Inhibitory activity against biological activities and antimicrobial activity against pathogenic bacteria of extracts from Hericium erinaceus (노루궁뎅이버섯 추출물의 생리활성 및 부패세균에 대한 항균효과)

  • The aim of this study is to investigate the biological activities of Hericium erinaceus. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of H. erinaceus extract was higher than positive control. The inhibitory activities of xanthin oxidase, ${\alpha}$-glucosidase, and hyaluronidase was measured as functional food activity, and inhibitory activities on collagenase, tyrosinase, and astringent effect as beauty food activity in water and ethanol extracts from H. erinaceus. In functional food activity, xanthin oxidase inhibitory activities at $50-200{\mu}g/mL$ phenolic concentration in ethanol extracts from H. erinaceus showed inhibitory activity in dose dependent manner. ${\alpha}$-Glucosidase inhibitory activities at $50{\mu}g/mL$ phenolic concentration showed high activity of higher than 80%. Inhibitory activities on hyaluronidase as anti-inflammation factor showed inhibition effect in dose dependent manner both in water and ethanol extracts. In beauty food activity, Inhibitory activities on collagenase at $200{\mu}g/mL$ phenolic concentration in water and ethanol extracts showed high activity to 65.09 and 58.38% dose dependently. Tyrosinase inhibitory activity in water extract showed 9.4-58.24%. Astringent activity as pore shrink effect in ethanol extracts also showed a very high activity of 18.94-100%. Antimicrobial activity on pathogenic bacteria was highly effective on Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus and Escherichia coli at 2.5 mg/mL or above. Therefore, the extracts from H. erinaceus can be used as a functional food and beauty food resources and natural antimicrobial agent on pathogenic bacteria in food.

Isolation and Characterization of Burkholderia cepacia EB215, an Endophytic Bacterium Showing a Potent Antifungal Activity Against Colletotrichum Species (탄저병균에 길항력이 우수한 식물내생세균 Burkholderia cepacia EB215의 분리 및 특성 규명)

  • In order to develop a new microbial fungicide using endophytic bacteria for the control of anthracnoses occurring on various crops, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth medium, their antifungal activities were tested for in vivo antifungal activity against cucumber anthracnose caused by Colletotrichum orbiculare. As the results, liquid cultures of 28 strains showed potent antifungal activities more than $90\%$ against cucumber anthracnose. At 3-fold dilutions of liquid cultures, 18 strains inhibited the development of cucumber anthracnose of more than $70\%$. They were further tested for in vivo antifungal activity against red pepper anthracnose caused by C. coccodes and in vitro antifungal activity against C. acutatum, a fungal agent causing red pepper anthracnose. Among 18 strains, a bacterial strain EB215 isolated from cucumber roots displayed the most potent antifungal activity against Colletotrichum species. It was identified as Burkholderia cepacia based on its physiological and biochemical characteristics, Biolog test and 16S rDNA gene sequence. It also controlled effectively the development of rice blast (Magnaporthe grisea), rice sheath blight (Corticium sasaki), tomato gray mold (Botrytis cinerea), and tomato late blight (Phytophthora infestans). Studies on the characterization of antifungal substances produced by B. cepacia EB215 are in progress.

Biological Control of Sesame Soil-born Disease by Antifungal Microorganisms (참깨 토양전염성병(土壤傳染性病)의 생물학적방제(生物學的防除))

  • In order to study the biological control of soil-borne disease of sesame, antagonistic isolates of Trichoderma , Bacillus sand streptomyces to Fusarium oxysporum and Rhizoctonia solani were isolated from the rhizosphere soils of sesame plants and some other habitats. Out of the isolates of microorganisms collected a strain of Trichoderma viride was selected as a biological control agent for the study and its effect on the control of damping-off and the seedling growth of sesame was investigated. The results obtained are as follows: 26 percents of Bacillus spp. isolated from the rhizosphere soil of sesame plants showed antagonism to two pathogenic fungi. Important species were B. Subtilis and B. polymyxa. Streptomyces species isolated from the rhizosphere soils of sesame lysed the cell wall of hyphae and conidia of F. oxysporum and reduced conspicuously the formation of macroconidia and chlamydospores of the fungus. 84 percents of Trichoderma spp. isolated from the rhizosphere soil of sesame plants were antagonistic to F. oxysporum and 60 percents of the isolates were antagonistic to both F. oxysporum and R. solani. Trichoderma viride TV-192 selected from antagonistic isolates of Trichoderma spp. was highly antagonistic to F. oxysporum and soil treatment with the isolate reduced notably damping-off of sesame. T. viride TV-192 showed better growth in crushed rice straw, barley straw and sawdust media than F. oxysporum. Sawdust was selective for the growth of T. viride. Supplementation of wheat bran and mixtures of wheat bran and sawdust inoculated with T. viride TV-192 in the soil reduced remarkably damping-off of sesame by F. oxysporum but high density of the fungus TV-192 caused the inhibition of seed germination and seedling growth of sesame. Inhibitory effects of Trichoderma species on seed germination and seedling growth of sesame were different according to the isolates of the fungus. Normal sesame seedlings on the bed treated with the fungus showed better growth than not treated seedlings.

  • Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a very serious disease in rice growing regions of the world. There are no effective ways of protecting rice from the disease. In this study, the bacteriophage (phage) mixtures infecting Xoo were investigated as biological control agent on BLB. The effects of pH, heat and ultraviolet on the stability of phages were investigated to check and increase the possibility of practical use in the field. Phages were rather stable between pH 5 and pH 10. The infectivity dropped sharply when the phages were incubated at $50^{\circ}C$ and more than 90% of the phages were inactivated after two minutes of ultraviolet treatment. The phages were stable for 7 days at the rice plant leaves, and the phages survived 10 times more than other treatments when mixed with skim milk. Although the skim milk increased the stability of the phages, the control efficacy was not effective. However, the phage mixtures reduced the occurrence of BLB when they were treated with Tecloftalam WP or Acibenzolar-S-methyl simultaneously. The results indicated that the Xoo phages could be used as an alternative control method to increase the control efficacy and reduce the use of agrochemicals.

  • Mycobacterium paratuberculosis is the etiologic agent of Johne's disease, a chronic inflammatory bowel syndrome in ruminants. The attempts to control or eradicate the disease were severely hampered by the inadequacies of present diagnostic methods. The first purpose of this study was to detect Johne's disease out of 577 cows in the province of Kyunggi, Chungchong, Gangweon and the second purpose was to compare the results of non-absorbed ELISA, absorbed ELISA, PCR, and conventional culture methods. The third purpose was to increase diagnostic specificity, accuracy and rapidity. When non-absorbed ELISA test was conducted with Mycobacterium paratuberculosis antigen, the prevalence of positive was 10.9%. To increase diagnostic specificity, absorbed ELISA test with Mycobacterium phlei was used. In this test, the positive prevalence was 1.7%. For the specific detection of Mycobacterium paratuberculosis, PCR was applied to bacterial culture obtained from fecal samples of cattle. The DNA sequences derived from IS900 were used to prepare DNA primers for detection and identification of Mycobacterium paratuberculosis by PCR. PCR for M paratuberculosis isolated from fecal cultures amplified specific target DNA. PCR was much more rapid than that obtained by conventional culture technique in diagnosis of Johne's disease.

Postharvest biological control of garlic blue mold rot caused by Pantoea agglomereans and its mode of action

  • To screen for potential biocontrol agents against postharvest disease of garlics caused by Penicillium hirsutum, a total of 933 isolates (432 fungi and 501 bacteria) were isolated from the rhizoshere or rhizoplane of garlics. Among them, Pantoea agglomerans isolate 59-4 (Pa 59-4) was selected for a potential biocontrol agent by in vivo wounded garlic bulb assay, When the spore suspension (10$\^$5/ spores/$m\ell$) of Penicillium hirsutum was co-inoculated with spore or cell suspension of each fungal or bacterial isolate on wounded garlics, the isolate highly suppressed disease development. Soaking garlic bulbs in the suspension of Pa 59-4 significantly reduced garlic decay from p. hirsutum. However, Pa 59-4 did not inhibit the mycelial growth of P. hirsutum in dual-culture with P. hirsutum on Tryptic soy agar. In order to elucidate mode of action of Pa 59-4 nutrient competition between Pa 59-4 and P. hirsutum was investigated using tissue culture plates with cylinder inserts containing defusing membrane reported by Janisiewicz et al. The results showed that Pa 59-4 effectively suppressed spore germination and mycelial growth of blue mold in the low concentration (0.5%) of garlic juice, but did not suppress those of blue mold in the higher concentration (5%) of garlic juice. This result suggests that the mechanism in biocontrol of garlic blue mold by Pa 59-4 may involve in nutrient competition with P. hirsutum on garlic bulbs.

