만성(慢性) Acohol 섭취(攝取) mouse에서 alcohol 대사(代謝) 효소활성(酵素活性)에 미치는 인삼(人蔘)의 영향(影響) (Effect of Ginseng on the Hepatic Alcohol Metabolizing Enzyme System Activity in Chronic Alcohol-Treated Mouse)

  • The present study was undertaken to investigate an effect of ginseng butanol fraction(total saponin) on the hepatic ethanol metabolism, we used experimental animals for the subject of study. When, in case of ADH and MEOS, ginseng butanol fraction was added, enzyme activity was increased in a small dose, and on the contrary, in a large dose, showed inhibitory effect. in catalase, the activity showed no significant effect by adding ginseng butanol fraction. In the light of kinetic aspect, when, in reaction mixture, ethanol and ginseng butanol fraction were concurrently added and reacted, Km value of ADH and MEOS was decreased. After pretreated with ginseng butanol fraction and inducement of acute toxic state by ethanol, the activities of ADH and MEOS were increased to an extent of about 25% compared to controls. But catalase activity was not significantly affected. In case that ginseng butanol fraction was given to mice fed with 5% ethanol instead of water for 60 days, the activities of ADH and MEOS were increased about 20% to 50% compared to ethanol-treated group. On the contrary, catalase activity was not affected. But blood concentrations of ethanol were decreased due to ginseng butanol fraction treatment. All these observations suggested that reduction of ethanol blood concentration should be dependent upon increased activities of ADH and MEOS. Thereby it suggests the recovery from alcohol intoxication can be prompted by treatment with ginseng.

Intake and blood concentrations of folate and their association with health-related behaviors in Korean college students

  • The purpose of this study is to assess folate intake, and serum and red blood cell (RBC) folate concentrations, and investigate the association between folate status and health-related behaviors among Korean college students. A total of 169 students, aged between 18 and 27 years, participated in this study. Dietary intake data were collected by trained interviewers using a 24-hour recall method for three non-consecutive days in 2009. Information on health-related behaviors was obtained by a self-administered questionnaire. Serum and RBC folate concentrations were measured by microbiological assay. The average intakes of folate were $456{\mu}gDFE$ and $347{\mu}gDFE$ in male and female students, respectively. While the average serum folate concentration was significantly lower in male students (8.9 ng/mL) compared to female students (12.5 ng/mL), RBC concentrations were not significantly different between male (398.6 ng/mL) and female students (405.3 ng/mL). In male students, low serum folate concentrations were associated with total folate intake less than the Estimated Average Requirement, non-use of folic acid supplements, smoking, alcohol drinking at least once a week and low physical activity. In female students, low serum folate concentrations were associated with smoking and alcohol drinking at least two drinks at a time and BMI ${\geq}25$. Alcohol drinking and low physical activity were also associated with low RBC folate concentrations in both male and female students. In order to improve folate nutritional status of college students, the practice of desirable health-related behaviors, such as non-smoking, moderate alcohol drinking, regular physical activity, and maintenance of healthy BMI should be encouraged along with consumption of folate-rich foods and supplements.

우리나라 여성의 임신 중 커피 음용 실태와 산모혈 및 제대혈 중 지질치 변화에 미치는 영향 (Coffee Consumption During Pregnancy in Korean and Effect upon Serum Lipids Level in Maternal and Umbilical Cord Blood)

  • To investigate the effect of coffee consumption during pregnancy on serum lipids in maternal and infants` umbilical cord blood, 76 cases of newborn infants and women delivered o f a child who had delivered at hospital located in Chung-Ju city from Feb. 1, 1996 to Aug. 31, 1996 were studied. The gestational ages of cases were 37 to 42 weeks and the average maternal ages were $28.3{\pm}4.63$ years old without any other medical or obstetric problems. Infants` Apgar scores and birth weights were decreased by above 3 cups per day of coffee consumption during pregnancy. No relationships between coffee consumption during pregnancy and maternal serum lipid levels were observed. However, serum total cholesterol and LDL-cholesterol levels in infants` umbilical cord were significantly increased with increasing coffee consumption during pregnancy. In contrary to coffee consumption, alcohol drinking habits affected maternal serum lipid levels but not serum lipid levels in infants` umbilical cord blood. Coffee consumption during pregnancy increased caffeine concentration in infants` umbilical cord blood. These results suggest that caffeine from maternal consumption of coffee can easily pass to the fetus and significantly affect serum lipids levels in infants` umbilical cord blood rather than those in maternal blood.

