Effects of Kwibitang on the Specific Immune Response after Immobilization Stress in C57BL/6 Mice (귀비탕이 C57BL/6 Mouse에 Stress 부하 후 특이적 면역반응에 미치는 영향)

  • Stress is known to influence the immune function via an effect on the central nervous system. To investigated the effects of Kwibitang water extract (KBT) on the specific immune response in C57BL/6 mice stressed by immobilization, we evaluated the changes in the cell viability, DNA fragmentation and subpopulation of thymocytes and splenocytes. KBT enhanced the cell viability of thymocytes and splenocytes decreased by immobilization stress. Also, KBT decreased DNA fragmentation of thymocytes and splenocytes increased by immobilization stress. KBT decreased the population of CD4/sup +/ cells and CD8/sup +/ cells in thymocytes and Thy1/sup +/ cells in splenocytes increased by immobolization stress, but increased the population of B220/sup +/ cells decreased by immobilization stress. In addition, KBT enhanced the production of ν-interferon and IL-2 decreased by immobilization stress. These results indicate that KBT may be useful for the prevention and treatment of stress via enhancement of the specific immune response

Troglitazone Lowers Serum Triglycerides with Sexual Dimorphism in C57BL/6J Mice

  • Thiazolidinediones (TZDs) are widely used antidiabetic drugs that activate the nuclear peroxisome proliferator-activated receptor ${\gamma}(PPAR{\gamma})$, and thereby improve the metabolic abnormalities linking hypertriglyceridemia to diabetes, hyperglycemia, insulin resistance, and cardiovascular disease. To determine whether the $PPAR{\gamma}$ ligand troglitazone regulates lipid metabolism with sexual dimorphism, we examined the effects of troglitazone on circulating lipids, body weight and the expression of hepatic genes responsible for lipid metabolism in both sexes of C57BL/6J mice. Compared to mice fed a low fat control diet, both sexes of mice fed a troglitazone-treated low fat diet for 14 weeks did not exhibit changes in body weight gain, serum total cholesterol, HDL-cholesterol and LDL-cholesterol levels. However, serum triglycerides were significantly reduced in both sexes of mice, although these effects were more pronounced among males. Furthermore, troglitazone regulated the expression of hepatic genes critical for lipid and lipoprotein metabolism, the magnitudes of which were much higher in males compared to females, as evidenced by results for increased acyl-CoA oxidase and decreased apolipoprotein C-III mRMA levels. These results suggest that $PPAR{\gamma}$ activator troglitazone may exert sexually dimorphic control of serum triglycerides in part through the differential activation of $PPAR{\gamma}$ in liver between male and female mice.

Effects of Inonotus Obliqua Extract on Blood Glucose Levels in Genetically Diabetic Mice (차가버섯 추출물이 당뇨생쥐 $(C57BL/KsJ^-m^{+/+}Lepr^{db})$의 혈당수준에 미치는 영향)

  • This study investigated the therapeutic effects of Inonotus obliqua extract on blood glucose, insulin, and other biochemical parameters in genetically diabetic mice $(C57BL/KsJ^-m^{+/+}Lepr^{db})$. The mice were divided into four groups-control, Chaga 1 (dose of 0.09 mg/kg of body weight), Chaga 5 (5 times of Chaga 1), and Chaga 10 (10 times of Chaga 1) - according to supplemented dose. Inonotus obliqua extract was orally administered to the animals for 6 weeks. The body and organ (liver and kidney) weights were not different among groups. Fasting blood glucose level was significantly lower in the Chaga 5 group compared with the control (p < 0.05). Hemoglobin A1c content was significantly lower in the Chaga 5 group compared with either the control and Chaga 1 group (p < 0.05). There was no significant difference in serum insulin level among groups. The glucose-6-phosphatase activity in liver was significantly the lowest in Chaga 10 group and was significantly lower in Chaga 5 group as compared with those of control and Chaga 1 groups. Therefore, the results of this study demonstrate that Inonotus obliqua extract alleviates many of the symptoms of diabetes in genetically obese mice and may offer a possibility as a therapeutic supplement for the normalization of blood glucose levels in human with hyperglycemia and have beneficial effects in patients with non-insulin-dependent diabetes mellitus.

