Background: Monoclonal antibodies (mAbs) recognizing Class III epitope of CD34 are essential for flow cytometric diagnosis of leukemia. Methods: 27H2 mAb was developed from a mouse alternatively immunized with human acute leukemia cell lines, KG1 and Molm-1. Using flow cytometric analysis of various leukemic cell lines and peripheral blood, immunohistochemical study of frozen tonsil, we characterized 27H2 mAb. Antigen immunoprecipitated with 27H2 mAb immunobloted with anti-CD34 mAb. A case of bone marrow sample of acute lymphoblastic leukemia (ALL) patient was obtained at CBNU Hospital. For epitope identification enzyme treatment with neuraminidase and O-sialoglycoprotein endopeptidase (OSGE) and blocking assay with known classIII mAb (HPCA-2) were done. Results: Only KG1 and Molm-1 revealed positive immunoreactivity. Immunohistochemical staining disclosed strong membranous immunoreactivity on high endothelial venules. Antigen immunoprecipitated by 27H2 mAb showed approximately 100 kDa sized band immunoblotted with anti-CD34 under non-reducing conditions. Epitope recognized by 27H2 mAb disclosed resistancy to both neuraminidase and OSGE treatment and completely blocked with known class III mAb preincubation. CD34 positive leukemic cells in BM of pre B cell ALL patient detected by FITC-conjugated 27H2 and HPCA-2 were identified with similar sensitivity. Conclusion: A novel murine mAb recognizing class III epitope of human CD34 with high affinity, which is useful for flow cytometric diagnosis of leukemia, was developed.
This study examined the effects of dietary ${\beta}$-cyclodextrin (${\beta}CD$) on the cholesterol of blood and tissues of swine. Thirty six male castrated swine ($Landrace{\times}Yolkshire{\times}Duroc$) weighing 50 kg were randomly assigned to one of four dietary groups until their weight reached 110 kg. The groups were: basal diet without ${\beta}CD$ (control) and basal diets containing 1.5%, 3.0%, or 5.0% ${\beta}CD$. Diets and water were offered ad libitum. No significant difference was found between treatments in terms of feeding performance measured by daily intake, daily weight gain, and feed efficiency. Addition of ${\beta}CD$ to the diets significantly reduced total lipid, triglyceride and total cholesterol levels in swine blood, particularly in the group receiving 5.0% ${\beta}CD$, which showed decreases (p<0.05) of 21.9%, 55.6% and 27.7%, respectively. Cholesterol levels in back fat, loin, belly and ham portions of swine fed ${\beta}CD$ significantly differed (p<0.05) from controls, especially in the 5.0% ${\beta}CD$-fed group, with reductions of 26.0%, 27.5%, 17.9% and 18.3%, respectively. These results suggested that the addition of ${\beta}CD$ to the diet of swine could reduce their body cholesterol by decreasing the migration of cholesterol through the blood.
This study was undertaken to investigate the effect of dietary Fe levels on Cd accumulation in Cd poisoned rats. Forty male weaning Sprague Dawley rats weighing 80-90g were divided into 4 groups(LFe : low Fe, LFeCd : low Fe and Cd, AFe : adequate Fe, AFeCd : adequate Fe and Cd) according to Cd administration(Cd : 0 or 50ppm in drinking water) and Fe levels (Fe : 6 or 40ppm in diet) for 12 weeks. The food intake and weight gain of LFe and LFeCd were significantly lower than those of AFe and AFeCd(p<0.01, p<0.001). The water intake was not affected significantly by Cd and Fe, therefore Cd intake was no significant difference between groups. The Cd accumulation of kidney in LFeCd was significantly higher than those of AFeCd(p<0.001). But the Cd accumulations of brain, liver and spleen were not significantly different between Cd groups and without Cd groups. The serum Cd content and urinary Cd excretion of LFeCd was significantly higher than those of AFeCd(p<0.01, p<0.01). But the fecal Cd excretion of LFeCd was significantly lower than that of AFeCd(p<0.001). The Cd retention, Cd retention rate, and apparent Cd digestibility of LFeCd were significantly higher than those of AFeCd(p<0.001, p<0.001, p<0.001). It was concluded that adequate Fe supplementation have protective effects on the long term Cd poisoning in rats.

