RGS3 Suppresses cAMP Response Element (CRE) Activity Mediated by CB2 Cannabinoid Receptor in HEK293 Cells (캐너비노이드 수용체 CB2의 신호전달작용에 미치는 RGS3의 억제적 효과)

  • RGS proteins have been identified as negative regulators of G protein signalling pathways and attenuate the activity of GPCR receptors. However, information on the regulatory effects of RGS proteins in the activity of cannabinoid receptors is limited. In this study, the role of RGS proteins on the signal transduction of the CB2 cannabinoid receptor was investigated in HEK293 cells co-transfected with CB2-receptors and plasmids encoding RGS2, RGS3, RGS4 and RGS5. Treatment of cells with WIN55, 212-2, a CB2 receptor agonist, inhibited forskolin-induced cAMP response element (CRE) activity in CB2-transfected HEK293 (CB2-HEK293) cells. This inhibitory effect of WIN 55, 212-2 on CRE activity was reversed by co-transfection of CB2-HEK293 cells with RGS3, but not with RGS2, RGS4 and RGS5. However, endogenous RGS3 protein knocked down by a small interfering siRNA targeting RGS3 gene enhanced inhibition of forskolin induced CRE activity via agonist induced CB2 receptor signal transduction. These results indicate the functional role of endogenous RGS protein in cannabinoid signaling pathways and define receptor-selective roles of endogenous RGS3 in modulating CRE transcriptional responses to agonist induced CB2 receptor activity.

Induction of Laccase from Wood-Rotting Fungi with 2,5-Xylidine (2,5-Xylidine을 이용한 목재부후균으로부터 Laccase 효소의 유도)

  • Some white-rot fungi, screened at the Laboratory of Forest Products Microbiological Chemistry, Chungbuk National University were cultured and added the inducer of laccase enzyme, 2,5-xylidine. The fungi named by CB-13, CB-20, CB-99, CB-100 and CB-123 strains showed positive results in the decolorization of aromatic compounds, carminic acid and Rhemazol brilliant blue R. Concerned to the inducing effect of 2,5-xylidine on laccase activity, CB-20, CB-100 and CB-123 strains showed very high activity by addition of 2,5-xylidine, whilst CB-13, CB-99 and CB-124 strains produced relatively high laccase enzymes, regardless of inducer addition. There were no any laccase activities on CB-25, CB-64 and CB-139, even in addition of inducer. It is confirmed that some screened fungi have decolorizing ability on aromatic compounds, carminic acid and Rhemazol brilliant blue R. Also, the addition of inducer, 2,5-xylidine, has increased the activity of laccase enzyme which is secreted from some white-rot fungi.

D-band Stacked Amplifiers based on SiGe BiCMOS Technology

  • This paper presents two 3-stage D-band stacked amplifiers developed in a $0.13-{\mu}m$ SiGe BiCMOS technology, employed to compare the conventional cascode topology and the common-base (CB)/CB stacked topology. AMP1 employs two cascode stages followed by a CB/CB stacked stage, while AMP2 is composed of three CB/CB stacked stages. AMP1 showed a 17.1 dB peak gain at 143.8 GHz and a saturation output power of -4.2 dBm, while AMP2 showed a 20.4 dB peak gain at 150.6 GHz and a saturation output power of -1.3 dBm. The respective power dissipation was 42.9 mW and 59.4 mW for the two amplifiers. The results show that CB/CB stacked topology is favored over cascode topology in terms of gain near 140 GHz.

  • Although 'Cinnamon' has been widely used for the food and biophamacy in the world, it's toxicity was not screened completely. Major component of 'cinnamon' is CB-OH and CB-PH. CB-PH has been reported to have antimutagenic effect. To investigate the toxicity of 2-o-Benzoylcinnama-Idehyde (CB-PH), repeated dose (4 weeks) oral toxicity test performed in SD rats. Results of repeated dose oral toxicity tests for 4 weeks (CB-PH; 500, 1000, 2000 mg/kg/day) suggested that the CB-PH treated group showed no significant toxicological findings with body weights, organ weights, hematological and histopathological findings. Therefore, these data indicated that the maximum tolerated dose of CB-PH was 2000 mg above/kg/day in the rats.

A Study on the Investment Efficiency of CB(Convertible Bond) (CB(전환사채)의 투자효율성에 관한 실증연구)

  • CB(Convertible bond) is mezzanine security that have the characteristics of bonds and stocks. From the perspective of investors, the purpose of the research is to empirically investigate the degree of investment efficiency of CB and to suggest efficient investment plans. The research method investigated the maturity interest rate, conversion price, and conversion date for CB, and then linked it with daily stock price fluctuations after the conversion date to determine the degree of investment efficiency and stock conversion effect of CB. As a result of the study, it was analyzed that the ratio of the conversion price exceeded days was only about 1/4 of the conversion date, so the investment efficiency was low. The conversion day yield was -6.3% on average and the maturity day yield was -5.2% on average, showing a minus return on average, which was calculated differently from investor expectations. It was analyzed that the number of stocks with a minus conversion day is 2.4 times greater than the number of plus stocks and 3.7 times more than the number of plus stocks with a minus maturity return, so the expected return on stock conversion of CB is low. The research contribution was derived from the problem that the expected rate of return of CB is not high, and it is that the investor's point of view when purchasing CB was established.