Modifying Effect of Diallyl Sulfide on Colon Carcinogenesis in C57BL/6J-ApcMin/+ Mice

  • Diallyl sulfide (DAS), a flavoring compound derived from garlic, is considered to have cancer chemopreventive potential in experimental animals and humans. This study was designated to examine possible chemopreventive effects of DAS on colon carcinogenesis using genetically engineered transgenic $Apc^{Min/+}$ mice, a well-established animal model for familial adenomatous polyposis (FAP) and sporadic colorectal cancer. Male C57BL/6J-$Apc^{Min/+}$ mice were divided into three groups. Animals of group 1 were placed on the basal diet (AIN-76A) as non-treated controls. Animals of groups 2 and 3 were given DAS-containing diets (in doses of 100 and 300 ppm, respectively). All mice were sacrificed at the end of week 10 of the experiment. Histopathological investigation revealed that the incidence of colonic polyps was decreased dose-dependently by 19% (13/16) in group 2 and by 32% (13/20) in group 3 compared to the 100% incidence (10/10) in group 1. The multiplicity of colonic polyps per mouse was also slightly decreased by DAS treatment ($1.88{\pm}0.35$ in group 2 and $1.63{\pm}0.36$ in group 3) compared to $2.00{\pm}0.39$ in group 1. On the other hand, there were no significant differences in the numbers of total polyps per mouse in the small intestine between the groups. Taken together, we suggest that DAS may exert promising inhibitory effects on colon carcinogenesis in the transgenic $Apc^{Min/+}$ mice.

The Inhibitory Effects of Liriopis Tuber on the Bleomycin-induced lung fibrosis in C57BL/6J mice (맥문동(麥門冬)이 C57BL/6J 생쥐의 Bleomycin 폐섬유화(肺纖維化)에 미치는 영향(影響))

  • Objectives : The aim was to identify the inhibitory effects of Liriopis Tuber on bleomycin-induced lung fibrosis by analysing the changes of imflammatory cell cytokines and SHI(Semiquantitative Histological Index), Materials and Methods : In this study fibrosis prone C57BL/6J mice were used. Control group was treated with blomycin(0.06mg/0.1 ml) by IT(intratracheal) instillation which is a popular method of inducing lung fibrosis and sample group took Liriopis Tuber water extract(38.0mg/10g body weight) orally for 14 days after IT instillation of blomycin. We measured the total and differential count of WBC, $IFN-{\gamma}$ & IL-4 in mice BALF and SHI(Semiquantitative Histological Index) from lung tissues of mice. BALF and lung tissues of mice were taken 14 days after IT instillation of blomycin. Results : In sample group total WBC count, proportion of neutrophil, SHI and IL-4 significantly(p<0.05) decreased, proportion of macrophage significantly(p<0.05) increased and proportion of lymphocyte, $IFN-{\gamma}$ did not decrease significantly. Conclusions : This study suggests that Liriopis Tuber has an inhibitory effects of pulmonary fibrosis by attenuation of inflammation and Th2 immune response. To determine whether this herbal medicine contribute to cure and prophylaxis of pulmonary fibrosis, further studies on the role of $IFN-{\gamma}$ relating to fibrosis are required.