The sensitivity to Cd of three ectomycorrhizal fungi, Paxillus involutus, Suillus bovinus and Rhizopogon subcaerulescens, was assessed and compared in terms of radial growth and dry mass production, using both agar and liquid culture. The radial growth of S. bovinus and R. subcaerulescens was significantly reduced at the lowest concentration (0.1mg Cd/L). The 50% effective concentration (EC$_{50}$) values calculated from radial growth rates of the ectomycorrhizal fungi showed that the sensitivity of the fungi to Cd was greatest in S. bovinus and lowest in R. subcaerulescens. Cadmium addition also significantly decreased dry mass production of the ectomycorrhizal fungi. The sensitivity of the fungi to Cd in terms of dry mass production, was greatest in S. bovinus and lowest in P. involutus. Higher growth rates of P. involutus and melanisation of R. subcaerulescens appeared to contribute to reduced Cd toxicity.
The paper describes on the implementation of marine casualty prediction model that is one of the main part of Korean MArine Casualty FOrecasting System (K-MACFOS). In this work, Cell Distributed Linear-In-the-Parameter (CD-LIP) model is proposed and discussed its usability with comparing Baltic model and revised LIP model. As evaluation results by regression analysis of variance, it is known that the CD-LIP model gives best performance to the marine casualty numerical D/B of the target sea area.
Tomato (Lycopersicon esculentum) seedlings exposed to various concentrations of $CdC1_2$ (0∼100 $\mu$M) in the nutrient solution for up to 9 days were analyzed with the seedling growth, $H_2O_2$ production, glutathione levels and activity changes of enzymes related to $H_2O_2$ removal. The growth of seedlings was inhibited with over 50 $\mu$M Cd, whereas the levels of $H_2O_2$ and glutathione were enhanced with Cd exposure level and time. Meanwhile, Cd exposure increased the activities of catalase (CAT) and glutathione reductase (GR) but decreased the activities of dehydroascorbate acid reductase (DHAR) and ascorbate peroxidase (APX) in both leaves and roots. These results suggest that the altered activities of antioxidant enzymes particularly involved in the $H_2O_2$ removal and the subsequent $H_2O$$_2$ accumulation could induce the Cd-induced phytotoxicity.
$BaMg_2Al_16O_27:Eu,\SrGa_2S_4:Eu$, and ZnCdS:Ag,Cl phosphors were chosen to produce blue, green, and red emissions, respectively, under excitation from a violet light emitting diode (LED). A four-band white LED was obtained by a combination of nonabsorbed violet emission from a violet LED and blue, green, and red emissions from $BaMg_2Al_16O_27:Eu,\SrGa_2S_4:Eu$, and ZnCdS:Ag,Cl phosphors. The luminescent properties of the four-band white LED were also discussed.
Cadmium is considered one of the most toxic, non biodegradable heavy metal for the human and animals. The purpose of the present study was to investigate the changes in biochemical parameters of blood and accumulation of cadmium in various tissue caused by various levels of dietary cadmium chloride ($CdCl_2$) in broiler chicks. $CdCl_2$ was administered through drinking water to broiler chicks. In spectral analysis, $CdCl_2$ treatment caused a significant increase in Glutamate pyruvate transaminase (GPT), creatinine and uric acid levels in all treated groups. Intriguingly, the GPT, creatinine, and uric acid levels were significantly higher at 75 mg/kg as compared to the groups treated with high doses (100, 125 and 150 mg/kg) of $CdCl_2$. Atomic Absorption Spectrophotometer (AAS) was used for the determination of Cd accumulation in kidney, liver and Breast muscles. AAS analysis revealed that Cd accumulation is increased in breast muscles as compared to liver and kidney at higher doses of Cd than 75 mg/kg.
