Comparative Analysis of Chemical Compositions and Antimicrobial Activities of Essential Oils from Abies holophylla and Abies koreana

  • The chemical compositions and antibacterial and antifungal activities of essential oils extracted from Abies holophylla and A. koreana were investigated. GC-MS analysis revealed that 38 compounds comprised 95.88% of the A. holophylla essential oil, with the main components being bicyclo[2.2.1]heptan-2-ol(28.05%), ${\delta}3$-carene(13.85%), $\alpha$-pinene(11.68%), camphene(10.41%), dl-limonene(7.61%), $\beta$-myrcene(7.11%), trans-caryophyllene(5.36%), and $\alpha$-bisabolol(3.67%). In the essential oil from A. koreana, 36 compounds comprised 98.67% of the oil, and the main compounds were bornyl ester(41.79%), camphene(15.31%), $\alpha$-pinene(11.19%), dl-limonene(8.58%), fenchyl acetate(5.55%), and $\alpha$-terpinene(2.29%). Both essential oils showed great potential of antibacterial activity against several bacteria tested, in the range of 2.2-$8.8{\mu}g$ per disc by the agar disc diffusion method, and minimal inhibitory concentration(MIC) values of 5.5-21.8 mg/ml by the microdilution method. Both oils showed very effective antifungal activities toward all pathogenic strains tested, including Candida glabrata, with MIC values in the range of 0.5-2.2 mg/ml. As a whole, A. koreana oil showed better antibacterial and antifungal properties than A. holophylla oil.

  • Seven monoterpenes, three sesquiterpenes, three phenolics and one flavonoid were isolated from the MeOH extract of Amomum xanthioides. Their structures were determined by spectroscopic methods to be caryophyllene oxide (1), bornyl acetate (2), nerolidol (3), spathulenol (4), (-)-borneol (5), (+)-5-endohydroxycamphor (6), vanillic acid (7), protocatechuic acid methyl ester (8), betulabuside A (9), (1R,4S,6R)-6-hydroxyfenchan-2-one-6-O-$\beta$-D-glucopyranoside (10), (1S,4R,6S)-6-hydroxybornan-2-one-6-O-$\beta$-D-glucopyranoside (11), (1R,2S,4S,5R)-angelicoidenol 2-O-$\beta$-D-glucopyranoside (12), 1-O-vanilloyl-$\beta$-D-glucopyranoside (13), and quercetin-3-rhamnopyranoside (14). Compounds 6-14 were isolated for the first time from this plant source. Compounds 3 and 4 exhibited moderate cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

  • Isolation of oils from leaves of Juniperus phoenicea and Juniperus oxycedrus was obtained by steam distillation extraction method. The compositions of essential oils (EOs) were studied by means of GC-MS and GC-FID, using the internal standard method and relative response factors. Around ninety eight compounds were determined in total, representing 98.25 g/100 g of EO of J. phoenicea and 98.48 g/100 g of EO of J. oxycedrus, respectively. The volatile leaf oils were dominated by the terpenic hydrocarbon fractions (79.87 g/100 g) and (61.27 g/100 g) characterized by high contents of α-pinene (64.6 g/100 g) and (54.0 g/100 g) in J. phoenicea and J. oxycedrus, respectively, as the main component. Also, the enantiomeric distribution of α-pinene, sabinene, camphene, δ-3-carene, β-pinene, limonene, linalool, terpinen-4-ol, bornyl acetate, and borneol in both oils is presented for the first time.

