Analysis of Gene-specific Molecular Markers for Biotic and Abiotic Stress Resistance in Tropically adapted Japonica Rice Varieties

  • Since 1992, the Rural Development Administration (RDA), Republic of Korea in collaboration with International Rice Research Institute (IRRI) has developed 6 japonica rice varieties(MS11, Japonica 1, 2, 6, 7 and Cordillera 4) that are adaptable to tropical regions. However, these varieties show moderate resistance or susceptibility to certain biotic and abiotic stress. The development of varieties with more stable forms of resistance is highly desirable, and this could be possibly achieved through rapid introgression of known biotic and abiotic resistant genes. In this study, we analyzed the allele types of major biotic stress resistant genes including Xa5, Xa13, Xa21 and Xa25 for bacterial leaf blight, Pi5, Pi40, Pish and Pita2 for blast, tsv1 for rice tungro spherical virus, and Bph6, Bph9, Bph17, Bph18 and Bph32 for brown planthopper by using gene-specific molecular markers. In addition, seed quality related genes Sdr4 for preharvest sprouting and qLG-9 for seed longevity were also analyzed. The results revealed that2h5 and Xa25 resistance alleles showed in all varieties while Pi5 resistance allele showed only in MS11. The Pish resistance allele were present in five varieties except for Japonica 1. Meanwhile, for the rest of the genes, no presence of resistance alleles found in six varieties. In conclusions, most of tropical japonica varieties are lack of the major biotic stress resistant genes and seed quality genes (Sdr4 and qLG-9). Moreover, the results indicated that rapid deployment of a few major genes in the current tropical japonica rice varieties is urgent to increase durability and spectrum of biotic stress resistance and also seed dormancy/longevity which are essential traits for tropical environments.

Effect of Biotic Substances on Isoflavone Content in Soybean Germination (Biotic 물질이 콩 발아 중 Isoflavone 함량에 미치는 영향)

  • For humans, soybean and soybean products are the main dietary sources of isoflvones, which are polyphenolic compounds that represent one of the most common categories of phytoestrogens. The objective of this study was to determine isoflavone concentrations in soybean cultivars during germination when treated with some biotic substances. Three soybean cultivars were germinated in replicated trials in 2018/2019 and their individual and total isoflavone concentrations were determined using HPLC (High-Performance Liquid Chromatography). Significant differences were observed in total isoflavone content were observed among cultivars regardless of years and treatments. 'Daepung2-ho' and 'Uram' had significantly higher total isoflavones than 'Pungsannaul-kong'. Differences among treatments were also significant for total isoflavone content. In 2018, with chitosan treatment, total isoflavone concentration ranged from 551.15 to 7584.07 ㎍ g-1, with an average of 2972.64 ㎍ g-1 across cultivars. In 2019, there was no significant difference among treatments in total isoflavone content. Regarding individual isoflavone concentrations, the malonyl-glucoside groups accounted for over 85% of the total isoflavone content, which is indicated that these groups play an important role with regard to isoflavone components in soybean seeds. The individual proportions in the total concentrations of isoflavones varied according to germination period and seed tissues. Glucosides and malonyl-glucosides showed differences in concentrations among seed tissues, aglycones were further accumulated as germination period was progressed. This study suggests that biotic substances have an impact on seed isoflavone content during germination. However, cultivars with consistently high or low isoflavone concentrations across biotic substance treatments were identified desspite differences in germination period and seed tissues, demonstrating that the genetic factor plays the most important role in isoflavone accumulation.

Water Environmental Assessment by the Species Biotic Index of Freshwater Fish in the Namdaecheon, Gangneung City (담수어류의 종생물지수를 이용한 강릉 남대천의 수환경 평가)

  • We investigated the water environmental assessment by the species biotic index (SBI) of freshwater fish at the Namdaecheon in Gangneung city, Gangweon-do during June and July in 2004 and June in 2005. In fish fauna, 29 species and 10 families were collected. Dominant species was Zacco platypus (relative abundance 15.33%) and subdominant species was Rhynchocypris steindachneri (13.13%). Species biotic index (SBI) in station 1, 2, 3, 4 was 1.70, 2.85, 1.00 and 3.39 respectively and water environmental grade by SBI was all very good. Station 5 was 4.13 in SBI and good grade. Station 6 was 4.47 and fairly good. Station 7 was 7.25 and poor. And station 8 was 8.10 and very poor grade. Results of water environmental grade by SBI and water quality grade were very similar in this stream.

