This study was conducted to evaluate the safety of herbicide-resistant rice, a genetically modified organism (GMO) developed by the Rural Development Administration, in Sprague-Dawley rats. The nutrient content of herbicide-resistant polished and brown cooked rice was compared with that of conventional Ilpum polished and brown cooked rice to assess composition equivalence. Compositional analysis was performed to measure proximates, fiber, and minerals before animal feeding. Growing male rats were fed one of the following four diets for six weeks: Ilpum polished cooked rice (IP) and Ilpum brown cooked rice (IB) as a non-GMO and herbicide-resistant polished cooked rice (GP) and brown cooked rice (GB) as a GMO. We checked clinical symptoms (anorexia, salivation, diarrhea, polyuria, anuria, fecal change) every day, food intake, change of body weight twice a week, and serum biochemistry and organ weights after 6 weeks of experimental feeding among the four groups. Nutrient content of the herbicide-resistant rice was similar to that of the non-transgenic control and was within the published range observed for non-transgenic rice. We could not find any significant difference in the above-mentioned items as the index to be checked in the animals fed the GMO. These results suggest that the nutrient content of genetically modified herbicide-resistant rice is compositionally equivalent to that of conventional Ilpum rice and that growing male rats fed herbicide-resistant rice are no different from those fed Ilpum rice, non-GMO for 6 weeks.
This study was carried out to estimate the minimum protein requirement for the Korean college girls, and to see the effect of carbohydrate intake on the minimum protein requirement in the rat. In the first experiment, three students attending Ewha Womans University were given protein free diet for 7 days while caloric intake were adjusted to maintain constant body weight. Endogenous nitrogen excretion was measured to estimate their minimum protein requirement. Second experiment was carried out in 3 parts to find out the effect of carbohydrate intake on the minimum protein requirement using 15 female, 50 days old rats weighing 120g. Inexperiment 2-1, the rats were fed protein free diet for 9 days and endogenous nitrogen excretion was measured to determine the minimum protein requirement of the rats. In experiment 2-2, the minimum casein requirement of the rats needed to maintain nitrogen equilibrium was estimated by the least squares regression method feeding 3 different levels of casein determined from the result of experiment 2-1. Finally, in experiment 2-3, the rats were given the minimum casein requirement for 7 days with 3 different levels of carbohydrate to find out the effect of carbohydrate intake on the nitrogen balance. The results of this study were as follows. Experiment I. Daily endogenous nitrogen losses of 3 college girls were 1.89g in urine, 1.10g in feces and 0.0108g from skin since the total daily endogenous nitrogen loss was 3.0g and the total daily nitrogen intake was 0.52g, the minimum nitrogen requirement of these subjects was about 2.48g/day (15.5g protein/day). Experiment II. Experiment 2-1: daily endogenous nitrogen losses of the rats fed protein free diet were $22.63{\pm}0.48\;mg$ through urine and $13.12{\pm}0.47\;mg$ in feces. The total daily endogenous nitrogen ouput was $35.71{\pm}1.27\;mg$ and the minimum protein requirement of the rats was 225mg/day in this study. Experiment 2-2: the minimum casein requirement needed to maintain the rats in nitrogen equilibrium was determined to be 340mg/day. Experiment 2-3: the rats were given 350mg/day casein with 3 different levels of carbohydrate (12,26 or 46 Cal by sugar-cornstarch mixture). The nitrogen balances were measured to be +0.0021, +0.0073 and +0.0143 respectively and there were significant differences among 3 groups.
Purpose: This study was conducted in order to investigate the protective effects of ethanolic extract of Acanthopanax koreanum Nakai (AE) against carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Methods: Male Sprague-Dawley rats were randomly divided into four groups in order to receive the following experimental diets with intraperitoneal injection of $CCl_4$ (2.0 mL/kg body weight, 20% solution 0.65 mL) for eight weeks (n = 8 per group): $CCl_4$ control (CON), $CCl_4$ + AE 1% (AE1), $CCl_4$ + AE 3% (AE3), or $CCl_4$ + acanthoic acid 0.037%, which is equivalent to AE 3% (AA). Results: Highest serum ALT activity and albumin level were observed in the $CCL_4$ control group, but showed a significant decrease by either AE or AA supplementation in a dose-dependent manner (p = 0.0063 and 0.0076, respectively). Both hemotoxylin and eosin staining and Masson's staining indicated remarkable prevention of $CCl_4$-induced liver damage in the AE3 group. $TNF{\alpha}$ and IL-6 production were significantly lowered in the AE treated groups, but not in the AA group (p = 0.0016 and p = 0.0002, respectively). The effects of AE3 were greater than those of AA for inflammation and liver toxicity biomarkers. Conclusion: Taken together, the results suggested that ethanolic extract of Acanthopanax koreanum Nakai provided hepatoprotective effects, leading to the reduction of inflammatory response. In addition, the effect of AE was superior to that of single compound AA.