Alcohol Intoxication and Glasgow Coma Scale Scores in Patients with Head Trauma

  • Purpose: Alcohol intoxication is commonly associated with traumatic brain injury (TBI), but the influence of alcohol on the Glasgow Coma Scale (GCS) score remains unclear. This study investigates the effects of blood alcohol concentration (BAC) on the GCS score in head trauma patients with alcohol intoxication. Methods: In total, 369 head trauma patients with alcohol intoxication in a 1-year period were retrospectively analyzed. The patients underwent head computed tomography and had a BAC ≥80 mg/dL. Patients were divided into TBI and non-TBI groups. Brain injury severity was further classified using the head Abbreviated Injury Score (AIS). The effects according to 5 BAC groups were examined. Results: The TBI group consisted of 64 patients (16.2%). The mean BAC was significantly higher in the non-TBI group (293.4±87.3 mg/dL) than in the TBI group (242.8±89.9 mg/dL). The mean GCS score was significantly lower in the TBI group (10.3±4.6) than in the non-TBI group (13.0±2.5). A higher BAC showed a significant association with a lower mean GCS score in the TBI group, but not in the non-TBI group. Above ≥150 mg/dL, higher BACs showed significant odds ratios for a lower GCS score. Conclusions: The influence of alcohol in patients with head trauma depended on the presence of a brain injury. An association between a higher BAC and a lower GCS score was only observed in patients with TBI. Therefore, if a severe brain injury is suspected based on a GCS evaluation in patients with alcohol intoxication, prompt diagnosis and intensive care should be performed without delay.

난소절제 랫트에서 에스트로겐 저하가 알코올 대사에 미치는 영향 (The Effect of Estrogen Deficiency on Ethanol Metabolism in Ovariectomized Rats)

  • This study was designed to examine the effect of estrogen deficiency on the metabolism of ethanol in ovariectomized rats. Female rats were assigned to an ovariectomy (OVX) and a sham (SHAM) surgery group. Gain body weight was greater in incresed in OVX group and especially uterus weight significantly decrease depending on the concentration of estrogen after 3 month of ovariectomy. Ethanol at the tolerative dose (6 g/kg) was injected to rats by oral administration to measure the concentration of ethanol in blood. The area under the blood concentration time curve (AUC) was significantly lower in OVX group than SHAM group. The significant decrease in AUC in OVX group indicates that the estrogen deficiency leads to changes of the factors related to ethanol metabolism. Activity of hepatic alcohol dehydrogenase was not significantly influenced by the ovariectomy and also the ethanol elimination rate in vivo was not different. Cytochrome P450 isozymes did not show any changes except CYP 1A1 and 2E1. Level of hepatic glutathione in OVX group was higher after treatment of ethanol. Therefore the reduction of AUC appears not to be directly associated with the difference of ethanol metabolizing enzyme, but to be related with the physical factors like body weight.

헛개과병추출물과 인삼열매추출물의 혼합 음료 섭취가 숙취해소에 미치는 효과 (Effects of mixed supplementation on Hoveni dulcis Thunb extracts and Ginseng-Berry extracts on hangover curves)