Comparative Analysis on the Application of Biotic Indices for Environmental Assessment of a Polluted Stream (Jinwi Stream) (오염하천(진위천)의 환경평가를 위한 생물지수간 적용성 비교분석)

  • Jinwi Stream is considerably polluted. The urban development in a Jinwi Stream basin can make state of aquatic ecosystem worse. However, researches for aquatic ecosystems in Jinwi Stream are insufficient. In this study, biotic indices of periphytic diatoms (DAIpo and TDI), benthic macroinvertebrates (EPT, KSI and ESB) and fish (IBI) were compared with the annual water quality data. Benthic macroinvertebrates indices showed highly significant correlations with concentrations of organic materials and nutrients, while DAIpo, TDI and IBI showed low correlations with them. In particular, ESB can be considered as an useful indicator that reflects the degree of diversity and abundance of biotic community as well as water quality. In polluted and disturbed streams as Jinwi Stream, DAIpo, TDI and IBI appeared to be not available for evaluating and discriminating the water quality, although they have been known as good indices in general streams.

A Study on Importance of Assessment Factors and Indicators of Natural Ecosystem for Environmentally Friendly Land Conservation (환경친화적 국토보전을 위한 자연생태계 평가요인 및 평가지표의 중요도에 관한 연구)

  • This study was carried out to offer the basic methodology of the system and model to objectively assess the natural ecosystem for environmentally friendly land conservation and present the alternative plan on establishing the environmental policy. The results of this study were as follows. We selected four assessment factors associated with biotic, abiotic, qualitative, and functional factors. Also, there were extracted fifty-six indicators including density, total nitrogen, hemeroby degree, and goods production. The assessment factor showed that biotic one was very important. The importance of indicators were analyzed that rare and endangered plant was important in biotic factor, in case of abiotic, qualitative, and functional factors, organic matter, landscape diversity, and conservation of ecosystem were greatly important. The results of factor analysis on the characteristics of indicators, classified biotic factor into six factors including a structural one, abiotic factor as a soil and physical one, qualitative factor as five ones including hierarchical one, and functional factor as public and conservational one. In the results of analysis on assessment model, R-square of biotic factor was 51.7%, those of abiotic, qualitative, and functional one were each 58.4%, 44.2%, and 39.3%, and statistical problem was no existence. In future, to develop the assessement model and methodology of sustainable natural ecosystem, we will have to achieve the integrated model and grouping by assessment factor.

The Selection of Suitable Site for Park and Green Spaces to Increase Accessibility and Biodiversity - In Case of Seongnam City - (접근성과 생물다양성 증진을 고려한 도시 공원·녹지의 필요지역 선정 - 성남시를 사례로 -)

  • Urban park and green space provide various functions. Among the functions, human benefit and increase of biodiversity are known to be important. Therefore, it is important to consider human and biotic aspect in the process of selecting suitable site for park and green space. However, there is insufficient research on both aspects. In this study, we used green network to analyze human and biotic aspect to select suitable site for park and green space in Seongnam City in Korea. To analyze the green network, we used accessibility for human aspect and used dispersal distance and habitat size for biotic aspect. We conducted least-cost path modelling using movement cost. In case of biotic aspect, GFS (generic focal species) is used to estimate habitat size and dispersal distance. To find out suitable site for park and green space, we used an overlay analysis method. As the result, old residential areas are shown have insufficient green network which needs park and green space. Furthermore, the green network for biotic aspect is insufficient in old residential areas comapred to green network for human aspect. The result of this study could contribute in planning of park and green space to maximize their functions.