소나무(Pinus rigida Miller) 잎 추출물의 휘발성 성분 (Volatile Compounds of Pine Needle(Pinus rigida Miller) Extracts)

  • This study was conducted to evaluate whether pine needle extracts can be used as tobacco flavors. Yield of essential oil, absolute and oleoresin extracted from pine needles is 0.07%, 1.20% and 6.08% respectively. The volatile compounds isolated from the three types of extracts were analyzed by gas chromatography(GC) and mass selective detector(MSD). Total 72 components were identified in the three type of extracts including 26 hydrocarbons, 16 alcohols, 13 esters, 9 acids, 4 phenols, 2 aldehydes and 2 ketones compounds. The major components were $\beta$-pinene, $\beta$-caryophyllene, $\delta$-cadinene and 4,5-dimethyl-1,3 -dioxol-2-one. There were 49 volatile components in the absolute, 44 components in the essential oil and 26 components in the oleoresin. The content of hydrocarbons and alcohols was higher in the essential oil extracted by simultaneous distillation extraction(SDE) than in others, while that of esters and acids was higher in the absolute than in others. Especially, phenols and ketones were identified only in the oleoresin. The components such as $\beta$-pinene, bornyl acetate, $\alpha$-terpineol and oxygenated terpenes have characteristic piney and fresh green odor. The contents of these components was higher in the essential oil and the absolute than in the oleoresin. Therefor, the essential oil and the absolute are expected to be more useful than the oleoresin as tobacco flavor.

Chemical and Antimicrobial Properties of Essential Oils from Three Coniferous Trees Abies koreana, Cryptomeria japonica, and Torreya nucifera

  • Three coniferous essential oils were extracted from Abies koreana, Cryptomeria japonica, and Torreya nucifera by hydrodistillation. The chemical composition of each oil was analyzed by GCMS, and their antimicrobial activities were tested against two bacteria and one yeast strains. Fortyseven compounds were identified from A. koreana oil, 39 from C. japonica, and 59 from T. nucifera. Main components of the essential oils were limonene (23.5%), bornyl acetate (17.9%), ${\alpha}-pinene$ (11.1%), and camphene (10.2%) in A. koreana, kaurene (26.3%), ${\gamma}-eudesmol$ (19.0%), elemol (6.9%), and sabinene (5.1%) in C. japonica, limonene (13.5%), ${\delta}-cadinene$ (10.5%), ${\alpha}-bisabolol$ (10.2%), and ${\alpha}-copaene$ (7.7%) in T. nucifera. Among the three coniferous trees tested, the essential oils of A. koreana exhibited higher and broader antimicrobial activity against the tested organisms than those of C. japonica and T. nucifera.

Monoterpenoids Concentration during Decomposition and Their Effect on Polysphondylium violaceum

  • The total monoterpenoid content of the pine litter layer and the availability of these compounds as inhibitors/stimulators on Polysphondylium violaceum of cellular slime molds were investigated. In order to determine the several monoterpenoids in the natural environment, we examined their concentrations in fresh, senescent, and decaying needles from 3 pine species (Pinus densiflora, P. thunbergii, P. rigida) by litter bag method. Total monoterpenoid content was highest in the fresh needles, but also remained relatively high in senescent needles. The effect of monoterpenoids identified from Pinus plants on the growth of P. violaceum was studied. We tested four concentrations (1, 0.1, 0.01, and $0.001\;{\mu}g/{\mu}L$) of each compound by using a disk volatilization technique. Each compound was treated after germination of spores of P. violaceum. All of the compounds at $1\;{\mu}g/{\mu}L$ concentration had a very strong inhibitory effect on cell growth of P. violaceum. Fenchone at all concentrations, myrcene, verbenone, bornyl acetate, and limonene at low concentrations stimulated the growth of P. violaceum. These results suggest that inhibitory or enhancing effects of selected monoterpenoids depend upon the concentration of the individual compound.

광릉쥐오줌풀, 넓은잎쥐오줌풀, 네팔산쥐오줌풀의 정유성분(精油成分)차이 (Differences of Essential Oil Content in Valeriana fauriei var. dasycarpa Hara, V. officinalis var lalfolia Miq and V. wallichii DC)