Biocontrol of Vegetables Damping-off by Bacillis ehimensis YJ-37 (Bacillus ehimensis YJ-37에 의한 채소류 모잘록병의 생물학적 방제)

  • Bacillus ehimensis YJ-37 was observed as a potential biological agent to control the occurrence of diseases and plant growth.promoting rhizobacteria (PGPR). Population density of B. ehimensis YJ-37 were higher 1.2~2 times in main roots and lateral roots than from nonrhizosphere soil and persisted around 10$^4$g root on the watermelon and radish root system upto 30 days after growing in pot condition. As a PGPR, B. ehimensis YJ-37 enhanced plant growth of watermelon and radish by soil treatment. The leaf area, hypocotyl length, root length and dry weight of radish were about 85, 33, 23 and 89% more than that of untreated plant, respectively. In case of watermelon were about 63, 27, 25 and 69% more than that of untreated plant, respectively. Biocontrol of damping-off in watermelon and radish caused by Rhizoctonia solani AG-4 and Pythium ultimum were carried out in pots using 3. ehimensis YJ-37. The results showed that might contribute to it's suppression of damping-off disease in field plants.

Effects of Collagen Tripeptide Supplement on Photoaging and Epidermal Skin Barrier in UVB-exposed Hairless Mice

  • Collagen tripeptide (CTP) is a functional food material with several biological effects such as improving dry skin and wound and bone fracture healing. This study focused on the anti-photoaging effects of CTP on a hairless mouse model. To evaluate the effects of CTP on UVB-induced skin wrinkle formation in vivo, the hairless mice were exposed to UVB radiation with oral administration of CTP for 14 weeks. Compared with the untreated UVB control group, mice treated with CTP showed significantly reduced wrinkle formation, skin thickening, and transepidermal water loss (TEWL). Skin hydration and hydroxyproline were increased in the CTP-treated group. Moreover, oral administration of CTP prevented UVB-induced MMP-3 and -13 activities as well as MMP-2 and -9 expressions. Oral administration of CTP increased skin elasticity and decreased abnormal elastic fiber formation. Erythema was also decreased in the CTP-treated group. Taken together, these results strongly suggest that CTP has potential as an anti-photoaging agent.

Molecular Discrimination of Cervidae Antlers and Rangifer Antlers

  • Cervi Parvum Cornu is widely used as a hemopoietic, tonifying, growth-promoting, cardiotonic, and immuno-modulating agent in Korea. In order to develop the quality control method of Cervi Parvum Cornu by the identification of the biological source or origin, the molecular approach was applied using PCR (polymerase chain reaction) and PCR-RFLF (PCR-restriction fragment length polymorphism) analysis. In the PCR analysis of the mitochondrial 12S rRNA gene and cytochrome b gene regions, no distinctive DNA bands from Cervidae (deer) antlers and Rangifer (reindeer) antlers were observed. However, when the amplified products in the mitochondrial cytochrome b gene region were subjected to restriction digestion with TaqI, Cervidae antlers showed an undigested state of 380 by band, differently from two bands of 230 by and 1S0 by from Rangifer antlers. Based on this finding, the base sequences of amplified PCR products in the range of mitochondria) cytochrome b gene from Cervidae antlers and Rangifer antlers were determined and subjected to restriction analysis by various endonucleases. The results showed that antlers from Rangifer species could be simply discriminated with other antlers from 8 Cervidae species (Chinese deer, Russian deer, Hong Kong deer, New Zealand deer, Kazakhstan deer, elk, red deer and Sika deer) by PCR-RFLP analysis using AtuI, HaeIII, HpaII or Sau3AI(MboI) as well as TaqI in the range of the mitochondrial cytochrome b gene.

First Record on the Exotic Parasitoids Anisopteromalus apiovorus Rasplus (Hymenoptera: Pteromalidae) in Korea (외래기생봉 권연벌레살이금좀벌(국명신칭), Anisopteromalus apiovorus Rasplus (Hymenoptera: Pteromalidae)의 한국 내 신 분포 기록)

  • An exotic parasitic wasp Anisopteromalus apiovorus of Pteromalidae was newly recorded in Korea. Specimens of this species have been collected in Gimhae of Korea since 2012. External morphology of this species was described in detail and diagnostic differences of this species from the related species and the host insect were also given. The funicle segments of female antennae are 1.2 to 1.5 times longer than those of Anisopteromalus calandrae, an sister species. Body color is gold-glossy black and with dense creamy-white hairs on the surface. This exotic species has been originated from the African continent including Democratic Republic of Congo and Cote d'Ivoire, and then intruded into other countries, for instance, Korea. This is the first distribution record of this species except the country of origin after original description as a new species. Although little is known about the life history or ecology of this species, we found that a new host of this small parasitic wasp is Lasioderma serricorne of coleoptera through present study. And this species possibly be a new agent of biological control for economic pests. It was presumed that the invasion of Anisopteromalus apiovorus into Korea was prior to November of 2012.