Assessment of Risk Based Pollution Level of Pb and Cd in Metal Contaminated Soils Using Biotic Ligand Model (Biotic ligand model에 근거한 중금속 오염지역의 Pb 및 Cd 위해오염도 평가기법 개발)

  • Risk based pollution level of Pb and Cd in metal contaminated soils depending on physicochemical properties of soil in a target site was assessed using biotic ligand model. Heavy metal activity in soil solution defined as exposure activity (EA) was assumed to be toxic to Vibrio fischeri and soil organisms. Predicted effective activity (PEA) determined by biotic ligand model was compared to EA value to calculate risk quotient. Field contaminated soils (n = 10) were collected from a formes area and their risk based pollution levels were assessed in the present study using the calculated risk quotient. Concentrations of Pb determined by aqua regia were 295, 258, and 268 mg/kg in B, H and J points and concentrations of Cd were 4.73 and 6.36 mg/kg in G and I points, respectively. These points exceeded the current soil conservation standards. However, risk based pollution levels of the ten points were not able to be calculated because concentrations of Pb and Cd in soil solution were smaller than detection limits or one (i.e., non toxic). It was because heavy metal activity in soil solution was dominant toxicological form to organisms, not a total heavy metal concentration in soil. In addition, heavy metal toxicity was decreased by competition effect of major cations and formation of complex with dissolved organic carbon in soil solution. Therefore, it is essential to consider site-specific factors affecting bioavailability and toxicity for estimating reliable risk of Pb and Cd.

SNAREs in Plant Biotic and Abiotic Stress Responses

  • In eukaryotes, membraneous cellular compartmentation essentially requires vesicle trafficking for communications among distinct organelles. A donor organelle-generated vesicle releases its cargo into a target compartment by fusing two distinct vesicle and target membranes. Vesicle fusion, the final step of vesicle trafficking, is driven intrinsically by complex formation of soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs). Although SNAREs are well-conserved across eukaryotes, genomic studies revealed that plants have dramatically increased the number of SNARE genes than other eukaryotes. This increase is attributed to the sessile nature of plants, likely for more sensitive and harmonized responses to environmental stresses. In this review, we therefore try to summarize and discuss the current understanding of plant SNAREs function in responses to biotic and abiotic stresses.

Assembly and Function of Seed Endophytes in Response to Environmental Stress

  • Seeds are colonized by diverse microorganisms that can improve the growth and stress resistance of host plants. Although understanding the mechanisms of plant endophyte-host plant interactions is increasing, much of this knowledge does not come from seed endophytes, particularly under environmental stress that the plant host grows to face, including biotic (e.g., pathogens, herbivores and insects) and abiotic factors (e.g., drought, heavy metals and salt). In this article, we first provided a framework for the assembly and function of seed endophytes and discussed the sources and assembly process of seed endophytes. Following that, we reviewed the impact of environmental factors on the assembly of seed endophytes. Lastly, we explored recent advances in the growth promotion and stress resistance enhancement of plants, functioning by seed endophytes under various biotic and abiotic stressors.

Induced Systemic Tolerance to Multiple Stresses Including Biotic and Abiotic Factors by Rhizobacteria (근권미생물에 의한 식물의 생물·환경적 복합 스트레스 내성 유도)

  • Recently, global warming and drastic climate change are the greatest threat to the world. The climate change can affect plant productivity by reducing plant adaptation to diverse environments including frequent high temperature; worsen drought condition and increased pathogen transmission and infection. Plants have to survive in this condition with a variety of biotic (pathogen/pest attack) and abiotic stress (salt, high/low temperature, drought). Plants can interact with beneficial microbes including plant growth-promoting rhizobacteria, which help plant mitigate biotic and abiotic stress. This overview presents that rhizobacteria plays an important role in induced systemic resistance (ISR) to biotic stress or induced systemic tolerance (IST) to abiotic stress condition; bacterial determinants related to ISR and/or IST. In addition, we describe effects of rhizobacteria on defense/tolerance related signal pathway in plants. We also review recent information including plant resistance or tolerance against multiple stresses ($biotic{\times}abiotic$). We desire that this review contribute to expand understanding and knowledge on the microbial application in a constantly varying agroecosystem, and suggest beneficial microbes as one of alternative environment-friendly application to alleviate multiple stresses.