  • 한국산(韓國産) 식물자원(植輪資源)을 발굴(發掘)하여 약용(藥用) 또는 향료자원(香料資源)으로서의 활용을 목적으로 국내(國內)에서 자생(自生)하고 있는 쥐오줌풀 중 광릉쥐오줌풀 및 넓은잎쥐오줌풀과 세계시장에서 유통(流通)되고 있는 네팔산(産) 쥐오줌풀과의 유효성분(有效成分) 차리(差異)를 조사(調査)하였다. 정유성분(精油成分) 함량(含量)은 광증릉쥐오줌풀이 1.7%, 넓은잎쥐오줌풀과 네팔산(産) 쥐오줌풀에서는 각각 0. 82% 및 0. 83% 이었다. GC 및 GC-MS 분석에 의해 광릉쥐오줌풀의 정유(精油)에서 47종(種), 넓은쥐오줌풀에서는 21종(種) 그리고 네팔산(産) 쥐오줌풀에서는 25종(種)의 성분(成分)을 동정하였다. 확인된 성분들 증 광릉쥐오줌풀의 정유(精油)에서는 bornyl acetate(전체(全體) 정유(精油)의 33, 8%), camphene (10. 8%), cedrol (4. 1%) , ${\alpha}-pinene$(3. 2%), 분자양(分子量) 222의sesquiterpene alcohol(3%)이 주성분(主成分)이었고 넓은잎쥐오줌풀에서는 borneol (62. 5%), ${\beta}-sesquiphellandrene$(6.8%) , spathulenol (2. 1%) 그리고 네팔산(産) 쥐오줌풀에서는 borneol(74.3%), {\alpha}-elemene(2. 7%)$${\beta}-sesquiphellandrene$(2.3%)이 주성분(主成分)이었다. 쥐오 줌풀의 약리효능(藥理效能)의 원인물질(原因物質)들 중 valeranone, valrrenal, $[\alpha}-kessyl\;acetate$는 넓은잎쥐오줌풀이나 네팔산(産) 쥐오줌풀보다는 광릉쥐오줌풀에 많이 함유되어 있었으며 kessoglycol diacetate는 세 시료 모두에서 검출(檢出)되지 않았다. Valepotriate계열(系列) 화합물의 함량(含量)은 네팔산(産) 쥐오줌풀이 광릉쥐오줌풀이나 넓은잎쥐오줌풀보다 많이 함유되어 있었고 광릉쥐오줌풀은 넓은잎쥐오줌풀에 비해 항산화첨성(抗酸化添性)이 높은 경향을 보였다.

Antibacterial Activity of Essential Oil from Abies holophylla against Respiratory Tract Bacteria

  • We extracted essential oils from four species (Pinus densiflora, Larix kaempferi, Pinus koraiensis, and Abies holophylla) in the family Pinaceae to investigate their antibacterial activities against respiratory tract bacteria (Klebsiella pneumoniae, Haemophillus influenzae, Streptococcus pyogenes, Streptococcus pneumonia, and Neisseria meningitides). Among the tested oils, that from A. holophylla was showed strong activity based on disc diffusion and broth medium dilution (minimum inhibitory concentration, MIC) assays. Qualitative analysis of A. holophylla oil was carried out by GC-MS; ${\alpha}$-pinene, camphene, ${\beta}$-pinene, 3-carene, limonene, bornyl acetate, borneol, ${\beta}$-caryophyllene, ${\alpha}$-caryophyllene, caryophyllene oxide, and ${\alpha}$-bisabolol were identified as its major constituents. Fractionation by silica gel chromatography was performed to analyze the active constituents of the crude oil. In particular, one fraction containing caryophyllene oxide as the major constituent showed stronger antibacterial activity than the crude oil of A. holophylla. Growth rates of bacterial strains exposed to fraction D were explored by optical density (OD600) measurements while morphology was examined by optical microscopy observations ( ${\times}1000$). OD600 of K. pneumoniae decreased from 0.2582 to 0.005 in response to treatment with fraction D at a MIC value of $0.31{\mu}{\ell}/m{\ell}$.