Purpose: Rice bran is a byproduct of the hulling of rice and contains a variety of bioactive components. Various studies have reported on the antioxidative, anticancer, immune-enhancing, and hypocholesterolemic effects of rice bran. However, few studies about the physiological activity of stabilized rice bran supplement on dietary intake of sugars is limited. The aim of this study was to investigate the effect of stabilized rice bran supplement on blood glucose in C57BL/6 mice fed a high sucrose diet. Methods: Animals were randomly divided into three groups respectively, and were fed a normal diet (ND group), a high sucrose diet (HSD group) or a high sucrose diet containing 20% stabilized rice bran (HSD-SRB group) for 12 weeks. Results: In the oral glucose tolerance test (OGTT), after seven weeks of feeding on the experimental diets, a significantly lower result was observed for HSD-SRB than for HSD at 30 and 60 minutes after oral administration in glucose solution (2 g/kg body weight). The incremental area under the curve (IAUC) of HSD-SRB was significantly lower than that of HSD. After 12 weeks, fasting blood glucose level of HSD-SRB was significantly lower than that of HSD. No significant difference in the serum insulin level was observed between HSD and HSD-SRB. However, HOMA-IR was significantly decreased in HSD-SRB compared to HSD. In addition, HOMA ${\beta}$-cell was significantly increased in HSD-SRB compared to HSD. Triglyceride in liver of HSD-SRB was significantly lower than that of HSD. Conclusion: Feeding diets containing 20% rice bran improved insulin resistance and insulin secretion by decreasing triglyceride in liver. Thus, rice bran has a positive effect on glycemic control. In addition, the results are expected to be utilized as a basis for human study and development of food products with added rice bran.
Purpose: The aim of this study was to assess the effects of guava leaf extract (GLE) supplementation on antioxidant enzyme activity and expression of endothelial nitric oxide synthase (eNOS) mRNA in ovariectomized rats. Methods: All animals were randomly assigned to four groups (n = 7 for each group): non-ovariectomized control (Sham), the ovariectomized control (OVX), ovariectomy + 150 mg/kg b.w. of GLE ($OVX{\cdot}GL$), and ovariectomy + 300 mg/kg b.w. of GLE ($OVX{\cdot}GH$). Treatment groups were administered GLE for 8 weeks every day. Results: Body weight gain was significantly reduced in the $OVX{\cdot}GL$ group compared with the OVX group (p < 0.05). The level of serum $17{\beta}$-estradiol (E2) was significantly lower in the OVX groups than the Sham group (p < 0.05). Serum triglyceride (TG) and HDL-cholesterol levels were not significantly different between all groups. However, serum total cholesterol (TC) level was significantly reduced in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Serum free fatty acid (FFA) level and liver TG level were significantly reduced in both $OVX{\cdot}GL$ and $OVX{\cdot}GH$ groups compared with the OVX group (p < 0.05). Furthermore, serum glutathione peroxidase (GPx) activity was significantly elevated in the GLE groups (p < 0.05). The mRNA expression level of GPx was not affected by ovariectomy. However, administration of GLE resulted in significantly increased nuclear factor erythroid 2-related factor (Nrf2) and catalase (CAT) mRNA expression levels in the liver (p < 0.05). In addition, liver nitric oxide (NO) level was significantly reduced in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Expression level of endothelial nitric oxide synthase (eNOS) was significantly elevated in the $OVX{\cdot}GH$ group compared with the OVX group (p < 0.05). Conclusion: These results suggest that GLE could have protective effects in OVX rats by stimulating eNOS expression and improving the antioxidant defense system.