Evaluation of host and bacterial gene modulation during Lawsonia intracellularis infection in immunocompetent C57BL/6 mouse model

  • Background: Proliferative enteritis caused by Lawsonia intracellularis undermines the economic stability of the swine industry worldwide. The development of cost-effective animal models to study the pathophysiology of the disease will help develop strategies to counter this bacterium. Objectives: This study focused on establishing a model of gastrointestinal (GI) infection of L. intracellularis in C57BL/6 mice to evaluate the disease progression and lesions of proliferative enteropathy (PE) in murine GI tissue. Methods: We assessed the murine mucosal and cell-mediated immune responses generated in response to inoculation with L. intracellularis. Results: The mice developed characteristic lesions of the disease and shed L. intracellularis in the feces following oral inoculation with 5 × 107 bacteria. An increase in L. intracellularis 16s rRNA and groEL copies in the intestine of infected mice indicated intestinal dissemination of the bacteria. The C57BL/6 mice appeared capable of modulating humoral and cell-mediated immune responses to L. intracellularis infection. Notably, the expression of genes for the vitamin B12 receptor and for secreted and membrane-bound mucins were downregulated in L. intracellularis -infected mice. Furthermore, L. intracellularis colonization of the mouse intestine was confirmed by the immunohistochemistry and western blot analyses. Conclusions: This is the first study demonstrating the contributions of bacterial chaperonin and host nutrient genes to PE using an immunocompetent mouse model. This mouse infection model may serve as a platform from which to study L. intracellularis infection and develop potential vaccination and therapeutic strategies to treat PE.

Anti-obesity effects of two herbal extracts in C57BL/6N mice fed high-fat diet

  • The objective of this study was to investigate the anti-obesity effects of adding Momordica charantia (MC) and Chrysanthemum zawadskii var. latilobum (CZ) extracts to drinking water on obesity-induced mice. A total of 84 eight-week-old C57BL/6N male mice with an initial body weight (BW) of 28.11 ± 1.39 g were used in this study. All treatments were fed a high-fat diet for d 28. Mice were randomly divided into seven drinking treatments (six replicate each treatment) based on the initial BW. Treatments are as follows: control (CON), normal tap water, MC 1, CON with 1% MC aqueous extract, MC 2, CON with 2% MC aqueous extract, CZ 1, CON with 1% CZ aqueous extract, CZ 2, CON with CZ aqueous extract (2%), MCZ 1, CON with 1% MC aqueous extract and 1% CZ aqueous extract, MCZ 2, CON with 2% MC aqueous extract and 2% CZ aqueous extract. During the entire period, the CZ 1, MCZ 1, and MCZ 2 significantly decreased (p < 0.05) gain to feed than CON. The CON significantly higher (p < 0.05) water intake than other treatments on d 0 to 14. The MCZ 1 significantly decreased (p < 0.05) relative (ratio of absolute organ weight to BW) organ weights, including retroperitoneal white adipose tissue (RWAT) weight and inguinal white adipose tissue (IWAT) weight, compared to CON. In conclusion, our study suggests that there was no significant difference in the anti-obesity effects between MC and CZ, and MCZ 1 has synergistic effects by regulating adipose tissue.

Effects of fermented blueberry liquid in high-fat diet-induced obese C57BL/6J mice (블루베리발효액이 식이유도 비만 Mice의 비만에 미치는 효과)

  • Purpose: The objective of the present study was to determine whether fermentation can increase the protective effects of blueberry liquid in a high-fat diet-induced obese mice model. Methods: Male C57BL/6J mice were fed a high-fat diet (HD, 60% fat, w/w,), HD supplemented with 10 ml/kg BW/day of blueberry liquid (BHD, blueberry high-fat diet), or HD supplemented with 10 ml/kg BW/day of fermented blueberry liquid (FBHD, fermented blueberry high-fat diet) for 10 weeks. Results: There were significant decreases in the body, epididymal adipose tissue, and liver weights of blueberry-fed groups compared to HD, whereas there were no significant differences in food intake among the groups. Furthermore, blueberry liquid groups, especially fermented blueberry liquid, significantly attenuated the contents of hepatic triglycerides and total cholesterol induced by HD. Serum LDL-cholesterol was significantly lower in the BHD and FBHD-fed groups, whereas FBHD significantly increased the serum HDL-cholesterol level compared to the control. Concentrations of aspartate transaminase, alanine transaminase, and leptins in serum were also reduced by blueberry liquid supplementation. The mRNA expression of hepatic acetyl CoA carboxylase was significantly reduced in both the BHD and FBHD groups compared to HD. Furthermore, FBHD altered the mRNA expression level of hepatic lipolysis genes. Conclusion: In conclusion, these results suggest that blueberry, especially fermented blueberry liquid, may improve obesity-related abnormalities.