  • 본 연구는 알코올 분해 및 간 손상 보호, 피로회복, 체력증진 등의 효과가 있는 헛개과병추출물과 인삼열매 복합추출물의 섭취가 숙취해소에 미치는 효과를 규명하는데 목적이 있다. 20세 이상 자발적으로 참여한 64명을 대상으로 무작위배정과 반복측정설계방법을 이용하여 블록무작위배정을 통해 집단 별 인원을 구성하였다. 집단 구성은 헛개과 병추출물+인삼열매 복합추출물(ARI 1000) 투여군, 헛개과병추출물투여군, 인삼열매추출물 투여군, 위약군을 각각 16명을 구성하였다. 호흡기알콜농도 변화결과 ARI 1000 투여군이 음주 1시간에서 헛개과병추출물투여군, 인삼열매추출물 투여군, 위약군에 비해 유의하게 낮았으며, 음주 2시간, 3시간에는 위약군에 비해 유의하게 낮았다(p<0.05). 혈중 알코올 농도결과 음주 2시간, 3시간 후에 ARI 1000 투여군이 헛개과병추출물투여군, 인삼열매추출물 투여군, 위약군에 비해 유의하게 낮았다(p<0.05). 결론적으로 음주 전 ARI 1000 섭취는 호흡기와 혈중 알코올 농도를 유의하게 감소시켜 숙취효과에 효능이 있는 것으로 생각된다.

국내 시판 숙취해소제 4종의 혈중 알코올 및 아세탈데히드 농도 감소 효능 비교 (Efficacy Verification of Four Hangover Cure Products for Reducing Blood Alcohol and Acetaldehyde Concentrations in Sprague - Dawley Rats)

  • 최근 알코올성 간손상이나 간건강에 효과가 있는 천연물 원료를 포함하는 숙취해소제들이 시판되고 있다. 하지만 간손상이나 간건강에 대한 기능성은 숙취해소와는 다르기 때문에 숙취해소제들의 숙취해소 효능 검증이 필요하다. 본 연구는 국내의 4가지 숙취해소제의 효능을 알코올에 의한 숙취유도 흰쥐 모델에서 비교 조사하고자 하였다. 4가지 샘플들은 각각 C, M, R, S로 명명되었다. 동물실험에서 숙취 완화 효능은 혈중 알코올 및 아세탈데히드 농도로 평가되기 때문에 각 샘플들의 효과를 혈중 알코올과 아세탈데히드의 농도와 AUC (Area Under the Curve)로 평가하였다. 알코올 투여는 유의적으로 흰쥐의 혈중 알코올과 아세탈데히드 농도를 증가시켰다. 샘플 C, M, R, S순으로 에탄올의 Cmax와 AUC 감소율은 각각 5.9%, 3.1%, 8.4%, 11.7%와 8.9%, 2.2%, 12.1%, 19.6%였다. 아세탈데히드의 경우, 같은 샘플 순서로 Cmax는 14.2%, 15.2%, 28.2%, 35.0% 감소하였으며, AUC는 21.6%, 7.5%, 22.4%, 29.9%의 감소율을 보였다. 결론적으로, 샘플 S, R, C, M순으로 4개의 샘플 모두 EtOH군에 대비하여 숙취가 해소되는 경향을 보였으나, 샘플 S만이 에탄올과 아세탈데히드의 혈중 농도와 AUC 모두에서 통계적으로 유의적인 감소율을 보였다. 따라서 이러한 실험결과는 시중에 판매되는 숙취해소제의 객관적인 효능 검증이 부족하다는 것을 시사한다.

인진(茵蔯)과 갈근(葛根)이 d-galactosamine, 급성(急性) alcohol중독(中毒) 및 $CCl_4$중독(中毒) 백서(白鼠)의 간손상(肝損傷)에 미치는 영향(影響) (Studies on the effects of PUERARIAE RADIX(葛根) and ARTEMISIAE HERBA(茵蔯) on experimental liver damages induced by alcohol, d-galactosamine and $CCl_4$)