Age and growth of the marbled rockfish, Sebastiscus marmoratus were studied based on 818 specimens caught from the coastal waters of Cheju Island from July 1992 to July 1993 and from June 1994 to July 1995. According to the monthly changes of marginal increment in each annual ring groups, the ring of otolith was formed in both female and male in February. Relationship between the total body length (TL) and the radius of otolith (R) was estimated, Growth of females was rather slower than that of male in total length; TL=-0.526+4.818R(r=0.847) for female, and TL=-1.895+ 5.239R (r=0.881) for male. The composition of herbicide-resistant brown rice with/without heating treatment was compared with those of conventional Ilpum brown rice with/without heating treatment to assess composition equivalence. Compositional analysis was performed to measure proximates, fiber, and minerals. The nutritional components of herbicide-resistant rice were similar to those of the nontransgenic control or were within the normal range of nontransgenic rice. Four groups of experimental male rats were fed one of the following diets for eight weeks: Ilpum brown rice (I) and its heated rice (IH) as non-GMO, and herbicide-resistant brown rice (G) and its heated rice (GH) as GMO- We checked clinical symptoms (anorexia, salivation, diarrhea, polyuria, anuria, fecal change), food intake, and water consumption every day, change of body weight once a week, and serum biochemistry and organ weights after 8 weeks of experimental feeding We did not find any significant differences in the above-mentioned items. These results suggested that genetically modified herbicide-resistant rice was compositionally equivalent to conventional Ilpum rice, and nutritional characteristics and safety of herbicide-resistant rice in old male rats treated for 8 weeks were not different from those of Ilpum rice, non-GMO.

Hypoglycemic effect of Prunus mume (PM) extract containing in Sangjinyangheul-tang and Hwangkeumtang, one of the diabetic herbal medicines, was determined by investigating insulin-like action, insulin sensitizing action and a-glucoamylase suppressing action. Insulin-like activity of 3T3-L1 fibroblast was not shown with the treatment of PM methanol extracts. However, treatment with 20% or 40% PM methanol extracts and differentiation inducers significantly decreased the differentiation of 3T3-L1 fibroblasts to adipocytes. A significant insulin sensitizing activity was observed in 3T3-L1 adipocytes, giving PM extracts (60%, 80% and 100%) with 1 ng/mL insulin to reach glucose uptake level increased by 50 ng/mL of insulin alone. In addition, 20% and 40% methanol extracts of PM suppressed the a-glucoamylase activity by 30% in vitro. However, there was no significant differences in the peak of serum glucose levels and area under the curve in Sprague Dawley male rats treated with PM ethanol extract or cellulose and 2 g maltose or dextrin/kg body weight. These data suggested that PM extracts contain effective insulin sensitizing compounds, lipid synthesis suppressing compounds and possibly a-glucoamylase suppressing compounds. Therefore, PM extracts are beneficial for anti-diabetic treatment in obese diabetic patients.

A computerized dietary prescription and nutritional counseling system for patients having hyperlipidemia has been developed using a personal computer. This system is composed of three programs. The first program is designed to investigate dietary history of patient, such as a dietary habit and a preference of food, to find out his incorrect dietary behavior and to give him some suggestions to correct dietary behavior. The second one is developed to analyze the energy and nutrients intake using 24-hour dietary recall method and also evaluate the status of dietary intake, especially the status of dietary fat. With these data, patient can replan pattern of his food intake including in-between-meal snack as well as regular meals. The third one is the diet and menu Planning program made using food exchange table. It provides the patient with a meal pattern suitable in his weight, activity and the other status of the body. Practicing these programs, patient with hyperlipidemia can help himself very conveniently in organizing his meal plan and in improving his dietary behavior.

This study was conducted to investigate the effect of dietary protein and fiber levels on the activities of ethanol metabolizing enzymes of liver in ethanol-treated rats. Sprague-Dawley male rats were fed on diets containing two levels of protein(7, 20%/kg diet) and pectin(5, 10%/kg diet). In ethanol experiments, ethanol(25% v/v) was administered by oral intubation(5g/kg body weight) at the same time once a day Control animals received an isocaloric dose of sucrose. The rats were sacrificed after 5 weeks of feeding periods. Alcohol dehydrogenase and microsomal ethanol oxidizing system activities of hepatic tissue were increased more in ethanol-treated groups than in control groups. Increment of activities predominated in normal protein normal fiber group. Aldehyde dehydrogenase activity was decreased in ethanol-treated groups and significantly decreased in normal Protein normal fiber group. Cytochrome P-450 content was significantly increased in ethanol-treated groups and Predominated in normal protein groups. Xanthine oxidase activity was increased in ethanol-treated groups, but not significantly except normal protein normal fiber group. Glutathione content tended to increase in proportion to level of dietary protein and was higher in normal fiber groups than in high fiber groups, whereas it was decreased by ethanol treatment. Lipid Peroxide content was significantly increased in low Protein normal fiber groups.
