metal mines in Korea and analyzed for As, Cd, Cu, Pb and Zn by atomic absorption spectrophotometer (AAS). The average contents of As, Cd, Cu, Pb and Zn in rice grain grown on the contaminated soils were 0.247, 0.174, 4.694, 0.804 and 16.78 mg/kg, respectively. These levels are higher than worldwide average concentrations. Assuming the rice consumption of 169 g/day by overall households in Korea, the estimated daily intakes from the rices were found to be 33, 48, and 63% for As, Cd, and Pb, respectively, of the acceptable daily intake (ADI) suggested by the FAO/WHO Joint Food Additive and Contaminants Committee.

Discoid lupus erythematosus (DLE) in a juvenile Alaskan Malamute dog

  • Discoid lupus erythematosus (DLE) is an immune-mediated skin disease which requires histopathology and immunohistopathology in both dogs and humans. A 10-month-old, intact female Alaskan Malamute presented for depigmentation, swelling, alopecia, erythema, and crusting on the bridge of the nose and the nasal planum. Cytological examination of nasal lesions revealed numerous cocci and neutrophils. Histopathological features included of infiltration of mononuclear cells at the dermoepidermal junction. Direct immunofluorescence tests and immunohistochemistry exhibited positive IgG, IgM, IgA, CD3, CD18, and CD79a on the epidermal basement membranes and around adnexal glands. This case indicates both T cells and B cells are related to mechanism of canine DLE. This case report describes advanced diagnostic tests and clinical outcome with immune suppressive therapy in a rare juvenile canine DLE case.

Co-existence of relapsing polychondritis and Crohn disease treated successfully with infliximab

  • Relapsing polychondritis (RP) is a rare, progressive immune-mediated systemic inflammatory disease of unknown etiology, characterized by recurrent inflammation of cartilaginous structures. Approximately 30% of RP cases are associated with other autoimmune diseases. However, the co-occurrence of RP and Crohn disease (CD) has rarely been reported. Herein, we present a 35-year-old woman diagnosed with RP and CD, who was refractory to initial conventional medications, including azathioprine and glucocorticoid, but who subsequently responded to infliximab (IFX). For both diseases, remission was sustained with IFX. There has been no previous report regarding the successful treatment of co-existing RP and CD with IFX.