Direct Headspace Sampling 방법을 이용한 당근의 휘발성 Terpenoids 분석에 관한 연구 (Studies on Measuring Volatile Terpenoids in Carrots Using the Direct Headspace Sampling Method)

  • 고품질 당근을 육성하기 위한 연구의 일환으로 당근의 휘발성 terpenoids를 GC 분석으로 측정하였다. 여러가지 휘발성 terpenoids 표준물질을 DHS(direct headspace sampling)방법으로 검출한 결과 ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, terpinolene 의 7가지 성분이 명확히 분리되었으나, ${\alpha}-phellandrene$${\beta}-myrcene$으로부터 분리되지 않았다. 또한 terpinen-4-ol, bornyl acetate, ${\alpha}-bisbolol$ 등 비등점이 높은 물질은 검출되지 않았다. 검출된 7가지 terpenoids 성분의 표준곡선은 결정계수$(r^2)$가 전부 0.99 이상으로서 고도로 유의한 표준곡선이었다. DHS 방법을 통한 본실험의 재현성을 측정한 결과, ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, limonene, ${\gamma}-terpinene$, terpinolene, total terpenoids의 변이 계수(C.V.)가 각각 6.8, 6.8, 8.4, 7.1, 3.8, 10.1, 7.1%로 $3.8{\sim}10.1%$의 범위를 나타내었다. 세계적으로 많이 재배되는 당근 65품종에 대한 ${\alpha}-pinene$, ${\beta}-pinene$, ${\beta}-myrcene$, ${\alpha}-terpinene$, limonene, ${\gamma}-terpinene$, terpinolene, total terpenoids를 측정한 결과, 각각 $0.28{\sim}2.48\;ppm$, $0.35{\sim}1.87\;ppm$, $0.56{\sim}1.51\;ppm$, 0 ppm, $0.59{\sim}1.84\;ppm$, $0.87{\sim}3.33\;ppm$, $5.15{\sim}35.81\;ppm$, $9.07{\sim}42.30\;ppm$으로서 품종에 따라 상당한 차이를 보였다. Total terpenoids 의 경우 65품종 중 5품종(7.7%)이 10 ppm미만이고, 15품종(23.1%)이 $10{\sim}11.99\;ppm$, 14품종(21.5%)이 $12{\sim}13.99\;ppm$, 9품종(13.8%)이 $14{\sim}15.99\;ppm$, 10품종(15.4%)이 $16{\sim}17.99\;ppm$, 4품종(6.2%)이 $18{\sim}19.99\;ppm$, 5품종(7.7%)이 $20{\sim}29.99\;ppm$, 3품종(4.6%)이 30 ppm 이상이었다. 전반적으로 일본에서 육성된 품종이 구미에서 육성되어 보급되고 있는 품종들에 비해 total terpenoids 함량이 낮은 편이었다.

미성숙 솔방울 열수추출물의 항균성 (Anti-bacterial Effects of Aqueous Extract Purified from the Immature Cone of Red Pine (Pinus densiflora))

  • Red pine (Pinus densiflora) is widely used traditional medicine, pharmacological and nutritional values from which the phytochemical compounds are derived. The present study was aimed to examine the antibacterial effects in the absence and presence of a immature red pine cone extract against 13 microorganisms. The components in the aqueous extract from immature red pine cone were identified by GC-MS. About 1.4% of total polyphenolic compound was measured in aqueous extract collected from immature red pine cone. Also, the high concentration of ${\beta}$-phellenandrene, ${\alpha}$-pinene, limonene, bornyl acetate and aldehyde was detected in total ion chromatograms. Of total 13 microorganisms, 4 microorganisms including Pseudomonas aeruginosa, Vibrio cholera, Listeria monocytogenes, Klebsiella pneumonia were effectively killed by aqueous extract of immature red pine cone. The highest anti-bacterial effect was detected in P. aeruginosa, followed by V. cholera, L. monocytogenes and K. pneumonia. In case of P. aeruginosa, the largest diameter of inhibition zone was maintained to 1/2 solution treated cells and slightly decreased at 1/4 and 1/8 solution treated cells. Also, in test used V. cholera and L. monocytogenes, the inhibition zone was strongly formed in only 1 and 1/2 solution treated cells, while K. pneumonia showed the very small diameter of inhibition zone in all concentrations. Therefore, these results suggested that the aqueous extracts of immature red pine cone should be considered as a new and potentially important anti-bacterial substrate to effectively prevent the microbial infection and penetration.
