Studies on a PR4 Gene for Breeding Disease Resistant Forage Crops (내병성 목초 품종개량을 위한 PR4 유전자의 연구)

  • Cytokinins are essential plant hormones that play crucial roles in various aspects of plant growth and development. By using mRNA differential display, we isolated a cytokinine-inducible cDNA encoding pathogenesis-related (PR) 4 from Arabidopsis amp1 mutant. The full-length PR4 cDNA, designated AtPR4, contains an open reading frame of 212 amino acids with calculated molecular mass of 22,900 Da and isoelectric point (pI) of 7.89. Genomic DNA blotting showed that the Arabidopsis genome has one copy of AtPR4. AtPR4 mRNA was induced by cytokinin and NaCl, but decreased by SA or JA treatment. PR proteins are induced in response to pathogen attack. Thus the AtPR4 gene isolated in this study may be a useful candidate for genetic engineering of forage crops for increased tolerance against pathogen.

Regeneration of Adventitious Shoots from Populus deltoides Bartr. (미류나무 엽육조직에서 식물체 재분화)

  • Adventitious organs (shoots and roots) from mature leaves of Populus deltoides (Eastern cottonwood) clones were induced on WPM (Woody Plant Medium) containing fructose as a carborn source and different combinations of either BA (Benzyl aminopurin) or TDZ (Thidiazuron) with NAA (Naphthaleneacetic acid). The morphologies of adventitious shoots were different depending on the kinds of growth regulators and the length of shoots induced on the medium containing BA were generally longer than that of TDZ. BA 0.5 mg/L and TDZ 0.01 mg/L combined with either 0.05 or 0.1 mg/L NAA showed the highest values in the frequency of regeneration (63% to 100%) the number of shoots per explant and in the organogenic sites per explant among tested three clones (A, B and C). Although there was a minor clonal difference in the development of adventitious shoots from leaf segments, their responses to growth regulators were very similar.

Relationships of Amylogram Characteristics and Table Quality in Waxy Corn Kernel (찰옥수수 립의 아밀로그램 분석과 식미 관련 특성과의 관계)

  • This study was carried out to compare botanical and amylogram characteristics including table quality in waxy corn kernel. The used materials in this experiment were producted and evulated at Corn Breeding Laboratory, Coll. of Agri. & Life Sci., in CNU. In botanical characteristics CNU H09-26 among used hybrids was highest in stem height as 228.5 cm, but that of CNU H09-30 was lowest. Ear height was highest in CNU H09-23 as 78.2 cm, but that of CNU H09-30 was lowest. Ear length among hybrids were also variable as 21.2 cm to 10.8 cm. in amylogram analysis CNU H09-23 hybrid was lowest in pasting temperature, while break down of this hybrid was highest These results appeared highly in table quality. Accordingly we thought that this hybrid will be adapted as a leading variety for edible waxy corn.

Genotype $\times$ Environment Interaction for Yield in Sesame (Sesamum indicum L.)

  • Application of genotype by environment ($G\;{\times}\;E$) interaction would be used for identifying optimum test condition of the varietal adaptation in the establishment of breeding purpose. Yield and yield components were used to perform additive main effect and multiplicative interaction (AMMI) analysis. Significant difference for $G\;{\times}\;E$ interaction were observed for all variable examined. For yield, 0.18 of total sum of squares corresponded to $G\;{\times}\;E$ interaction. Correlation analysis was carried out between genotypic scores of the first interaction principal component axis (IPCA 1) for agronomic characters. Significant correlations were observed between IPCA 1 for yield and capsule bearing stem length (CBSL), number of capsule per plant (NOC). The biplot of grain yield means for IPCA1 which accounted for 34% of the variation in total treatment sums of squares showed different reaction according to $G\;{\times}\;E$ interaction, genotypes and environments. Taegu showed relatively lower positive IPCA1 scores, and it also showed smaller coefficient variation of yield mean where it is recommendable as a optimal site for the sesame cultivar adaptation and evaluation trial. In case of variables, Yangbaek and M1 showed relatively lower IPCA1 scores, but the score direction showed opposite each other on the graph. Ansan, Miryang1, Miryang4, and Miryang6 seemed to be similar group in view of yield response against IPCA1 scores. These results will be helpful to select experimental site for sesame in Korea to minimize $G\;{\times}\;E$ interaction for the selection of promising genotype with higher stability.

  • This study was carried out to propose the new procedure to apply Habitat Evaluation Procedure(HEP) of target species using delphi technique, which is suitable to develop endangered species with few researches and ecological knowledges. To identify habitat quality of specific species in development project site, we can develop habitat model and create habitat suitability maps. In this study, we select the Common Kestrel(Falco tinnunculus) as target species in four areas(Paju, Siheung, Ansan, Hwaseong) which is located near the Seoul metropolitan area. The Delphi technique was selected to get the reliable information on the species and habitats requirements. Through the delphi approach, seven habitat components were determined as suitable variables for the Common Kestrel: density($n/km^2$) of small mammals, area($km^2$) of bare-grounds, pasturelands and riparian, and open area(%), spatial distribution and area of croplands, landscape diversity, breeding sites(tall trees, cliffs, high-rise buildings), and the length of shelf. Habitat variables used in this model were classified into two categories: % of suitable land-cover type(open areas, croplands, pasturelands, wetlands, and baregrounds) and the quality of feeding sites(within 250m from edges of woodlands). Habitat quality of the Common Kestrel was assessed against occurred sites derived from the nationwide survey. Predicted habitat suitability map were closely related to the observed sites of the endangered avian species in the study areas. With the habitat suitability map of the Common Kestrel, we assess the environmental impacts with habitat loss after development project in environmental impact assessment.

