Pectobacterium, which causes soft rot disease, is divided into 18 species based on the current classification. A total of 225 Pectobacterium strains were isolated from 10 main cultivation regions of potato (Solanum tuberosum), napa cabbage (Brassica rapa subsp. pekinensis), and radish (Raphanus sativus) in South Korea; 202 isolates (90%) were from potato, 18 from napa cabbage, and five from radish. Strains were identified using the Biolog test and phylogenetic analysis. The pathogenicity and swimming motility were tested at four different temperatures. Pectolytic activity and plant cell-wall degrading enzyme (PCWDE) activity were evaluated for six species (P. carotovorum subsp. carotovorum, Pcc; P. odoriferum, Pod; P. brasiliense, Pbr; P. versatile, Pve; P. polaris, Ppo; P. parmentieri, Ppa). Pod, Pcc, Pbr, and Pve were the most prevalent species. Although P. atrosepticum is a widespread pathogen in other countries, it was not found here. This is the first report of Ppo, Ppa, and Pve in South Korea. Pectobacterium species showed stronger activity at 28℃ and 32℃ than at 24℃, and showed weak activity at 37℃. Pectolytic activity decreased with increasing temperature. Activity of pectate lyase was not significantly affected by temperature. Activity of protease, cellulase, and polygalacturonase decreased with increasing temperature. The inability of isolated Pectobacterium to soften host tissues at 37℃ may be a consequence of decreased motility and PCWDE activity. These data suggest that future increases in temperature as a result of climate change may affect the population dynamics of Pectobacterium.

The consumption of Gwamegi, a semi-dried saury Cololabis saira, and its vegetable sides has increased owing to its availability online. Therefore, this study investigated the microbial contamination levels in Gwamegi and its accompanying vegetable sides bought online by measuring total viable bacteria, coliforms, Escherichia coli, Staphylococcus aureus and fungi. The total viable bacteria ranged from 3-5 log CFU/g. The fungi in Gwamegi and garlic were 3.4 and 3.9 log CFU/g, respectively. The positive rate of bacterial contamination was 100% (2-3 log CFU/g) in Gwamegi, cabbage Brassica rapa subsp. pekinensis, and green chili Capsicum annuum, whereas the contamination positive rate was 80% and 60% (< 2 log CFU/g) in chives Allium ascalonicum L. and garlic A. sativum L., respectively. The positive rates of E. coli were 0%, 20%, 60%, and 40% in Gwamegi, green chili, cabbage, and chives, respectively. The contamination levels of E. coli were 1-2 log CFU/g. S. aureus was detected at < 1 log CFU/g in all raw materials. The data on microbial contamination levels may be used for microbial risk assessment of Gwamegi and vegetables for controlling the level of microbial contamination and securing microbiological safety.

Baechu (Brassica campestris var. pekinensis) was salted in 29.6% brine at $30^{\circ}C$ for 4 hours on mobile automatic salting equipment (MASE) with brine circulation four times, and the quality of MASE salted kimchi was compared with that of general factory salted kimchi (control, 12% brine, $10^{\circ}C$, 16 hr) for 5 weeks. Salinity, acidity and number of lactic acid bacteria of MASE kimchi were higher than those of control at 2.5%, 1.17%, and 8.38 log CFU/g, compared to 1.5%, 1.00%, and 2.68 log CFU/g, respectively, whereas reducing sugar content and texture were not significantly different. Overall quality of sensory evaluation was higher than 4.0 in MASE kimchi compared to lower than 4.0 in the control, and taste was significantly higher (P<0.01). The quality of kimchi salted at high salt concentration and high temperature for a short time was higher than those salted at low salt concentration and low temperature for a long time. Higher productivity in the Baechu salting process can be achieved by shortening salting time with MASE.