  • Recent survey shows that chronic liver disease such as chronic hepatitis, liver cirrhosis and hepatoma is the third leading causes for death in Korea. In oriental medicine, viral hepatitis is related to Hwangdal(黃疸) and alcoholic liver disease is related to Joosang(酒傷). ARTEMISIAE HERBA and PUERARIAE RADIX have long been used in treating those symptoms. This study was done to evaluate the effect of AR1EMISIAE HERBA and PUERARIAE RADIX on viral and alcoholic hepatitis. ARTEMISIAE HERBA and PUERARIAE RADIX were decocted respectively with water and followed by vaccum evaporation. The solution was diluted to adequate concentration. Sprague-Dawley rats were used in this experiment. Each group was given PUERARIAE RADIX or ARTEMISIAE HERBA solution orally and CCl4, d-galactosamine or alcohol was given orally 30 minutes later. After 24 hours of starvation, blood samples were taken to check serum GOT, GPT, LDH and ALP activities, TC, TG, glucose and BUN levels. These results show that ARTEMISIAE HERBA has better effect on liver injury induced by d-Galactosamine than PUERARIAE RADIX and that both ARTEMISIAE HERBA and PUERARlAE RADIX have good effect on acute alcoholic liver disease while in the liver injury induced by $CCl_{4}$, PUERARIAE RADIX has better inhibitory effect on serum AST, ALT and ALP levels and ARTEMISIAE HERBA has better inhibitory effect on serum total cholesterol and triglyceride. And the result that high concentration group has better effect shows these effects are concentration-dependent. Further study on the mechanism of these herbs is still required.

다이설피람-에탄올 반응에 의한 저체온증 1례 (A case of Hypothermia Resulting from Disulfiram-Ethanol Reaction)

  • Disulfiram (tetraethylthiuram disulphid) is used in the treatment of chronic alcoholism since it causes an unpleasant aversive reaction to alcohol. It works by inactivating hepatic aldehyde dehydrogenase, leading to pronounced rise in the acetaldehyde concentration when ethanol is metabolized. Acetaldehyde causes alcohol sensitivity, which involve vasodilation associated with feeling of hotness and facial flushing, increased heart rate and respiration rates, lowered blood pressure, nausea, headache. One of its metabolites, diethyldithiocarbamate (DDC) can inhibit the enzyme dopamine $\beta$-hydroxylase (DBH), this may account for the profound refractory hypotension and hypothermia seen with the disulfiram-ethanol reaction (DER), resulting from norepinephrine depletion. This report is presents the case of a patient we met, who presented with hypothermia caused by the disulfiram-ethanol reaction, and along with a brief review of the subject.

급성 알코올 중독에서 헛개나무 추출물을 포함한 식품 조성물의 보호 효과 (Protective Effects of Food Including Hovenia dulcis on Acute Alcohol Intoxication)

  • 본 연구는 기존에 알코올 해독 기능이 우수한 것으로 알려진 헛개나무 열매 추출물과 헛개나무 열매 추출물을 주성분으로 하여 매실 및 댕댕이나무 열매 추출물이 혼합되어진 새로운 식품 조성물의 항알코올 기능에 대한 효능을 비교 분석함으로써 새로운 알코올 독성 경감 소재를 개발하고자 하였다. 새로운 식품 조성물인 SAC-1을 흰쥐에게 투여한 후 알코올을 섭취시켰을 때 혈중 알코올 농노의 저하가 헛개나무 추출물에 비하여 매우 우수한 효능이 있는 것으로 나타났으며, 간손상의 지표인 GOT, GPT에 있어서 대조군에 비하여 낮은 값을 나타내어 알코올에 의한 간손상을 경감시킬 수 있는 것으로 생각된다. 또한, 조직에서 산화적 손상으로 효능을 살펴본 결과는 지질과산화물의 생성을 혈장과 간 조직에서 모두 억제하는 것으로 나타났으며, total glutathione의 감소를 억제하는 것으로 나타나 SAC-1은 알코올성 조직 손상에서 충분한 보호 작용을 나타낼 수 있었던 것으로 생각된다. 특히 SAC-1은 헛개나무 열매 추출물 단독 사용에 비해서 더 높은 활성을 나타낸 것으로 나타났으며, 이것은 SAC-1이 알코올 해독에 좋은 천연 성분을 함유하고 있는 결과로 추측된다. 이상의 결과에서 SAC-1은 알코올로 인해 유도되어지는 조직 손상 및 숙취 증상으로부터 조직을 보호할 수 있는 능력이 있는 것으로 생각된다.