Effects of Pipe Network Composition and Length on Power Plant Waste Heat Utilization System Performance for Large-scale Horticulture Facilities (발전소 온배수를 적용한 대규모 시설원예단지용 난방시스템의 열원이송 배관 재질 및 거리에 따른 성능평가)

  • Korean government plans to establish large-scale horticulture facility complexes using reclaimed land in order to improve the national competitiveness of agriculture at the government level. One of the most significant problems arising from the establishment of those large-scale horticulture facilities is that these facilities still largely depend on a fossil fuel and they require 24 h a day heating during the winter season in order to provide the necessary breeding conditions for greenhouse crops. These facilities show large energy consumption due to the use of coverings with large heat transmission coefficients such as vinyl and glass during heating in the winter season. This study investigated the applicability of waste heat from power plant for large-scale horticulture facilities by evaluating the waste heat water temperature, heat loss and energy saving performance as a function of distance between power plant and greenhouse. As a result, utilizing power plant waste heat can reduce the energy consumption by around 85% compared to the conventional gas boiler, regardless of the distance between power plant and greenhouse.

Rooting Ratio of Hemp Varieties (대마 품종의 삽목 증식율)

  • In order to obtain basic information for pure line breeding of hemp which is effective on the pure line preservation and propagation, three varieties were investigative from 1995 to 1996 at Mokpo Experiment Station, Honam Agriculture Experiment Station, and cutting parts, cutting times, cutting length and storage duration after cutting of plant were observed. Higher rooting ratio were observed in cutting of shoot and later branch, in July 5, optimum harvesting time and 5 days after harvesting time, in shorter storage duration,0 and 2 days. Also there were the highest rooting ratio increased by from 12% to 16% in shorter cutting of 10cm than 15cm long cutting in shoot, lateral branch and stem cuttings. Hemp cv. Korea showed higher rooting ratio than the other varieties, Japan and Turkey. Therefore, it was concluded that hemp, a dioecious plant, could be effectively propagated for the pure lines by cutting method, especially in cv. Korea.

Taxonomic Status of Acheilognathus sp. (Cyprinidae) found in the Geum River, Korea (금강에서 발견된 Acheilognathus sp. (Cyprinidae)의 분류학적 위치)

  • To examine the taxonomic status of Acheilognathus sp. specimens from the Geum River, morphological and genetical characteristics of A. sp., A. yamatsutae and A. majusculus were investigated and compared in detail. Specimens of A. sp. could be distinguished from the other two species by the combination of some morphological characters such as nuptial color, vertebrae, gillrakers and etc. Males of A. sp. had red bands on the outer margin of dorsal and anal fins and a white band on the outer margin of ventral fin in breeding season. A. sp. had larger maximum body length and somewhat more vertebrae than A. yamatsutae, and had fewer gillrakers than A. majusculus. A. sp. appeared as a monophyletic group with A. majusculus and A. cyanostigma based on genetic analysis. In addition, it had even more close relationship with other congeners than A. yamatsutae. Therefore it is presumed that A. sp. from the Geum River may be a distinct species in genus Acheilognathus.

Suppression of tobamovirus movement toward upper leaves in the tomato plant over-expressing a maize calreticulin (옥수수 calreticulin 과발현 토마토에서 tobamovirus의 상엽 이동 억제)

  • To ascertain the effect of over-expressed maize calreticulin in tomato plant on tobamovirus movement in addition to validating potentiality of the gene (ZmCRT) as a means for the virus-resistance resource, four ZmCRT-expressing homozygous lines were generated from the T0 plants as using an Agrobacterium-mediated transformation, nucleic acid analyses, and a conventional breeding method. Of them, a line was subjected to the bioassay for tolerances to tobacco mosaic virus-U1 (TMV-U1) and tomato mosaic virus (ToMV) followed by RT-PCR and a chlorophyll fluorescence quenching analyses. Both transgenic plants transcribing ZmCRT and wild-type plants showed no symptom by 20 days after viruses inoculation, however the photosystem II quantum yield parameter measured from the upper leaves of ToMV-inoculated plants revealed that ZmCRT transgenic plants have higher photosynthetic ability than wild-type ones at that time, which indirectly implies that over-expressed ZmCRT product acts as a barrier to the cell-to-cell and/or systemic movement of ToMV. Moreover, ZmCRT transgenic plants showed remarkably longer shoot length than wild-type ones in 40 days after TMV-U1 or ToMV inoculation each, which might be resulted from higher photosynthetic ability during the phase not yet showing any external symptoms. Collectively, over-expressed ZmCRT protein in tomato plants is able to interrupt the systemic movement of infected TMV-U1 and ToMV even though not perfect.

Molecular Characterization of the Soybean L-Asparaginase Gene Induced by Low Temperature Stress

  • L-asparaginase (EC 3.5.1.1) catalyzes the hydrolysis of the amide group of L-asparagine, releasing aspartate and $NH_4{^+}$. We isolated a low temperature-inducible cDNA sequence encoding L-asparaginase from soybean leaves. The full-length L-asparaginase cDNA, designated GmASP1, contains an open reading frame of 1,258 bp coding for a protein of 326 amino acids. Genomic DNA blotting and fluorescence in situ hybridization showed that the soybean genome has two copies of GmASP1. GmASP1 mRNA was induced by low temperature, ABA and NaCl, but not by heat shock or drought stress. E. coli cells expressing recombinant GmASP1 had 3-fold increased L-asparaginase activity. A possible function of L-asparaginase in the early response to low temperature stress is discussed.