A tobamovirus, Ribgrass mosaic virus (RMV), was identified newly from chinese cabbage (Brassica campestris L. pekinensis) in Korea. Virus disease incidence of RMV on chinese cabbage was 37.9% in alpine area on August in 1993. RMV induced the symptoms of necrotic ring spots, necrotic streak on midrib and malformation. RMV, Ca1 and Ca3 isolate, could infect 35 species out of 45 plants including Chenopodium amaranticolor. Physical properties of RMV Ca1 isolate were very stable as 10.8 over for dilution end point, $95^{\circ}C$ for temperature inactivation point and 18 weeks for longevity in vitro. RMV had the soil transmission rate of 75.0% for the chinese cabbages, 'Chunhawang' and 'Seoul' cultivars. The purified virions of RMV had the typical ultraviolet absorption spectrum of maximum at 260 nm and minimum at 247 nm. RMV of Ca1 isolate was related serologically with antisera of Tobacco mosaic virus (TMV)-Cym, TMV-O and Pepper mottle virus, but not related with antiserum of Odontoglossum ring spot virus. coat protein gene of RMV-Ca1, sized 473 nucleotides, encoded 158 amino acid residues. Nucleotide identity of RMV-Ca1 CP gene was 96.4% with RMV-Shanghai (GenBank accession No. of AF185272) from China and 96.0% with RMV-Impatiens (GenBank accession No. of AM040974) from Germany. Identity of amino acids between RMV-Ca1 and the two RMV isolates was 96.8%. Specific three primers were selected for rapid and easy genetic detection of RMV using Virion Captured (VC)/RT-PCR method.
Turnip mosaic virus (TuMV) is an infectious viral pathogen on the cruciferous crops, predominantly Chinese cabbage (Brassica campestris subsp. pekinensis) and radish (Raphanus sativus). On the basis of the symptom development in selective differential hosts from indicator host species, Chinese cabbage and Korean radish inbred lines, the representative eight isolates of TuMV were divided into two major groups/or six types. Group I includes Th 1, Ca-ad7, and Cj-ca2-1 isolates, while group II includes the other isolates (rg-pfl, r 9-10, Rhcql-2, Stock and Mustard). According to the molecular phylogenetic analysis, these isolates, however, divided into two groups and two independent isolates. Phylogenetic analysis indicated that four isolates (Tu 1, r9-10, Stock and Rh-cql-2) formed a distinct phylogenetic group, and the other two isolates (Ca-ad7 and Cj-ca2-1) also formed another group. Mustard and rg-pfl isolates did not seem to have any relationship with these two groups. Taken together, these results indicated that virulence differentiation on host plants, molecular phylogenetic analysis of the nucleotide and the deduced amino acid of TuMV coat proteins did not show any relationship. The multi-resistant lines, Wonyae 20026 and BP058 in Chinese cabbage represent valuable genetic materials that can be used for crucifer breeding programs on TuMV resistance, but not in Korean radish.
xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed. Chinese cabbage grown during autumn season is confronted with drought conditions for a certain period, especially during the early growth stage. In this study, we investigated the effects of drought stress on plant growth characteristics, as well as free amino acid, carotenoid, and proline in Chinese cabbage. Chinese cabbage seeds (Bulam Plus) were germinated, and all the seedlings were transplanted into plastic containers (28 cm diameter ${\times}$ 22 cm high) containing a commercial growth medium. The soil water content was measured and maintained at 10% for the drought-stressed plants and at 30% for the control plants, for three weeks. The results revealed that plant growth parameters were lower in the drought-stressed plants than in the control plants. The total free amino acid content tended to decrease in both drought-stressed and control plants with time. The total free amino acid content was found to be lower in the drought-stressed plants than in the control plants and the proline content was unaffected. Moreover, at three weeks after treatment, carotenoid content in drought stressed plants was significantly higher than that in the untreated plants. We believe that our study makes a significant contribution to the literature because the effects of drought stress on plant growth parameters, free amino acid, carotenoids, and proline accumulation in autumn growing cultivar of Chinese cabbage have not been widely studied in Korea, and our study provides valuable information in this regard, as Chinese cabbage is consumed throughout the year in Korea.