  • Waste tire management and recycling have grown to be significant issues because they bring up a global environmental concern. Thus, turning recycled waste tires into useful products may help tackle the environmental issue. This research aims to study and compare the effect of recycled carbon black (rCB) and commercial carbon black (CB) at certain 15 vol. % of filler loading on the mechanical, thermal, morphology and electrical properties of epoxy/CB composites. For this project, epoxy resin, diethyltoluenediamine (DETDA), recovered carbon black (rCB) and commercial carbon black (CB) graded N330, N550, N660 and N774 were mixed and compared accordingly to the formulation determined. The CB content was dispersed in the epoxy matrix using the mechanical mixing technique. The distribution and dispersion of CB in the epoxy matrix affect the characteristics of the conductive composites. rCB content at 15 vol% was selected at fixed content for comparison purposes due to the optimum value in electrical conductivity results. The flexural strength results followed the sequence of rCB>N774>N660>N550>N330. As for electrical conductivity results, epoxy/N330 exhibited the highest conductivity value, while the others achieved a magnitude of X10-3 due to the highest external surface area of N330. In terms of thermal stability, epoxy/N330 and epoxy/N774 were slightly more stable than epoxy/rCB.

Optimization of Post-Activation Systems to Improve the Embryonic Development in Porcine Parthenogenesis and Somatic Cell Nuclear Transfer

  • This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB ($7.5{\mu}g/mL$), CB ($7.5{\mu}g/mL$) + CHX ($10{\mu}g/mL$), CB ($7.5{\mu}g/mL$) +DC ($0.4{\mu}g/mL$), and CB ($7.5{\mu}g/mL$) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

Antiviral Activity of Gallic Acid against Coxsackievirus B3 and Coxsackievirus B4 (Gallic acid의 Coxsackievirus B3와 Coxsackievirus B4에 대한 항바이러스 효과)

  • Viral infections are capable of inducing reactive oxygen species (ROS) production in the infected cells and antioxidants have been reported to have antiviral activities against many viruses. In this study, an antiviral assay using the cytopathic effect (CPE) reduction method revealed that gallic acid possesses good anti-coxsackievirus B3 (CB3) and coxsackievirus B4 (CB4) activities, reducing the formation of visible CPE. However, ribavirin did exhibit weak anti-CB3 and CB4 activities and was unable to prevent CPE. Therefore, we conclude that the inhibition of CB3 or CB4 production by gallic acid may be due to its general action as an antioxidant.

Synthesis and biological evaluation of tricarbonyl technetium labeled 2-(4-chloro)phenyl-imidazo[1,2-a]pyridine analog (99mTc-CB257) as a TSPO-binding ligand

  • In our previous study, tricarbonyl $^{99m}Tc$-labeled TSPO-binding ligand, named $^{99m}Tc$-CB256, having positively charge (+1) was investigated but did not show promising results in in vivo environment despite of a nanomolar binding affinity for TSPO. Because the overall positively charge of $^{99m}Tc$-CB256 would likely interrupt its target protein uptake, we herein designed the neutral tricarbonyl-$^{99m}Tc$ labeled TSPO-binding ligand ($^{99m}Tc$-CB257, 1). $^{99m}Tc$-CB257 was prepared by the facile incorporation of the $[^{99m}Tc(CO)_3]^+$ into a N-(hydroxycarbonylmethyl)-2-picoly moiety in CB257. The radiochemical yield of $^{99m}Tc$-CB257 after HPLC purification was $54.1{\pm}2.4%$ (decay corrected, n = 3). The authentic Re-CB257 (2) was synthesized by using $(NEt_4)_2[Re(CO)_3Br_3]$ in 69.0% yield. The binding affinity of 2 for TSPO was measured in leukocyte and showed approximately 280 times higher than that observed for the positively charged (+1) ligand, Re-CB256 ($K_i=0.57{\pm}0.06nM$ versus $159.3{\pm}8.7nM$, respectively). Our results indicated that 1 can be considered potentially as a new SPECT radiotracer for TSPO-rich cancer and provides the foundation for further in vivo evaluation related with abnormal TSPO-overexpression environments.

Effect of Cytochalasin B Treatment on the Improvement of Survival Rate in Vitrified Pig Oocyte

  • To improve survival rates of vitrified pig oocytes, the treatment of cytoskeletal stabilizer on an appropriate time is one of the possible approaches. However, the exact treatment timing and effect of cytoskeletal stabilizer such as cytochalasin B (CB) is not well known during oocyte vitrification procedures. Thus, the present study was conducted to determine optimal treatment timing of CB during vitrification and warming procedures. In experiment 1, the survival rates of the postwarming pig oocytes were analyzed by fluorescein diacetate (FDA) assays with 4 classifications. In results, post-warming oocytes showed significantly (p<0.05) decreased number of alive oocytes (31.8% vs. 86.4%) compared to fresh control. In detail, the significant difference (p<0.05) was found only in strong fluorescence (18.2% vs. 70.5%) not in intermediate fluorescence groups (13.6% vs. 15.9%). In experiment 2, CB was treated before (CB-Vitri) and after (Vitri-CB) vitrification. In results, group of Vitri-CB showed significantly (p<0.05) higher (91.6%) survival rates compared to group of CB-Vitri (83.7%), significantly (p<0.05) and comparable with group of Vitri Control (88.7%) by morphological inspection. In FDA assay results, group of Vitri-CB showed significantly (p<0.05) higher (44.2%) survival rates compared to groups of CB-Vitri (36.7%) and Vitri Control (35.1%). In conclusion, the increased survival rates of post-warming pig oocyte treated with Vitri-CB method are firstly described here. The main finding of present study is that the CB treatment during recovery could be helpful to refresh the post-warming pig oocyte resulting its improved survival rates.