The Effect of EH-HA(Ephedrae Herba Herbal Acupuncture) at ST36 on OVA-induced Asthma in Mice (마황(麻黃) 약침(藥鍼)의 천식억제(喘息抑制) 및 면역조절효과(免疫調節效果)에 대(對)한 실험적(實驗的) 연구(硏究))

  • Background : Recently, so many people are suffered from the allergic or auto-immune disease, and the representative disease is just Allergic Asthma. It is because human immune function has been decreased. Many treatments were done to treat this disease, and many methods were studied to increase immune function and to suppress the asthma. But, the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture) has not been studied in detail. To study the effects of EH-HA, we injected EH-HA at Joksamni(ST36) of C57BL/6 mice. Objectives : The purpose of this study is to investigate the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture). EH-HA was done at Joksamni(ST36) of the mice with ovalbumin-induced asthma. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks. Two experimental groups were treated with different concentrations(1%, 0.1%) of EH-HA at Joksamni(ST36) for the later 8 weeks(3times/week). Results : 1. The lung weight of the group treated with EH-HA decreased significantly compared with that of control group. 2. The total cells in lung, total leukocytes and eosinophils in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the group treated with EH-HA in photomicrographs decreased significantly compared with those of control group. 4. The concentrations of IL-13, IgE, IL-4 in serum and IL-4 in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 5. The numbers of $Gr-1^+/CD11b^+\;cells,\;CD3e^-/CCR3^+\;cells,\;CD4^+\;cells,\;CD8^+\;cells,\;CD3e^+/CD69^+\;cells\;and\;IgE^+/B220^+\;cells$ in lung of the group treated with EH-HA decreased significantly compared with those of control group. 6. In RT-PCR, the mRNA expression of IL-4, IL-5 and IL-13 in the group treated with EH-HA decreased compared with those of control group. Conclusion : These results suggested that EH-HA at Joksamni(ST36) in C57BL/6mice may be effective to OVA-induced asthma of C57BL/6 mice.

The Inhibitory Effects of Sansayukbokhap-bang(SSYBHB) on the Obese-Mouse Induced High Fat Diet (산사육복합방(山査肉複合方)이 생쥐의 비만억제에 미치는 영향)

  • Objectives In order to investigate the effects of Sansayukbokhap-bang (SSYBHB) on the hematological and histological changes. Methods C57BL/6 mice were fed with high fat diet. C57BL/6 mice were divided into four groups and fed for 15weeks. Results 1. The body weight of SSYBHB intake mice was significantly lower than high fat diet group. 2. The final increase of body weight was decreased significantly. 3. The levels of ALT, AST, total cholesterol, LDL-Cholesterol, triglyceride, Leptin were decreased significantly. 4. The levels of creatinine were decreased but did not show significance. 5. The level of HDL-cholesterol and the expression of ${\beta}$3AR mRNA gene in 3T3-L1 Adipocytes were increased significantly. 6. Adipocytes' size was decreased significantly. 7. The expression of ${\beta}$3AR mRNA gene, Leptin mRNA gene and serotinin mRNA gene in Adipocytes tissue was decreased significantly. Conclusion Based on these results, it is proved that SSYBHB is effective on the therapy of obesity by referring to obese-gene and obese inhibitory. So, it is espected that the clinical application of SSYBHB can help the treatment of obesity.