This study investigated the applicability of horticultural media with recycled coir substrates the growth of Chinese cabbage (Brassica campestris L. ssp. Pekinensis) and lettuce (Lactuca sativa L.) crop. The six different types of coir based substrates were A, Coir 45: Perlite 35: Vermiculite 12: Zeolite 8 (%), B, Coir 55: Perlite 25: Vermiculite 12: Zeolite 8 (%), C, Coir 65: Perlite 15: Vermiculite 12: Zeolite 8 (%), D, Coir 75: Perlite 5: Vermiculite 12: Zeolite 8 (%), E, Coir 85: Perlite 5: Vermiculite 5: Zeolite 5 (%) and F, nursery media (control). The pH and Electric conductivity of the horticultural nursery media were 6.06-7.00 and $0.45-1.10dS/m^{-1}$, respectively. The nursery media containing coir substrates had higher level of Total N, Ca, K, Mg and P than those without coir. Additionally, it was observed that the growth of Chinese cabbage was the best on D (containing 75% coir) while that of lettuce was the best on E (containing 85% coir). In general, when substrates containg a higher percentage of coir were used, the growth of Chinese cabbage and lettuce was ideal. Additionally, the P, Ca, and Mg content in both plants was not significantly altered by the amount of coir present in the media. However, with an increase in the amount of coir substrate, the chlorophyll, N, and K content was increased. After harvesting, there was no significant difference in the chemical properties of the horticultural nursery media of both plants. Thus, it can be suggested that, coir substrate after a single use could be recycled as horticulture nursery media.
Nitrogen (N) is a vital element in growing crops and is essential for improving the yield and quality of crops. Thus, N fertilizer is the most widely used fertilizer and the primary N input source in soil-crop systems. Inorganic fertilizers such as urea are known to improve crop productivity and increase soil fertility. However, application with excessive amounts can interfere with crop growth and accelerate soil acidification. For these reasons, the use of organic fertilizers, which mainly contain organic nitrogen, has gradually increased worldwide. Therefore, this study evaluated the effects of N fertilizer on the growth of Chinese cabbage including its functional compounds glucosinolates (GSLs). For the cultivation of Chinse cabbage, inorganic fertilizer was used for urea, and organic fertilizers were divided into conventional and biochar-based fertilizers. The growth parameters of Chinese cabbage treated by organic fertilizers was better than those of the inorganic fertilizers. Additionally, it was found that their co-application was more efficient. However, their GSL contents were lower with the application of the organic fertilizers. The characteristics of the experimental soil also changed according to the type, amounts and co-application of fertilizers. Therefore, this study presents the basis for an eco-friendly method that can increase the functionality and productivity of Chinese cabbage compared to conventional cultivations.
Kimchi cabbage is widely consumed in Korea, with the popularity of this pickled vegetable dish growing internationally due to its health benefits. In this study, the physical (size, color), functional (antioxidant activity, total polyphenol, and flavonoid content), and nutritional (water- and fat-soluble vitamins) characteristics of two new kimchi cabbage varieties, namely red and gold kimchi cabbages (RKC and GKC, respectively), were analyzed and compared with those of the common kimchi cabbage (CKC). There were no significant differences in the thickness or length of the three kimchi cabbages, although RKC had the narrowest outer leaves among the three varieties (11.94 cm). Regarding chromaticity, yellowness was highest in GKC (29.86), whereas redness was highest in RKC (9.31). Furthermore, RKC had the highest recorded vitamin B6 and B9 (1,288.5 ㎍/100 g and 776.7 ㎍ dietary folate equivalent/100 g, respectively). On the other hand, the fat-soluble vitamins vitamin A (β-carotene) and K (Phylloquinone) were both highest in GKC (907.1 ㎍/100 g and 712.2 ㎍/100 g, respectively). Generally, all kimchi cabbage samples contained high levels of vitamin E (1.8-4.9 mg α-tocopherol equivalent/100 g). RKC attained the highest antioxidant activity and total polyphenol and total flavonoid contents among the three kimchi cabbages. These results show that gold and red kimchi cabbage can be used as raw materials in the food-processing industry.
